Development of Non-Destructive Sorting Technique for Viability of Watermelon Seed by Using Hyperspectral Image Processing (초분광 영상기술을 이용한 수박종자 발아여부 비파괴 선별기술 개발)

  • Seed viability is one of the most important parameters that is directly related with seed germination performance and seedling emergence. In this study, a hyperspectral imaging (HSI) system having a range of 1000-2500 nm was used to classify viable watermelon seeds from nonviable seeds. In order to obtain nonviable watermelon seeds, a total of 96 seeds were artificially aged by immersing the seeds in hot water ($25^{\circ}C$) for 15 days. Further, hyperspectral images for 192 seeds (96 normal and 96 aged) were acquired using the developed HSI system. A germination test was performed for all the 192 seeds in order to confirm their viability. Spectral data from the hyperspectral images of the seeds were extracted by selecting pixels from the region of interest. Each seed spectrum was averaged and preprocessed to develop a classification model of partial least square discriminant analysis (PLS-DA). The developed PLS-DA model showed a classification accuracy of 94.7% for the calibration set, and 84.2% for the validation set. The results demonstrate that the proposed technique can classify viable and nonviable watermelon seeds with a reasonable accuracy, and can be further converted into an online sorting system for rapid and nondestructive classification of watermelon seeds with regard to viability.

A Review of Current Status and Applications of Korean Industrial Standards (KS) on the Floor Slip Resistance Testing (바닥의 미끄럼 시험에 관한 한국산업표준(KS) 현황 및 적용 실태)

  • Although various laws and regulations have been put in place to prevent slips and falls, many accidents still occur. In this study, the root cause of slips and falls not decreasing were investigated. There are five types of slip resistance test methods in the Korean Industrial Standards (KS). Namely, KS F 2375:2016, KS F 2601:2020, KS F 2602:2016, KS L 1001:2020, and KS G 5821-1:2020 are listed. These test methods are cited in building certification standards (BF and G-SEED), construction specifications, and other documents that specify slip safety criteria. As a result of the investigation, a number of errors in KS regulations and legal text, errors in the manufacture and operation of slip testers, and errors in the use of measured values were found. These errors threaten the public life safety and disrupt industrial sites, and must be corrected immediately.

Assessment of Genetic Relationship among Watermelon Varieties Revealed by ISSR Marker (Inter-simple sequence repeat (ISSR) marker를 이용한 수박의 품종간 유연관계 분석)

  • Inter-simple sequence repeat (ISSR) analysis were used to assess genetic diversity among 18 genotypes of watermelon (Citrullus lanatus Thunb.) including breeding lines and commercial varieties. The 21 ISSR primers selected from 100 primers were showed the amplification of 105 reproducible fragments ranging from about 200 bp to 5000 bp. A total of 58 DNA fragments were polymorphic with an average 2.7 polymorphic bands per primer. The polymorphic primers were divided into 18 anchored primers and 3 non anchored primers. All of the anchored primers were di-nucleotide repeat motif, and was more polymorphic than non anchored primers. Eighteen watermelon genotypes were classified into two large groups. Clustering was in some accordance with the division of fruit shape into 18 watermelon. Therefore, ISSR markers may be suitable for variety discrimination and for constructing a linkage map of watermelon.

Mutagenic and Antimutagenic Effects of Hemp Seed Oil Evaluated by Ames Salmonella Testing (삼종실유의 항돌연변이 효과)

  • We examined the in vitro mutagenic and antimutagenic effects of hexane-extracted hemp (Cannabis sativa L. subsp. sativa var. sativa) seed oil (HO) with and without S9-mediated metabolic activation, using the TA98 and TA100 Salmonella Typhimurium strains. The MTT assay revealed no cytotoxicity in HepG2 cells for HO quantities $\leq400g$/mL. In the mutagenicity test, revertant colonies did not exceed spontaneous colonies in number. Colony numbers did not increase in either strain after HO treatment, with or without metabolic activation. HO showed no mutagenic effects and did not induce a mutation in either strain. In the antimutagenicity test, HO reduced the number of mutated colonies induced by 4NQO in both strains. The inhibition rates of HO (TA98, 21-91%; TA100, 21-85%) indicated a potent reduction in mutagenicity induced by 4NQO. HO showed no significant mutagenicity and may have antimutagenic effects, as assessed by Ames testing.

  • In this study, the feasibility of hyperspectral reflectance imaging technique was investigated for the discrimination of viable and non-viable lettuce seeds. The spectral data of hyperspectral reflectance images with the spectral range between 750 nm and 1000 nm were used to develop PLS-DA model for the classification of viable and non-viable lettuce seeds. The discrimination accuracy of the calibration set was 81.6% and that of the test set was 81.2%. The image analysis method was developed to construct the discriminant images of non-viable seeds with the developed PLS-DA model. The discrimination accuracy obtained from the resultant image were 91%, which showed the feasibility of hyperspectral reflectance imaging technique for the mass discrimination of non-viable lettuce seeds from viable ones.

Survey on Seed-borne Fungi in Some Important Crops of Korea (한국산(韓國産) 주요작물(主要作物)의 종자전염성(種子傳染性) 진균(眞菌)에 관(關)한 조사(調査))

  • One hundred and sixteen seed samples of 16 different crops were tested by routine seed health testing methods. Seeds of rice, soybean and barley were found to be associated with 18, 15 and 7 species of fungi, respectively. Discoloration and spotting of various kinds were observed in rice seeds. Epicoccum purpurascens, Pyricularia oryzae and Trichoconiella pedwickii were higher in these discoloured than in normal seeds of rice. Macrophomina phaseolina, the charcoal-rot fungus is a new record for soybean in Korea. Seeds of sesame were infected by 13 fungi Alternaria sesami, A. sesarmicola and Corynespora cassiicola were detected predominantly. Twenty one species of fungi were detected in seed samples of 12 commercial vegetable crops. Alternaria, Colletotrichum and Fusarium were found predominantly. Some seed samples of commercial vegetable crops were heavily infected with pathogenic fungi and showed heavy seed and seedling rot.

One-step Multiplex RT-PCR Method for Simultaneous Detection of Seed Transmissible Bacterium and Virus Occurring on Brassicaceae Crop Seeds (십자화과 작물 종자에서 종자전염 세균 및 바이러스 동시 검출을 위한 One-step Multiplex RT-PCR 방법)

  • The aim of this research was to develop specific and sensitive PCR-based procedures for simultaneous detection of economically important plant pathogenic bacteria and seed borne virus in commercial Brassicaceae crop seeds, Xanthomonns campestris pv. campestris (Xcc) and Lettuce Mosaic Virus (LMV). Bacterial and virus diseases of Brassicaceae leaves are responsible for heavy losses. PCR with arbitral primers: selection of specific primers, performance of PCR with specific primers and determination of the threshold level for pathogens detection. To detect simultaneously the Xcc and LMV in commercial Brassicaceae crop seeds (lettuce, kohlrabi, radish, chinese cabbage and cabbage), two pairs of specific primer (LMV-F/R, Xcc-F/R) were synthesized by using primer-blast program (http://www.ncbi.nlm.nih.gov/tools/primer-blast/). The multiplex PCR for the two pathogens in Brassicaceae crop seeds could detect specifically without interference among primers and/or cDNA of other plant pathogens. The pathogen detection limit was determined at 1 ng of RNA extracted from pathogens. In the total PCR results for pathogen detection using commercial kohlrabi (10 varieties), lettuce (50 varieties), radish (20 varieties), chinese cabbage (20 varieties) and cabbage (20 varieties), LMV and Xcc were detected from 39 and 2 varieties, respectively. In the PCR result of lettuce, LMV and Xcc were simultaneously detected in 8 varieties.

Change of Germination Rate for Chili Pepper and Chinese Cabbage Seed in Relation to Packaging Materials and Storage Conditions over 10 Years (보관용기 및 저장조건에 따른 고추 및 배추종자의 10년간 발아율 추이)

  • Seed viability is affected by storage conditions and rapid loss of viability in storage is the major cause for low germination. This study was carried out to examine the effect of packaging materials and storage temperature on seed germination rate over 10 years in two species (Capsicum annuum L. and Brassica rapa L. ssp. pekinensis) and determine effective storage conditions for maintaining seed viability. Seeds were packaged in aluminum poly pouches under vacuum, polyethylene bottles, and paper bags containing silica gel and stored under one of two controlled conditions ($15^{\circ}C$, RH 40% or $5^{\circ}C$, RH 30%) or at ambient condition. Seed germination was recorded at 6-month intervals for 10 years. The seeds of both species showed no decline in viability until 6.5 years at 15 or $5^{\circ}C$, irrespective of packaging materials. However, under ambient conditions, the seeds of chili pepper and Chinese cabbage in paper bags lost viability after 4 and 5 years, respectively. By contrast, seeds of both species in vacuum-aluminum poly pouches exhibited a 99% germination rate after 6 years under ambient conditions. Pepper seeds in the vacuum-aluminum poly pouches maintained a 93% germination rate after 10 years in ambient conditions. These results indicated that a special seed storage facility for maintaining viability of chili pepper and Chinese cabbage seed might not be essential and seed testing would not be necessary for 10 years, if chili pepper and Chinese cabbage seeds were packed in ambient/vacuum-aluminum poly pouches or $5^{\circ}C$/vacuum-aluminum poly pouches.

Adaptive Random Testing for Integrated System based on Output Distribution Estimation (통합 시스템을 위한 출력 분포 기반 적응적 랜덤 테스팅)

  • Adaptive Random Testing (ART) aims to enhance the performance of pure random testing by detecting failure region in a software. The ART algorithm generates effective test cases which requires less number of test cases than that of pure random testing. However, all ART algorithms currently proposed are designed for the tests of monolithic system or unit level. In case of integrated system tests, ART approaches do not achieve same performances as those of ARTs applied to the unit or monolithic system. In this paper, we propose an extended ART algorithm which can be applied to the integrated system testing environment without degradation of performance. The proposed approach investigates an input distribution of the unit under a test with limited number of seed input data and generates information to be used to resizing input domain partitions. The simulation results show that our approach in an integration environment could achieve similar level of performance as an ART is applied to a unit testing. Results also show resilient effectiveness for various failure rates.

Studies on the development of seed disinfectant in non-mercurious compounds (비유기수은 종자소독제개발에 관한 연구)

  • Tests were made to find new non-mercurious seed disinfectants that were best for rice seeds. For these experiments four seed samples carr)?ing natural infection of Pyricularia oryzae, Helminthosporium oryzae, and Fusarium moniliforme were used and the following fungicides were used; Zinc Omadine, Sodium Omadine, Panoctine, Tecto-F, Benlate-T, Homai, Sisthane, $P_{242}$, Busan 30, Tecto-Wp and Terracoat Zn. Blotter method and water agar plate method used in the laboratory and growing-on test used in greenhouse. Results have shown that Sisthane, $P_{242}$ and Sodium Omadine have equal or superior effect to organic mercury compound against P. oryzae, H. oryzae, and F. moniliforme. Benlate T and Homai are effective against blast and Bakanae disease, but are inferior to organic mercury compound against brown spot disease. Busan 30 and Panoctine are effective against blast and brown spot disease, but have moderately inferior effects against F. monilifome. It is considered that the recommendable testing methods of seed treatment were blotter method for P. oryzae and water agar plate method for H, oryzae and F. moniliforme according to the experimental results obtained. No phytotoxicity against seed germination and seedling growth were observed when treated with disinfectants before germination of seeds.
