A Survey of the Brassica rapa Genome by BAC-End Sequence Analysis and Comparison with Arabidopsis thaliana

  • Brassica rapa ssp. pekinensis (Chinese cabbage) is an economically important crop and a model plant for studies on polyploidization and phenotypic evolution. To gain an insight into the structure of the B. rapa genome we analyzed 12,017 BAC-end sequences for the presence of transposable elements (TEs), SSRs, centromeric satellite repeats and genes, and similarity to the closely related genome of Arabidopsis thaliana. TEs were estimated to occupy 14% of the genome, with 12.3% of the genome represented by retrotransposons. It was estimated that the B. rapa genome contains 43,000 genes, 1.6 times greater than the genome of A. thaliana. A number of centromeric satellite sequences, representing variations of a 176-bp consensus sequence, were identified. This sequence has undergone rapid evolution within the B. rapa genome and has diverged among the related species of Brassicaceae. A study of SSRs demonstrated a non-random distribution with a greater abundance within predicted intergenic regions. Our results provide an initial characterization of the genome of B. rapa and provide the basis for detailed analysis through whole-genome sequencing.

  • Tandem GMAW is one of the high performance welding process and used in many industries to increase the productivity. An evaluation is presented of the mechanical properties and microstructural characteristics of the Tandem GMAW and conventional Single GMAW welds in 30mm thickness 490MPa grade steel by comparison method. Welding sequence and bead with and hight was kept, avoiding the effect of the bead shape and welding sequence. Tension, bending, hardness and Charpy impact test results of Tandem GMAW met the requirement of specification and showed similar distribution with conventional Single GMAW. Volume fraction of ferrite phase in weld metal showed little difference between Tandem GMAW and Single GMAW

Genomic Organization of Heat Shock Protein Genes of Silkworm Bombyx mori

  • The Hsp 20.8 and Hsp 90 cDNA sequence retrieved from NCBI database and consists of 764 bp and 2582 bp lengths respectively. The corresponding cDNA homologus sequences were BLAST searched in Bombyx mori genomic DNA database and two genomic contigs viz., BAAB01120347 and AADK01011786 showed maximum homology. In B. mori Hsp 20.8 and Hsp 90 is encoded by single gene without intron. Specific primers were used to amplify the Hsp 20.8 gene and Hsp 90 variable region from genomic DNA by using the PCR. Obtained products were 216 bp in Hsp 20.8 and 437 bp in Hsp 90. There was no variation found in the six silkworm races PCR products size of contrasting response to thermal tolerance. The comparison of the sequenced nucleotide variations through multiple sequence alignment analysis of Hsp 90 variable region products of three races not showed any differences respect to their thermotolerance and formed the clusters among the voltinism. The comparison of aminoacid sequences of B. mori Hsps with dipteran and other insect taxa revealed high percentage of identity growing with phylogenetic relatedness of species. The conserved domains of B. mori Hsps predicted, in which the Hsp 20.8 possesses ${\alpha}-crystallin$ domain and Hsp 90 holds HATPase and Hsp 90 domains.

  • In this paper, an integrated fault section detection and isolation strategy is proposed based on the application of the Distribution Automation System(DAS) utilizing advanced IT and communication technologies. The Feeder Remote Terminal Unit(FRTU) has been widely used to collect data in the Korean distribution system. The achieved data is adopted in this method for detecting multiple fault types. Especially in the case of single phase-to-ground fault, the fault section is detected by comparison of the zero-sequence current phase angle. The test results have verified the effectiveness of the proposed method in a radial distribution system through extensive simulations in Matlab/Simulink. Furthermore, a communication-based demo system identical to the simulation model has been developed, and it can be applied as an online monitoring and control program for fault section detection and isolation.

  • Three methods(the stepped sine method, the statistical method(random excitation method) and the maximum-length sequence(MLS) method) for head-related transfer functions(HRTFs) are experimentally compared in view point of accuracy and efficiency. First, the stepped sine method has high signal-to-noise ratio, but low efficiency. Second, the statistical method is fast measurement speed, but weak to noise than the other methods. Finally, the MLS method shows both good efficiency and high signal-to-noise ratio, but it needs additional software or equipment such as MLS signal generator. For comparison of measurement accuracy, HRTFs of KEMAR dummy are measured for various azimuths and elevations. Error norms for magnitude and phase of HRTFs are defined and calculated for the measured HRTFs. The calculated error norms show that the methods give similar results in magnitude and phase except a little phase difference in the MLS method.

Identification of Entomopathogenic Fungus, Beauveria spp. F-101 Isolated from Thecodiplosis japonensis Using Internal Transcribed Spacer Sequence

  • For the development of the alternative control system against the major forest pests, Beauveria spp. F-101, isolated from a dead larva of Thecodiplosis japonensis, was selected because this isolate showed high pathogenicities against T. japonensis and Acantholyda parki. Beauveria spp. F-101 had irregular clustered conidio-phores and conidia borne on a distinctive apical zigzag extension, and it showed typical characteristic of the genus, Beauveria in morphology. For molecular based-identification, the ribosomal ITS region of Beauveria spp. F-101 was amplified with ITS1 and ITS4 primers, and cloned into pGEM- T Easy vector. The amplified PCR product was 569 bp in size and completely sequenced. The similarities of the cloned ITS sequence were 99 % and 97% to those of B. bassiana and B. brongniartii, respectively. In comparison to other species among the genus Beauveria, the ITS region of Beauveria spp. F-101 showed a similarity of 95% to B. amorpha, 95% to B. tenella, 89% to B. vermiconia and 69% to B. alba, respectively. In addition, in comparison to different genus, it had 95% similarities to Cordyceps militaris and 91% to Paecilomyces tenuipes. Accordingly, the current result suggests that Beauveria spp. F-101 was a variant of B. bassiana and it seems to be a new isolate considering sequence variation in ITS region.

  • In this paper, the performance comparison of the TR-UWB System using Wavelet Pulse, DPSS pulse, and second Gaussian pulse in multi-user environment is analyzed. The data signal and the reference signal is sent with some time intervals in TR-UWB system. At this time the two signals are through the same channel. In the receiver the reference signal is used as a template. This advantage results in demodulation without channel estimation and low complexity of it. However the conventional TR-UWB system based on a Gaussian signal in a multi-user environment has the disadvantage of poor performance due to the interferences between the users. To overcome this disadvantage, DPSS (Discrete Prolate Shperoidal Sequence) is used to reduce the interferences between the users. We propose the system with multiresolution function of orthogonal wavelet reducing the interferences, which has a better performance in multi-user environment when they are sent in transmission side.

Nucleotide Sequence of the Penicillin G Acylase Gene from Bacillus megaterium and Characteristics of the Enzyme (Bacillus megaterium에서 발견된 Penicillin G Acylse 유전자의 염기서열과 그 효소의 특성)

  • The complete nucleotide sequence of the cloned pga gene encoding the penicillin G acylase of Bacillus megaterium ATCC 14945 and its 5'- and 3'-flanking regions was determined. The sequence revealed only one large open reading frame (2,406 hp) of the penicillin G acylase (pga) gene. Upstream from ATG of the pga gene, there was a putative ribosome binding site, Shine-Dalgarno sequence. The promoter-like structure, - 10 and - 35 sequences, was also found. Following the stop codon, TAG, a structure reminiscent of the E. coli rho-independent transcription terminator was present. The amino acid sequence was deduced from the nucleotide sequence. The molecular mass of the polypeptide was 91,983 Da. There was a potential signal sequence in its amino-terminal region. A comparison of its deduced amino acid sequence with other characterized penicillin G acylases and the result of SDS-polyacrylamide gel electrophoresis of the purified enzyme showed that a precursor polypeptide of 92 kDa was processed into two dissimilar ${\alpha}$ and ${\beta}$-subunits of 25 and 61 kDa.

  • The inter-laboratory comparison test which is demanded on the authorized test of ISO/IEC 17025 is the program to guarantee the confidence of test result. This paper, as a part of the inter-laboratory comparison test between South Korea and China, is refered to the characteristic of friction coefficient of disk brake lining for rolling stock according to disk type. Brake tests were carried out under constant brake force and operating sequence by using dynamo-tester according to disk and disk lining types. To establishing the confidence of test result, we calculated A type uncertainty about friction coefficient and investigated the factors about the variation of friction coefficient.

Methods of pairwise comparisons and fuzzy global criterion for multiobjective optimization in structural engineering

  • The method of pairwise comparison inherently contains information of ambiguity, fuzziness and conflict in design goals for a multiobjective structural design. This paper applies the principle of paired comparison so that the vaguely formulated problem can be modified and a set of numerically acceptable weight would reflect the relatively important degree of multiple objectives. This paper also presents a fuzzy global criterion method ($FGCM_{\lambda}$) included fuzzy constraints that coupled with the objective weighting rank obtained from the modified pairwise comparisons for fuzzy multiobjective optimization problems. Descriptions in sequence of this combined method and problem solving experiences are given in the current article. Multiobjective design examples of truss and mechanical spring structures illustrate this optimization process containing the revising judgement techniques.
