A comparative analysis of characteristics and antioxidant capacity of Korean mulberries for efficient seedling cultivation

  • Mulberry exhibits unique characteristics and functionalities across various components, including the roots, branches, leaves, and fruits. However, despite numerous studies on mulberry, research on this plant at the seedling stage is insufficient. Therefore, this study aimed to compare the suitability for seedling cultivation and antioxidant effects of four Korean mulberry cultivars, namely, Daesim, Suhong, Simgang, and Cheongsu. In terms of seed weight, germination rate, and growth rate, Daesim was the most suitable cultivar for seedling production. Polyphenol and flavonoid content analysis showed that all cultivars, except for Cheongsu, showed the highest phenolic content at the 2-week seedling stage. Similarly, antioxidant assays using 2,2'-azino-di-3-ethylbenzthiazoline sulfonic acid (ABTS) and 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radicals revealed that the antioxidant effect of all cultivars, except for Cheongsu, increased with cultivation at 2- 4- and 6-week. However, mulberry seedlings had a slower reaction rate against DPPH radical removal than mulberry leaves. In addition, ABTS radical scavenging activity showed a different correlation with polyphenol content. This phenomenon may be due to the different polyphenol compositions between mulberry leaves and seedlings. The results of this study suggest that mulberry seedlings exhibit different bioactivities from mulberry leaves, and component analysis is required in further research.

Characteristics of Seedling Establishment and Yield of Platycodon grandiflorus by Ridge Width and Mulching Materials (이랑폭과 피복재에 따른 도라지의 입모율 및 수량특성)

  • This study was carried out to investigate the seedling establishment and yield of direct-sown Platycodon grandiflorus seeds cultured by three ridge width, 0.4, 0.8, and 1.2 m and four mulching materials, rice straw, rice bran, black plastic film, and no mulching in upland. Seedling establishment rate (70%) was the highest in rice straw mulched plot of 500 seeds/$m^2$ sowed and root yield after one year was also the highest in rice straw mulched treatment and followed by rice bran, no mulching, and black plastic film treatment. In this result, ridge width 120cm and rice straw mulching combined treatment was best for getting the highest seedling establishment ratio and seedling numbers per area. However, ridge width and mulching materials should be considered soil moisture content and weed population for saving labor cost.

Influence of Thickness of Styrofoam-Panel on Tobacco Seedling Growth in Newly Developed Floating System (새로 개발된 부상형 육묘에 있어서 스티로포움 판의 두께가 연초 묘 생육에 미치는 영향)

  • The inluence of styrofoam-panel thickness of floating frame on the seedling growth of flue-cured tobacco(Nicotiana tabacum L.) in the newly developed direct-seed float system was investigated . Floating frame used instead of styrofoam-tray consisted of styrofoam-panel (106×108cm) for floating, eight holes(1.Sx2S.0 cm) to uniform the water, and nutrition supply far seedling and water and nutrition absorption cloth placed on the styrofoam-panel. Each floating-frame may lay eight plastic-trays on it. Eight grades of styrofoam-panel thickness from 26 mm to 46mm and two kinds of media were used in the greenhouse system. Dry cells were found from the styrofoam-panel thickness of 43mm in carbonized chaff, compost and original soil rate of L3-3(v/v) media, and of 37mm in peat and perlite rate of 8-2(v/v) media. The thinner styrofoam-panel produced more tender and succulent seedling with the more trunk and the less root weight. When considering the appearance of dry cell, seedling growth, producing healthy tobacco seedlings, and utilization of styrofoam-panel for two kinds of medias the ideal styrofoam-panel thickness were suggested to be around 34mm for flue-cured tobacco reeling production in the newly developed direct-seeding float system. Priming Effect of Rice Seeds on Seedling Establishment under Adverse Soil Conditions

  • An experiment was carried out to find out the priming effects of rice seeds, Oryza sativa L. (cv. Ilpumbyeo) on. the seedling establishment and early emergence under excess soil moisture conditions. Seeds were primed by soaking in -0.6 MPa polyethylene glycol (PEG) solution at $25^{\circ}C$ for 4 days. The primed seeds were sown in soils with various soil moistures (60, 80, 100, 120, and 140% field capacity) at 17 and $25^{\circ}C$, respectively. Germination and emergence rates, plumule height, and radicle length of primed seeds were higher than those of untreated seeds at any soil moisture and temperature examined. The time from planting to 50% germination ($T_{50}$) of primed seeds was less than that of untreated seeds by 0.9~3.7 days. Germination rate, emergence rate, plumule height, and radicle length were highest at the soil moisture of 80% field capacity among the soil moistures. Priming effects of rice seeds on germination and emergence rates were more prominent under the unfavorable soil moistures (60, 100, 120, and 140% field capacity) than those under the optimum soil moisture condition (80% field capacity). However, priming effects on seedling growth were greater at near optimum soil moisture compared with too lower or higher soil moistures. Therefore, these findings suggest that priming of rice seeds may be a useful way for better seedling establishment under the adverse soil conditions.

Comparison of Physiological Characteristics, Stomata and DNA Content between Seedling and 5-year-old Somatic Plant (Somatic Embryo Derived-plant) in Liriodendron tulipifera (백합나무 5년생 실생묘 및 체세포묘 (체세포배 유래 식물체) 간의 생리적 요인, 기공 및 DNA 함량 비교)

  • Field performance of somatic plants (somatic embryo derived-plants) of yellow-poplar (Liriodendron tulipifera) produced from somatic embryogenesis was compared with that of seedlings at age 5. In comparison of photosynthetic rate (seedling, $10.67{\mu}mol$ $CO_2m^{-2}s^{-1}$; somatic plant, $9.04{\mu}mol$ $CO_2m^{-2}s^{-1}$), stomatal conductance rate (seedling, 0.2 $H_2Om^{-2}s^{-1}$; somatic plant, 0.166 $H_2Om^{-2}s^{-1}$) and respiration rate (seedling, 1.71 mmol $H_2Om^{-2}s^{-1}$; somatic plant, 1.513 mmol $H_2Om^{-2}s^{-1}$), no significant differences were found between plants. The seedlings were a little higher in comparison of stomatal density (seedling, $23.33/mm^2$; somatic plant, $22.43/mm^2$), length (seedling, $25.83{\mu}m$; somatic plant, $23.46{\mu}m$) and width (seedling, $15.87{\mu}m$; somatic plant, $15.3{\mu}m$). In comparison of DNA content of the leaves using flow cytometry, no differences in ploidy level were found between the seedlings and somatic plants.

Effect of Planting Season and Vegetation Competition on Growth Performances of Containerized Seedlings of Pinus densiflora (식재시기 및 조림지 하층식생 조건이 소나무 용기묘의 생장에 미치는 영향)

  • The objective of this study was to find optimal planting season of the effect of vegetation competition around planting tree. This study evaluates the possibility of extending the planting period of containerized seedling of Pinus densiflora (1-0 year). We planted seedling at three different seasons (spring, summer, fall) at 18 plots in 3 locations. Planting sites were composed of vigorous vegetation (Site A) and less vigorous vegetation (Site B). For over 3 years, the study investigated survival rate, growth of root collar diameter and height, and biomass of containerized seedling of P. densiflora. In all sites, containerized seedling of P. densiflora showed high survival rate in summer planting. Height and root collar diameter of containerized seedling of P. densiflora were the highest in summer planting, but in the initial period after planting growth was not good. After time passed, growth rate was increased. Site B showed better growth than Site A. Biomass was the highest in summer planting and also the highest in Site B. These results suggest that planting period can be extended by using containerized seedling and vegetation control in the initial is very important for survival and growth of containerized seedling.

Effects of $GA_3$ on Seed Germination and Seedling Survival Rate of Acanthopanax senticosus Maxim. (가시오갈피의 $GA_3$처리에 따른 종자발아와 유묘생존)

  • This experiment was carried out to investigate the effects of $GA_{3}$ and cold stratification as presown treatments on seed germination, seedling emergence and final survival rate of Acanthopanax senticosus Maxim. Seeds collected 145 days after-ripening period followed by 10 days of cold stratification was effective in promoting germination. Dehisced seeds treated with 500 ppm of $GA_{3}$ for 3 days was also effective in promoting germination. However, seedling emergence rate remained low in both treatment. Seedling emergence rate was higher for seeds germinated in Heungnong-Bio and Klasman-Bio than in vermiculite, perlite, vermiculite-perlite mixture, or sand. After 40 days of cold stratification, seedling emergence was significantly higher in the 500 ppm $GA_{3}$ treatment than nontreatment for both dehisced and non-dehisced seeds. However, for dehisced seeds, $GA_{3}$ treatment before sowing resulted in decreased final seedling survival rate.

Improvement of Seedling Stand and Lodging Prevention in Direct Seeded Rice (벼 직파재배(直播栽培) 입묘율향상(立苗率向上)과 도복경감(倒伏輕減))

  • The results of recent researches for improvement of seedling stand in direct seeded rice on the dry paddy in Korea were summarized as the following ; a variety to be cultivated should be chosen the characteristics of high percentage germination under low temperature, shorter period of shoot emergence, and better growth of the mesocotyl and shoots. Meanwhile, there was 40% increase in seedling stand at the treatment of removal of the seed awn under using the drill seeder. After seeding the rice seed covered with soil of 3cm depth was better seedling emergence and also there was the hightest seedling emergence at the 70% of moisture content of the soil. In addition, the application of the Release containing GA 10% enabled to increase the seedling stand and furthermore it was effective under deep seeding depth. The optimum seeding date should be seeded around May 10 when mean air temperature is above 12-13$^{\circ}C$ so that may establish more less 70% in seedling stand. Based on an appropriate seedling stand of 150/$m^2$, the optimum seeding rate was 5kg/10a. It was the best in seeding method using drill seeder and the most desirable recommended seeding method was the drill seeder in terms of seedling stand. In order to improve seedling stand water management was more effective in canal irrigation and in drainage at 6hr after irrigation following by the seeding process. On the other hand, for the increase of seedling stand under flooded condition a variety might have characters being better germination at low concentration of dissolved oxygen and vertically deeper growing of the crown root. Also, seedling stand was able to increase with the seed coating of $CaO_2$in the flooded soil. It was possible to be seeded on the early part of May being mean air temperature of avove 10$^{\circ}C$ and the optimum seeding rate was 5kg/10a. For an effective water management water would be flooded up to 3cm depth for 2-3 weeks after seeding. The rice plant grown under the direct seeded cultivation might be not so much strong in lodging resistance compared to that grown under the transplanting and moreover direct seeded rice cultivation under flooded condition would be more weak growth of the rice plant than that on dry paddy. Meanwhile, the lodging would be affected by the seeding rate, the soil depth after seeding. and seeding method even in the same variety. In particular, roots in the lodging pattern of direct seeded rice cultivation under flooded condition were largely distributed on the soil surface so that resulted easily in the lodging. In general, the lodging resistance would be greater as seeding rate and amount of N fertilizer application are lower and soil depth after seeding is higher. Among the introduction of different seeding method the high ridged drill seeding method on dry paddy soil resulted in the lowest in the lodging index and also it was lower in the drill seeding method than in the scattering seeding method under flooded condition. In case of more than 150 seedlings per $m^2$ there was a severe lodging due to high lodging index at the 3rd and 4th internodes. The effective lodging prevention was able to at the treatment of the Inabenfide at 45 days before heading and the Uniconazol at 15 days before heading which caused the shortage by 10-15cm in culm length. Also, fertilizer management using split application of nitrogen would be contributed the reduction of lodging at the rate of 20-30-20-20-10%(basal-5th leaf stage-7th leaf stage-panicle initiation stage-heading stage) on the dry paddy soil.

Growth Characteristics of Veronica rotunda var. subintegra (Nakai) T.Yamaz. according to Several Cultivation Conditions (몇 가지 재배조건에 따른 산꼬리풀의 생육특성)

  • This study was aimed to establish the most effective approach for the cultivation of Veronica rotunda var. subintegra (Nakai) T.Yamaz. plants, which was expected as new ornamental plants. We conducted an experiment using plug seedlings, varied the seeding container type and seeding rate. We also varied seedling quality, planting container, soil type, and shading ratio. Seedling quality was used seedlings produced from different seeding containers and seeding rates. The seedling quality were seeding growth using 162, 200, and 288 trays, and seedings rate was used seedlings produced by sowing 1, 2, 4 and 6 seeds. As a result, 162 trays of seedlings were suitable for use in this study. Plants grown with one seed per cell in individual cells exhibited increased individual growth, but those grown with four seeds per cell exhibited better growth for the whole plant. According to seedling quality, seedlings produced in the 162 trays or with four seeds per cell showed higher growth than other seedlings. In the cultivation of V. rotunda var. subintegra (Nakai) T.Yamaz., seedling growth increased depending on container capacity for both shoot and root parts. Container material had no significant impact on seedling growth. Seedlings grew the best on horticultural substrate, and showed better growth on mixed soils with high decomposed granite content than on peatmoss.

Germination, Seedling Emergence, and Growth of Burcucumber Affected by Maturity and Size (안동대목 종자의 크기와 등숙 정도에 따른 발아.유묘 출현 및 생장)

  • Burcucumber (Sicyos angulatus L.), used as a medicinal or rootstock for cucurbitaceous crops have shown high variation in seed maturity and size. This study was carried out to determine the effect of seed maturity and size on seed germination, seedling emergence and growth of burcucumber, Seeds collected from a native site were divided into small, medium and large ones after sorted to mature and immature ones. Their germination, seedling emergence and growth were done in indoor and a green house. Mature seeds showed higher germination and seedling emergence rate than immature ones. Regardless of their maturity, medium. and small seeds showed higher germination rate than large ones. Medium and large seeds, however, had the greatest and the least seedling emergence, respectively. Seedling height, number of true leaves areas of cotyledons and true leaves except hypocotyl length were increased with increased seed size although were not affected by the maturity. Cotyledon, leaf, hypocotyl, root and their total dry weights were greater in large mature seeds than large immature ones while increased with increased seed size in both maturities.
