Eye irritation, primary skin irritation and skin sensitization tests for Aloewhite were tested in New Zealand White rabbits and Hartley guinea pig. In primary skin irritation test of male New Zealand White rabbits, body weights were not significantly changed and Primary Irritation Index (PII) was O.47, indicating Aloewhite as mildly irritating material. In ocular irritation test, any injury on iris, conjunctival membrane, and cornea in New Zealand White rabbits was not observed. No injuries of the ocular mucous membrane were also recorded. Skin sensitization was tested in guinea pig after intradermal and epicutaneous induction and graded 1 with zero % sensitization rate. These results indicate that Aloewhite was not considered to be irritant in test organs of animals.
Clinical evaluation of contact sensitization to 2, 4-dinitro-chlorobenzene [DNCB] was performed in 2 groups: group A [30 patients with non-malignant disease] and group B [30 patients with bronchogenic carcinoma]. Initial sensitization was elicited out by applying 2, 000 ug of DNCB to skin surface of the both group A and B. Subsequently a relatively weak challenge dose, 200 ug of DNCB, was applied 14 days later, showing the satisfactory results of sensitization with minimizing non-specific irritative inflammatory skin response. Delayed cutaneous hypersensitivity reactions shown by spontaneous flare phenomena appeared at the challenge site, and they were assessed 48 hours later. The reaction were graded from +1 to +4 according to the degree of flare or vesicular reaction. The results were as follows: 1. 28 cases [93%] of group A, however, only 18 cases [67%] of group B exhibited delayed cutaneous hypersensitivity reaction to DNCB contact sensitization [P<0.02]. 2. Of group A, the delayed cutaneous hypersensitivity reactions above +2 of DNCB score were 25 cases [83%], meanwhile 11 cases [37%] in group B [P<0.001]. 3. Undifferentiated carcinomas showed highest incidence of anergy to DNCB contact sensitization in the all histologic types of group B. 4. In group B, 8 [42%] of 19 cases who react to DNCB were resectable, whereas only 2 [18 %] of 11 cases who failed to react to DNCB were resectable for curative cancer surgery. These study suggests that cellular immune reaction of group B was depressed remarkably comparing with that of group A.

Repeated administration of psychostimulants such as amphetamines increases locomotor activity in rodents. These drugs, including methamphetamine, enhance dopaminergic neurotransmission and result in hyper-locomotion and behavioral sensitization. It is well known that the existence of a complex balance between the cholinergic and dopaminergic systems in the central nervous system. Thus, behavioral sensitization by methamphetamine may be related to the expression of the M1 muscarinic acetylcholine receptors gene. The present study investigated the changes of M1R mRNA in hyperlocomotor activity and behavioral sensitization by methamphetamine (2 mg/kg) in mice. Our results showed that M1R mRNA expression was increased in the frontal cortex and the hippocampus region (the CA2 region) in the acute methamphetamine administered group compared to the saline administered group. In the chronic group, M1R mRNA expression was increased in the frontal cortex ill1d the hippocampus regions (CA2 and DG regions) in melt1amphetamine administered group compared to saline control group. These results indicate that acute or chronic treatment of mathamphetamine leads to the region-specific changes in mRNA expression levels of M1R. Therefore, Therefore, the present result suggests that M1R may play a role in modulating of methamphetamine-induced behavioral sensitization in mice.
Lateral giant (LG)-mediated escape response of crayfish is sensitized by natural traumatic events. Such sensitization has previously been shown to be associated with increased transmission between primary afferents and sensory interneurons at the cholinergic synapse of LG escape reflex circuit. In the present study, it was firstly investigated as to whether transmission is also altered at other synapses of the LG-escape reflex circuit by traumatic shock-induced sensitization. Evidence that traumatic shock also directly affects the excitability of lateral giants is now provided by the finding that traumatic shock produces a significant reduction of the time needed for LG to recruit its contralateral homologue, which is defined as commissural delay. Octopamine, a naturally occurring neuromodulator in the crayfish nerve cord, has also been shown to enhance transmission at the cholinergic synapse between primary afferents and sensory interneurons, and has been conjectured to mediate sensitization. Like traumatic shock, $octopamine\;(10^{-5}-5{\times}10^{-4}\;M)$ also enhanced the efficacy of commissural transmission between lateral giants, as indicated by a significant reduction of commissural delay. This effect was blocked by an octopamine antagonist phentolamine, suggesting a specific action of octopamine on the octopamine receptor present on LGs. These observations suggest that both traumatic shocks and octopamine may cause a rather broad alteration in the excitability of the crayfish nervous system that contributes to the sensitization of the LG escape response.
The inhibitory effects of (-)-epigallocatechin gallate (EGCG), a major compound of green tea, on the development of locomotor sensitization, conditioned place preference (CPP) and dopamine receptor supersensitivity induced by the repeated administration of morphine were investigated in mice. A single administration of morphine produces hyperlocomotion. The repeated administration of morphine develops sensitization, a progressive enhancement of locomotion, which is used as a model for studying the craving and drug-seeking behaviors characterizing addiction, and CPP, which is used as a model for studying drug reinforcement, respectively. EGCG inhibited morphine-induced hyperlocomotion, sensitization and CPP. In addition, EGCG inhibited the development of postsynaptic dopamine receptors supersensitivity, which may be an underlying common mechanism that mediates the morphine-induced dopaminergic behaviors such as sensitization and CPP. Apomorphine (a dopamine agonist)-induced climbing behaviors also were inhibited by a single direct administration of EGCG These results provide evidence that EGCG has anti-dopaminergic activity, as inhibiting the development of dopamine receptor supersensitivity and apomorphine-induced climbing behaviors. Therefore, it is suggested that green tea may be useful for the prevention and therapy of these adverse actions of morphine.

Repeated administration of all addictive drugs, including nicotine, can produce sensitization of extracellular dopamine levels in the nucleus accumbens and behavioral sensitization in rat, as evidenced by an enhanced locomotor response and increased dopamine release in brain to a subsequent injection of the drug. In order to investigate the effect of Zizyphus jujuba extract on repeated nicotin-induced sensitization, rats were given repeated injection of saline or nicotine (0.4 mg/kg s.c., twice a day for 7 d), followed by one challenge injection on the 4th day after the last daily injection. Systemic challenge with nicotine (0.4 mg/kg s.c.) and a direct local challenge of 3 mM a larger increase in locomotor activity and extracellular dopamine release in the nucleus accumbens in nicotine-pretreated rats than in saline-pretreated rats, respectively. Zizyphus jujuba extract significantly decreases locomotor activitiy and dopamine release in the nucleus accumbens induced by a nicotine challenge. These results suggest that Zizyphus jujuba extract may attenuate nicotine-induced neurochemical and behavioral sensitization and may be effective in suppressing compulsive nicotine-seeking behavior.
The purpose of the present study was to investigate differences in the sensitizing potential of Rhus Veniciflua(Rhus-II), when tested by the guinea pig maximization test(GPMT) and Freund's complete adjuvant test(FCAT) with an identical, intradermal induction concentration. A new grading classification of the sensitization potential is proposed. The GPMT was conducted according to OECD guideline $\#406$, using a multiple-dose design and test results were analysed with logistic regression analysis. During the induction stage, we injected intradermally each three site 0.1 ml(l mg/animal) test material, 0.1 ml complete Freund's adjuvant and 0.lml the test agent emulsified in the adjuvant. 7 days later, we induced weak sensitization with $10\%$ sodium lauryl sulfate(SLS) and applide 1ml(l0mg/animal) test agent topically on the same site and made a tight occlusion. 14 days later we challenged with 1 ml(l 0mg/animal) of test material on the flank and observed ant 24 hours and 48 hours later. The results were also observed $0\%$ at 24 hours challenge. The results observed 48 hours after challenge were the identical. These data indicated that, although Rhus-II is a no contact allergen. It was reported that the skin sensitization by Rhus-II was not detected the skin sensitization in the guinea pig maximization test (GPMT). Consequently, it was confirmed that Rhus-II had no contact allergic sensitization in guinea pig maximization test.

The effect of subcutanecus injection of Corynebacterium parvum($700{\mu}g$) on cellular and humoral immune responses when given at various time relative to sheep red blood cell(SRBC) sensitization were studied by the evaluation of Arthus, delayed-type hypersensitivity(DTH), rosette forming cell, hemagglutinin and hemolysin reactions. Arthus reactivity(3 hours) developed in control mice and test mice pretreated with C. parvum 8 days prior to intravenous sensitization with SRBC were similar. However, there was slight depression of reactivity when C. parvum was given subcutaneoutly(s.c.) 4 or 2 days prior to SRBC sensitization. Arthus reactivity was significantly depressed when C. parvum was given s.c. either at the same time as, or 2 days later than, antigen. DTH reaction was net depressed significantly when C. parvum was injected 8 or 2 days prior to SRBC sensitization or at the same time as antigen. In contrast DTH was significantly augmented when C. parvum given s.c. 4 days prier to SRBC sensitization. DTH was depressed when C. parvum was given s.c. 2 days after antigen. No significant change occurred in rosette forming percetages of spleen cell when C. parvum was given s.c. 8, 4 or 2 days before SRBC sensitization. In contrast, a significant reduction in percentages of rosette forming cell occurred when C. parvum was given s.c. either at the same time as, or 2 days later than, antigen. Serum hemaggulutinin and hemolysin titers were not significantly affected by subcutaneous injection of C. parvum regardless of time relative to SRBC sensitization. However, mercaptoethanol-resistant hemaggulutinin and hemolysin(IgG) titers were somewhat augmented when C. parvum was given 2 days after antigen. It is concluded from these results that depending on the time and route of inoculation, C. parvum can enhance or depress immune responses in mice, suggesting the time and route of C. parvum inoculation is an important point of concern about clinical use of C. parvum for the treatment of cancer.

In this study, some considerations have been suggested in developing on-site techniques to evaluate the sensitization of stainless steels. Electrochemical potentiokinetic reactivation (EPR) technique is known to be a candidate tool for field applications since it enables quantitative assessment in reasonable test time, compared to oxalic etching (ditch) technique. The on-site application of the test method imposes additional restrictions on the selection of the test method (for example, minimum surface preparation requirement, insensitivity to testing temperature, etc.). The EPR and etching techniques have been compared in order to sensitization of stainless steel structures. It has been widely reported that the maximum sensitivity in the welded structure of stainless steel is shown at heat-affected zone (HAZ) than weldments with cast structure. In this work, sectioned weldments and external surfaces were investigated to reveal the degree of sensitization by the etching and the results were compared with those of EPR test. The EPR test showed little sensitivity to surface roughness and test temperature.
