Effect of Fertilizer N Application and Bradyrhizobium sp. (Vigna) Inoculation on Symbiotic N2 Fixation of Peanut at Newly Reclaimed Soil (신개간지(新開墾地)에서 질소시비(窒素施肥)와 근류균(根瘤菌) 접종(接種)이 땅콩의 질소고정(窒素固定)에 미치는 영향(影響))

  • This experiment was carried out to study the effects of Bradyrhizobium moculation and fertilizer N application on symbiotc $N_2$ fixation of peanut (Arachis hypogaea L. cv. Younghotangkong) at newly reclaimed soil. Treatments consisting of B. sp. (Vigna) HCR-3 and HCR-46 with control and five levels of nitrogen (0, 4, 8, 16, 32kg/10a) were arranged with split design of four repetitions. The results obtained were as follows. The number of viable Bradyrhizobium in rhizosphere was decreased to 20 days after sowing and thereafter it was remarkably increased, until full ripe stage of pods, decreased as fertilizern rates increased. Nodulation and nitrogenase activity were reduced with increasing levels of fertilizer, whereas top dry matter and nitrogen accumulation where increased. Seed yield of Bradyhizobium inoculation under the fertilizer N zero level was similar to that N : 8kg/10a under uninoculation. It suggested that Bradyrhizobium inoculation could replace the effect of nitrogen application.

  • Effective strains of cowpea bradyrhizobia JB7 $nal^rspe^r$ and CB756 $str^rrif^r$, antibiotic-resistant variants of JB7 and CB756, respectively, were used to examine changes of rhizosphere populations and nodule occupancy. Populations of each strain increased gradually in the rhizosphere, reaching a maximum of about $10^8$ cells per root system. Nodule number increased as the density of inoculum increased from $10^2$ cells to $10^8$ cells per seed. Inoculation with liquid suspension resulted in the formation of more nodules than the peat slurry or granule inoculation. When JB7 $nal^rspe^r$ and CB756 $str^rrif^r$ were introduced in equal numbers in inoculum mixtures the former consistantly occupied the majority of nodules with all three groundnut cultivars used. There was no difference in yield between nitrogen treatments and inocultation treatments.

Development of PCR and TaqMan PCR Assays to Detect Pseudomonas coronafaciens, a Causal Agent of Halo Blight of Oats

  • Pseudomonas coronafaciens causes halo blight on oats and is a plant quarantine bacterium in many countries, including the Republic of Korea. Using of the certificated seed is important for control of the disease. Since effective detection method of P. coronafaciens is not available yet, PCR and TaqMan PCR assays for specific detection of P. coronafaciens were developed in this study. PCR primers were designed from the draft genome sequence of P. coronafaciens LMG 5060 which was obtained by the next-generation sequencing in this study. The PCR primer set Pc-12-F/Pc-12-R specifically amplified 498 bp from the 13 strains of P. coronafaciens isolated in the seven different countries (Canada, Japan, United Kingdom, Zimbabwe, Kenya, Germany, and New Zealand) and the nested primer set Pc-12-ne-F/Pc-12-ne-R specifically amplified 298 bp from those strains. The target-size PCR product was not amplified from the non-target bacteria with the PCR and nested primer sets. TaqMan PCR with Pc-12-ne-F/Pc-12-ne-R and a TaqMan probe, Pc-taqman, which were designed inside of the nested PCR amplicon, generated Ct values which in a dose-dependent manner to the amount of the target DNA and the Ct values of all the P. coronafaciens strains were above the threshold Ct value for positive detection. The TaqMan PCR generated positive Ct values from the seed extracts of the artificially inoculated oat seeds above 10 cfu/ml inoculation level. PCR and TaqMan PCR assays developed in this study will be useful tools to detect and identify the plant quarantine pathogen, P. coronafaciens.

Antifungal Activities of Crude Extractum from Camellia semiserrata Chi (Nanshancha) Seed Cake Against Colletotrichum musae, Colletotrichum gloeosporioides and Penicillium italicum in vitro and in vivo Fruit Test

  • Antifungal activities of crude extractum of Nanshancha Seed Cake (NSC), to inactivate postharvest pathogens were investigated. Highest inhibitory rate was found against C. musae, C. gloeosporioides and C. papaya P.Henn, which was much stronger than that by tea saponin. Compared to tea saponin, effects of NSC extractum was relatively weak and similar on C. gloeosporioides Penzig and P. italicum. In an in vivo study, best controlling effects by NSC extractum was found with banana anthracnose disease development, which showed no inhibitory effects by tea saponin. NSC extractum controlled in vitro C. musae growth through directly inhibiting germination rate and germ tube elongation, and causing distortation, rupture and indentation of C. musae mycelium. In banana fruit subject to C. musae inoculation, higher PAL, POD, GLU and CHT activity was observed in banana fruit treated with crude NSC extractum than that of water control fruits. Current study proved the best controlling effects of crude NSC extractum in C. musae in vitro and in vivo development, which through direct inhibition of C. musae growth and increasing defense system of the banana fruit.

Sensory Characteristics of Citrus Vinegar fermented by Gluconacetobacter hansenii CV1 (제주 감귤식초 발효균주 선발)

  • Citrus juice, a concentrate manufactured by the Jeju Provincial Corporation, was converted into vinegar orderly by alcohol and acetate fermentation. The juice with a 6-fold dilution by distilled water was used as the sole nutrient source throughout the experiments. The diluted juice contained 12.96Brix of total sugar, $0.632\%$ of total acid and $20.23{\mu}g/ml$ of hesperidin. Naringin was not detected from the juice. Citrus wine having $5.6\~6.3\%$ alcohol was produced from the diluted juice after 3 days of fermentation at $28^{\circ}C$. A kind of citrus-malomelo-yeast CMY-28 was used for the wine fermentation. The wine was successfully fermented for 8 days at $30^{\circ}C$ after inoculation of seed vinegar which contained active cells of acid producing bacteria CV1. The inoculum size of the seed vinegar was controlled to $10\%$(v/v) of the citrus wine. The wine was converted into vinegar by the fermentation process. Citrus vinegar, the final fermentation product, was colored with very thin, radish-yellow and was transparent. It's acidity ranged between $5.8\~6.2\%$ of that of acetic acid. The vinegar attained the best score by sensory test among several natural fruit vinegars. It was clear from the results that high quality citrus vinegar could be produced from concentrated citrus juice. However, the fermentation conditions should be improved to reduce the amount of reducing alcohol.

  • Occurrence of sheath rot rice caused by Acrocylindrium oryzae on indica varieties, IR lines and IR varieties was light to moderate in the field, whereas generally none of the recommended japonica varieties showed the disease. In general, percentage infection in the field agreed with that obtained from seed inoculation tests made in the laboratory. Culture filtrates of the fungus inhibited the sprouting of seeds of rice, barley, wheat, rye and rape. A new variety, Tongil obtained from a hybrid between japonica and indica was inhibited significantly more than was Jinheung. Conidial germination of Pyricularia oryzae was inhibited by the filtrate while no inhibitory effects were found on conidia of Cochliobolus miyabeanus.

Characteristics and suitability of various cereal grains in spawn production of button mushroom

  • Spawn is the vegetative growth of the mycelium and serves as the inoculum or seed for cultivating mushrooms. Various cereal grains, such as wheat, millet, barley, sorghum, brown rice, rye, and oat were assessed to compare their characteristics and suitability for spawn production of button mushroom (Agaricus bisporus). The mycelial growth rates, density, and the number of completely colonized grains were measured from the twentieth day of inoculation. Wheat grains showed fastest mycelial growth with 8.4 cm followed by rye, oat, barley with 8.2, 7.5 and 7.3 cm, respectively. In the mycelial density, foxtail millet, barley, and sorghum were best compared with that of wheat grains. Especially, the number of grains which were completely colonized by mycelia were greatest in foxtail millet with 5,123 grains followed by proso millet, and wheat with 3,052 and 914, respectively. Based on the results obtained, barley, foxtail millet, and sorghum grains would be appropriate substituting for wheat grain in spawn production of button mushroom.

Antibody-Mediated Resistance to Rhizomania Disease in Sugar Beet Hairy Roots

  • Agrobacterium rhizogenes-mediated transformation of sugar beet hairy roots expressing single-chain variable fragment (scFv) was exploited to evaluate the efficacy of four antibody-based constructs for interfering with the Beet necrotic yellow vein virus infection. The scFv specific to a major coat protein of virus, p21, was targeted to various cellular compartments including the cytosol (pIC and pICC constructs), apoplast (pIA), and mitochondrion (pIM). After mechanical virus inoculation, most of the hairy root clones expressing scFv in the cytosol displayed low virus titers while the majority of transgenic hairy root clones accumulated antibody in outer membrane of mitochondria or apoplast were infected. This hairy root system provided an efficient and rapid approach to initially investigating root disease resistance like rhizomania prior to transform whole recalcitrant plants such as sugar beet.

  • Lines bred for resistance to Phytophthora capsici by incorporation of resistance to P. capsici in PI201234 or CM334 into 'Subi' and 'Chilseong', land races in Youngyang, and 'Punggak', a land race in Cheongdo in Gyeungbuk province, and lines bred for fortification of one of them above with resistance to viral complex, and tolerant selections from another landrace collection from Punggak (KC268) were evaluated for resistance to P. capsici by inoculation at seedling stage. Almost all the breeding lines showed high level of resistance to P. capsid and selections from KC268 showed tolerance or moderate resistance to P. capsid. The selected plants were grown in a net cage in an outdoor nursery for seed production. Utilization of the lines in breeding was discussed.

  • In this study, colloidal Ag solution was spouted on the surface of the inorganic pigment using the hybridizer system and the spray nozzle. Then, the surface of the inorganic pigment was modified by titanium dioxide in order to possess antibacterial ability. Nano-sized colloidal Ag was made by using a seed sol method in this study. It was confirmed that the size of particle per unit weight becomes enlarged, as the addition of $AgNO_{3}$ increased, and as the time of reaction increased, in the manufacturing process of nano-sized colloidal Ag. The antibacterial measurement of the inorganic pigment showed that the growth of fungus was reduced as the reaction time increased. It was measured that the antibacterial activity was excellent at fixed time frame, after the antibacterial ability appeared in $5{\sim}7$ hours of the antibacterial inoculation experiment. The experiment of titanium dioxide's Photocatalyst effect showed $60{\sim}70%$ efficiency in about 80 minute reaction time of the dissolution results regarding measurements of benzene. It was shown that more than 90% of the dissolution efficiency was achieved in the reaction time of about 30 minute.
