Phylogenetic implication of seed coat sculpturing in subtribe Agrimoniinae (Rosaceae) (장미과 짚신나물아족 종피형태의 계통분류학적 고찰)

  • Seed morphology in the subtribe Agrimoniinae (Rosaceae) was examined using scanning electron microscopy to identify distinct characters and evaluate their evolution in a phylogenetic framework for five genera in the subtribe: Agrimonia L., Aremonia Neck. ex Nestl., Hagenia J.F. Gmel., Leucosidea Eckl. & Zeyh., and Spenceria Trimen. All genera have one or two mature achenes in a fruiting hypanthium. In the seed coats, the cell shape, size, wall features, and sculpturing vary across genera. Of most significance is the presence of papillae structures in both Agrimonia and Aremonia. Through the mapping of papillae features onto phylogenetic trees, either one or two changes in seed coats are hypothesized. The phylogenetic tree inferred from four nuclear and six chloroplast regions of sequence data suggests that at least two steps of papillae sculpturing on seed coats are required. On the other hand, in the phylogenetic tree of a low-copy nuclear gene, one independent evolutionary step is postulated to explain the current character states. In the latter hypothesis, the seed coat sculpturing also supports a monophyletic relationship for cosmopolitan Agrimonia and European endemic Aremonia. The seed coat sculpturing provides valuable information for inferring phylogenetic relationships at the generic level in the subtribe Agrimoniinae.

Effect of Safflower and Safflower Seed Extract on Osteogenic Differentiation of MC3T3E1 Cells (홍화, 홍화씨 추출물이 MC3T3E1 세포의 골분화 과정에 미치는 영향)

  • Objectives This study investigated the effect of purified safflower (Carthamus tinctorius Linne) and safflower seed (Carthamus tinctorius L. seed; CS) extract, using hot water and ethanol extract methods , on the osteogenic differentiation of MC3T3E1 cells.Methods The safflower and safflower seed were extracted with hot water and ethanol. The samples were concentrated by a rotary evaporator and then freeze-dried using a freeze-dryer. The MC3T3E1 cells were propagated and maintained in DMEM (Gibco) containing 10% FBS and a 1% antibiotic antimycotic solution. To induce osteogenic differentiation, the cells were treated for 14 days with DMEM with 10 mM β-glycerophosphate and 50 μM ascorbic acid. Extract doses were confirmed by the results of an MTT assay, and treatment of the extracts was performed in a differentiation medium every two days. The ALP staining and activity were tested after osteogenic differentiation for five days, and after 14 days, osteogenic differentiation was determined by alizarin red S staining. The mRNA expressions of osteogenic-related genes were quantified using quantitative real-time PCR.Results In the results of the MTT assay, all concentrations of safflower extracts had no toxicity in the MC3T3El cells. But in the groups of 100 ng/ml and 200 ng/ml concentrations of safflower seed extracts, the cell viability was significantly reduced by up to 40-50%. So we fixed the treatment concentration of the extract at 50 ng/ml. In the ALP and alizarin red S staining, all extract groups increased osteogenic differentiation compared with the control group. The water-safflower extract group showed the highest mRNA level of Alp, Runx2, and Dlx5 genes. The mRNA level of Ocn, an osteogenic gene related to late-stage differentiation, in the ethanol-safflower extract group increased the mineralization more significantly than in other groups.Conclusions These data suggest that the extract of safflower increases the osteoblastic differentiation activates of MC3T3E1 cells like the extract of safflower seed. The water-extract and ethanol-extract of safflower have effects on different stages of osteogenesis in MC3T3El. Not only safflower seed but also safflower will be useful therapeutic reagents for age-associated chronic diseases such as osteoporosis.

  • A disposable password is necessary to avoid any danger by the use of a static password and reinforce the user's authentication. In order to prevent personal information from being exposed, OTP generation algorithm is regarded as important. The OTP generation algorithm we suggest in this thesis generates 256-bit-size OTP Data by using Seed value and Time value. This value that the generated OTP Data are arranged with a matrix and a 32-bit-value is extracted on an irregular basis becomes the final value. We can find out that the more OTP generation frequency we have, the lower probability of clash tolerance we get in our suggested algorithm, compared to the previous algorithm.

  • The need for information security increases interests on cipher algorithms recently. Especially, a large volume of data transmission over high-band communication network requires faster encryption and decryption techniques for real-time processing. It would be a good solution for this problem that we implement the cipher algorithm in forms of hardware circuits. Though some previous researches use this approach, they focus only on repeatedly executing the core part of the algorithm to minimize the hardware chip size, while most cipher algorithms are inherently parallel. In this paper, we propose a new design for the SEED block cipher algorithm developed by KISA (Korea Information Security Agency) in 1998 as Korean standard cipher algorithm. It exploits the parallelism of the algorithm basically and implements it in a pipelined fashion. We described the design in VHDL program and performed functional simulations on the program, and then found that it worked correctly. In addition, we synthesized it and verified that it could be implemented in a single FPGA chip, implying that the new design can be Practically used for the actual hardware implementation of a high-speed and high-performance cipher system.

The Safety and Pharmacokinetics of Cyanidin-3-Glucoside after 2-Week Administration of Black Bean Seed Coat Extract in Healthy Subjects

  • We analyzed the pharmacokinetics of C3G on data from twelve subjects, after 2-week multiple dosing of black bean (Phaseolus vulgaris, Cheongjakong-3-ho) seed coat extract, using the mixed effect analysis method (NONMEM, Ver. 6.2), as well as the conventional non-compartmental method. We also examined the safety and tolerability. The PK analysis used plasma concentrations of the C3G on day 1 and 14. There was no observed accumulation of C3G after 2-week multiple dosing of black bean seed coat extract. The typical point estimates of PK were CL (clearance)=3,420 l/h, V (volume)=7,280 L, Ka (absorption constant)=9.94 $h^{-1}$, ALAG (lag time)=0.217 h. The black bean seed coat extract was well tolerated and there were no serious adverse events. In this study, we confirmed that a significant amount of C3G was absorbed in human after given the black bean seed coat extract.

Systematic Studies of 12S Seed Storage Protein Accumulation and Degradation Patterns during Arabidopsis Seed Maturation and Early Seedling Germination Stages

  • Seed storage proteins (SSPs) are important for seed germination and early seedling growth. We studied the accumulation and degradation profiles of four major Arabidopsis 12S SSPs using a 2-DE scheme combined with mass spectrometric methods. On the 2-DE map of 23 dpa (days post anthesis) siliques, 48 protein spots were identified as putative full-length or partial $\alpha$, $\delta$ subunits. Only 9 of them were found in 12 dpa siliques with none in younger than 8 dpa siliques, indicating that the accumulation of 12S SSPs started after the completion of cell elongation processes both in siliques and in developing seeds. The length and strength of transcription activity for each gene determined the final contents of respective SSP. At the beginning of imbibition, 68 SSP spots were identified while only 2 spots were found at the end of the 4 d germination period, with $\alpha$, subunits degraded more rapidly than the $\alpha$ subunits. The CRC $\delta$ subunit was found to degrade from its C-terminus with conserved sequence motifs. Our data provide an important basis for understanding the nutritional value of developing plant seeds and may serve as a useful platform for other species.

Position Effects on Abortion of Reproductive Characters in Vicia cracca (Leguminosae) (등갈퀴나물 ( Vicia cracca : Leguminosae ) 번식기관의 발육정지에 대한 위치효과)

  • The position effects on the abortion of flowers, fruits, and ovules were examined in relation to the resource limitation hypothesis using a temperate legume species, Vicia cracca. Fifty plants were randomly sampled from a natural population located at the Belmont Conservation Site in Belmont, MA, USA. Due to indeterminant growth and acropetal flowering of this species, the position of flower or fruit on the plant was able to be coded by the combination of architectural levels such as plant half (lower and upper half) and inflorescence half (lower and upper half) within each plant half. Overall, only 17.3% of the flowers on an inflorescence began to develop into fruits, while overall 36.0% of the fruits initiated failed to mature. Consequently, 11.5% of the flowers successfully matured into fruits. The mean flower number per inflorescence differed significantly among plants as well as between plant halves. Although the probability of fruit initiation was higher in the lower halves of both plant and inflorescence than in the upper halves of the latter, fruit abortion rate differed significantly only between plant halves. The overall mean seed set was 33.6%, implying that about two out of six ovules in a fruit matured into seeds. The seed set was independent of both plant identity and architectural level unlike other reproductive characters examined. When the ovule developmental stage was examined on data pooled over all fruits, the proportion of ovules in varying developmental stages decreased in order of early abortion, seed maturation, late abortion, and seed predation (48.4, 34.9, 12.2, and 4.5%, respectively). A within-fruit ovule position was also used as a class variable for the analysis of position effects on ovule development. All architectural levels considered exerted singificant influence over the ovule development. In particular, ovule development was strongly affected by the within-fruit ovule position. Ovules in both extreme ends within a fruit tended to abort early, while those in the middle position were more likely to mature into seeds. The strong position effects detected from the flowering to seed maturation stage were interpreted as an indication of competitive interaction among reproductive characters which are largely constrained by plant architecture.

Evaluation of Estrogenic Activity of Pumpkin Seed Extract using Recombinant Yeast Assay (재조합 효모를 이용한 호박씨 추출물의 에스트로겐 활성 평가)

  • The aim of this study is to evaluate the estrogenic activity of Cucurbita pepo seed extract which includes ${\beta}-sitosterol$ and other phytosterols. Sample was extracted from Cucurbita pepo seed by supercritical carbondioxide method and resuspended with ethanol. Estrogenic activity was measured by recombinant yeast assay which detects estrogenic activity using recombinant yeast with high level of estrogenic receptor. However, estrogenic activity of pumpkin seed extract was not found in this study. Based on this data, pumpkin seed extract will not cause estrogenic disturbance.

Prosopis juliflora invasion and environmental factors on density of soil seed bank in Afar Region, Northeast Ethiopia

  • The aims of the study were to analyze (1) the effects of Prosopis juliflora (Prosopis) on the spatial distribution and soil seed banks (SSB) diversity and density, (2) the effects of environmental factors on SSB diversity and density (number of seeds in the soil per unit area), and (3) the effects of animal fecal droppings on SSB diversity, density, and dispersal. Aboveground vegetation data were collected from different Prosopis-infested habitats from quadrats (20 × 20 m) in Prosopis thickets, Prosopis + native species stand, non-invaded woodlands, and open grazing lands. In each Prosopis-infested habitats, soil samples were collected from the litter layer and three successive soil layer, i.e., 0-3 cm, 3-6 cm, and 6-9 cm. Seeds from soil samples and animal fecal matter were separated in the green house using the seedling emergence technique. Invasion of Prosopis had significant effects on the soil seed bank diversity. Results revealed that the mean value of the Shannon diversity of non-invaded woodlands was being higher by 19.2%, 18.5%, and 11.0% than Prosopis thickets; Prosopis + native species stand and open grazing lands, respectively. The seed diversity and richness, recovered from 6-9-cm-deep layer were the highest. On the other hand, the density of Prosopis seeds was the highest in the litter layer. About 156 of seeds/kg (92.9%) of seeds were germinated from cattle fecal matter. However, in a small proportion of seedlings, 12 of seeds/kg (7.1%) were germinated from shot fecal matter. Thus, as the seeds in the soil were low in the study areas, in situ and ex situ conservation of original plants and reseeding of persistent grass species such as Cynodon dactylon, Cenchrus ciliaris, Chrysopogon plumulosus, and Brachiaria ramosa are recommended.

Effects of Feeding the Mixture of Lineseed and Sunflower Seed Oil on the Lipid Components and Fatty Acid Compositions of Liver in Dietary Hyperlipidemic Rats (아마인유와 해바라기 종자유의 혼합급이가 식이성 고지혈증 흰쥐의 간장 지질성분 및 지방산 조성에 미치는 영향)

  • This study was carried out to investigate the effect of the feeding mixture of linesed oil, rich in n-3 PUFA and the sunflower seed oil, rich in n-6 PUFA on the lipid metabolism in the dietary hypprlidemic rats. After male Sprague-Dawley rats were induced hyperlipidemia by feeding the diet containing lard, butter, and cholesterol for 3 weeks, then they were fed with the diet containing lard 3.0% and butter 12.0% for control, the mixture in different proportion of both linseed oil and sunflower seed oil, and antihyperlipidemic durgs for 2 weeks. Analysis of the lipid component and the fatty acid composition of the liver showed following results. Concentration s of the total cholesterol and phospholipid in liver were significantly higher in group 2 (olive oil 12.0%) and lower in the other groups than in the control group, especially lower in groups 3 (cholestyramine 2.0%) and 9 (sunflower seed oil 12.0%) . Concentration of triglyceride was lower in the other groups except group 4 (liparoid), especially lowe rin group 9 than in the control group. In the fatty acid composition of liver lipids, C18:2 was the major fatty acid. Contents of n-6 PUFA increased , while those of n-3 PUFA decreased in groups composition of the test lipids. From the data on concentration s of total cholesterol. Phospholipid and triglyceride in liver, we concluded that the feeding mixed with 3.0% lard and 12.0 % sunflower seed oil were most effective for the improvement of the live lipids. The fatty acid composition in liver lipids were affected by the fatty acid composition of the test lipids.
