Study on Renal Anemia - A Double Tracer Study on Iron Metabolism and Red Cell Life Span in Chronic Renal Diseases using Radioactive Iron ($^{59}Fe$) and Chromium($^{51}Cr$) - (신성빈혈(腎性貧血)에 관(關)한 연구(硏究) - 만성신질환(慢性腎疾患)의 철대사(鐵代謝) 및 적혈구수명(赤血球壽命)에 관(關)하여 -)

  • The ferrokinetics and red cell life spans of the patients with chronic glomerulonephritis were investigated by the double tracing method using radioactive iron ($^{59}Fe$) and chromium ($^{51}Cr$). According to the serum NPN levels, the patients were subdivided into 3 groups: Group 1. 6 patients, had the levels below 40 mg/dl Group 2. 6 patients, had the levels between 41 mg/dl to 80 mg/dl Group 3. 10 patients, had the levels above 80 mg/dl The results were as follows: 1) Red blood cell-, hematocrit- and hemoglobin values were moderately reduced in patients with normal serum NPN levels, while markedly reduced in patients with elevated serum NPN levels. 2) The plasma volume was increased, while the red cell volume was decreased in patients with elevated serum NPN levels, hence, total blood volume was unchanged. 3) The serum iron level was slightly reduced h patients of groups 1 and 2, while was within the normal ranges in patients of group 3. 4) i) In patients with normal serum NPN levels, the plasma iron disappearance rate, red cell iron utilization rate, red cell iron turnover rate, daily red cell iron renewal rate, circulating red cell iron and red cell iron concentration were within the normal ranges, while the plasma iron turnover rate was slightly reduced. ii) In patients with elevated serum NPN levels, the plasma iron disappearance rate was delayed, while the plasma iron turnover rate was within the normal ranges. The red cell iron utilization rate, red cell iron turnover rate and circulating red cell iron were decreased and the period in which the red cell iron utilization rate reachd its peak was delayed in Group 3 patients. The daily red cell iron renewal rate and the red cell iron concentration were unchanged. iii) The mean red cell life span was within the normal ranges in patients with normal serum NPN levels, while was shortened in patients with elevated serum NPN levels.

Protective Effect of Plantago asiatica L. Extract Against Ferric Nitrilotriacetate (Fe-NTA) Induced Renal Oxidative Stress in Wistar Rats (차전초 추출물을 투여한 랫드에서의 Fe-NTA 유발 산화스트레스에 대한 신장보호 효과)

  • Plantago asiatica L. (PA), which is widely distributed in Korea, Japan and China, has traditionally been used as a popular folk medicine for the treatment of liver diseases. A variety of activities of PA was reported, that is hepatoprotective, anti-inflammatory, anti-glycation and anti-oxidant effect. Ferric nitrilotriacetate (Fe-NTA) is a potent nephrotoxic agent and has been reported to induce renal proximal tubular necrosis. In the present study, pre-treatment with PA extract (PAE) in Wistar rat followed by Fe-NTA i.p. treatment (13.5 mg Fe/kg body weight) was performed to detect the renal protective effect of PAE. Only Fe-NTA treated group showed increases in the level of serum blood urea nitrogen (BUN) and serum creatinine (Cr), and renal tissue malondialdehyde (MDA), product of lipid peroxidation. Moreover, the level of biomarkers indicate the antioxidants status, reduced glutathione (GSH), glutathione-S-transferase (GST) and glutathione reductase (GR) were decreased. However, PAE pre-treated group showed decreases in the levels of serum BUN, serum Cr and renal tissue MDA in concentration dependent manner and increases in the level of GSH, GST and GR. These results are significantly different (p < 0.05) to the other groups. Our data suggest that PAE may be used as an chemopreventive material against Fe-NTA-mediated renal oxidative stress.

Effects of Smoking, Drinking, Exercise on Body Composition, Nutrient Intakes and Serum Lipids in Male High School Students (흡연, 음주, 운동이 남자 고등학생의 체성분, 영양소 섭취 및 혈청 지질 양상에 미치는 영향)

  • This study was done to investigate effects of smoking, drinking and exercise on body composition, dietary intakes, serum lipids of male high school students in Seoul. Body Composition was analyzed by the Bioelectrical Impedance Fatness Analyzer. Dietary survey was conducted by 24-hour recall method and nutrient intakes were analyzed by the Computer Aided Nutritional analysis program for professional (CAN-Pro). All data were statistically analyzed by SAS PC package program. Mean and standard error were examined for each items. The significant difference was examined by student's t-test. Body composition data showed that protein mass, mineral mass and lean body mass of regular exercisers were significantly higher (p < 0.05) than those of irregular exercisers. Energy and protein intakes of male high school students were 1868 kcal/day (69.2% RDA), 68 g/day (91.3% RDA), respectively. Ca, Fe and Vitamin B$_2$ intakes were lower than 2000 RDA. Analysis of serum lipids showed that serum levels of HDL-cholesterol of drinkers were significantly higher (p < 0.05) than those of nondrinkers. Overall results imply that smoking and alcohol drinking of adolescent males may not much influence on serum lipids levels because pack-years of smoking or the amount of alcohol drinking was not serious enough to be harmful to health.

A Study on SOD Activity and Serum Antioxidant Mineral Concentrations in Obese Adolescents (비만 청소년의 SOD 활성도 및 혈청 항산화무기질 농도에 관한 연구)

  • The purpose of this study was to investigate the relationship between obesity and, erythrocyte SOD (superoxide dismutase) activity and serum antioxidant mineral (Fe, Zn, Cu, Mn and Se) concentrations of adolescents. Subjects were assigned to one of two groups such as obese ($BMI{\geq}25$, 32 boys, 24 girls) and normal group (18.5 < BMI < 23, 27 boys, 30 girls) Subjects were evaluated based on anthropometric measurements, 24-hr dietary recalls and blood analysis. The mean age of the total subjects was 13.8 years. The mean weight (p < 0.001), BMI (p < 0.001) and body fat (p < 0.001) of obese were higher than those of normal group. There was no significant difference in nutrient intake between obese and normal groups. SOD activity of obese group was not significantly different from normal groups, in both males and females. However, in the males, serum Cu concentration of obese were significantly lower than those of normal group. In the females, Serum Mn concentration of obese were significantly lower then those of normal group. In the correlation analysis, BMI of the subjects had significantly negative correlations with serum Cu, Zn and Mn. To summarize the results, increase of obesity may lead to decrease of serum antioxidant minerals such as Cu, Zn and Mn.

Effect of Copper Supplementation on Mineral Utilizations in Rats (흰쥐에 있어 구리 보충이 무기질 대사에 미치는 양향)

  • The purpose of this study was to investigate the effect of copper supplementation on mineral utilizations in rats. Intakes, excretions, and tissue levels of Fe, Zn, Cu, Ca, Mg, Na, K were compared in rats fed diet with 100, 200, 400% of copper requirement(1Cu, 2Cu and 4Cu, respectively) for 6 weeks. The feed intake was significantly higher in 2Cu compared to 1Cu group, however, body weight gain was not significantly different among the three groups. While the serum level of Fe was significantly decreased, that of Cu was significantly elevated by Cu supplementation. The liver and kidney levels of Mg and Na, intakes of all the minerals, urinary excretions of Fe, Cu, Ca and Mg were significantly higher in 2Cu and/or 4Cu compared to 1Cu. The fecal excretions, retentions and absorptions of all the minerals were not affected by Cu supple mentation. In summary, these results suggest that dietary Cu supplementation has no effect on mineral utilizations, but effect on distribution in tissues of rats.

A Study on Correlation among Physical Fitness, Diet Intakes and Serum Lipid in Middle Aged Women. (중년여성의 체력, 식이섭취와 혈청지질과의 상관관계에 대한 연구)

  • This study was performed to investigate the correlations among physical anthropometric measurement, diet intakes, physical fitness test, and serum lipids in eight sedentary women(control group), and twenty seven exercising women(aerobic groups), aged 35~45 years. The results of the study are as follows : 1) systolic blood pressure(SBP) and diastolic blood pressure(DBP) of aerobic groups were lower than serf entary. Pulse rate was decreased according to the length of aerobic periods. Muscular endurance and jump power of all subjects were stronger than standard range, and those of aerobic exercising groups were stronger than control group. Muscular power of the upper half of the body In all subjects were lower than standard range. 2) Daily calorie intakes of all subjects were in the ring: of recommended dietary allowances (RDA) for Koreans. But daily protein Intakes were higher than RDA. daily cholestrol intakes of all subjects were slightly less than 200~300 mg: which is normal consumption of Koreans. 3) There was a significant positive correlation between serum 7G and daily protein intakes. It was considered that excessive protein intake converted to serum 7G. There was not a significant correlation between serum TC and dietary TC. Therefore, dietary TC seemed to have not much effect on serum TC. Physical fitness was negatively related to serum TG, TC, LDL-C, but positively related to HDL-C, HDL-C /TC. It was found that the serum lipids could be changed better according to improving the physical fitness. There were significant positive correlations between SBP and serum TC, LDL-C. (According to this results), the higher SBP, the higher serum TC, LDL-C tended to be. There was a significant positive correlations between sit up and weightloss nth exercise r=0.7(p<0.001), push ups and weightless with exercise r=0.5(p<0.001). It was considered that muscle strength could be improved according to the weightloss. 4) The purpose of exercise in aerobic groups was rather promoting health than reducing body fatness. Most of al1 subjects (83.2%) drank caffeine-contained beverage therefore, we should concern about that absorption of Fe could be Interrupted and blood pressure could be elevated by caffeine.

A Study on Nutrient Intake, Anthropometric Data and Serum Profiles among High School Students Residing in Seoul (서울 지역 고등학생의 영양소 섭취와 체위 및 혈액성상)

  • This study was performed to investigate the anthropometric data, serum profiles, and nutrient intakes of high school students who are living in Seoul. Anthropometric data showed that mean height and weight were 174.1 cm and 66.6 kg in 99 male and 161.4 cm and 54.0 kg in 116 female students. Mean BMl for male and female students were 21.9 and 22.3 kg/$m^2$, and subjects with BMI > 25 kg/$m^2$ were 14 and 7%, respectively. Total cholesterol, HDL- and LDL-cholesterol of subjects were 149.6$\pm 3.1,\; 46.4 \pm 1.3,\; 86.4 \pm 2.3$,/TEX> mg/dl for males and 169.2 $\pm 3.1,\;50.7 \pm 1.0,\; 100.9 \pm 2.7 mg/dl$ for females, respectively. Hemoglobin (Hb) and total iron binding capacity (TIBC) were 12.8 $\pm 0.1 g/dl, \;437.3 \pm 7.2\mug/dl\; in\; male\; and\; 11.9 \pm 0.1 g/dl,\; 439.4 \pm 5.7\mug/dl$ in female, respectively. Based on Hb classification, 45.6% of male and 45.0% of female students fell into anemic condition. Intakes of energy, Ca, Fe, Zn, riboflavin, and folate were below the Korean RDA. In particular Ca (male and female, respectively, 64.6 and 64.3% RDA) and Fe (male and female, respectively, 78.6 and 64.3% RDA) intakes were extremely low. Serum triglyceride was positively significantly correlated with weight (r =0.22), BMI (r =0.279), waist circumference (WC, r =0.235), triceps skin-fold thickness (TSF, r =0.197) and obesity index (OI, r =0.279). Hb concentration was positively correlated with height (r + 0.387), weight (r =0.349), BMI (r =0.191) and waistlhip ratio (WHR, r =0.380). As for the correlation between serum profiles and nutrient intakes, the Hb concentration was positively correlated with energy (r =0.163), protein (r =0.149), Na (r =0.153), vitamin A (r =0.165), thiamin (r =0.201) and niacin (r =0.192, p < 0.01). These result suggest that the prevalence of Ca and Fe deficiency of student is high and dietary guideline for prevent anemia in this age needs to contain the adequate intake of energy and its related vitamins.

The Effect of Extracts of Berberis koreana Bark by Lactobacillus Fermentation on the Concentration of Serotonin and Melatonin in the Serum of Treated ICR Mice (유산균 발효된 매자나무 추출물이 마우스 혈청중의 Melatonin 및 Serotonin의 함량에 미치는 영향)

  • This study was to investigate the effect of fermentation extracts on the concentration of serotonin and melatonin in the serum of the ICR mice. The ICR mice were divided into water control group, lactobacillus fermentation solution including (Lactobacillus paracasei and Bifidobacterium longum B6) control group, positive control group (milk and doxylamine succinate), negative control group (caffein) and the groups treated with the extracts of Berberis koreana bark (WE: water extracts, FE-L.P: fermentation extracts of Lactobacillus paracasei, FE-B.L: fermentation extracts of Bifidobacterium longum B6). After ten-day feeding treatment, the mean concentration of serotonin for water control, WE, FE-L.P and FEB. L group was 134.72, 183.01, 232.09 and $223.78 ng/m{\ell}$, respectively. The mean concentration for FE-L.P and FE-B.L group were approximately 66% larger than that for water control group. The mean concentration of melatonin for water control, WE, FE-L.P and FE-B.L group was 76.92, 106.66, 157.56 and $141.81pg/m{\ell}$, respectively. The mean concentration of melatonin for FE-L.P and FE-B.L group were also larger than that for water control group. Our results indicated that the fermentation extracts of Berberis koreana bark have relatively greater potential to induce secretion of serotonin and melatonin. Therefore, the fermentation extracts have antidepressant effect.

Relation among Mineral (Ca, P, Fe, Na, K,Zn) Intakes, Blood Pressure, and Blood Lipids in Korean Adults (일부 성인남녀의 무기질 (Ca, P, Fe, Na, K, Zn) 섭취와 혈압 및 혈중 지질과의 관련성)

  • The purpose of this study was to investigate the relation among mineral intakes, blood pressure, and blood lipids for health management of Korean adults over 20 years old. Subjects were recruited from the district where they lived, which included rural (Yoju, n = 137) , coastal (Taean, n = 100) , and urban district (Suwon, n : 117) . hnthropornekical measurement, blood pressure measurement, dietary intake assessment using 24-hour recall method, and blood collection and analysis of blood lipids were conducted. The average age, height, weight, and BMI were 54.1 years, 168.5 cm, 67.3 kg, and 24.5 kg/$m^{2}$ for men and 53.9 years,153.8 cm,59.2 kg, and 25.0 kg/$m^{2}$ for women, respectively. The mean daily intakes of total food and energy were 1219.1 g and 1740.9 kcal for men and 1071.3 g and 1432.6 kcal for women. The mineral intakes of men and women were 448.5 mg and 400.4 mg for calcium,955.3 mg and 803.7 mg for phosphorus, 13.3 mg and 11.1 mg for iron, 4302.8 mg and 3393.3 mg for sodium,2588.0 mg and 2264.9 mg for potassium, and 8.9 mg and 7.3 mg for zinc. The daily intakes of energy, calcium, zinc, vitamin $B_{2}$, and vitamin I were $76.1\%$, $60.1\%$, $73.1\%$, $68.6\%$, and $80.4\%$ of RDAs, respectively. The intakes of energy, calcium, zinc, vitamin $B_{2}$ and I were not meet Korean RDAs. Especially, calcium, zinc, and vitamin $B_{2}$ were less than $75\%$ of RDAs. Blood pressure of the subjects was 128.3/75.5 mmHg for men and 124.1/73.7 mmHg for women. Serum lipids of men and women were 180.2 mg/dL and 184.9 mg/dL for total cholesterol, 160.8 mg/dL and 137.6 mg/dL for triglyceride,41.5 mg/dL and 44.7 mg/dL for HDL-cholesterol,106.6 mg/dL and 112.7 mg/dL for LDL-cholesterol, and 3.5 and 3.3 for atherogenic index. While serum triglyceride and AI of men was significantly higher, HDL-cholesterol was lower than those of women. In the relation among mineral intakes, blood pressure, and serum lipids, we found a significantly negative correlation of intakes of calcium, calcium/phosphorus, calcium density with systolic blood pressure. There were significantly negative correlation between intake of calcium/phosphorus and serum cholesterol, calcium density and serum cholesterol, sodium intake and serum HDL-cholesterol. Based on these results, we concluded that high intakes of calcium, calcium/phosphorus and low sodium intake were associated with favorable blood pressure and serum lipid profiles in adult men and women. Effect of Iron(II)-ascorbate Complex on Protein and DNA of Phages (파아지 단백질 및 DNA에 대한 2가철-아스코르빈산착체의 영향)

  • The inactivating effect of iron(II)-ascorbate complex (Fe-Asc) on various phages was previously reported. This paper describes the molecular target in the phage virion attacked by Fe-Asc. The effect of Fe-Asc on protein was investigated with bovine serum albumin and the structural protein of phage J1. There were no differences in the SDS-polyacrylamide gel electrophoresis (patterns of these two proteins when either they were treated) with Fe-Asc or not. Also, there were no changes in the amino acid composition and ultraviolet spectrum of the proteins. The effects of Fe-Asc on DNA was investigated with pUC18 DNA, M13mpB DNA and ${\lambda}$ DNA as well as DNA from phage J1. Fe-Asc caused initially nicking of the subsequently form of pUC18 DNA to yield the open circular form and then subsequently the linear form. Strand breaks were also confirmed with M13mp8 DNA and ${\lambda}$ DNA as well as J1 DNA. The results indicate that the strand breaks in phage DNA could be responsible for the inactivation of phages by Fe-Asc.
