Selection of Early and Late Flowering Robinia pseudoacacia from Domesticated and Introduced Cultivars in Korea and Prediction of Flowering Period by Accumulated Temperature

  • The objectives of this study were to select early, late, and abundant flowering trees of black locust from domesticated and introduced cultivars, and prediction of flowering period by calculation of accumulated temperature in spring. Four cultivars (Debreceni-2, Pusztavacs, Jaszkiseri, and Rozsaszin AC) from Hungary and a cultivar from Beijing, China, were introduced, propagated by seed and planted in a seed orchard. For domesticated black locust, 63 cultivars from 10 locations throughout the country were selected and propagated by root cutting. Criteria for selection of domesticated cultivars were abundant flowering, long flowering period, or abundant nectar production with, if possible, straight stems. Accumulated temperature was calculated from data of a nearby weather station by accumulating daily maximum temperature minus 5 degree Celsius from January 1 up to the date reaching 880 degrees. Daily mean temperature was also used to calculate accumulated temperature up to the date reaching 450 degrees. The percentages of two-year and three-year-old flowering trees propagated by root cutting were higher than that of trees propagated by seeds, while four-year-old trees all flowered regardless of propagation methods. Among the domesticated cultivars, all the cultivars from Ganghwa showed abundant flowering with highest nectar production of 6.5 ul per flower, which was 100% more than other domesticated cultivars and 50% more than Debreceni-2 cultivar with highest nectar production among the introduced cultivars from Hungary. At the end of the eight years of observations, two trees of Debreceni-2 cultivars and a tree from Beijing, China were selected for early flowering trees which flowered 2 to 3 days earlier than average trees, while a tree of Debeceni-2 and three trees from Bejing were selected for late flowering trees which flowered 2 to 3 days later than average trees. It is possible to extend the flowering period of black locust by 4 to 6 days by planting early and late flowering cultivars together. Abundant flowering trees were unable to be selected due to severe damages by leaf gall midges which killed many trees and reduced the crown size of the remaining trees in the seed orchard, and which were first found in Korea in 2001 and now damaging most of the black locust forests in Korea. The prediction of flowering period by accumulated temperature indicated that black locust flowered to a peak when accumulated daily maximum temperature reached 880 degrees Celsius, and when daily mean temperature reached 450 degrees.

Phylogenetic Relationship of Araliaceae in Korea by Seed Morphological Characteristics (종자 외부형태학적 특성에 의한 한국산 두릅나무과(Araliaceae) 식물의 종간 유연관계)

  • The purpose of this study is to describe and compare the morphological characteristics of seeds of 12 taxa of Araliaceae, and to evaluate their possible use for taxonomic considerations. For light microscope observations and measurements, the seeds were observed using Image Analyzing System. The observations were made on twenty randomly selected seeds of each species. Obtained data were statistically processed using analysis of variance. Principal Components Analysis indicated four groups of characters, the genera Aralia, Eleutherococcus, Panax and the others, which explained 65.47% of the total variation. As a results of Cluster Analysis using the eleven variables, 12 species of Araliaceae were also discriminated into four groups. Eleutherococcus senticosus and E. gracilistylus were closely related, which is well supported by the results from recent molecular studies. Also, the genera Dendropanax and Eleutherococcus were closely related in terms of seed characters.

Comparison of the Characteristics of Seed Germination and the First Stage of Growth in Cynanchum wilfordii (Maxim.) by Different Light Conditions (광조건별 백수오의 종자 발아 특성 및 초기생육 비교)

  • This study was carried out to compare the characteristics of seed germination and the first stage of growth in Cynanchum wilfordii (Maxim.) by different light conditions such ass fluorescent light, dark and light emitting diode (LED). There was not much difference from the germination rate of C. wilfordii (Maxim.) in different light conditions for 10 days. The highest of germination rate occurred over 91.11 percentage by LED red condition at $26^{\circ}C$. Also mean germination velocity and promptness index were represented the highest results of 2.3 ea/day and 52.67 under red light, respectively. But mean germination time under both LED red and blue light was represented low data that it takes over three days for germinating of seed. Even though handling both LED red and blue lights, the germination uniformity was not constant. These results may help our understanding of variations in germination characteristics for C. wilfordii (Maxim.) seeds treated under different light conditions.

Effect of Irradiation on Growth, Bulblet Formation, and Germination of Pollen and Seed of Several Lily Cultivars (나리 생장, 자구 발달 및 화분과 종자발아에 미치는 방사선의 영향)

  • To build up data for mutation breeding of lily, sensitivity to irradiation of bulb, seed and pollen was investigated. The bulbs of seven cultivars including 'New Wave' were very sensitive to gamma rays. Non-irradiated bulbs produced plants with bloom as normal. However, irradiated bulbs showed retarded growth and did not flower. Bulbs of 'New Wave' and 'Tiny Dino' irradiated with 125Gy gamma rays survived, but their growth rate dropped considerably. Part or whole leaves of the other cultivars except 'New Wave' and 'Tiny Dino' withered. Moisture content of individual scale was 72~78% depending on cultivar. Bulblet formation from untreated scales was 100%. Among all irradiated scales only 'Siberia' scales showed 8.5% bulblet formation when irradiated with 50Gy gamma rays. Pollen germination as affected by gamma ray irradiation had no consistent tendency. When dosage of $F_1$ 'Augusta' seeds reached 300Gy, their survival ratio was drastically decreased. Bulblet formation in vitro was observed only in non-irradiated seeds. Lethal dose ($LD_{50}$) of seed was less than 100Gy. Height and width of $F_1$ 'Augusta' bulblets in vitro irradiated with gamma rays decreased as dosage increased regardless of the culture method. The percentage shoot formation was 100% in non-irradiated bulblets and 15~60% in irradiated ones.

Single Nucleotide Polymorphism (SNP) Discovery and Kompetitive Allele-Specific PCR (KASP) Marker Development with Korean Japonica Rice Varieties

  • Genome resequencing by next-generation sequencing technology can reveal numerous single nucleotide polymorphisms (SNPs) within a closely-related cultivar group, which would enable the development of sufficient SNP markers for mapping and the identification of useful genes present in the cultivar group. We analyzed genome sequence data from 13 Korean japonica rice varieties and discovered 740,566 SNPs. The SNPs were distributed at 100-kbp intervals throughout the rice genome, although the SNP density was uneven among the chromosomes. Of the 740,566 SNPs, 1,014 SNP sites were selected on the basis of polymorphism information content (PIC) value higher than 0.4 per 200-kbp interval, and 506 of these SNPs were converted to Kompetitive Allele-Specific PCR (KASP) markers. The 506 KASP markers were tested for genotyping with the 13 sequenced Korean japonica rice varieties, and polymorphisms were detected in 400 KASP markers (79.1%) which would be suitable for genetic analysis and molecular breeding. Additionally, a genetic map comprising 205 KASP markers was successfully constructed with 188 $F_2$ progenies derived from a cross between the varieties, Junam and Nampyeong. In a phylogenetic analysis with 81 KASP markers, 13 Korean japonica varieties showed close genetic relationships and were divided into three groups. More KASP markers are being developed and these markers will be utilized in gene mapping, quantitative trait locus (QTL) analysis, marker-assisted selection and other strategies relevant to crop improvement.

Current Status and Direction of Weed Management According to Cropping Systems (작부체계에 따른 잡초관리 연구 동향과 방향)

  • The present study was conducted to propose future research directions for weed management by examining the current trends of research on weed occurrence according to cropping systems. The cropping systems are developed for the efficient use of arable land, and the weed flora changes according to the management practices of a given cropping system. In particular, weed occurrence can be reduced by altering the soil environment. In addition, cultivation methods, such as tillage, affect the weed seed bank in the soil, thus altering the pattern of weed occurrence. Here, we propose three weed management practices according to the cropping system. First, it is necessary to develop a model that can classify weed species by analyzing young seedlings and can predict the flora in the field. Second, it is important to manage the cropping system history and establish a database of agricultural information, which can be linked to meteorological and geographic data. Third, it is critical to estimate the weed occurrence and soil seed bank dynamics, based on which a cropping system platform and digitalization technology can be developed. In the future, the prediction of weed occurrence and control according to the cropping system will contribute to sustainable agriculture by reducing the use of herbicides and solving the problems of resistant weeds.

Protein of Major Germplasms for High-Protein Wheat Breeding (밀 주요 교배모본의 단백질 특성)

  • Studies to obtain a fundamental data to improve the quantity and quality of wheat protein and to select the excellent breeding materials suitable for various uses of it were performed on the focuses of protein content, amino acid composition, and its related quality. The range of seed protein content of crossing block consisted with 319 varieties or lines was from 9.9% to 17.6% with the average of 13.2%. The minimum protein content was showed by Wonkwang, and the maximum by Chungnamjaerae and Suwon 210. Among the Korean native varieties or breeding lines of crossing block, at least twelve lines or varieties were selected as the high-protein wheats. In the effects of environmental conditions, the variations of protein content by cultivating locations and year were showed out to be up to 3.6%, and 3.3%, respectively. But the varietal characteristics of wheat protein were seemed to remain constantly. In the results of amino acid analysis, those related with bread quality, except proline were improved with the increase of seed protein. But the improvement of nutritional quality did not agree with the protein content increase of seed. The total content of amino acid essential to adults was very low with 266.6-273.3mg per one gram of protein compare to the 350mg (free from the amino acid of tryptophan) recommended by FAO/WHO. Especially the limiting amino acid such as methionine, lysine and threonine were critically deficient. The correlation between protein contents and sedimentation values was obtained r=0.656$\^$**/ of average notwithstanding widly different environmental conditions, and in some condition the value was obained about to r=0.956$\^$**/.

  • Korean perilla (Perilla frutescens Britten var. Japonica Hara) has been used in various ways in food since before the Unified Silla period, and it has not been systematically studied due to its long cultivation history. However, the global perilla oil market is expected to reach $1,854.55 million by 2031, up from $981.6 million in 2023, with a CAGR of 10.6%. In the bio industry, bio cosmetics refer to cosmetics that contain natural ingredients based on biotechnology, but there is still no clear academic definition. As consumers' interest has recently focused on raw materials and ingredients, interest in substances based on natural substances that enhance skin metabolism is increasing. Despite the importance of natural extracts produced domestically, there has not been much research on domestic perilla seed as a cosmetic raw material and material. This study was conducted based on previous papers and literature studies conducted over the past five years on perilla seed after oil extraction, and it was found that perilla seed contains a large amount of phenolic compounds with excellent radical scavenging ability, and thus it was possible to find out the antioxidant and anti-inflammatory effects, whitening effects, and anti-obesity effects of skin beauty. Therefore, this study is expected to be used as basic data for various studies on perilla, a natural extract, as a bio-industry cosmetic material.

Use of Microsatellite Markers Derived from Genomic and Expressed Sequence Tag (EST) Data to Identify Commercial Watermelon Cultivars (수박 시판 품종의 식별을 위한 Genomic과 Expressed Sequence Tag (EST)에서 유래된 Microsatellite Marker의 이용)

  • This study was carried out to construct a DNA profile database for 102 watermelon cultivars through the comparison of polymorphism level and genetic relatedness using genomic microsatellite (gMS) and expressed sequence tag (EST)-microsatellite (eMS) markers. Sixteen gMS and 10 eMS primers showed hyper-variability and were able to represent the genetic variation within 102 watermelon cultivars. With gMS markers, an average of 3.63 alleles per marker were detected with a polymorphism information content (PIC) value of 0.479, whereas with eMS markers, the average number of alleles per marker was 2.50 and the PIC value was 0.425, indicating that eMS detects a lower polymorphism level compared to gMS. Cluster analysis and Jaccard's genetic distance coefficients using the unweighted pair group method with arithmetic average (UPGMA) based on the gMS, eMS, and combined data sets showed that 102 commercial watermelon cultivars could be categorized into 6 to 8 major groups corresponding to phenotypic traits. Moreover, this method was sufficient to identify 78 out of 102 cultivars. Correlation analysis with Mantel tests for those clusters using 3 data sets showed high correlation ($r{\geq}0.80$). Therefore, the microsatellite markers used in this study may serve as a useful tool for germplasm evaluation, genetic purity assessment, and fingerprinting of watermelon cultivars.

Study on the Habitat Environment of Sulf Clam, Tresus Keenae (왕우럭조개의 서식환경에 관한 연구)

  • For the aquaculture industrialization of surf clam (Tresus keenae), it is important to basic data on the marine environment of the habitat of surf clam (T. keenae). In this study, we investigated the marine environment of habitat of surf clam (T. keenae) and sought to basic data for the preparation of surf clam (T. keenae) for artificial seed production. The water temperature of the habitat of surf clam (T. keenae) was the lowest in winter and appeared high in summer. The salt concentration showed it range from 31.2 to 33.9 psu. The pH showed it range from 7.69 to 8.70, with high pH in winter and low pH in summer. The dissolved oxygen(DO) was showed it range from 6.20 to 10.24 mg / L and the autumn was relatively higher than the spring and winter. The species composition of phytoplankton was about 30 to 40 species, and most of them were diatoms. The abundance of seasonal phytoplankton showed it range from 23.5 to 61.3 cells / ml, showing seasonal differences. The expression of dominant species also showed a difference depending on the season. As for the particle size composition of the sediment, sandy silt was the most distributed. Flow velocities appeared at 50-80 cm / s in the southeast direction at ebb tide and at 60-100 cm / s in the northwest direction at flood tide. The results of this study can be used as basic data for providing knowledge about the habitat and marine environment of surf clam (T. keenae) and for studying shellfish that inhabit the sedimentary layer.
