Zn2+ PVC-based Membrane Sensor Based on 3-[(2-Furylmethylene)amino]-2-thioxo-1,3-thiazolidin-4-one

  • The 3-[(2-furylmethylene)amino]-2-thioxo-1,3-thiazolidin-4-one (FTT) was used as an excellent ionophore in construction of a $Zn^{2+}$ PVC-based membrane sensor. The best performance was obtained with a membrane composition of 30% poly(vinyl chloride), 62% nitrobenzen (NB), 3% FTT and 5% sodium tetraphenyl borate (TBP). This membrane sensor shows very good selectivity and sensitivity towards $Zn^{2+}$ over a wide variety of cations, including alkali, alkaline earth, transition and heavy metal ions. The membrane sensor revealed a great enhancement in selectivity coefficients for $Zn^{2+}$ ions, in comparison to the previously reported $Zn^{2+}$ membrane sensors. Theoretical studies also showed the selective interaction of TFF and $Zn^{2+}$ ions. The proposed membrane sensor exhibits a Nernstian behavior (with slope of 29.3 ${\pm}$ 0.3 mV per decade) over a wide concentration range (1.0 ${\times}$ $10^{-6}$-1.0 ${\times}$ $10^{-2}$) with a detection limit of 8.5 ${\times}$ $10^{-7}$ M (52 ng mL$^{-1}$). It shows relatively fast response time, in the whole concentration range ($\lt$ 20 s), and can be used for at least 10 weeks in a pH range of 3.0-7.0. The proposed membrane sensor was successfully used in direct determination of $Zn^{2+}$ ions in wastewater of industrial zinc electroplating companies, and also as an indicator electrode in titration with EDTA.

A High-performance Lane Recognition Algorithm Using Word Descriptors and A Selective Hough Transform Algorithm with Four-channel ROI (다중 ROI에서 영상 화질 표준화 및 선택적 허프 변환 알고리즘을 통한 고성능의 차선 인식 알고리즘)

  • The examples that used camera in the vehicle is increasing with the growth of the automotive market, and the importance of the image processing technique is expanding. In particular, the Lane Departure Warning System (LDWS) and related technologies are under development in various fields. In this paper, in order to improve the lane recognition rate more than the conventional method, we extract a Normalized Luminance Descriptor value and a Normalized Contrast Descriptor value, and adjust image gamma values to modulate Normalized Image Quality by using the correlation between the extracted two values. Then, we apply the Hough transform using the optimized accumulator cells to the four-channel ROI. The proposed algorithm was verified in 27 frame/sec and $640{\times}480$ resolution. As a result, Lane recognition rate was higher than the average 97% in day, night, and late-night road environments. The proposed method also shows successful lane recognition in sections with curves or many lane boundary.

Voltammetric Determination of Copper(II) Using Glassy Carbon Electrodes Modified with Nafion-DTPA-Glycerol

  • A glassy carbon electrode(GCE) modified with nafion-DTPA-glycerol was used for the highly selective and sensitive determination of a trace amount of Cu$\^$2+/. Various experimental parameters, which influenced the response of nafion-DTPA-glycerol modified electrode to Cu$\^$2+/, were optimized. The copper(II) was accumulated on the electrode surface by the formation of the complex in an open circuit, and the resulting surface was characterized by medium exchange, electrochemical reduction, and differential pulse voltammetry, A linear range was obtained in the concentration range 1.0${\times}$10$\^$-8/M∼1.0${\times}$10$\^$-6/M Cu(II) with 7 min preconcentration. Further, when an approximate amount of lead(II) is added to the test solution, nafion-DTPA-glycerol modified glassy carbon electrode has a dynamic range of 2 orders magnitude(1.0${\times}$10$\^$-9/M∼1.0${\times}$10$\^$-7/M). The detection limit(3 $\sigma$) was as low as 5.0${\times}$10$\^$-6/M(0.032ppb). The interferences from other metal ions could be reduced by adding KCN into the sample solutions. This method was applied to the determination of coppe,(II) in certified reference material(3.23${\times}$10$\^$-7/M, 21ppb), sea water(9.50${\times}$10/sup-7/M, 60ppb). The result agrees satisfactorily with the value measured by Korea Research Institute of Standard and Science.

Correlationship of Vertical Distribution for Ammonia Ion, Nitrate Ion and Nitrifying Bacteria in a Fixed Bed Nitrifying Biofilm

  • The vertical distributions of nitrifying bacteria in aerobic fixed biofilm were investigated to evaluate the relationship between nitrification performance and microbial community at different HRT. Fluorescent in situ hybridization (FISH) and portable ion selective microelectrode system were adopted to analyze microbial communities and ions profiles according to the biofilm depth. Cilia media packed MLE (Modified Ludzack-Ettinger) like reactor composed of anoxic, aerobic I/II was operated with synthetic wastewater having COD 200 mg/L and $NH_4{^+}$-N mg/L at HRT of 6 hrs and 4 hrs. Total biofilm thickness of aerobic I, II reactor at 4 hrs condition was over two times than that of 6 hrs condition due to the sufficient substrate supply at 4 hrs condition (6 hrs; aerobic I 380 ${\mu}m$ and II 400 ${\mu}m$, 4 hrs; aerobic I 830 ${\mu}m$ and II 1040 ${\mu}m$). As deepen the biofilm detection point, the ratio of ammonia oxidizing bacteria (AOB) was decreased while the ratio of nitrite oxidizing bacteria (NOB) was maintained similar distribution at both HRT condition. The ratio of AOB was higher at 4 hrs than 6 hrs condition and $NH_4{^+}$-N removal efficiency was also higher at 4 hrs with 89.2% than 65.4% of 6 hrs. However, the ratio of NOB was decreased when HRT was reduced from 6 hrs to 4 hrs and $NO_2{^-}$-N accumulation was observed at 4 hrs condition. Therefore, it is considered that insufficient HRT condition could supply sufficient substrate and enrichment of AOB in all depth of fixed biofilm but cause decrease of NOB and nitrite accumulation.

  • A new electrochemical sensor to selectively determine epinephrine was developed and its analytical characteristics has been investigated. A glassy carbon electrode was modified with Ni(II)-macrocyclic complex which has electrocatalytic effect. It was further modified with physiologically suitable and negatively charged polyuretane benzyl L-glutamate(PUBLG). The present electrode showed long term stability and it could be applied to the selective determination of epinephrine in urine sample with various coexisting compounds. Under the optimum experimental conditions the linear range was $8.0\;{\times}\;10^{-7}\;-\;2.0\;{\times}\;10^{-4}\;M$ and the limit of detection was $1.0\;{\times}\;10^{-7}\;M$. The recovery of epinephrine in urine sample diluted 5 times with buffer solution was $101.5({\pm}3.2)%$ for 6 measurements.

  • This paper presents a robust viewpoint estimation algorithm for Moving Parallax Barrier mobile 3D display in sudden illumination changes. We analyze the previous viewpoint estimation algorithm that consists of the Viola-Jones face detector and the feature tracking by the Optical-Flow. The sudden changes in illumination decreases the performance of the Optical-flow feature tracker. In order to solve the problem, we define a novel performance measure for the Optical-Flow tracker. The overall performance can be increased by the selective adoption of the Viola-Jones detector and the Optical-flow tracker depending on the performance measure. Various experimental results show the effectiveness of the proposed method.

  • Microbeam is a new avenue of radiation research especially in radiation biology and radiation protection. Selective irradiation of an ionizing particle to a targeted cell organelle may disclose such mechanisms as signal transaction among cell organelles and cell-to-cell communication in the processes toward an endpoint observed. Bystander effect, existence of which is clearly evidenced by application of the particle microbeam to biological experiments, suggests potential underestimation in the conventional risk estimation at low particle fluence rates, such as environment of space radiations in ISS (International Space Station). To promote these studies we started the construction of our microbeam facility (named as SPICE) to our HVEE Tandem accelerator (3.4 MeV proton and 5.1 MeV $^4$He$\^$2+/). For our primary goal, "irradiation of single particle to cell organelle within a position resolution of 2 micrometer in a reasonable irradiation time", special features are considered. Usage of a triplet Q magnet for focussing the beam to submicron of size is an outstanding feature compared to facilities of other institutes. Followings are other features: precise position control of cell dish holder, design of the cell dish, data acquisition of microscopic image of a cell organelle (cell nucleus) and data processing, a reliable particle detection, soft and hard wares to integrate all these related data, to control and irradiate exactly determined number of particles to a targeted spot.

Fabrication of Low-cost and Flexible Potassium Ion Sensors based on Screen Printing and Their Electrochemical Characteristics (스크린 프린팅 기반 저가형의 플렉서블 칼륨 이온 센서 제조 및 이의 전기화학적 특성)

  • A low-cost and flexible potassium ion (K+) sensor was fabricated through a screen-printed process. Uniform and conformal coating of conductive inks was verified by scanning electron microscopy and optical microscopy measurements. The K+-sensors showed a high sensitivity, fast response time, and low detection limit. The sensitivity of K+-sensor was similar to that of both mechanically normal and bent states. The K+-sensor exhibited a good reproducibility with no hysteresis effect and excellent long term stability. In addition, the K+-sensor showed an excellent selectivity for K+ concentrations in the presence of other interfering cation ions. Successful measurements of K+ concentrations in sports drink samples were demonstrated by comparing K+ concentration values from K+-sensor to those of using a commercial K+-meter.

  • In this work, we prepared porous WO3 nanofibers (NFs) functionalized by bio-inspired catalytic $Cr_2O_3$ and $Co_3O_4$ nanoparticles as highly sensitive and selective $H_2S$ gas sensing layers. Highly porous 3-dimensional (3D) NFs networks decorated by well-dispersed catalyst NPs exhibited superior $H_2S$ gas response ($R_{air}/R_{gas}$ = 46 at 5 ppm) in high humidity environment (95 %RH). In particular, the sensors showed outstanding $H_2S$ selectivity against other interfering analytes (such as acetone, toluene, CO, $H_2$, ethanol). Exhaled breath sensors using $Cr_2O_3$ and $Co_3O_4$ catalysts-loaded $WO_3$ NFs are highly promising for the accurate detection of halitosis.

Genetic Characterization of Molecular Targets in Korean Patients with Gastrointestinal Stromal Tumors

  • Purpose: Gastrointestinal stromal tumors (GISTs) frequently harbor activating gene mutations in either KIT or platelet-derived growth factor receptor A (PDGFRA) and are highly responsive to several selective tyrosine kinase inhibitors. In this study, a targeted next-generation sequencing (NGS) assay with an Oncomine Focus Assay (OFA) panel was used for the genetic characterization of molecular targets in 30 Korean patients with GIST. Materials and Methods: Using the OFA that enables rapid and simultaneous detection of hotspots, single nucleotide variants (SNVs), insertion and deletions (Indels), copy number variants (CNVs), and gene fusions across 52 genes relevant to solid tumors, targeted NGS was performed using genomic DNA extracted from formalin-fixed and paraffin-embedded samples of 30 GISTs. Results: Forty-three hotspot/other likely pathogenic variants (33 SNVs, 8 Indels, and 2 amplifications) in 16 genes were identified in 26 of the 30 GISTs. KIT variants were most frequent (44%, 19/43), followed by 6 variants in PIK3CA, 3 in PDGFRA, 2 each in JAK1 and EGFR, and 1 each in AKT1, ALK, CCND1, CTNNB1, FGFR3, FGFR4, GNA11, GNAQ, JAK3, MET, and SMO. Based on the mutation types, majority of the variants carried missense mutations (60%, 26/43), followed by 8 frameshifts, 6 nonsense, 1 stop-loss, and 2 amplifications. Conclusions: Our study confirmed the advantage of using targeted NGS with a cancer gene panel to efficiently identify mutations associated with GISTs. These findings may provide a molecular genetic basis for developing new drugs targeting these gene mutations for GIST therapy.
