LPS로 유도된 RAW264.7세포주에서 황금뿌리 물추출물의 항염증활성 (Anti-inflammatory activity of Scutellaria Baicalensis root extract in lipopolysaccharide-induced RAW 264.7 cells)

  • Scutellaria baicalensis water extract (SWE)는 지질 다당류 LPS로 유도된 RAW 264.7 세포에서 NO 및 전 염증성 사이토카인인 TNF-α의 생성을 세포 독성을 유발하지 않고 유의하게 억제하였다. 또한, SWE는 iNOS 및 COX-2의 단백질발현을 농도의존적으로 감소시켰으며, ERK, JNK, p38과 같은 MAPKs 계열의 인산화 발현 수준을 조사한 결과 JNK와 p38의 발현 수준을 감소시켰다. 이는 SWE가 p38 인산화를 억제함으로써 iNOS, COX, 그리고 TNF-α와 같은 전 염증성 사이토카인의 발현을 감소시키며 결론적으로 NO의 생성을 억제시킨다는 결과를 도출할 수 있었다. 본 연구는 항염증 효능 검증뿐 아니라 염증대사기전의 주요인자를 탐색함으로써 황금의 기능성 소재로써의 가능성을 시사한다.

  • Candida 바이오필름은 숙주조직과 의료기기의 표면에 자라는 자가-조직화된 미생물의 군락으로 전통적인 항진균제에 대한 저항성이 높게 나타난다. 황금(Scutellaria baicalensis)의 뿌리는 극동지방에서 의료용 목적으로 널리 사용되어 왔다. 본 연구의 목적은 10 C. albicans 임상 분리균주에 의해 형성된 바이오필름에 대한 황금의 수용성 추출물의 효과를 평가하고, 항바이오필름 활성에 대한 메커니즘을 알아보는 것이다. 바이오필름에 대한 효과는 XTT 환원분석법을 사용하였으며, 조사된 모든 균주에 대한 대사활성은 MIC에서 유의하게 감소($57.7{\pm}17.3$%)하였다. 황금추출물은 (1,3)-${\beta}$-D-글루칸 합성효소의 활성을 저해하였고 C. albicans의 형태에 대한 황금의 효과는 글루칸 합성의 억제로 인한 생장의 변화와 관련이 있었다: 대부분의 세포는 둥글고 팽창되었으며 세포벽이 진하게 염색되거나 파열되었다. 항캔디다 활성은 살진균성이었고, 황금은 C. albicans를 $G_0/G_1$기에 머물게 했다. 데이터는 황금이 목표가 되는 균류에 다중의 치명적인 효과를 내며, (1,3)-${\beta}$-D-글루칸 합성효소의 활성을 저해함을 통해 궁극적으로는 세포벽의 파열과 죽음에 이르게 한다는 것을 나타낸다. 따라서 황금은 바이오필름과 관련된 캔디다의 감염을 치료하고 제거하기 위한 항진균제 개발 후보 물질로서의 가능성을 가진다.

An Optimization of Flavonoid Production from the Suspension Culture of Scutellaria baicalensis Georgi Cells

  • Flavonoid production by suspended cells of Scutellaria baicalensis Georgi was studied and the medium was optimized for cell growth and baicalin production. In SH medium the flavonoid production was not closely associated with the cell growth. A modified SH medium, FPM, was therefore designed for enhanced baicalin production. In FPM, both cell growth and baicalin production were increased by 1.5 times and 1.67 times than in the original SH medium, respectively. The increases could be attributed to the increased metabolic activities involved in the flavonoid biosynthesis as represented by enhanced activities of phenylalanine ammonia-lyase.

  • Objectives : The present study was designed to investigate corelation between mix proportion of Scutellaria baicalensis (SB) and Coptis chinensis (CC) on lipopolysaccharide (LPS)-induced TYPE-1 interferon production. Methods : I examined TYPE-1 interferon, interferon regulating factor (IRF)-1,7 and interleukin(IL)-10 production on LPS-induced RAW264.7 cells to evaluate inhibitory effect of mix proportion of SB and CC using real time PCR. Results : Mixture of SB and CC regulated TYPE-1 interferon and IRF-1,7 mRNA expression with SB dose dependent manner, while maintained IL-10 mRNA expression on LPS-induced RAW264.7 cells. Conclusion : In mixture of SB and CC, SB plays a key role in reducing TYPE-1 interferon through inactivation IRF-1,7. Furthermore mixture of SB and CC maintained IL-10 mRNA level. Collectively, this results suggest that SB confer beneficial effects in autoimmune diseases clinically.

황금의 세포배양에 의한 Baicalin 생산 연구 (Production of Baicalin by Cell Culture of Scutellaria baicalensis)

  • Callus was derived from the leaves of Scutellaria baicalensis Georgi. The productions of baicalin in the cultivated callus under various conditions, especially, the effects of the light sources and temperature were studied. In this experiment, the callus cultivated at $25^{\circ}C$ showed higer production of baicalin than the callus cultivated at $21^{\circ}C$ and $23^{\circ}C$. The illumination of light(fluorescence and UV) accelerated generally the growth of callus and the production of baicalin during the cultivation for three weeks. But, the illumination of light more than three weeks reduced the rate of production of baicalin in the callus.

It has been used in traditional medicine formula from long time ago in the east Asia. It is constituted of the active flavone ingredients such as baicalin. baicalein. DTF(Di-methyl Tetra -hydroxy Flavone), wogonin. wogonoside. $\beta$- Sitosterol. etc.. General purified fractions of Scutellaria baicalensis has the high portion of the baicalin which has the problem of narrow antimicrobial spectrum and compatibility against cosmetic formula. Now. we has been develop the new purificaton process of Scutellaria baicalensis that has the high rate of DTF content, which is improved in antimicrobial activity and cosmetics compatibility. So. we have assure that it is the potent preservative against various cosmetic formula.

  • Objective : The present study aimed to evaluate the change of the content of 7 active components in decoctions produced by various proportional pairs of Coptis chinensis Franch and Scutellaria baicalensis Georgi in 'Mixed decoction (MD)' and 'Single decoction mixture (SDM)'. Methods : The samples of MDs were prepared by decocting C. chinensis : S. baicalensis with the ratios of 10 g:10 g, 10 g:20 g, and 20 g:10 g. Those of SDMs were prepared by blending each single decoction from C. chinensis and S. Baicalensis with the ratios of 1:1, 1:2, and 2:1. The samples were evaluated by high-performance liquid chromatography with statistical analyses. Results : The analytical methods, which were optimized and validated, were reliably applied to present research. The content of all components in both MDs and SDMs at C. chinensis : S. baicalensis = 1:1 ratio were reduced compared with single herb decoction. The components from each compositional herb in MDs were proportionally increased with the ratio of original herb increased, but inversely proportional to paired herb. The contents of components in MDs were significantly lower than those in SDMs at all ratios, except for high content of baicalin at C. chinensis : S. baicalensis = 2:1. Conclusion : It was concluded that MDs and SDMs as well as the proportions of herbs could affect the contents of the components from original herbal medicines. These results provide the information for the quality control of herbal medicine combined C. chinensis with S. baicalensis.

  • To overcome the limit of morphological method for classification of herbal drug, a novel method to discriminate its origin using pyrolysis mass spectrometry-multivariate analysis was developed. This method was applied successfully to Scutellaria baicalensis Georgi, one of the most popular herbal drug in oriental countries. The ethylacetate soluble fractions were prepared by sonication from pulverized roots of S. baicalensis which were collected from various regions including Korea and China, and subjected to direct insertion probe (DIP) mass spectrometry to achieve mass spectra of pyrolizates of extracts. The probe temperature was elevated from $30^{\circ}C$ to $320^{\circ}C$ at increasing rate $64^{\circ}C/min$, and the average mass spectrum calculated from total ion chromatography (TIC) was obtained. The relative peak intensities versus m/z were subjected to SAS program, and the training set (9 from Korea origin and 22 from China origin) was clustered two groups as its origin. In the test set, 11 samples among total 13 test sample were successfully classified according to their origin by developed method with accuracy of 85%.

Control of Feed Rate Using Neurocontroller Incorporated with Genetic Algorithm in Fed-Batch Cultivation of Scutellaria baicalensis Georgi

  • To enhance the production of flavonoids [baicalin, wogonin-7-Ο-glucuronic acid (GA)], which are secondary metabolites of Scutellaria baicalensis Georgi(G.) plant cells, a multilayer perceptron control system was applied to regulate the substrate feeding in a fed-batch cultivation. The optimal profile for the substrate feeding rate in a fed-batch culture of S. baicalensis G. was determined by simulating a kinetic model using a genetic algorithm. Process variable profiles were then prepared for the construction of a multilayer perceptron controller that included massive parallelism, trainability, and fault tolerance. An error back-propagation algorithm was applied to train the multiplayer perceptron. The experimental results showed that neurocontrol incorporated with a genetic algorithm improved the flavonoid production compared with a simple fuzzy logic control system. Furthermore, the specific production yield and flavonoid productivity also increased.
