Rice blast caused by the filamentous fungus Magnaporthe oryzae, is arguably the most devastating rice disease worldwide. Development of a high-throughput and reliable field blast resistance evaluation system is essential for resistant germplasm screening, resistance genes identification and resistant varieties breeding. However, the occurrence of rice blast in paddy field is easily affected by various factors, particularly lack of sufficient inoculum, which always leads to the non-uniform occurrence and reduced disease severity. Here, we described a procedure for adequately inducing the occurrence of rice seedling blast in paddy field, which involves pretreatment of diseased straw, initiation of seedling blast for the first batch of spreader population, inducing the occurrence of the second batch of spreader population and test materials. This procedure enables uniform and consistent infection, which facilitates efficient and accurate assessment of seedling blast resistance for diverse rice materials.
The control activity of isoprothiolane and tricyclazole mixed with carbosulfan, and probenazole by the nursery treatment was performed against rice leaf and neck blast caused by Magnaporthe grisea. In the paddy field, three fungicides showed good activities against leaf blast 3 months after nursery treatment. Especially the activity of tricyclazole against leaf blast gradually increased by the laps of time to 85.5%, which was assessed at 6 September,2003. Although the control value of isoprotholane and tricyclazole mixed with carbosulfan against neck blast was 47.5% and 61.1%, respectively, probenazole showed a very high activity against not only leaf blast but also neck blast, of which that was 91.2%. No phytotoxicity was observed in all the treatments after transplanting rice seedling in the paddy field. Based on these results, three systemic fungicides tested in this study showed such a good potential that they might be used to formulate the nursery treating granule.
To investigate the mutagenecity of sodium azide (SA) and to select mutants having various agronomic characteristics in rice (Oryza sativa L. ssp. japonica), dry seeds of rice variety 'Suweon 472' were treated with 0, 0.5, 1.0, 2.0, and 4.0 mM SA solutions prepared in 0.1 M phosphate buffer (pH 3.0). Germination rate, seedling height and sterility were investigated in $M_1$ generation and chlorophyll mutations were observed in $M_3$ generation. Germination rate and seedling height decreased as the increase of SA concentration in $M_1$ generation, the maximum seed sterility (40.8%) was found at 4.0 mM SA concentration. Chlorophyll mutants were occurred in $M_3$ generation and the frequency calculated on a line basis was 13.5% at the same treatment. Many kinds of mutations for morphological and agronomic characters were observed and mutations with short culm and glabrous leaf were frequently found in $M_3$ generation. Interestingly, five mutant lines resistant to blast or bacterial blight (BB) were selected and evaluated with several isolates in $M_3$ generation although Suweon 472 has been known to be susceptible to blast and BB. These mutants showed all resistance to seven isolates of blast and a total of 76 lines among 2,567 lines evaluated showed resistant to race K1 of bacterial blight. Two mutant lines (440172 and 41272) showed different reaction to BB isolates from the other resistant mutants. A few kinds of endosperm mutants were also identified and most of them were waxy mutants.

The objective of this research was to determine controlling effects on rice blast (Pyricularia oryzae) in rice plants by using plant extracts from different extraction methods (water, boiling water, fermentation, and ethanol) from 38 agricultural materials of traditional agriculture. Rice blast was completely suppressed by 3% ethanol extracts in Rheum palmatum roots, and suppressed 97% and 77% by 10% ethanol extracts in onion bulb and pine tree leaves, respectively in a laboratory test. However, other agricultural materials showed low effect on suppression of rice blast. Additionally, in a seedling test, rice injury of two cultivars (Ilmibyeo and Hopyoungbyeo) against rice blast was reduced 40-71%, 29-63%, and 23-63% by 5 and 10% ethanol extracts in Rheum palmatum roots, onion bulb, and pine tree leaves, respectively, compared with non-treated controls. Rice injury of two cultivars (Ilmibyeo and Hopyoungbyeo) against rice blast was reduced by 21-55%, 23-46%, and 5-39% in response to Rheum palmatum roots, onion bulb, and pine tree leaf applications at 100, 200 and $400g/m^2$ at 0 day after seeding, respectively, compared with non-treated controls. Rice plants did not show any leaf injuries and growth reduction after treatments of the Rheum palmatum roots, onion bulb, and pine tree leaf extracts or soil application. Thus, the above materials may be used for controlling rice blast in organically produced rice fields.
Seven hundred and forty eight rice cultivars were screened for drought tolerance at seedling stage in a special green house. A number of cultivars such as Wonkiljo, Hanyangdo, YR 52-$_{x}\textrm{BF}_7-67, Satominori, Mamoriaka, Akanomochi 114, Thkebenimochi, IR 937-55-3, IR2735-F_3B-6-2, IR 2871-53-2, $_{x}\textrm{BFI76}^9/Dwan, Khoo lliok Mali-4-2-105, Os6, Palawan, IRAT10, TOS 2583 and H-l05 seemed to be tolerant to drought at seedling stage. There was negative correlation between drought resistance score and growth of plant height and water content of culm and leaf after drought treatment, and highly positive correlation between resistance to blast and resistance to drought at seedling stage. There was no relationship between drought resistance and germination speed, plant height before drought treatment.

Blast resistance of one hundred and thirty-one rice cultivars bred in Korea was tested with thirty Korean isolates and twenty-two Philippines isolates using three screening methods. In the blast nursery conducted in Korea and in the Philippines, average disease index of rice cultivars were 4.6 and 2.2, respectively. Seventy-nine cultivars showed different resistance reaction in Korea and in the Philippines, and 19 cultivars showed the same resistant reaction in two locations. In the seedling test, Korean blast isolates displayed different levels of virulence. 93-093, a Korean isolate, was compatible with 90 cultivars whereas 97-057 showed a compatible reaction with 13 cultivars. Twenty-three cultivars showed high level of resistance against Korean and Philippines isolates but Chucheongbyeo, Heugnambyeo, and Manmibyeo showed susceptible reaction to all blast isolates. Through the sequential planting test in Korea and in the Philippines, Palgongbyeo and Seomjinbyeo displayed durable resistance, and Nagdongbyeo and Gihobyeo showed high level of disease infection over the planting time. These results indicate that blast isolates collected in two countries have different genetic background and number of compatible isolates should be considered in definition the durability of rice cultivar to rice blast.
Using the milled, blast infected leaves (BIL) as an inoculum source on the screening for the resistance to blast of rice plant was a simple and useful technique. The temperature with high (25^\circ C\sim 35^\circ C) and low (15^\circ C\sim 28^\circ C) and the water stressed or not, was conditioned of to the inoculation with the BIL to the test varieties in seedling stage. In low temperature, most of the varieties were more infected with blast, however the Indica-Japonica hybrids were more infected in high temperature conditions. The water stressed was more infected with blast than the not stressed. The interaction of variety with water stress was not so much as that of variety with temperature. Resistant reaction to blast (BIL) was not affected by the temperature and water stress, but the moderately resistant or susceptible one was much affected by them. Inoculum of BIL was virulent to the newly bred Indica-Japonica hybrid cultivars such as Tongil, Nopung, etc, but not virulent to the Japonica cultivars such as Nongbaek, Jinheung, etc. The discrete, mixed or variable lesions were observed mainly in the moderately resistant or susceptible cultivars such as Kanto 51, Yashiromochi, Ishikari-shiroke, etc.
Exposure of seedling of rice (Oriza sativa cv.Dongin) to cold stress ($6^{circ}C$, 7day) induced differential gene expression. Differentially expressed polyadenylated RNA induced by low temperature were isolated and identified from the leaves of rice (Oriza sativa cv.Dongin) seedling by using the technique, differential display of reverse transcription through polymerase chain reaction (DDRT-PCR). Four bands of cDNAs were differentially displayed on the PAGE gel through DDRT-PCR, and among them three bands were those of overexpressed genes while one band was of an underexpressed gene One of the overexpressed cDNA was characterized. The size of the DDRT-PCR product was found to be about 200 bp. The sequence of the cloned DNA was compared with those of GenBank through a BLAST E-Mail server, and it was found to have no homologies in the nucleotide sequence with that of any known DNA: therefore, it was designated as RC101 The expression of the cold-stress induced-gene, RC101, was sustained with Northern Blot analysis by using the cloned DDRT-PCR product as a probe.
The effects of climatic factors on organisms lire variable and complex, and it, however, can be interpreted in terms of those on the distribution and those on the population densities. The distribution of an organism may largely be determined by the temperatures, except some temporal organisms which are depended on the air mass movements. Population density of an organism is determined by various climatic factors, such as previous winter temperature, temperature of growing season and rainfall. The start of growing season of the rice plants has been shifted to earlier since last decade in Korea. This may mean that the overall climatic condition during the growing season might be considerably different from those in past years, and such a difference in climatic conditions might have close relation with the recent status of the diseases and insect pests through direct effects on the physiology and population dynamics of the organisms, as well as through on the biotic associations of the pest organisms. The white back planthopper and brown planthopper have become the key insect pests in Korea in recent years. They are migratory and have high reproductive pontentials and more generations than average residential insects. The synchronization of the migrants and physiological condition of the rice plants seems to be the important factors in relation to the recent outbreaks of these insects; the high reproductive rate can be obtained with the growth stage of rice being 30-50 days after transplanting. The modication of the microclimate associated with high plant density and some other introduced new cultural techniques also have some relation with the outbreak. The key diseases of the rice are the blast disease, sheath blight and the bacterial leaf blight. For the rice blast, the seedling blast and leaf blast during the early growing season and the neck blast, have become more serious, the former may be related to hotbed nursery and the later may be related to the high humidity in early August, and synchronization of the heading time which has been shifted to early part from middle or late part of August. In general, for the rice diseases, the development of the new races have been the most serious which are largely resulted from the introduction of the new varieties, but it also seems to be related with the prolonged periods of the favorable condition associated with the shifted growing seasons. In general, the diseases and insect pest problems have become much more variable and complex, and control measures should be based on the thorough knowledge of the ecology of the pest organisms, that is, effects of various environmental factors on the disease cycle; spore release, spore deposition, infection, colonization and sporulation of the disease organisms, and those on the development, reproductive potentials, dispersal, age specific responses of the insects. The well organized real-time pest management systems, such as alfalfa weevil management system developed at the Purdue University in U.S., is the prime importance for the implementation of the pest management principles.
Due to fast industrialization and reduced agricultural population, there has been increased farm mechanization to reduce the labor requirement. In rice production, mechanical transplanting has been increasingly popular due to the heavy labor requirement in hand transplanting and development of convenient transplanters. For mechanical transplanting, rice seedlings is grown in boxes with limited soils under super dense population, thus short period of exposure to unfavorable temperature and poor water management would cause severe damage to rice seedlings such as seedling damping off and the similar physiological disorders. Several chemicals were evaluated for their effectiveness to control the disease and disorders, and other effects as plant growth stimulants. 3-hydroxy-5-methylisoxazole-a soil fungicide, Metalozyl-a fungicide which controls mildew, SF 8002-the composite of above two chemicals, and Isoprothiolane-a fungicide which controls rice blast were found to be effective controling seedling damping off and similar physiological disorders, and improvement of physiological characteristics of the seedlings such as the amount of bleeding sap, rooting ability, negative geotrophism, and regrowth after cutting. The results indicated that the chemicals will be very effective raising healthy seedlings even under unfavorable environments by the improvement of physiological activities of seedlings and preventing seedling damping off and the similar physiological disorders.
