Features of Selective Attention shown by Difference of Space Type in Department Stores - Focused on Observation Features Over Observation Time - (백화점 공간의 유형 차이에 나타난 선택적 주의집중 특성 - 주시시간의 경과에 나타난 주시특성을 중심으로 -)

  • For this research with the objects of spaces in two department stores which can be referred to as representative facility of commercial space, observation test has carried out to estimate how much visitors rivet their eyes to the display of shops. In addition, to find out what effect the difference among the department types has on the selective attention to space element, the observation time was applied as a medium for estimation. The followings are the result from analyzing the observation frequency and the observation intensity feature of each section where the characteristics of design could be found at attention. First, both images of A and B had concentrative dominant-observation at left shops. In case of Image A, Customers began to observe the right shops very attentively after 25 seconds, and with Image B, the attentive observation at right and left took place alternatively after 35 seconds. In other words, regardless of the characteristics of shop displays, the left shops were observed first while in case of the observation after the early and middle time-frame the characteristics of shops were found to have effects on observation. Second, the normal observation showed some difference among attention sections over time while on the whole both images of A and B had the same highly attentive observation at the middle space. Accordingly, it could be concluded that the middle space was playing a faithful role as background for commercial spaces. Third, the ignorant observation, which is the opposite to the attentive observation, was found different between the images of A and B. When the ignorant observation is considered to have intentionality, it will be possible to set up the display which may attract the attention aggressively by the process of figuring out the characteristics of ignored shops.

An Automated Water Nitrate Monitoring System based on Ion-Selective Electrodes

  • Purpose: In-situ water quality monitoring based on ion-selective electrodes (ISEs) is a promising technique because ISEs can be used directly in the medium to be tested, have a compact size, and are inexpensive. However, signal drift can be a major concern with on-line management systems because continuous immersion of the ISEs in water causes electrode degradation, affecting the stability, repeatability, and selectivity over time. In this study, a computer-based nitrate monitoring system including automatic electrode rinsing and calibration was developed to measure the nitrate concentration in water samples in real-time. Methods: The capabilities of two different types of poly(vinyl chloride) membrane-based ISEs, an electrode with a liquid filling and a carbon paste-based solid state electrode, were used in the monitoring system and evaluated on their sensitivities, selectivities, and durabilities. A feasibility test for the continuous detection of nitrate ions in water using the developed system was conducted using water samples obtained from various water sources. Results: Both prepared ISEs were capable of detecting low concentrations of nitrate in solution, i.e., 0.7 mg/L $NO_3-N$. Furthermore, the electrodes have the same order of selectivity for nitrate: $NO_3{^-}{\gg}HCO_3{^-}$ > $Cl^-$ > $H_2PO_4{^-}$ > $SO{_4}^{2-}$, and maintain their sensitivity by > 40 mV/decade over a period of 90 days. Conclusions: The use of an automated ISE-based nitrate measurement system that includes automatic electrode rinsing and two-point normalization proved to be feasible in measuring $NO_3-N$ in water samples obtained from different water sources. A one-to-one relationship between the levels of $NO_3-N$ measured with the ISEs and standard analytical instruments was obtained.

  • Efficient ditopic receptor, uranyl(II) N,N'-(ethylenedioxy)benzenebis(salicylideneimine) (3) for beryllium ion has been obtained upon functionalization of 1,2-ethylenedioxybenzene (1) with a uranyl-salphen (salphen = N,N'-phenylenebis(salicylideneimine)) unit. Binding affinities of the receptor, 3 in AN-DMSO (v/v 95:5) solution have been measured for alkali and alkaline earth metal ions by conductometry comparing 1. The results showed that both monotopic 1 and ditopic receptor 3 were selective for $Be^{2+}$ ions over other cations, while especially 3 that can complex both with cations (coordinated to basic oxygen of ethylenedioxybenzene) and anions (coordinated to the Lewis acidic uranyl center) results in an increase of the stability constants by a factor of $10^{2.42}$ with respect to 1. Furthermore, the $Be^{2+}$-3 interactions are demonstrated by $^1H$ NMR experiments in highly polar solvent medium, DMSO-$d_6$. Higher selectivities were also observed for $Be^{2+}$ when the ditopic receptor, 3 was incorporated into PVC membranes and tested as ion selective electrodes at neutral pH.

  • This study focuses on analyzing a subset of consumer activities (especially social medium) in their wedding preparation. The focus of this study is on wedding online communities and understanding the impact of communities on selective behavior for information-acceptance. Data was compiled based on female consumers in their 20s and 30s who actively participate in online community forums. It included female consumers preparing for their weddings and individuals interested in weddings. A total of 211 questionnaires were collected from January $10^{th}$ to $23^{rd}$ in 2012. The online communities were identified from Naver, Yahoo, and online search portals; subsequently, they were rank-sorted based on number of members, visitors, and forum posts. We identified four different characteristic based on the findings from the analysis of on-line wedding communities. The characteristic of these communities were divided into sharing experience, functionality of the webpage, informativeness, and interactivity; consequently, use of these online communities is based on trust and significant personal relationships between the members online. Out of the four different community characteristics, sharing experience was found to have a greater impact for the selective behavior of wedding dresses and information- acceptance than the functionality of the webpage, interactivity, and informativeness. We conclude that trust in information provided by members with marriage process experience is the foremost important factor in the behavior of individual consumers wit iexplore.exe -extoff hout marriage process experience. Therefore, the impact of these online communities catering to would-be brides is based on the trust of posters and how well it is articulated.

Agrobacterium- mediated Genetic Transformation and Plant Regeneration of Sweetpotato (Ipomoea batatas) (Agrobacterium 매개에 의한 고구마 형질전환 및 식물체 재분화)

  • Transformed sweetpotato (Ipomoea batatas (L.) Lam. cv. Yulmi) plants were developed from embryogenic calli following Agrobacterium tumefaciens-mediated transformation. A. tumefaciens strain EHA105/pCAMBIA2301 harboring genes for intron $\beta$-glucuronidase (GUS) and kanamycin resistance. Transient expression of GUS gene was found to be higher when embryogenic calli were co-cultivated with Agrobacterium for 2 days. The co-cultured embryogenic calli transferred to selective MS medium containing 1mg/L 2,4-D, 100mg/L kanamycin, and 400mg/L claforan. These embryogenic calli were subcultured to the same selection medium at 4 weeks interval. Kanamycin-resistant calli transferred to hormone-free MS medium with kanamycin gave rise to somatic embryos and then converted into plantlets in the same medium. Southern blot analysis confirmed that the GUS gene was inserted into the genome of the sweetpotato plants. A histochemical assay revealed that the GUS gene was preferentially expressed in the leaf, petiole, and vascular tissue and tip of root.

Agrobacterium-mediated Transformation of Eleutherococcus sessiliflorus using Embryogenic Calli and the Regeneration of Plants (오갈피(Eleutherococcus sessiliflorus)의 배형성 세포를 이용한 고빈도 형질전환 및 재분화)

  • We have developed a reliable and high-frequency genetic transformation and regeneration system via somatic embryogensis of Eleutherococcus sessiliflorus. Embryogenic callus obtained from seed were co- cultivated with Agrobacterium tumefaciens strain EHA101/pIG121Hm harboring genes for intron-$\beta$-glucoronidase(GUS), kanamycin and hygromycin resistance. Following co-cultivation, two types of samples(fine embrogenic calli and early globular embryo clusters) were cultivated on Murashige and Skoog(MS) medium containing 1 mg/L2.4-D for 3day in dark. Transient expression of GUS gene was found to be higher in the early globular embryo clusters than in the embryogenic calli. Also, co-cultivated period affected expression of GUS gene; the best result was obtained when globular embryo clusters were co-cultivated with Agrobacterium for 3 days. Subsequently, this callus transferred to selective MS medium containing 1mg/L2.4-D, 50mg/L kanamycin or/and 30mg/L hygromycin and 300mg/L cefortaxime. These embryogenic calls were subcultured to the same selection medium at every 2 weeks intervals. Approximately 24.5% of the early globular embryos co-cultivated with Agrobacterium for 3days produced kanamycin or/and hygromycin-resistant calli. Transgenic somatic embryos were converted into plantlets in half strength MS medium supplemented with 3mg/L GA$_3$ kanamycin and were confirmed by GUS histochemical assay and polymerase chain reaction analysis. Genomic Southem blot hybridization confirmed the incorporation of NPT II gene into the host genome.

Production and Characterization of Nitrate Reductase Deficient Mutants in Petunia parviflora

  • Nitrate reductase deficient (NR) mutant lines were selected indirectly by their resistance to 100mM chlorate in cell cultures of P. parviflora. A total of 585 chlorate resistant lines were confirmed by a second passage on a high concentration of chlorate. Frequency of spontaneous mutation was $9.7{\times}10^{-7}$ in 3 month old suspension-cultured cells, and in non-selective media containing amino acids as sole nitrogen source. The frequency of mutation could be increased up to 11-fold by culture for 12 months. Out of 40 randomly selected calli, 22 were fully deficient in NR. The rest of the clones contained a decreased level of NR activity. Further characterization was carried out in 13 mutant lines which were fully deficient in NR and in 5 mutant lines containing residual (0-7.0%) NR activity, as compared to wild-type cells cultured on the same medium. The $NR^-$ mutants were tentatively classified as defective in the NR apoenzyme (nia-type; 11 mutant lines including the 5 with residual NR activity) or in the molybdenum cofactor (cnx-type; 7 mutant lines) by the XDH activity. The cnx-type could be further classified into two groups. In one group (5 mutant lines) of these, the NR activity could be partially restored by nonphysiologically high (1.0mM) molybdate in the culture medium. Both types of $NR^-$ mutants were unable to grow on minimal medium containing nitrate as sole nitrogen source, but grew well on amino acids. They also proved to be extremely sensitive to the standard medium ($MSP_1$) containing nitrate and ammonium. Shoot regeneration was obtained only in the $NR^-$ mutants, which contained residual NR activity, but they so far have failed to grow into plants.

Evaluation of Mechanical Characteristic and Biological Stability of Dental Alloys by the Manufacture Method (가공방법에 따른 치과용 합금의 기계적 특성 및 생물학적 안정성 평가)

  • The material of the dental prosthesis was required bio-compatibility for biological, chemical, and physical stabilities. This study was conducted the stability evaluation of mechanical, biological characteristics through comparing Co-Cr alloy(SC group), Ti alloy(ST group) made by the selective laser melting method with Co-Cr alloy(CC group), Ni-Cr alloy(CN group) made by the casting method. Modulus of elasticity for mechanical characteristic evaluation was measured by the tensile test. And we conducted the release material analysis using lactic acid-NaCl solution for the evaluation of biological stability and were observed cytotoxicity through the content of this release medium. Taking these results into account, the Co-Cr alloy made by the selective laser melting method was observed modulus of elasticity higher than he Co-Cr alloy made by the casting method. And the Co-Cr alloy made by the selective laser melting method had more superior biological stability than the other groups as the result of cytotoxicity evaluation through the release material analysis. By this results, we think that alloys made by the selective laser melting method can be applied as materials for making the dental prosthesis.

Semi-selective Medium for Monitoring Colletotrichum acutatum Causing Pepper Anthracnose in the Field (고추 탄저병균Colletotrichum acutatum 의 포장 밀도 조사를 위한 반선택 배지의 확립 및 활용)

  • It was confirmed that anthracnose pathogen, Colletotrichum acutatum, could specifically grow on PDA amended with $100\mu\textrm{g}$ /ml of ampicillin and tetracycline, and 100 $100\mu\textrm{g}$/ml of mixture with carbendazim and diethofencarb. There was a positive correlation between the number of colony enumerated on semi-selective media and the disease severity on pepper fruits caused by C. acutatum. Using semi-selective media for C. acutatum, the number of pathogen on soil and plant debris infected by anthracnose pathogen was investigated. In plant debris, the colony number of C. acutatum was more than in soil. For the identification of colony appeared on semi-selective media, 10 isolates were selected randomly. They were identified as C. acutatum through PCR using C. acutatum-specific primer.

Isolation of the Phytophthora Root Rot Pathogen of Atractylodes macrocephala, Phytophthora drechsleri, and Bioassay of the isolates with Seedlings (백출의 역병균 분리 및 유묘를 이용한 병원성 정정 방법의 확립)

  • Using semi-selective (SS-) and selective (Jee-) medium, we identified the pathogens isolated from the symptomatic plants and soils collected from different locations, such as Suwon, Andong, and Youngju, as P. drechsleri, which is Phytophthora root rot causal agent of A. macrocephala. At $25^{\circ}C$, these isolates were grown faster on 10% V8A (V8 juice agar) medium than on PDA (potato dextrose agar) with hyphal swelling, but no growing was observed at below $5^{\circ}C$ and over $40^{\circ}C$. In order to identify the pathogenicity of each isolate, seedlings of A. macrocephala were inoculated with mycelium -zoospore suspended inoculum, which was prepared by culturing on 10% V8A medium and homogenizing in distilled water. By this method, wide ranges of pathogenicity were observed as follows; $5.0%{\sim}26.4%$ of disease severities concerning the lesion areas of the top plants and $23.5%{\sim}72.2%$ of disease incidences. Therefore, this was considered as a efficient method to identify the pathogenicity of P. drechsleri in large scale screening. P-A200073, isolated from soils in Andong, and P-9755, from the root of symptomatic plant of A. macrocephala in Suwon, showed the highest degree of pathogenicity to the seedlings. By these isolates, lesion areas and disease incidences of the inoculated seedlings were occurred $26.4%{\sim}63.2%$ and $25.1%{\sim}72.2%$, respectively. However, no symptoms were observed in uninoculated control. Same pathogens were reisolated from roots and lower stems of the inoculated plants, but not from leaves.
