새로운 효소반응기를 이용한 요소의 연속·자동화 정량 (Continuous Automated Determination of Urea Using a New Enzyme Reactor)

  • 새로운 효소반응기를 쓰는 요소 정량용 연속${\cdot}$자동화 장치의 감응성질을 조사하였다. 효소반응기는 지지체인 nylon-6입자(42∼48 mesh)를 teflon관(안지름 2mm, 길이 20cm)에 충전시키고, 이 지지체의 표면에 공유결합제인 glutaraldehyde로 urease를 고정화시켜서 제작하였다. 연속${\cdot}$자동화장치는 효소반응기, 기체투석기 및 지시전극인 관형 PVC-nonactin막 암모늄 이온 선택성 전극을 차례로 연결하여 만들었다. 이 장치를 써서 요소를 정량할 때 감응특성은 다음과 같다. 곧 직선감응 농도범위는 $5.5{\times}10^{-6}$~$2.4{\times}10^{-3}M$, 감응기울기는 57.8 mV/decade, 검출한계는 $1.5{\times}10^{-6}M$, 효소반응기의 전환백분율은 80.8%이었다. 효소반응기의 최적 완충용액은 0.01M Tris-HCl 완충용액(pH 7.0∼7.8)과 0.01M 인산염 완충용액(pH 6.9∼7.5)이었고, 수명은 150일 정도였다. 또한 다른 생리활성물질의 방해는 없었다.

혼합 약한 분류기를 이용한 AdaBoost 알고리즘의 성능 개선 방법 (A Method to Improve the Performance of Adaboost Algorithm by Using Mixed Weak Classifier)

  • The weak classifier of AdaBoost algorithm is a central classification element that uses a single criterion separating positive and negative learning candidates. Finding the best criterion to separate two feature distributions influences learning capacity of the algorithm. A common way to classify the distributions is to use the mean value of the features. However, positive and negative distributions of Haar-like feature as an image descriptor are hard to classify by a single threshold. The poor classification ability of the single threshold also increases the number of boosting operations, and finally results in a poor classifier. This paper proposes a weak classifier that uses multiple criterions by adding a probabilistic criterion of the positive candidate distribution with the conventional mean classifier: the positive distribution has low variation and the values are closer to the mean while the negative distribution has large variation and values are widely spread. The difference in the variance for the positive and negative distributions is used as an additional criterion. In the learning procedure, we use a new classifier that provides a better classifier between them by selective switching between the mean and standard deviation. We call this new type of combined classifier the "Mixed Weak Classifier". The proposed weak classifier is more robust than the mean classifier alone and decreases the number of boosting operations to be converged.

  • 감도가 높고 선택적인 추출-분광광도법은 미량의 시메티딘의 측정을 위해 개발되어 왔다. 이 방법은 클로로포름안에 브로모티몰 블루(BTB)와 함께 이온 쌍으로 시메티딘의 추출에 기초했다. 그리고 417 nm에서 흡광도를 측정하고 있다. pH, BTB의 농도, 클로로포름의 부피, 섞는 시간과 같은 다른 변수들의 효과를 연구하였다. 또한, 추출에서 간섭이온의 효과도 연구하였다. 보정곡선은 0.9997의 상관계수와 함께 0.25~8 ug/mL의 범위에서 선형이다. 3Sb에 기초한 검출한계는 0.14 ug/mL이고 시메티딘의 0.1과 4.0 ug/mL의 10번 측정에 대한 상대표준편차는 3.2와 1.49%이었다. 제안된 방법은 좋은 회수와 함께 약제 샘플에서 시메티딘의 측정에 적용되었다.

Kojic Acid, a Potential Inhibitor of NF-$textsc{k}$B Activation in Transfectant Human HaCaT and SCC-13 Cells

  • The activation of NF-$\kappa$B induced by kojic Acid, an inhibitor of tyrosinase for biosynthesis of melanin in melanocytes, was investigated in human transfectant HaCaT and SCC-13 cells. These two keratinocyte cell lines transfected with pNF-$\kappa$B-SEAP-NPT plasmid were used to determine the activation of NF-$\kappa$B. Transfectant cells release the secretory alkaline phosphatase (SEAP) as a transcription reporter in response to the NF-$\kappa$B activity and contain the neomycin phosphotransferase (NPT) gene for the dominant selective marker of geneticin resistance. NF-$\kappa$B activation was measured in the SEAP reporter gene assay using a fluorescence detection method. Kojic Acid showed the inhibition of cellular NF-$\kappa$B activity in both human keratinocyte transfectants. It could also downregulate the ultraviolet ray (UVR)-induced activation of NF-$\kappa$B expression in transfectant HaCaT cells. Moreover, the inhibitory activity of kojic Acid in transfectant HaCaT cells was found to be more potent than known antioxidants, e.g., vitamin C and N~acetyl-L-cysteine. These results indicate that kojic Acid is a potential inhibitor of NF-$\kappa$B activation in human keratinocytes, and suggest the hypothesis that NF-$\kappa$B activation may be involved in kojic Acid induced anti-melanogenic effect.

첨가제 및 패턴인식에 의한 후막 SnO2 가스센서의 선택성 향상 (The Enhancement of Selectivity in Thick Film SnO2 Gas Sensors by Additives and Pattern Recognition)

  • Sn $O_2$ 가스센서는 낮은 농도의 가연성 가스 및 유독 가스를 표면 저항의 변화로부터 탐지할 수 있으나, 가스 선택성이 부족하다는 단점을 가지고 있다. 이러한 단점을 보완하기 위해서는 가스반응기구의 규명과 같은 기초이론 연구와 함께 선택성이 우수한 센서재료의 개발 및 적절한 신호처리방법의 적용이 필요하다. 본 논문에서는 Sn $O_2$ 표면에서 일어나는 에탄올 (C$_2$ $H_{5}$OH)과 아세토니트릴($CH_3$CN)의 촉매산화반응을 가스크로마토그래피 분석을 통해 확인하였다. PdCl$_2$가 첨가된 Sn $O_2$ 센서는 에탄올과 아세토니트릴에 대하여 높은 감도를 보였고, 반면에 La$_2$ $O_3$가 첨가된 Sn $O_2$ 센서는 에탄올에 대해서는 높은 감도를, 그리고 아세토니트릴에 대해서는 낮은 감도를 보였다. 이들 두 센서재료 개발 및 패턴인식기법적용을 통하여 아세토니트릴에 대한 선택성을 크게 증가시킬 수 있었다. 아세토니트릴에 대한 최소 탐지농도는, 공기 중에서는 15 ppm이었고, 다른 방해가스와 함께 존재할 경우에는 20 ppm에서 100 ppm 정도로 나타났다.

Implant Anneal Process for Activating Ion Implanted Regions in SiC Epitaxial Layers

  • The mechanical strength of silicon carbide dose nor permit the use of diffusion as a means to achieve selective doping as required by most electronic devices. While epitaxial layers may be doped during growth, ion implantation is needed to define such regions as drain and source wells, junction isolation regions, and so on. Ion activation without an annealing cap results in serious crystal damage as these activation processes must be carried out at temperatures on the order of 1600$^{\circ}C$. Ion implanted silicon carbide that is annealed in either a vacuum or argon environment usually results in a surface morphology that is highly irregular due to the out diffusion of Si atoms. We have developed and report a successful process of using silicon overpressure, provided by silane in a CAD reactor during the anneal, to prevent the destruction of the silicon carbide surface, This process has proved to be robust and has resulted in ion activation at a annealing temperature of 1600$^{\circ}C$ without degradation of the crystal surface as determined by AFM and RBS. In addition XPS was used to look at the surface and near surface chemical states for annealing temperatures of up to 1700$^{\circ}C$. The surface and near surface regions to approximately 6 nm in depth was observed to contain no free silicon or other impurities thus indicating that the process developed results in an atomically clean SiC surface and near surface region within the detection limits of the instrument(${\pm}$1 at %).

표면 검출과 볼륨 확장을 이용한 삼차원 물체의 선택 분할 (Selective Segmentation of 3-D Objects Using Surface Detection and Volume Growing)

  • 삼차원 볼륨 영상으로부터 대상 물체를 분할하는 것은 가시화 또는 볼륨 측정을 위해서 매우 중요한 단계이다. 본 논문에서는 볼륨 가시화를 위해 널리 사용되는 르보이 필터링 방법을 개선하여 물체의 표면을 검출하는 방법을 제시한다. 그리고 형태학적 연산자를 이용하여 완전히 닫힌 표면을 생성하고 볼륨 확장 알고리즘에 의해 물체를 선택적으로 분할한다. 제시된 방법은 합성된 삼차원 구 영상과 심혈관 조영영상에 적용되었다. 이 방법을 합성된 구 영상을 사용하여 기존의 브로이 필터링과 정량적으로 비교한 결과 제시한 방법이 복셀 오차면에서 더 우수하였다. 또한 심혈관 영상을 사용하여 시각적으로 비교한 결과 역시 제시한 방법이 더 정확하였다. 본 논문에서 제시한 방법은 삼차원 영상처리에서 자주 함께 사용되는 분할, 가시화, 측정을 쉽게 연계할 수 있기 때문에 볼륨 영상의 분할을 위해 매우 효고적이다.

  • 사람 아이치바이러스(Aichivirus A; AiV-A)는 positivesense single-strand RNA 비외피 바이러스로 지난 10년 동안 하수, 강, 지표 및 지상의 다양한 물환경에서 전 세계적으로 검출이 보고되고 있다. 지하수 등 물환경에서 AiV-A 진단을 위한 고감도 및 특이성이 우수한 방법의 개발이 요구됨에 따라, 본 연구에서는 기존 및 신규 설계된 프라이머 세트를 기초로 역전사(RT) 및 이중 중합효소연쇄반응이 가능한 조합을 개발하였다. 개발한 방법을 국내 음용 지하수 시료에 적용 및 평가하였으며, 그 결과 지하수 시료에서 AiV-A를 성공적으로 검출 및 동정할 수 있는 RT-nested PCR primer sets가 선정되었고 후속적으로 동정할 수 있는 절차가 고안되었다. 본 연구 결과는 지하수 등 물 환경에서 AiV-A 오염을 탐지하기 위한 모니터링 시스템 마련에 기여할 것으로 기대된다.

Imprinted Graphene-Starch Nanocomposite Matrix-Anchored EQCM Platform for Highly Selective Sensing of Epinephrine

  • In this paper, an electrochemical sensor for epinephrine (EP), a neurotransmitter was developed by anchoring molecularly imprinted polymeric matrix (MIP) on the surface of gold-coated quartz crystal electrode of electrochemical quartz crystal microbalance (EQCM) using starch nanoparticles (Starch NP) - reduced graphene oxide (RGO) nanocomposite as polymeric format for the first time. Use of EP in therapeutic treatment requires proper dose and route of administration. Proper follow-up of neurological disorders and timely diagnosis of them has been found to depend on EP level. The MIP sensor was developed by electrodeposition of starch NP-RGO composite on EQCM electrode in presence of template EP. As the imprinted sites are located on the surface, high specific surface area enables good accessibility and high binding affinity to template molecule. Differential pulse voltammetry (DPV) and piezoelectrogravimmetry were used for monitoring binding/release, rebinding of template to imprinted cavities. MIP-coated EQCM electrode were characterized by contact angle measurements, AFM images, piezoelectric responses including viscoelasticity of imprinted films, and other voltammetric measurements including direct (DPV) and indirect (using a redox probe) measurements. Selectivity was assessed by imprinting factor (IF) as high as 3.26 (DPV) and 3.88 (EQCM). Sensor was rigorously checked for selectivity in presence of other structurally close analogues, real matrix (blood plasma), reproducibility, repeatability, etc. Under optimized conditions, the EQCM-MIP sensor showed linear dynamic ranges ($1-10{\mu}M$). The limit of detection 40 ppb (DPV) and 290 ppb (EQCM) was achieved without any cross reactivity and matrix effect indicating high sensitivity and selectivity for EP. Hence, an eco-friendly MIP-sensor with high sensitivity and good selectivity was fabricated which could be applied in "real" matrices in a facile manner.

Discovery of a new primer set for detection and quantification of Ilyonectria mors-panacis in soils for ginseng cultivation

  • Background: Korean ginseng is an important cash crop in Asian countries. However, plant yield is reduced by pathogens. Among the Ilyonectria radicicola-species complex, I. mors-panacis is responsible for root-rot and replant failure of ginseng in Asia. The development of new methods to reveal the existence of the pathogen before cultivation is started is essential. Therefore, a quantitative real-time polymerase chain reaction method was developed to detect and quantify the pathogen in ginseng soils. Methods: In this study, a species-specific histone H3 primer set was developed for the quantification of I. mors-panacis. The primer set was used on DNA from other microbes to evaluate its sensitivity and selectivity for I. mors-panacis DNA. Sterilized soil samples artificially infected with the pathogen at different concentrations were used to evaluate the ability of the primer set to detect the pathogen population in the soil DNA. Finally, the pathogen was quantified in many natural soil samples. Results: The designed primer set was found to be sensitive and selective for I. mors-panacis DNA. In artificially infected sterilized soil samples, using quantitative real-time polymerase chain reaction the estimated amount of template was positively correlated with the pathogen concentration in soil samples ($R^2=0.95$), disease severity index ($R^2=0.99$), and colony-forming units ($R^2=0.87$). In natural soils, the pathogen was recorded in most fields producing bad yields at a range of $5.82{\pm}2.35pg/g$ to $892.34{\pm}103.70pg/g$ of soil. Conclusion: According to these results, the proposed primer set is applicable for estimating soil quality before ginseng cultivation. This will contribute to disease management and crop protection in the future.
