Six commercial rootstock varieties were tested for resistance to bacterial wilt (Ralstonia solanacearum) by dipping the roots of seedlings in the bacterial suspension at seedling stage. 'Shincheonggang' and 'B-Blocking' ranked top and second in resistance to bacterial wilt, respectively. For disease index of the grafted plants, 'Dotaerang' grafted onto 'Shincheonggang' was the least, followed by 'Ultra', 'Fighting', 'B-Blocking', 'Special', and 'Connection' grafts. Thus, disease development on the grafted plants was correlated with resistance of rootstocks. The effect of rootstocks on horticultural characteristics including days to flower of scion was minimum or negligible. In correlation between horticultural characteristics of rootstocks and scions grafted thereon, statistically significant negative correlation was observed in days to flower and fruit height but no significant correlation was observed in yield per plant, fruit diameter and soluble solid content in the fruits. Thus, selection of resistant rootstock varieties that support the growth and yield of scion well is necessary for effective control of the disease and yield increase.
Rearing condition and species of microalgae on growth and survival of the sunray surf clam, Mactra chinensis larvae were investigated for artificial seedling production. The larvae of M. chinensis on higher temperature showed high growth and low survival and was grown over $230{\mu}m$ in shell length 14 days after hatching, but low growth as $151.1{\mu}m$ at $18^{\circ}C$. The larvae of M. chinensis on salinity showed highest daily growth and survival as $11.3{\mu}m$ and 65.8% at 30 psu, respectively. The optimum of water temperature and salinity for the larval rearing were $23^{\circ}C$ and 30-35 psu over 25 psu at least. The density of larval rearing was below 10 per 1 ml in rearing seawater for elevating the development rate from D-shaped to settled (metamorphosing) stage. The larvae fed the mixed diet of Isochrysis galbana, Pavlova lutheri, Isochrysis sp. (green), Chlorella ellipsoidea showed highest growth and survival. The larvae fed the single diet of I. galbana, Isochrysis sp. (green) showed high growth and survival. But the larvae fed the single diet of P. lutheri and C. ellipsoidea showed low growth and survival. The optimum diet of larvae of M. chinensis was over two species of microalgae included I. galbana, Isochrysis sp. (green) for elevating the high growth and survival.

This study was conducted to clarify the effects of the different mixing ratios of substrate mixtures based on peat moss and the concentration of nutrient solution on the growth of tomato (Lycopersicon esculentum Mill.) seedlings. Substrates such as peat moss, rice hull, carbonized rice hull, decomposed sawdust, perlite and granular rock wool were mixed and used. The concentration of nutrient solution were adjusted to EC $0.5{\sim}1.5mS/cm$. The volumetric moisture contents became higher as peat moss mixed were much more. Total porosities in all substrate mixtures were over 80%, and pH in substrate mixtures became lower as the volume of peat moss mixed higher. Mixing ratios of substrates suitable for the production of tomato seedlings with the higher quality were peat moss:rice hull:carbonized rice hull:decomposed sawdust:perlite=25:10:25:20:20(v/v). The plant growth was not significant among the different substrate mixtures. However, plant growth such as plant height, leaf area, and total dry weight became significantly increased as EC increasing.

Virus-free stocks was produced by the combination of the heat treatment of virus infected plant and shoot-tip grafting (STS). To produce virus-free stocks, the plants infected with citrus viruses were used for virus-free stock production using the modified method of STG in thermotherapy at $40^{\circ}C$ for 16 hours in the light, and at $30^{\circ}C$ for 8 hours of darkness for 4 weeks. Trifoliate orange (P. trifoliata) were used as rootstock seedling for STG. Percentages of virus-free stocks against citrus tristeza virus (CTV), satsuma dwarf virus (SDV) and citrus tatter leaf virus (CTLV) were $75.7\%,\;100.0,\%\;82.6\%$ respectively. Shoot tip size for successful STG were as small as possible. Less than $0.3\;\cal{mm}$ of shoot tips gave the hight efficiency of virus free plants but survival rates were low. And, survival rate after shoot-tip culture was analyzed and the rates were dependant on the cultivars; Yuzu cultivar showed the hight survival rate ($74.6\%$) and early satsuma mandarin (Iwasagi) was $13.3\%$ as the lowest cultivar. But citrus trees were not succeed to grown, turned brown, and died.
The possible role of nitric oxide (NO)-induced cell division was investigated to explain the physiologycal effects of a NO donor, sodium nitroprusside (SNP) on the growth of primary leaves in chinese cabbage seedling plants. Exogenous treatment of SNP to chinese cabbage plants for 8 days at different concentrations (0, 200, 500 and 1000 ${\mu}M$) affected the leaf growth in a concentration-dependent manner, showing a maximum growth at $200\;{\mu}M$. In accordance with leaf growth responses, the chlorophyll and soluble protein contents increased strongly to 142% and 134% of control at $200\;{\mu}M$ SNP, respectively. However, a very little decrease in chlorophyll and a 13%> decrease in protein were observed at $1000\;{\mu}M$ SNP. In addition, the content of DNA and RNA also increased maximumly to 142% and 139% of the control at $200\;{\mu}M$ SNP, respectively, whereas they decreased to 80% and 84% of the control at $1000\;{\mu}M$ SNP. With respect to the development of enzymes related to cell wall synthesis, $200\;{\mu}M$ SNP led to the maximum activities in both phenylalanine ammonia-lyase (212% of the control) and guaiacol peroxidase (134% of the control). However, the activities of both enzymes were not modified significantly at $1000\;{\mu}M$ SNP. In conclusion, these results suggest that the enhancement of leaf growth in chinese cabbage plants by SNP at the effective concentration was probably due to the NO ability in the induction of cell division.
M1 plants which were produced from seed soaking in chemical mutagen, EMS or NaN$_3$, appeared wide morphorogical variations such as dwarf, albino, twisted leaf, white streaked leaf, and purpled stem. In mutants of reproductive organs, there were monoecious plants such as female-flower plant and male-flower plant, multiple spikes, and steriled plants among M1 plants. Also, barren stalk was increased significantly in M1 plants. Ear bagging at ear initiation stage prevented seed set on cob in normal plants. In spite of ear bagging, M1 plants which had cobs with seed set was 3.9-11.2% of stalks developed from seeds soaking with mutagens, but only three or four kernels could be matured on a cob. Ear bagging after mutagen injection into initiating ear produced 5.1-10% in cobs with seed set, but only 1.7-6.3 kernels could be matured. Cobs removed silk at four hours after artificial pollination increased the rate of cobs with seed set to 27%. Microscopic observation confirmed that ontogeny of kernels matured from ear bagging and mutagen treatment would be both adventitious and diplosporous apomictic reproduction. Chromosome set of M2 seedling was found to be diploid type in chromosomal counting of root tip. As M$_2$ plants showed an uniform appearence within each lines and their CV of plant height were ranged 4-6% in each lines, we concluded that they were apomictic progeny. But we could not find any marker traits combined with apomixis.
The variety Tong-il from the IR667 lines have been reported as a variety resistant to the smaller brown planthopper, Laodelphax striatellus Fallen, in Korea. The resistance of Tong-il to the insect, however, have been mainly evaluated from a viewpoint of plant reaction of rice at the seedling stage. In this study, the nature of resistance of Tong-il to the insect was furthermore evaluated from the standpoints of feeding preference, antibiosis and plant tolerance in comparison with the resistant varieties Karsamba Red ASD-7 and Vellailangalayan. and the susceptible variety Jinheung which have been reported to the insect. The smaller brown planthopper much more prefered Tong-il for feeding than the resistant varieties ASD-7 and Vellailangalayan. The degree of antibiosis of Tong-il to the smaller brown planthopper was significantly lower than ASD-7 and Vellailangalayan. The results showed significantly lower nymphal mortality and higher adult emergence in Tong-il and Jinheung than other two resistant varieties. The degree o( tolerance of Tong-il to the insect was also lower than ASD-T and Vellailangalayan. Only. Tong-il seemed to be relatively more tolerable than Jinheung. The reason of showing resistant reaction of Tong-il in the mass screening test might be related with relatively lower feeding preference and more tolerance than Jinheung and other susceptible varieties.
In this study, the reattachment processes of small and medium size of spot of Mytilus coruscus and M. edulis were observed. The small spats (Shell length, SH, $3{\sim}7mm$) of hard shelled mussel, M. coruscus showed 100% reattachment rate within 7 minutes after the detachment of byssus thread and 100% survival rate. While large spats (SH 25 mm) showed 85% reattachment rate within 5 hours and 100% survival rate for 24 hours. The reattachment rate of M. edulis (SH 30 mm) was higher than that of M. coruscus (SH, 28 mm). The thread consisted of three identifiable structures; adhesive disc, adhesive thread and adhesive root. The adhesive disk and adhesive thread of M. coruscus were larger and thicker than those of M. edulis, whereas the thickness of adhesive root was the other way round. Further studies are required to identify the relationship between the structure of byssus thread and attachment ability. This study suggests that the sizes of spats could be an important factor in determining the timing of removing and reattaching mussel seedling for aquaculture or releasing to the sea.
To substitute fromzed and freeze-dried food for live food in the development of the artificial seedling production of bivalve larvae, the dietary value of live T. suecica was compared with those of freeze-dried T. suecica, frozed T. suecica, live T. suecica (50%)+frozen T. suecica (50%), and live T. suecica (50%)+freeze-dried T. suecica (50%) in the rearing of oyster (Crassotrea gigas) and hen clam (Mactra chinensis) larvae, and manila clam (Tapes philipninarum) spats. Oyster larvae fed live T. suecica showed the highest growth (shell hight $231.9^{\mu}$m) and survival rate (72.6%) and those fed freeze-dried T. suecica showed the lowest growth (shell height $168.9^{\mu}$m) and survival rate (35.3%). However, in the hen clam larvae, there were not significantly different among diet group in growth and survival rate. The small spats of manila clam fed live t. suecica or live T. suecica (50%)+freeze-dried T. suecica (50%) showed higher growth and survival rate than those fed other diet group. In the case of large spats of manila clam, live T. suecica and live T. suecica (50%)+frozen T. suecica (50%) showed better growht. But, survival rates were not different among diet groups. Dietary valuse of frozen and dried T. suecica were different on species and growth stage, and frozen and freeze-dried T. suecica can be partially used as substitute food for T. suecica live T. succica in shellfish hatchery.
This study was carried out to obtain the fundamental data for the mass seedling production of grunt, Hapalogenys nitens in terms of the natural spawning and some characteristics of the eggs spawned. The wild grunt were reared at indoor tanks for three years. The adults spawners were 34.0∼44.0 cm (38.6$\pm$4.0 cm, n=7) in total length, 1.00∼2.23 kg (1.62$\pm$0.50 kg, n=7) in body weight. Spawning were observed 9 times from September 22 to October 1, 2000 and 37 times from August 22 to October 3, 2001, with a water temperature range of 19.8$\pm$28.5$^{\circ}C$. The total number of eggs collected was 2.29${\times}$10$^{7}$ (1.7${\times}$10$^{3}$/ml). The relative proportion of floating eggs to total eggs was 41.7%. The fertilization rate of floating eggs was ranged between 85.0 and 99.9% and the hatching rate was ranged between 2.9 and 93.0%. Fertilized eggs were buoyant and spherical in shape, and were 0.85∼0.98 mm in diameter. Each egg contained 1-5 oil globules which were, 0.18∼0.25 mm in diameter. The incubation time from fertilization to blastodisc formation was 10 minutes, to blastula was 3 hours, and to the hatched larvae at 26$^{\circ}C$ was 20 hours 30 minutes. The newly hatched larvae attained total length of 1.81$\pm$0.18 mm. The time required from fertilization to hatching was 31∼34 hours at 23$^{\circ}C$ and 17∼20 hours at 29$^{\circ}C$.
