Identification of Quantitative Trait Loci Associated with Seed Size and Weight in Soybean

  • Small seed size is one of the major traits of soybean cultivars for sprouts with regard to high sprout yield. This study was conducted to identify quantitative trait loci (QTL) for seed size and weight in a set of F 6 seeds of 89 lines derived from a cross between 'Pureunkong', a soybean cultivar developed for sprouts and 'Jinpumkong 2', a soybean cultivar with no beany taste in seed due to the lack of lipoxygenases. The genetic map of 25 linkage groups with a total of 98 markers including RFLP, RAPD, SSR and classical markers was constructed from this F/sbu 5/-derived population and was used for QTL analysis. 'Pureunkong' was significantly smaller (P<0.01) than 'Jinpumkong 2' in seed size and seed weight. Genetic variation was detected and transgressive segregation was common in the population for these traits. Seven DNA markers including opT14-1600 in LG A2, opF02-400 in LG B2, Satt100, opC09-700, opG04-730 and opQll-650 in LG C2, and opY07-1100 & 1000 in LG(unknown) were significantly associated and accounted for 4.7 to 10.9% and 5.1 to 10.1 % of the phenotypic variation in seed size and seed weight, respectively. 'Pureunkong' alleles increased seed size and seed weight at the all four significant marker loci on the LG C2. These marker loci in LG C2 were closely linked and were presumed to be a single QTL. Overall, at least three independent QTLs from 3 linkage groups (A2, B2, and C2) were putatively involved in the control of seed size and seed weight.

Basic Seed Stock Maintenance and Multiplication in Indian Tropical Tasar Silkworm Antheraea mylitta Drury-A Strategic Approach

  • Daba ecorace of Antheraea mylitta Drury (Lepidoptera: Saturniidae), the semi domesticated Indian tropical tasar silkworm being reared outdoor; the egg and silk yields are dependent of genotype environment interaction. The insufficient maintenance and multiplication of its P4 seed stock need a coherent as well as scientific strategy to safeguard breed potential, being commercially applied ecorace. The sort-out lines of P4 stock studied over five generations highlighting on commercial trait up gradation suits for a tropical crop season, revealed enhanced performance. The line with high pupal parents (T2) shown improved fecundity (12.9%) and the line with high shell parents (T3) recorded higher shell weight (40.0%) and silk ratio (24.1%). While, the line of high pupal female and high shell male (T4) reveal enhancement in fecundity (9.0%), egg hatching (14.1%), shell weight (50.0%), silk ratio (35.2%) and absolute silk yield (52.0%) indicating the need and role of varied basic seed stock lines. The approach could improve economically vital egg fecundity and cocoon shell weights besides balancing them in same line for commercial operation. The progressive show of lines (T1 to T4) along successive generations (G1 to G5), in spite of passing through seed crop (Jul-Aug) and commercial crop (Sep-Nov) seasons emphasize their compatibility. The study infers that the strategic plan of combining preferred parental phenotypes, methodical selection for desired commercial trait(s) through generations with best possible genotype environment interaction has enriched P4 stock with elevation in needy trait(s) besides assuring choice of suitable lines for seasons and regions and timely replenishment of basic seed of Daba ecorace.

QTL Analysis of Soybean Seed Weight Using RAPD and SSR Markers

  • Soybean [Glycine max (L.) Merr.] seed weight is a important trait in cultivar development. Objective of this study was to identify and confirm quantitative trait loci (QTLs) for seed weight variation in the F2 and F2:3 generations. QTLs for seed weight were identified in F2 and F2:3 generations using interval mapping (MapMaker/QTL) and single-factor analysis of variance (ANOVA). In the F2 plant generation (i.e., F3 seed), three markers, OPL9a, OPM7a, and OPAC12 were significantly (P<0.01) associated with seed weight QTLs. In the F2:3 plant row generation (i.e., F4 seed), five markers, OPA9a, OPG19, OPL9b, OPP11, and Sat_085 were significantly (P<0.01) associated with seed weight QTLs. Two markers, OPL9a and OPL9b were significantly (P<0.05) associated with seed weight QTLs in both generations. Two QTLs on USDA soybean linkage group C1 and R were identified in both F2 and F2:3 generations using interval mapping. The linkage group C1 QTL explained 16% of the variation in seed weight in both generations, and the linkage group R QTL explained 39% and 41% of the variation for F2 and F2:3 generation, respectively. The linkage group C2 QTL identified in F2:3 generation explained 14.9% of variation. Linkage groups C1, C2 and R had previously been identified as harbouring seed size QTLs. The consistency of QTLs across generations and populations indicates that marker-assisted selection is possible in a soybean breeding program.

  • Plants belonging to the category of 2n apomixis or agamospermy form embryos and seeds without the processes of normal meiosis and syngamy. Seeds produced in this way have identical genotype of their maternal parent. Three different types of agamospermy are recognized: diplospory, apospory, and adventitious (adventive) embryony. $F_1$ hybrid cultivars cannot be used as seed sources in the next ($F_2$) generation because this generation would be extremely variable as a result of genetic segregation. Hybrid vigor is also reduced in the $F_2$ generation. Therefore, parental stocks for hybrid seed production need to be maintained and cross must be continuously repeated. Agamospermic 2n apomixis would make it possible to fix the genotype of a superior variety so that clonal seeds faithfully representing that genotype could be continuously and cheaply produced independent of pollination. That is, $F_1$ hybrid seeds could be produced for many generations without loss of vigor or genotype alteration. Production of apomictic $F_1$ hybrid seed would be simplified because line isolation would not be necessary to produce seed or to maintain parental lines, and the use of male-sterile lines could be avoided. Overall, apomixis would enable a significant reduction in hybrid seed production costs. Additionally, the production of clonal seed is not only important for seed propagated crops, but also for the propagation of heterozygous fruit trees and timbers. Clonal seed would help avoid costly and time-consuming vegetative propagating methods that are currently used to ensure the large-scale production of these plants. Apomixis is scattered throughout the plant kingdom, but few important agricultural crops possess this trait Therefore, most research to date has centered on introgressing the trait of apomixis into agricultural crops such as wheat, maize, and some forage grasses from wild distant relatives by traditional cross breeding. The classical breeding approach, however is slow and often impeded by many breeding barriers. These problems could be surmounted by taking mutagenesis or molecular approach. Arabidopsis thaliana is a tiny sexually reproducing plant and is convenient in constructing and screening in molecular researches. Male-sterile mutants of Arabidopsis are particularly suitable genetic background for mutagenesis and screening for apomictic mutants. Molecular approaches towards isolating the genes controlling the apomictic process are feasible. Direct isolation of genes conferring apomixis development would greatly facilitate the transfer of this trait to wide variety of crops. Such studies are now in progress.

Quantitative trait loci (QTLs) detection for plant regeneration ability from seed culture in rice (Oryza sativa L.)

  • Quantitative trait loci (QTLs), which were related to the ability of callus induction and plant regeneration in seed culture of rice, were analyzed using a mapping population from a cross between the rice cultivars 'Samgang' (tongil type) and 'Nagdong' (japonica). A tongil type rice cultivar, 'Samgang' showed lower frequency (20%) of plant regeneration than that (35%) of japonica rice, 'Nagdong'. Transgressive segregations were observed for the ability of callus induction and plant regeneration from the seed-derived calli of 58 doubled haploid (DH) lines. The ability of plant regeneration of 58 doubled haploid lines showed a continuous distribution with comparatively wide range (10.0 to 66.7%) of variation. Composite interval mapping analysis was used to identify the QTLs controlling callus induction and plant regeneration ability. Four significant QTLs, qCWS6, qCWS8, qCWS9 and qCWS11, associated with callus weight per seed were detected on chromosomes 6, 8, 9, and 11 with LOD values of 3.30, 2.60, 2.70 and 2.43, explaining 36% of the total phenotypic variation. Three significant QTLs, qPR1, qPR6, and qPR11, for the ability of plant regeneration were located on chromosome 1, 6, and 11 at LOD score of 2.25, 2.15 and 2.55, accounting for 24 % of the total phenotypic variation. The present study should be useful for improving the efficiency of plant regeneration in tissue culture of indica rice by means of marker-assisted selection.

한국 서울 식생의 번성자로서 팥배나무의 형질 변화 양상 (Changes in Leaf and Reproductive Traits of Mountain Ash (Sorbus alnifolia) as Urban Flourisher in the Seoul Metropolitan, South Korea)

  • 도시 생태계에서 우점 수종의 형질 특성은 분획화 및 고립된 식생, 그리고 생물상호작용 감소 등 환경 변화에 대한 종의 형질 적응과 생태계 기능에 대한 통찰력을 제공한다. 우리는 한국의 서울 중심에서 남쪽 방향의 4개 산지에서 환경 요인 (지리, 기상 및 토양 속성)을 포함하여 모두 40개체 팥배나무 (Sorbus alnifolia)에서 측정한 4개 유형의 잎 형질 (Leaf area, Specific leaf area, Leaf dry mass content, Leaf shape index)과 7개 유형의 번식 형질 (Fruit width, Fruit length, Fruit shape, Fruit dry weight, Fruit dry matter content, Seed weight 및 Seed ratio)의 평균과 분산 양상, 그리고 형질 조합에 대한 다변량 분석을 실시하였다. 측정된 환경 요인은 조사 장소의 개별성을 나타내었지만, 도심 식생이 더 건조하고 생장도일이 길었다. 도심에서 팥배나무의 잎은 작고 무거워지며, 열매는 길쭉하고 가벼운 종자를 생산하여, 오랜 도시화 영향에 따른 형질 반응들이 관찰되었다. 우리의 연구는 식생 파편화 및 고립화에 따른 생물 상호작용 감소를 나타내는 도심 환경에서 팥배나무 집단의 성장 및 번식 기제의 변화를 확인하였다. 우리는 축소된 생물 상호작용 규모, 그리고 대기 오염 같은 부정적 환경 요인이 많은 도시에서 핵심 수종의 기능과 지속성에 관해 제한적이지만 뚜렷한 생태 정보를 제공한다.

RAPD Loci for Seed Protein and Oil Content in Soybean (Glycine max)

  • Seed protein and oil content is important trait in the soybean. Both seed protein and oil content in this plant species is inherited quantitatively. A 68-plant $F_2$ segregation population derived from a mating between Mercury and PI 467.468 was evaluated with random amplified polymorphic DNA (RAPD) markers to identify QTL related to seed protein and oil content. Marker OPB12 was found to be associated with differences in seed protein content. Four markers, OPA09b, OPM07b, OPC14, and OPN11b had highly significant effects on seed oil content. By interval mapping, the interval between marker OPK3c and OPQ1b on linkage group 13 contained a QTL that explained 25.7% variation for seed oil content.

  • Abiotic stresses such as high/low temperature, drought, salinity, and submergence directly or indirectly influence the physiological status and molecular mechanisms of rice which badly affect yield. Especially, the low temperature causes harmful influences in the overall process of rice growth such as uneven germination and the establishment of seedlings, which has become one of the main limiting factors affecting rice production in the world. It is of great significance to find the candidate genes controlling low-temperature tolerance during seed germination and study their functions for breeding new rice cultivars with immense low-temperature tolerance during seed germination. In this study, 120 lines of Cheongcheong/Nagdong double haploid population were used for quantitative trait locus analysis of low-temperature germinability. The results showed significant difference in germination under low different temperature conditions. In total, 4 QTLs were detected on chromosome 3, 6, and 8. A total of 41 genes were identified from all the 4 QTLs, among them, 25 genes were selected by gene function annotation and further screened through quantitative real time polymerase chain reaction. Based on gene function annotation and level of expression under low-temperature, our study suggested OsGPq3 gene as a candidate gene controlling viviparous germination, ABA and GA signaling under low-temperature. This study will provide a theoretical basis for marker-assisted breeding.

겨울 유채의 적정수확시기 결정에 관한 연구 (Determination of the Optimum Time of Harvest in Winter Rape)

  • Field experiment was conducted to determine the optimum harvesting time in winter rape (Brassica napus L.) by investigating the percent oil, 1, 000 seed weight, seed yield, dehiscent pod ratio and oil yield at 46, 50, 54, 58, 62, 66 and 70 days after flowering. Variation of all characters with days after flowering could be explained significantly by second degree polynomial equations. Percent oil and 1, 000 seed weight increased until 62 days after flowering and thereafter these traits decreased, while seed yield increased to 58 days after flowering and thereafter this trait decreased. This controversy was due to the drastic increase in dehiscent pods beyond 58 days after flowering which brought loss in seed yield. These results suggest that optimum harvesting time is 58 days after flowering and it should not be later than 60 days after flowering.
