Bacterial leaf blight of carrots caused by Xanthomonas hortorum pv. carotae (Xhc) is an important worldwide seed-borne disease. In 2012 and 2013, symptoms similar to bacterial leaf blight were found in carrot farms in Jeju Island, Korea. The phenotypic characteristics of the Korean isolation strains were similar to the type strain of Xhc. Pathogenicity showed symptoms on the 14th day after inoculation on carrot plants. Identification by genetic method was multi-position sequencing of the isolated strain JJ2001 was performed using four genes (danK, gyrB, fyuA, and rpoD). The isolated strain was confirmed to be most similar to Xhc M081. Furthermore, in order to analyze the genetic characteristics of the isolated strain, whole genome analysis was performed through the next-generation sequencing method. The draft genome size of JJ2001 is 5,443,372 bp, which contains 63.57% of G + C and has 4,547 open reading frames. Specifically, the classification of pathovar can be confirmed to be similar to that of the host lineage. Plant pathogenic factors and determinants of the majority of the secretion system are conserved in strain JJ2001. This genetic information enables detailed comparative analysis in the pathovar stage of pathogenic bacteria. Furthermore, these findings provide basic data for the distribution and diagnosis of Xanthomonas hortorum pv. carotae, a major plant pathogen that infects carrots in Korea.

Purple-discoloration of the uppermost leaves has been observed in some soybean cultivars in recent years. The purpose of this study was to characterize the novel phenotypic changes between the uppermost and middle leaves via multiple approaches. First, quantitative trait loci mapping was conducted to detect loci associated with the novel phenotype using 85 recombinant inbred lines (RILs) of the 'Daepung' × PI 96983 population. 180K SNP data, a major quantitative trait locus (QTL) was identified at around 60 cM of chromosome 6, which accounts for 56% of total phenotypic variance. The genomic interval is about ~700kb, and a list of annotated genes includes the T-gene which is known to control pubescence and seed coat color and is presumed to encode flavonoid 35-hydroxylase (F3'H). Based on Hyperspectral imaging, the reflectance at 528-554 nm wavelength band was extremely reduced in the uppermost leaves compared to the middle (green leaves), which is presumed die to the accumulation of anthocyanins. In addition, purple-discolored leaf tissues were observed and compared to normal leaves using a transmission electronic microscope (TEM). Base on observations of the cell organelles, the purple-discolored uppermost leaves had many pigments formed in the epidermal cells unlike the normal middle leaves, and the cell wall thickness was twice as thick in the discolored leaves. The thickness of the thylakoid layer in the chloroplast the number of starch grains, the size of starch all decreased in the discolored leaves, while the number of plastoglobule and mitochondria increased.

Silybum marianum is an annual or biennial plant from the Asteraceae family. It can grow in low-nutrient soil and drought conditions, making it easy to cultivate. From the seed, a specialized plant metabolite called silymarin (flavonolignan complex) is produced and is known to alleviate the liver from hepatitis and toxins damages. To infer the phylogenetic placement of a Korean milk thistle, we conducted a chloroplast assembly and annotation following by a comparison with existing Chinese reference genome (NC_028027). The chloroplast genome structure was highly similar with an assembly size of 152,642 bp, an 153,202 bp for Korean and Chinese milk thistle respectively. Moreover, there were similarities at the gene level, coding sequence (n = 82), transfer RNA (n = 31) and ribosomal RNA (n = 4). From all coding sequences gene set, the phylogenetic tree inference placed the Korean cultivar into the milk thistle clade; corroborating the expected tree. Moreover, an investigation the tree based only on the ycf1 gene confirmed the same tree; suggesting that ycf1 gene is a potential marker for DNA barcoding and population diversity study in milk thistle genus. Overall, the provided data represents a valuable resource for population genomics and species-centered determination since several species have been reported in the Silybum genus.

Soybean (Glycine max (L.) Merr.) is a representative food resource. To preserve the integrity of soybean, it is necessary to protect soybean yield and seed quality from threats of various pests and diseases. Riptortus pedestris is a well-known insect pest that causes the greatest loss of soybean yield in South Korea. This pest not only directly reduces yields but also causes disorders and diseases in plant growth. Unfortunately, no resistant soybean resources have been reported. Therefore, it is necessary to identify the distribution and movement of Riptortus pedestris at an early stage to reduce the damage caused by insect pests. Conventionally, the human eye has performed the diagnosis of agronomic traits related to pest outbreaks. However, due to human vision's subjectivity and impermanence, it is time-consuming, requires the assistance of specialists, and is labor-intensive. Therefore, the responses and behavior patterns of Riptortus pedestris to the scent of mixture R were visualized with a 3D model through the perspective of artificial intelligence. The movement patterns of Riptortus pedestris was analyzed by using time-series image data. In addition, classification was performed through visual analysis based on a deep learning model. In the object tracking, implemented using the YOLO series model, the path of the movement of pests shows a negative reaction to a mixture Rina video scene. As a result of 3D modeling using the x, y, and z-axis of the tracked objects, 80% of the subjects showed behavioral patterns consistent with the treatment of mixture R. In addition, these studies are being conducted in the soybean field and it will be possible to preserve the yield of soybeans through the application of a pest control platform to the early stage of soybeans.

The present study was conducted to analyze agronomic characteristics of 8 ecotypes of Arundinella hirta (A. hirta) and the correlation among them. Changes in phenotypic characteristics of 8 ecotypes were measured at equal intervals of time from May to September. Among ecotypes, Jangsoo-1 has the highest plant height (172.33 cm), number of leaves (9.00) and leaf length (55 cm) while the ecotype Youngduk has the highest leaf width (1.57 cm), fresh mass (26.63 g), dry mass (7.06 g), number of spikelets per spike (53.33), amount of seeds per spike (0.74 g) and amount of seeds per 10 spikes (7.23 g). The ecotype Jinju-1 has the shortest plant height (119 cm) and leaf number (6.33), while Okgye-2 has shortest leaf length (30.67 cm), leaf width (0.93 cm), fresh mass (12.60 g), dry mass (3.30 g), spike length (30.33 cm), spikelet per spike (39.67), amount of seeds per spike (0.61 g) and amount of seeds per 10 spikes (6.00 g). Correlation coefficients were estimated among the studied agronomic characteristics which showed positive and significant association with each other. In the present study, the agronomic data collected would be useful to understand the potential of A. hirta as a forage resource and helpful in selecting the high-yielding genetic resource for future forage improvement.

Buckwheat (Fagopyrum esculentum), which is a traditional Korean crop, has been known as a health food due to its rich nutrition. This study was conducted to evaluate the change in flavonoid content of flowers and seeds during post-flowering growth of Korean tartary buckwheat variety 'Hwanggeummiso', with the aim of providing basic data for the development of functional food and feed additive. Tartary buckwheat took 69 and 99 days from the sowing date to reach the flowering and maturity stages, respectively. As a result of examining the flavonoid components of each part of tartary buckwheat, chlorogenic acid, rutin, and isoquercitrin of flowers increased from the flowering period on 22 May (0 days after flowering) to 42 days after flowering, while quercetin increased until 21 days after flowering and then decreased thereafter. In seeds, chlorogenic acid, rutin, and isoquercitrin were most abundant at the time of seed-bearing on 14 days after flowering, and showed a decreasing tendency thereafter. On the other hand, quercetin showed a tendency to increase until 21 days after flowering and then decrease. Overall, the flavonoid content was higher in flowers than in seeds, with rutin being particularly prominent. Based on this, the possibility as food materials and feed additives was confirmed using buckwheat produced in Korea.

Water dropwort (Oenanthe stolonifera DC.) is a perennial herbaceous plant that grows wild throughout Korea. As of 2020, 24,819 tons (51.7% of open field, 48.3% of facility) are produced in 1,278 ha (59.9% of open field, 40.1% of facility) nationwide. Water dropwort, which is rich in nutrients such as vitamins and iron, is mainly cultivated by vegetative propagation method using local traditional species, however, seed propagation and breeding of cultivars are insufficient so far. Since securing, propagating and continuous characterization of various genetic resources are required to breed new cultivars, this study was conducted to compare the main characteristics of domestic genetic resources and to improve their utilization. Growth characteristics such as plant height, fresh weight, plant type, and flowering date were investigated for the 89 varieties of genetic resources owned by Jeollabuk-do Agricultural Research and Extension Services after vegetative propagation in the individual pots. Also, the morphological image information of leaves and flowers was constructed. Genetic resources were collected nationwide and originated in 30 regions including Hwaseong, Siheung and Jeju. Their plant types could be classified into straight, intermediate and creeping types according to their morphological characteristics, and at this time, the number of intermediate types accounted for the largest proportion. Flowering was carried out under high-temperature and long-day conditions in summer. According to the flowering date, they could be classified into early, middle and late flowering varieties, and at this time, the middle flowering varieties occupied the largest proportion. As a result of the investigation of vegetative growth characteristics, varieties with long plant height, heavy fresh weight and thick stem thickness were identified. Along with this result, it is thought that classification and selection of genetic resources for various purposes will be possible through additional investigations such as analysis of components and antioxidant activity. Moreover, it is judged that such results can be used as basic data for breeding new water dropwort cultivars in the future.
Objective: The role of preoperative overt hepatic encephalopathy (OHE) in the neurophysiological mechanism of cognitive improvement after liver transplantation (LT) remains elusive. This study aimed to explore changes in sub-regional thalamic functional connectivity (FC) after LT and their relationship with neuropsychological improvement using resting-state functional MRI (rs-fMRI) data in cirrhotic patients with and without a history of OHE. Materials and Methods: A total of 51 cirrhotic patients, divided into the OHE group (n = 21) and no-OHE group (n = 30), and 30 healthy controls were enrolled in this prospective study. Each patient underwent rs-fMRI before and 1 month after LT. Using 16 bilateral thalamic subregions as seeds, we conducted a seed-to-voxel FC analysis to compare the thalamic FC alterations before and after LT between the OHE and no-OHE groups, as well as differences in FC between the two groups of cirrhotic patients and the control group. Correction for multiple comparisons was conducted using the false discovery rate (p < 0.05). Results: We found abnormally increased FC between the thalamic sub-region and prefrontal cortex, as well as an abnormally decreased FC between the bilateral thalamus in both OHE and no-OHE cirrhotic patients before LT, which returned to normal levels after LT. Compared with the no-OHE group, the OHE group exhibited more extensive abnormalities prior to LT, and the increased FC between the right thalamic subregions and right inferior parietal lobe was markedly reduced to normal levels after LT. Conclusion: The renormalization of FC in the cortico-thalamic loop might be a neuro-substrate for the recovery of cognitive function after LT in cirrhotic patients. In addition, hyperconnectivity between thalamic subregions and the inferior parietal lobe might be an important feature of OHE. Changes in FC in the thalamus might be used as potential biomarkers for recovery of cognitive function after LT in cirrhotic patients.
Alluvial beds are intimately associated with electrical properties related to soil types, including clay mineral content, porosity, and water content. The hydraulic property governs water movement and storage in alluvial beds. This study revealed electrical resistivity and hydraulic properties in space and time in relation to the hydrogeological data, groundwater pumping, and rainfall infiltration into the alluvial bed located in Daesan-myeon, Changwon City. An electrical resistivity survey with electrode spacings of 2 and 4m using a dipole-dipole array indicates that electrical resistivity changes in the alluvial bed depend on groundwater pumping and rainfall events. Additionally, rainfall infiltration varies with hydraulic conductivity in the shallow zone of the alluvial bed. The 2 m electrode spacing survey confirms that electrical resistivity values decrease at shallow depths, corresponding with rainfall and increased water content in the soil, indicating rainfall infiltration approximately 1-2 m below the land surface. The 4m electrode spacing survey reveals that hydraulic conductivity (K) values and electrical resistivity (ρ) values display an inverse relationship from the surface to the water table (approximately 9 m) and at deeper levels than the water table. Notably, ρ values are impacted by pumping around the depth of the water table at 9 m. This study suggests that time-lapsed electrical resistivity surveys in space and time could be effective tools for detecting the impact of rainfall and pumping, as well as hydraulic conductivity in shallow alluvial beds.

Ecosystem-disturbing plant species pose a significant threat to native ecosystems due to their high reproductive capacity, making it essential to monitor their distribution and develop effective mitigation strategies. Consequently, it is crucial to enhance the evaluation of the impacts of these species in environmental impact assessments by incorporating scientific evidence alongside qualitative assessments. This study introduces a dispersal model into the species distribution model to simulate the potential spread of ecosystem-disturbing plant species, reflecting their ecological characteristics. Additionally, we developed mitigation scenarios and quantitatively calculated reduction rates to propose effective mitigation strategies. The species distribution model showed a reliable AUC (Area Under the Curve) of at least 0.890. The dispersal model's results were also credible, with 31 out of 34 validation coordinates falling within the predicted spread range. Simulating the impact of the spread of ecosystem-disturbing plant species over the next five years revealed that one project site had potential habitats for Ambrosia artemisiifolia, necessitating robust mitigation measures such as seed removal. Another project site, with potential habitats for Symphyotrichum pilosum, indicated that physical removal methods within the site were effective due to the species' relatively short dispersal distance. These findings can serve as fundamental data for project executors and reviewers in evaluating the impact of the spread of ecosystem-disturbing plant species during the planning stages of projects.
