Seedborne Fungi and Fungicide Seed Treatment of Ginseng

  • The incidence of fungi and their possible contribution to low vigour were examined in a collection of ginseng (Panax quiquefolius) seed from Ontario. When examined after one winter of stratification in the field and storage at 4f for five months in the laboratory, the collection exhibited low vigour (plant stand 16.7% of seeding rate six weeks after seeding) and high incidence (94%) of discolored or soft seed. Fungi isolated (and incidence) from 1,304 endosperm halves recovered from surface-sterilized seed were, in order of abundance, Fusarium rostrum (22.2%), Chaetomium crispuum (14.3%), Funriud oxysporum (9.0%), Fusarium sdani (9.0%), iwmor sp. (8.4%), Alternaria sp. (8.1%), Zowieua lucotricha (7.8%), Cylindruarpn sp. (0.9%), Fusarium avenacmm (0.9%), and Vdudla iliata (0.4%). Most of these fungi, including known and potential pathogens of ginseng (species of Alerraria, Cylindrocarpon, Fusarium, and Trichodirma), were associated with both healthy and diseased seed. Application of Benlate (benomyl), Thiram (thiram), or UBI-2584 (tebuconazole) to seed caused slight to pronounced reduction in emergence and did not significantly affect plant stand six weeks after seeding. The study demonstrated the high level of infection by fungi, including known and potential pathogens of the cry, in an arbitrary collection of ginseng seed from commercial sources, and the phytotoxicity of the fungicides tested when applied to moist stratified seed. The lack of efficacy of the fungicides precluded determination of the contribution of seedborne fungi to low vigour of the seed.

Morphological Characters and Seed Transmission of Bipolaris panici-miliacei Causing Leaf Spot of Common Millet (기장 점무늬병균 (Bipolaris panici-miliacei)의 형태적 특징과 종자전염)

  • Bipolaris panici-miliacei, Cercospora fusimaculans, Fusarium moniliforme and Rhizoctonia solani were pathogenic fungi detected from 5 seed samples of common millet (Panicum miliaceum). Morphological characters of B. panici-miliacei were as follows. Conidiophores were dark olivaceous brown, simple, cylindrical, geniculate, and septate. Conidia were fusoid, dark olivaceous brown, tapering gradually toward the ends, straight to slightly curved, 3~13 distoseptate, and 29.4~155.4$\times$10~26 ${\mu}{\textrm}{m}$ in size with dark hilum included within the contour of the basal cell. Seed infection with B. panici-miliacei caused seed rotting, coleoptile spot, and seedling blight of common millet plants. According to the inoculation experiments, B. panici-miliacei showed strong virulence on the young seedlings of common millet, but very weak virulence on the young seedlings of rice (Oryza sativa) and foxtail millet (Setaria italica).

Development of Molecular Markers to Detect Diaporthe spp. from Decayed Soybean Seeds

  • Soybean is one of the world's most widely cultivated food crops, and soybean seeds are supplied from national seed resources in Korea. However, the transmission of seed-borne diseases through infected soybean seeds is problematic. Among these diseases, soybean seed decay is caused by Diaporthe spp. Infecting the pods, and the infected seeds show rotting symptoms. Most diseased seeds are removed during the selection process; however, it is difficult to distinguish infected seeds that do not display symptoms. Hence, a sequencebased method was devised to screen Diaporthe-infected seeds. Based on the nuclear ribosomal internal transcribe spacer (ITS) region of the pathogen, a primer was designed to distinguish the infection from other soybean seed pathogens. As a result of the comparison between healthy and Diaporthe-diseased seeds by using the primers, Diaporthe was detected only in the diseased seeds. Therefore, it is possible to distribute healthy soybean seeds by detecting Diaporthe-diseased seeds at the genetic level using the Diaporthe-specific primers.

Development of non-destructive measurement method for discriminating disease-infected seed potato using visible/near-Infrared reflectance technique (광 반사방식을 이용한 감염 씨감자 비파괴 선별 기술 개발)

  • Pathogenic fungi and bacteria such as Pectobacterium atrosepticum, Clavibacter michiganensis subsp. sepedonicus, Verticillium albo-atrum, and Rhizoctonia solani were the major microorganism which causes diseases in seed potato during postharvest process. Current detection method for disease-infected seed potato relies on human inspection, which is subjective, inaccurate and labor-intensive method. In this study, a reflectance spectroscopy was used to classify sound and disease-infected seed potatoes with the spectral range from 400 to 1100 nm. Partial least square discriminant analysis (PLS-DA) with various preprocessing methods was used to investigate the feasibility of classification between sound and disease-infected seed potatoes. The classification accuracy was above 97 % for discriminating disease seed potatoes from sound ones. The results show that Vis/NIR reflectance method has good potential for non-destructive sorting for disease-infected seed potatoes.

Fluctuations in the Outbreak of Jujube(Ziziphus jujuba Miller) Witches'-broom Disease (대추나무 빗자루병의 발생량 변화)

  • Infection rate of witches'-broom disease (WB) was observed yearly in transplanted jujube seedlings for 7 years. Seedlings of‘Hongan’grafted either on the rootstock from seed (RS) or on the rootstock from rooting (RR) were planted in separate farms. Occurrence of the disease was determined 4 years after transplanting with the witches'-broom symptom. Only 1 seedling of RS was infected with WB, while 56 seedlings of RR showed WB. Yearly infection rates of WB were observed in several varieties of jujube including‘Boeun’,‘Keumsung’,‘Moodeung’,‘Bokjo’,‘Koori’(10 trees for each in Cheongjoo), and‘Hongan’(165 trees in Boeun). In most varieties, WB first appeared in the third year from transplanting, and in the seventh year, more than 80% of the total tree showed WB. The yearly infection rate of newly infected trees was almost. stable during the first years. However, the ratio sharply increased from the sixth year. The accumulated infection rate of WB increased double, year by year from the third to seventh year. Between Cheongjoo and Boeun, no difference in infection rates was found.

Effect of Soil and Tuber Disinfection on Potato Common Scab(S. scabies) in Jeju Field Conditions (감자 더뎅이병 화학적 방제)

  • It was conducted to find out the effect of chemical treatment on seed tubers and soil to control potato common scab in Jeju Island in 2001. Although the treatment of soil disinfection with Dazomet Gr. (30kg/10a) was more effective comparing to the treatment of seed disinfection and the untreated, the control value was merely 40.5%. However, the combined treatment of soil and seed disinfection was considerably more effective to reduce the percentage of infection and infection area, and increase the production of marketable tubers and also increase the control value. Therefore, the combined treatments are demanded to reduce the scab incidence in Jeju field conditions.

Seed Transmission of Tomato yellow leaf curl virus in White Soybean (Glycine max)

  • Tomato yellow leaf curl virus (TYLCV) infection of the common bean (Phaseolus vulgaris) has been reported, but soybean (Glycine max) has not previously been identified as a TYLCV host. Five cultivars of white soybean were agro-inoculated using an infectious TYLCV clone. At 30 days post-inoculation, they showed infection rates of 25% to 100%. Typical TYLCV symptoms were not observed in any inoculated plants. To examine whether TYLCV was transmitted in soybean seeds, DNA was isolated from bundles of five randomly selected seeds from TYLCV-inoculated soybean plants and amplified with a TYLCV-specific primer set. With the exception of one bundle, all bundles of seeds were verified to be TYLCV-infected. Virus dissemination was also confirmed in three of the 14 bunches. Viral replication was also identified in seeds and seedlings. This is the first report demonstrating that soybean is a TYLCV host, and that TYLCV is a seed-transmissible virus in white soybean.

Rice Seed Infection of Fusarium spp. at Paddy Field and Selection of Seed Disinfectants (포장(圃場)에서 Fusarium속균(屬菌)의 종자(種子) 감염(感染)과 소독제(消毒劑)의 선발(選拔))

  • F. roseum "Graminearum" and F. moniliforme were isolated from hulls of seeds collected from fields. F. roseum "Graminearum" was isolated from brown rice of all the varieties tested. F. moniliforme was isolated from Nagdongbyeo, Seomjinbyeo, Sinsunchalbyeo, Milyang 23, Baegyangbyeo and Samnambyeo. F. moniliforme was isolated from the most of the seeds damaged by sparrow and it formed sporodochia of this fungus. Five species including F. moniliforme were isolated from plant hopper collected from paddy fields in September. To select seed a disinfectant, twelve fungicides were tested and P242 was one of the best effective fungicidetested. After seed treatment with the fungicide, F. moniliforme was not isolated from hulls but was frequently isolated from brown rices.

Studies on the Potato Virus X and Potato Leaf Roll Virus for Disease-free Seed Potato Production (무병종서 생산을 위한 감자X바이러스 및 엽권바이러스에 관한 연구)

  • A series of experiment was carried out to study on the production of disease-free seed potatoes at the Alpine Experiment Station from 1960 to 1968, which initiated a study of comparison on degeneration of plain warm region and high altitude products and the effect of latent potato virus X (PVX) and potato leaf roll virus(PLRV) on degeneration. Particular observations were made on some aspect of the nature of potato virus disease and its control such as concentrations of PVX, range of host plants, physical properties such as concentrations of PYX, range of host plants, physical properties and carrying effect of insects, by investigating 9 different areas of the main potato producing regions (Kimhae, Taegu, Choongju, Taejoen, Suwon, Kwangju, Chonju, Cheju and Chinju). Highly purified anti-serum was separated and tested for control of the virus disease and also various method of prevention and control of PLRV were observed, using cultivation of sprouted seed tubers, early harvesting method, and systemic chemicals. The results obtained are summarized as follows; 1. Potato yield in the plain region decreased by 32.8~66.3% in the first year cultivation of seed potatoes from colder region, and the rate of virus infection was 92.9 to 95.4%. 2. Plants of three families including, 20 species were susceptible to the PVX, and among the plants Salvia officinalis of a habits only was the carrier while the symptom of Digitalis purpurea of Screphulariaceae was masked. Necrosis and ring spot was occurred in most pJants of the Solanaceae and ring spot symptom also was observed in Nicotiana tabacum L. var. White Burley and in N. glutinosa. 3. The 8$C_2$ strain of virus had the following physical properties; thermal inactivation point, 68-$72^{\circ}C$ : dilution inactivation point, above 1, 000, 000 dilution: ageing in vitro, 240-360 days: and ageing in dry plant tissue, 30 days. 4. Myzus persicae and Oxya spp. did not transmit the 8$C_2$ strain of potato virus. 5. Virus was purified through the ammonium sulphate isolating method, and higher titer value, 1/2048 was obtained through anti-serum test. 6. Inhibition Chenopodiacae on the virus infection of potato was remarkable, and inhibition of local lesion host also was observed. 7, By earlier planting of sprouted seed tubers, growth period could be prolonged by 10 to 12 days. 8. Earlier harvest decreased much the rate of virus infection of seed potatoes. 9. According to the results of aphid control trial using systemic soil insecticides at Kangnung and Taekwanlyung, PSP 204, Disyston and Thimet was effective to aphid control. In particular, control effect of twice treatments of PSP 204 was great. 10. Treatmental effect of those chemicals lasted about 60-70 days. However, single foliar application of emulsified chemicals was not effective to potato virus control. 11. The effect of PSP 204, Disyston, and Thimet on the control of potato leaf roll virus was great, particularly in the case of two treatments of PSP 204, at Kangnung as well as at Taekwanlyung. Higher negative correlationship between the control effect of potato leaf roll virus and potato yield was observed showing the value r=-0.85 at Kangnung, and r=-0.87 at Taekwanlyung. 12. Differences in the control effects among PSP 204, Disyston, and Thimet was not noticed.

Screening of Seed Treatment Agents against Leaf Blight and Black Root Rot for Carrot Organic Cultivation (당근 유기재배를 위한 검은잎마름병과 검은무늬병 종자소독제 선발)

  • This study was conducted to control the seed borne pathogens (Alternaria spp.) of carrot and to examine the seed germination rate by using 13 environment friendly agricultural materials instead of conventional chemicals for organic cultivation. The growth inhibiting effects on pathogens showed the different responses according to each agricultural material and effective 7 materials against seed borne pathogens were selected. Among 7 materials, the carrot seeds sterilized with plant extracts, Tanger Stop and Land Saver were not germinated at all. The germination rate of seeds sterilized with other materials showed the similar levels with reference chemical (Benlate-T) and non-treated seeds. Infection rates of seeds sterilized with seaweed extract, Bellopper for controlling A. radicina and plant extract, Ssial-100 for A. dauci were similar or low infection rate compared with reference chemical. The germination rate and root length of seeds sterilized with Bellopper and Ssial-100 was similar or superior to those of chemical sterilized seeds. The optimal condition seemed to be a little bit different depending on the concentration of materials. As a result, the sterilization of carrot seeds by using the environment friendly materials could be effectively utilized as a technology to inhibit the infection of seed-borne pathogens.
