Determination of Aflatoxins Using High-Performance Liquid Chromatography and Fluorescence or UV Absorbence Detection (HPLC에 의한 aflatoxin 분석법에 관한 연구 형광 및 자외선 흡광 검출의 비교)

  • A comparison was made of two detection methods(UV absorbence detection and fluorescence detection with pre-column derivatization, with trifluoroacetic acid) coupled with HPLC for the simultaneous determination of aflatoxin $B_1, B_2, G_1$ and $G_2$. A good separation of the four aflatoxins was achieved on a reversed-phase $C_{18}$ column (30 cm x 3.9 mm) with methanol-acetonitrile-water(20+20+60) for absorbence detection or acetonitrile-water(25+75) for fluorescence detection at the flow rate of 1.0 ml/min. The calibration graphs were linear over the ranges 100 ppb-1 ppm for $B_1/G_1$ and 30~300 ppb for $B_2/G_1$ with absorbence detection, and 1~500 ppb for $B_1/G_1$ and 0.3~150 ppb for $B_2/G_2$ with fluorescence detection. The correlation coefficients were greater than 0.94 and 0.99 for absorbance detection and for fluorescence detection, respectively. The detection limit was 100 ng for $B_1/G_1$ and 30 ng for $B_2/G_2$ with absorbence detection, and 1 ng for $B_1/G_1$ and 0.3 ng for $B_2/G_2$ with fluorescence detection. Recovery rates of aflatoxin $B_1, B_2, G_1$ and $G_2$ added to yeast-extract sucrose broth medium were 66.6%, 59.4%, 67.5% and 59.2%, respectively, for absorbence detection and 82.9%, 71.5%, 80.0% and 69.3%, respectively, for fluorescence detection. The four aflatoxins in culture medium were quantitatively detected by the two methods. The aflatoxins in the rice sample were not detected the absorbence detection method, but were below 10 ppb using the fluorescence detection method. Analysis of aflatoxins by both the absorbence and fluorescence methods coupled with HPLC showed acceptable linearity and good recovery. The absorbence detection was less timeconsuming and safer for treatment. The fluorescence detection was more elective and sensitive though elevated $B_1$ and $G_1$ contents were determined from the TFA-induced conversion of $B_1$ to $B_{2a}$ and $G_1$ to $G_{2a}$.

  • Extraction of quaternary ammonium compounds (choline and betaine) from plant samples (spinach) using ion exchange resin (AG1, OH form) is a very simple and inexpensive approach. However, it is very hard to determine amounts of choline and betaine simultaneously using high-performance liquid chromatography-ultraviolet (HPLC-UV) detection. Unlike choline, betaine has low molar absorptivity in UV-visible (UV-Vis) region, which makes it difficult to carry out UV-Vis detection of betaine. The mixture of quaternary ammonium compounds (choline and betaine) was derivatized using 2-bromo acetophenone as a derivatizing agent. As a result, choline did not react with the derivatizing agent, whereas betaine formed a betaine derivative. This betaine derivative exhibited detectable UV absorption with baseline separation between choline and the betaine derivative. Thus, with this method, choline and betaine can be determined simultaneously by using the HPLCUV method through one-step derivatization, which is an easy, sensitive, and reliable method.

  • Edge detection is an important process in low level image processing. But many proposed methods for edge detection are not very robust to the image noise and are not flexible for different images. To solve the both problems, an algorithm is proposed which eliminate the noise by mean shift algorithm in advance, and then adaptively determine the double thresholds based on gradient histogram and minimum interclass variance, With this algorithm, it can fade out almost all the sensitive noise and calculate the both thresholds for different images without necessity to setup any parameter artificially, and choose edge pixels by fuzzy algorithm.

Quantitative Real-Time PCR Assay for Detection of Paenibacillus polymyxa Using Membrane-Fusion Protein-Based Primers

  • Paenibacillus polymyxa is known to be a plant-growth-promoting rhizobacterium. The present study describes a quantitative polymerase chain reaction (qPCR) assay for the specific detection and quantitation of P. polymyxa using a primer pair based on the sequence of a membrane-fusion protein for the amplification of a 268 bp DNA fragment. This study reports that the qPCR-based method is applicable for the rapid and sensitive detection of P. polymyxa and can be used as an alternative method for agricultural soil monitoring.

Development of Species-Specific PCR Primers for the Detection of Streptococcus sobrinus

  • This study was undertaken to develop species-specific forward and universal reverse PCR primers for the detection of Streptococcus sobrinus. These primers target the variable regions of the 16S ribosomal RNA coding gene (rDNA) and their specificity was tested against 10 strains of S. sobrinus strains and 20 different species of oral bacteria using serial dilutions of the purified genomic DNA of S. sobrinus ATCC $33478^T$. Our data show that species-specific amplicons were obtained from all the S. sobrinus strains tested but not from other species. Both direct and nested PCR could detect as little as 400 pg and 4 fg of genomic DNA from S. sobrinus ATCC $33478^T$, respectively. This result suggests that these PCR primers are highly specific and sensitive and applicable to the detection of S. sobrinus.

  • In this study, laser ablation atomic fluorescence (LAAF) spectroscopy has been applied for a nanometer-scale surface analysis of Na-doped polymethyl methacrylate (PMMA). LAAF spectroscopy is a new sensitive element detection technique which involves atomizing of a sample by the laser ablation and detection of ablated plume by laser-induced fluorescence (LIF) spectroscopy. Using this technique in the detection of Na atoms with Na-doped PMMA, a detection limit is obtained as 36 fg for single laser shot. Further, the depth distribution in the sample is measured with a very high spatial resolution using a two-layer PMMA sample by observing the shot-by-shot LIF intensity from the Na atoms.

  • A new chemosensor based on a self-assembled system has been devised to detect Hg2+ions efficiently. We demonstrated that the amphiphilic building blocks consisting of pyrene and boronic acid (1) aggregate in aqueous solutions and provide an outstanding sensing platform for sensitive detection. The self-assembled 1 exhibited high selectivity and sensitivity for Hg2+ion detection via fluorescence quenching, where the Hg2+ion detection ensued from a fast transmetallation of 1. The Stern-Volmer (SV) quenching constant for its fluorescence quenching by Hg2+ions was approximately 1.58 × 108 M-1. In addition, self-assembled 1 exhibited excellent sensing abilities at nano-molar concentration levels when tap water and freshwater samples were contaminated with of Hg2+ ions.

  • In this paper, a robust traffic light detection method is proposed by using vision sensor and DGPS(Difference Global Positioning System). The conventional vision-based detection methods are very sensitive to illumination change, for instance, low visibility at night time or highly reflection by bright light. To solve these limitations in visual sensor, DGPS is incorporated to determine the location and shape of traffic lights which are available from traffic light database. Furthermore the geometric relationship between traffic light and vision sensor is used to locate the traffic light in the image by using DGPS information. The empirical results show that the proposed method improves by 51% in detection rate for night time with marginal improvement in daytime environment.

  • An object detection algorithm using a modified IDM(Image Differential Method) is proposed for detecting an object in a level crossing area. The conventional object detection method using LASER light has the deadzone that it cannot detect small objects, while the object detection method using image data in a level crossing area can detect such small objects. But the image data in a level crossing area can be changeable easily because the data is outdoor and sensitive to such surrounding environments as the change of the sun beam, the shadow of cars, and so on. So we resolve these problems by adding the normalization and the process for shadow of the image data in a level crossing area to the basic IDM(Image Differential Method).

  • A sensitive method for detection of Hepatitis A virus (HAV) by utilizing gold-DNA probe on an array was developed. Amino- modified oligodeoxynucleotides at the 5' position were arrayed on an activated glass surface to function as capture probes. Sandwich hybridization occurred among capture probes, the HAV amplicon, and gold nanoparticle-supported oligonucleotide probes. After a silver enhancement step, signals were detected by a standard flatbed scanner or just by naked eyes. As little as 100 fM of HAV amplicon could be detected on the array. Therefore, the array technology is an alternative to be applied in detection of HAV due to its low-cost and high-sensitivity.
