동백잎 추출물의 신생혈관 및 세포부착 억제작용과 그 기전 (Anti-angiogenic and Anti-cell Adhesion Effects and Their Mechanism with the Extract of Camellia japonica Leaf)

  • 동백잎의 열수 추출물이 신생혈관 생성억제 효과가 강하게 나타남으로써 이들 추출물에 대한 독성 시험을 HUVECs를 사용하여 검토한 바는 200 ug/mL에서도 독성이 없는 것으로 나타났으며, 1.5, 3.0, 15 및 30 ug/mL으로 농도가 증가함에 따라 각각 30.7%, 38.5%, 53.8% 그리고 70.0%의 신생혈관 생성억제율을 보였다. 세포부착 저해효과는 C. japonica leaf (CJL)의 농도가 50, 100, $200{\mu}g{/well}$으로 증가할 때 E-selectin이 46.7%, 66.7% 그리고 86.76%, VCAM-1이 23.0%, 61.5% 그리고 84.6%, ICAM-1이 11.0%, 55.5% 그리고 88.8%로 나타났다. C. japonica leaf (CJL)의 성분 증가에 따라 발현이 감소되는 것을 보아 농도가 증가함에 따라 cell adhesion의 저해 효과가 높아짐을 알 수 있었다. 신호전달의 기전규명은 western blot으로 행하였으며 CJL의 농도가 증가함에 따라 밴드의 발현이 약해지는 것을 관찰할 수 있다. 따라서 신호전달 분자인 VEGFR-2, $\beta$-catenin, Pl3-K는 CJL에 의해 신호전달이 차단되는 것을 볼 수 있고, 이는 NF-${\kappa}$B를 억제함으로서 신생혈관 생성을 저해하는 것으로 확인되었다. 따라서 동백잎은 신생혈관 생성에 의존하고 있는 것으로 알려진 암 등의 치료와 암전이의 억제, 류마치스성 관절염, 그리고 항비만제제로서 개발될 수 있음을 시사한다.

사람의 피부상피세포에서 황색포도상구균의 독소인자인 Staphylococcal Protein A의 염증반응 촉진효과 (Stimulatory Effect of Staphylococcal Protein A on Inflammatory Response in Human HaCaT Keratinocytes)

  • 황색포도상구균은 사람에게서 염증을 동반한 다양한 형태의 국소적 또는 전신적 감염을 일으키는 주요 병원균이며, 황색포도상구균에서 풍부하게 발현되는 Staphylococcal protein A (SPA)는 염증의 활성화나 면역 반응의 회피와 관련된 균력인자로서 작용할 수 있다. 본 연구에서는 사람의 HaCaT 피부상피세포에서 재조합 SPA 단백질을 이용하여 염증반응에 대한 효과를 조사하기 위해서 pET-28a 발현벡터시스템을 이용하여 성공적으로 재조합 SPA 단백질을 제작하였고, 이 단백질(2 ${\mu}g$/ml)을 6, 12 및 24시간 처리한 HaCaT 피부상피세포에서 RT-PCR 및 ELISA를 이용하여 염증관련 부착인자 및 사이토카인의 발현을 분석하였다. SPA 처리 후 6시간에서 24시간까지 E-selectin, ICAM-1, MCP-1, IL-6 및 IL-8의 발현이 현저하게 증가함을 확인하였다. 또한 SPA는 HaCaT 피부상피세포에 대한 U937 단핵구의 부착력을 증진시켰다. 따라서, 본 연구의 결과는 SPA가 HaCaT 피부상피세포의 염증반응을 촉진시킨다는 사실을 보여주었으며, 황색포도상구균에 의한 피부염증질환에 있어서 중요한 병원성인자로서의 역할을 수행한다는 사실을 시사해준다.

혈관내피세포에서 우방자(牛蒡子) 에탄올 추출물의 항염증 효과 (Anti-Inflammatory Effect of Ethanol Extract from the Seeds of Arctium Lappa L. in Vascular Endothelial Cells)

  • Objectives: The seeds from Arctium lappa have been considered for its various pharmacological properties, which include anti-carcinogenic, anti-inflammatory, anti-diabetic, and anti-viral activities. Methods: In the present study, we investigated the anti-inflammatory effect of the ethanol extract from the seeds of Arctium lappa L (EAL) on cytokine-induced vascular inflammation in human umbilical vein endothelial cells (HUVEC). Results: Pretreatment with EAL significantly decreased tumor necrosis factor alpha ($TNF-{\alpha}$)-induced cell adhesion molecules expression such as intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and endothelial-selectin (E-selectin) in a dose-dependent manner. Cell adhesion assay showed that pretreatment with EAL suppressed HUVEC-monocyte adhesion by $TNF-{\alpha}$ over $1{\mu}g/ml$ concentration. We investigated the involvement of nuclear transcription factor kappa-B ($NF-{\kappa}B$) in $TNF-{\alpha}$-induced vascular inflammation. $NF-{\kappa}B$ p65 nuclear expression was induced by $TNF-{\alpha}$, however, pretreatment with EAL was attenuated that nuclear translocation. In cytoplasm, EAL was also attenuated $TNF-{\alpha}$-induced decrease of inhibitor of ${\kappa}B-{\alpha}$ ($I{\kappa}B-{\alpha}$) expression. Moreover, EAL significantly decreased $TNF-{\alpha}$-induced production of intracellular reactive oxygen species (ROS). Conclusions: Taken together, our findings suggest that seeds of Arctium lappa L could be a therapeutic herb for prevention of cardiovascular diseases throughout the inhibition of vascular endothelial inflammation.

TNF-α로 유도된 혈관내피세포의 혈관염증에 미치는 오적산(五積散)의 억제 효과 (Inhibitory Effects of Ojeoksan on TNF-α-induced Vascular Inflammation in Human Umbilical Vein Endothelial Cells)

  • Objectives : Ojeoksan, originally recorded in an ancient Korean medicinal book named "Donguibogam" and has been used for the treatment of circulation disorder of blood which was called blood accumulation (血積) in Korean medicine. Therefore, this study was carried out to investigate the beneficial effect of OJS on vascular inflammation in HUVECs. Methods : We evaluated the effect of OJS on the expression of cell adhesion molecules and protective role in HUVEC stimulated by TNF-${\alpha}$ by using Western blot. Results : Pretreatment with OJS decreased the adhesion of HL-60 cells to TNF-${\alpha}$-induced HUVEC. OJS suppressed TNF-${\alpha}$-induced expression level of cell adhesion molecules such as intracellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1(VCAM-1), and endothelial cell selectin (E-selectin). Moreover, OJS significantly decreased TNF-${\alpha}$-induced production of intracellular reactive oxygen species (ROS); and inhibited the phosphorylation of $I{\kappa}B-{\alpha}$ in the cytoplasm compared to the experimental group. Pretreatment with OJS inhibited the trans-location of NF-${\kappa}B$ p65 to the nucleus. OJS also inhibited phosphorylation of MAPKs compared to the experimental group. OJS significantly increased the protein expression of Nrf2 and HO-1. Conclusions : Ojeoksan has a protective effect on vascular inflammation, and might be a potential therapeutic agent for early atherosclerosis.

Antioxidant and Anti-inflammatory Activities of Ethanol Extract from Leaves of Cirsium japonicum

  • Antioxidant and anti-rheumatoid activities of Cirsium japonicum leaf extract (CJLE) were investigated in this study. CJLE had similar DPPH radical scavenging activity and reducing power to ascorbic acid and several flavonoids. Rheumatoid arthritis (RA) is a chronic inflammatory tissue-destructive disease, partly related with functions of hyaluronidases (HAases) and collgenases. CJLE ($1,000\;{\mu}g/mL$) had approximately 60.7 and 31.9% inhibition of HAase and collagenase activity, respectively. Also, CJLE inhibited lipopolysaccharide (LPS)-induced nitrite production in a dose-dependent manner, and CJLE ($1,000\;{\mu}g/mL$) suppressed approximately 70% of LPS-induced nitrite production effectively in RAW 264.7 macrophage cells. CJLE had inhibitory effects on the adherence of monocytic THP-1 to human umbilical vein endothelial cell (HUVEC) monolayers to the basal level. Inhibitory effect of CJLE on the adhesion was caused by suppression of tumor necrosis factor-a-upregulated expression of vascular cellular adhesion molecule-1 (VCAM-1) and E-selectin. We expect that CJLE may alleviate the inflammatory process in rheumatoid synovium, and these findings will raise the possibility of the usage of C. japonicum as a traditional pharmaceutical of anti-rheumatoid arthritis.

Anti-inflammatory functions of purpurogallin in LPS-activated human endothelial cells

  • Enzymatic oxidation of commercially available pyrogallol was efficiently transformed to an oxidative product, purpurogallin. Purpurogallin plays an important role in inhibiting glutathione S-transferase, xanthine oxidase, catechol O-methyltransferase activities and is effective in the cell protection of several cell types. However, the anti-inflammatory functions of purpurogallin are not well studied. Here, we determined the effects of purpurogallin on lipopolysaccharide (LPS)-mediated proinflammatory responses. The results showed that purpurogallin inhibited LPS-mediated barrier hyper-permeability, monocyte adhesion and migration and such inhibitory effects were significantly correlated with the inhibitory functions of purpurogallin on LPS-mediated cell adhesion molecules (vascular cell adhesion molecules, intracellular cell adhesion molecule, E-selectin). Furthermore, LPS-mediated nuclear factor-${\kappa}B$ (NF-${\kappa}B$) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) releases from HUVECs were inhibited by purpurogallin. Given these results, purpurogallin showed its anti-inflammatory activities and could be a candidate as a therapeutic agent for various systemic inflammatory diseases.

Anti-cell Adhesion Effect of PLM-f74 with U937 Cell from Hallophilic Enterobacteria and Identification of Strain

  • Fermented materials with enterobacteria isolated from fusiform fish, have strong anti-angiogenesis effect and anti-cell adhesion effect. PLM-f74 got from 74th fraction of size exclusion chromatography from fermented material, showed strong anti-cell adhesion effect between HUVECs and U937 monocytic cell. Adhesion of U937 cell to HUVEC stimulated with IL-1b was clearly inhibited by PLM-f74 in a dose-dependent manner by 12.1, 21.2, 50.9, and 78.2%, when U937 cells treated with each of the PLM-f74 and stimulated with PMA (100 mg/L) was added onto untreated and unstimulated HUVECs, adhesion was observed by 15.8, 31.9, 70.8, and 102%, when both cell types were pretreated with PLM-f74, the adhesion was prominently decreased by 83.7, 99.2, 110, and 120.8%, with 0.74, 3.7, 7.4, and 18.5ug/mL of PLM-f74, respectively. PLM-f74, also, reduced IL-1-stimulated HUVEC expression of adhesion molecules, VCAM-1, ICAM-1, and E-selectin dose-dependently by ELISA method.

The protective effect of Prunella vulgaris ethanol extract against vascular inflammation in TNF-α-stimulated human aortic smooth muscle cells

  • Atherosclerosis, which manifests as acute coronary syndrome, stroke, and peripheral arterial diseases, is a chronic inflammatory disease of the arterial wall. Prunella vulgaris, a perennial herb with a worldwide distribution, has been used as a traditional medicine in inflammatory disease. Here, we investigated the effects of P. vulgaris ethanol extract on TNF-${\alpha}$-induced inflammatory responses in human aortic smooth muscle cells (HASMCs). We found that P. vulgaris ethanol extract inhibited adhesion of monocyte/macrophage-like THP-1 cells to activated HASMCs. It also decreased expression of intercellular adhesion molecule-1, vascular cell adhesion molecule-1, E-selectin and ROS, No production in TNF-${\alpha}$-induced HASMCs and reduced NF-${\kappa}B$ activation. Furthermore, P. vulgaris extract suppressed TNF-${\alpha}$-induced phosphorylation of p38 mitogen-activated protein kinase (MAPK) and extracellular signal-regulated kinase (ERK). These results demonstrate that P. vulgaris possesses anti-inflammatory properties and can regulate TNF-${\alpha}$-induced expression of adhesion molecules by inhibiting the p38 MAPK/ERK signaling pathway.

Anti-platelet Activity of Tissue-cultured Mountain Ginseng Adventitious Roots in Human Whole Blood

  • Present study investigated the effects of the 70% ethanol extracts of tissue-cultured mountain ginseng (TCMG), Korean red ginseng (KRG), and Panax ginseng (PG) on agonist-induced platelet aggregation and activation in human whole blood. The $IC_{50}$ values for TCMG, KRG, and PG were 1.159, 3.695, and 4.978mg/mL for collagen-induced aggregation, 0.820, 2.030, and 4.743mg/mL for arachidonic acid-induced aggregation, and 1.070, 2.617, and 2.954 mg/mL for ADP-induced aggregation, respectively. Also, this study assessed the effects of the most active extract, TCMG, on markers of platelet activation by determining receptor expression on platelet membranes in healthy subjects, including expression of GPIIb/IIIa-like (PAC-1) and P-selectin (CD62), by flow cytometry. A significant decrease in PAC-l expression (p=0.018) was observed in the presence of TCMG. These results show that TCMG has potent anti-platelet activity.

Clustering for Home Healthcare Service Satisfaction using Parameter Selection

  • Recently, the importance of big data continues to be emphasized, and it is applied in various fields based on data mining techniques, which has a great influence on the health care industry. There are many healthcare industries, but only home health care is considered here. However, applying this to real problems does not always give perfect results, which is a problem. Therefore, data mining techniques are used to solve these problems, and the algorithms that affect performance are evaluated. This paper focuses on the effects of healthcare services on patient satisfaction and satisfaction. In order to use the CVParameterSelectin algorithm and the SMOreg algorithm of the classify method of data mining, it was evaluated based on the experiment and the verification of the results. In this paper, we analyzed the services of home health care institutions and the patient satisfaction analysis based on the name, address, service provided by the institution, mood of the patients, etc. In particular, we evaluated the results based on the results of cross validation using these two algorithms. However, the existence of variables that affect the outcome does not give a perfect result. We used the cluster analysis method of weka system to conduct the research of this paper.
