Large-Scale Screening of the Plant Extracts for Antifungal Activity against the Plant Pathogenic Fungi

  • Plants produce chemicals of immense diversity that provide great opportunities for development of new antifungal compounds. In search for environment-friendly alternatives to the fungicide of current use, we screened plant extracts obtained from more than eight hundred plant materials collected in Korea for their antifungal activity against the model plant pathogenic fungus, Magnaporthe oryzae. This initial screening identified antifungal activities from the eleven plant extract samples, among which nine showed reproducibility in the follow-up screening. These nine samples were able to suppress not only M. oryzae but also other fungal pathogens. Interestingly, the plant extracts obtained from Actinostemma lobatum comprised five out of eight samples, and were the most effective in their antifungal activity. We found that butanol fraction of the A. lobatum extract is the most potent. Identification and characterization of antifungal substances in the A. lobatum extracts would provide the promising lead compounds for new fungicide.

Isolation and Structure Elucidation, Molecular Docking Studies of Screlotiumol from Soil Borne Fungi Screlotium rolfsii and their Reversal of Multidrug Resistance in Mouse Lymphoma Cells

  • A new compound namely (13-(3,3-dihydroxypropyl)-1,6-dihydroxy-3,4-dihydro-1H-isochromen-8(5H)-one (1) was isolated from an ethyl acetate extract of the borne fungi Screlotium rolfsii. Its chemical structure was elucidated by spectroscopic analysis. Screlotiumol 1 were evaluated for their effects on the reversion of multidrug resistant (MDR) mediated by P-glycoprotein (P-gp) of the soil borne fungi. The multidrug resistant P-glycoprotein is a target for chemotherapeutic drugs in cancer cells. In the present study rhodamine-123 exclusion screening test on human mdr1 gene transfected mouse gene transfected L5178 and L5178Y mouse T-cell lymphoma which showed excellent MDR reversing effect in a dose dependent manner against mouse T-lymphoma cell line. Moreover, molecular docking studies of compound-1 also showed better results as compared with the standard. Therefore the preliminary results obtained from this study suggest that screlotiumol 1 could be used as a potential agent for the treatment of cancer.

  • These studies were carried out to detect the presence of mycotoxin fungi and detoxification of toxins in various kinds of grain and foodstuffs in Korea. The experiments were divided into three parts: mycologic, toxicologic and electron microscopic study. The resuIts were summarized as follows: 1. From the 210 various, samples(local grains, 150 samples; rice-cakes, 50 samples; meju, 10 samples), 1,205 colonies or fungi were isolated. In 1,127 of 1,205 colonies, it was possible to identify 21 genera. Among the identified strains, the predominant genera were Aspergillus sp.(28.5%), Penicillium sp.(27.1%) and Mucor sp.(7.8%). 2. In cytotoxicity test on HeLa cells, 25 strains showed severe toxic effects among the 240 strains of experiments. 3. In histopathologic test on mice, 21 strains showed severe toxic effect to mouse liver cells among the 240 tested strains. 4. In electron microscopic studies of HeLa cells and mouse liver cells from animal which had been treated with crude toxin, the liver cells showed the cytoplasmic change: dillatation or vacuolization of endoplasmic reticulum, swelling of mitochondria and disappearance of mitochondrial cristae, increased number of lipid and glycogen particles. Nucleus and nucelar envelope alterations, were also noted. 5. In the detoxification study with ultraviolet irradiation, 90% of the toxic substances were denatured by the ultraviolet light irradiation for 24 hours. 6. As a mass screening, the cytotoxicity test of HeLa cells and histopathologic study of mice liver cells treated with culture filtrates, employed and feasible to detect mycotoxin producing fungi.

Screening and Evaluation of Streptomyces Species as a Potential Biocontrol Agent against a Wood Decay Fungus, Gloeophyllum trabeum

  • Two-hundred and fifty-five strains of actinomycetes isolated from soil samples were screened for their antagonistic activities against four well-known wood decay fungi (WDF), including a brown rot fungus, Gloeophyllum trabeum and three white rot fungi Donkioporia expansa, Trametes versicolor, and Schizophyllum commune. A dual culture assay using culture media supplemented with heated or unheated culture filtrates of selected bacterial strains was used for the detection of their antimicrobial activity against four WDF. It was shown that Streptomyces atratus, S. tsukiyonensis, and Streptomyces sp. greatly inhibited the mycelial growth of the WDF tested compared with the control. To evaluate the biocontrol efficacy of S. atratus, S. tsukiyonensis, and Streptomyces sp., wood blocks of Pinus densiflora inoculated with three selected Streptomyces isolates were tested for weight loss, compression strength (perpendicular or parallel to the grain), bending strength, and chemical component changes. Of these three isolates used, Streptomyces sp. exhibited higher inhibitory activity against WDF, especially G. trabeum, as observed in mechanical and chemical change analyses. Scanning electron microscopy showed that cell walls of the wood block treated with Streptomyces strains were thicker and collapsed to a lesser extent than those of the non-treated control. Taken together, our findings indicate that Streptomyces sp. exhibits the potential to be used as a biocontrol agent for wood decay brown rot fungus that causes severe damage to coniferous woods.

농산폐자원의 미생물학적 이용에 관한 연구 (제오보) 섬유소분해효소 생산 곰팡이의 분리 및 선별 (Studies on The Microbial Utilization of Agricultural Wastes (Part 5) Isolation and Selection of Cellulase Producing Fungi)

  • 곰팡이를 이용하여 섬유소분해효소를 생산할 목적으로 전국각처에서 수집한 102종의 시료로부터 총 221주의 곰팡이를 분리하였다. 이들 분리균주의 섬유소분해능을 여과 지붕괴법 ? 액화법 및 cup method로서 1차 36균주를 선별하였고 1차 선별된 균주를 탄소원을 변경하여 배양한뒤 활성을 추정하여 2차로 15균주를 선별하였다. 15균주를 개선된 배지에서 배양한뒤 $C_1$, $C_{x}$ 및 filter paper-activity을 측정하여 최종적으로 $C_1$$C_{x}$ -Cellulase의 활성이 강한 균주인 Aspergillus sp. strain No. AS-9, Penicillium sp. strain No. KI-1-2 Trichoderma sp. strain No. KI-7-2, KI-7-5, 및 KI-4-IB을 선별하였다. 이상 최종적으로 선별된 균주중의 strain No. KI-4-1-1B 조효소액으로 ammonia 처리볏짚을 당화시킬 때 24시간 작용으로 26%가 당화되었다.

Enzyme Amplification System Immunoassay에 의한 시설원예산물의 Deoxynivalenol 생성곰팡이의 검색 (Screening of Deoxynivalenol Producing Fungi from Greenhouse Horticulture by Enzyme Amplification System Immunoassay)

  • 시설원예산물 중 DON 생성능이 있는 균주의 오염 여부를 조사하기 위해 진주를 비롯한 서부 경남 일원과 경북 안동 근교에서 총 192점의 시료를 분리하여 균 분리를 실시한 결과 Fusarium속 103균주를 분리하였다. 분리된 균을 YES배지로 $28^{\circ}C$, 15일 배양한 후 ethyl acetate로 추출하여 thin layer chromatography(TLC)법으로 확인한 결과 DON 생성 균주 8개가 확인되었다. Enzyme amplification system immunoassay법을 이용하여 DON 생성 균주 8개를 정량한 결과 검출범위는 $0.007-1.21\;{\mu}g/mL$ 이었다. 이 중 남해 토양에서 분리한 32-D-3 균주가 $1.21\;{\mu}g/mL$로 가장 높았다.

버섯 중 항균물질의 검색 및 개발에 관한 연구 -그람음성균 및 곰팡이에 대한 항균물질의 검색(2보)- (Study on the Screening and Development of Antibiotics in the Mushrooms -The Screening of Bacterial and Fungal Antibiotics in Basidiomycetes (II)-)

  • 98종의 한국산버섯의 그람음성균에 대한 항균활성을 검색한 결과 20종의 버섯의 80% 에탄올(E) 및 증류수(H) 추출물이 Klebsiella pneumoniae, Proteus mirabilis, Serratia marcescens, Shiegella sonnei, Shiegella dysenteiae, Salmonella typhi, Salmonella typhimurium 및 Pseudomonas aeruginosa에 대하여 항균력을 나타내며, 특히 R. sinuatus, S. luteus, B. umbriniporus, 그리고 A. tabescens의 E 추출물은 다른 버섯에 비하여 여러 종류의 그람음성균에 대하여 다양한 항균활성을 보였다. Abortiporus biennis, Phellinus gilvus, 및 Polyporus dispansus의 E 추출물은 S. typhi에 대하여 강력한 항균력을 나타내었으며 이들의 MIC는 $10\;{\mu}g/ml$이었다. 25종 버섯의 곰팡이균에 대한 항균활성을 검색한 결과 Armillariella tabescens의 E 추출물이 Trichopyton mentagrophytes에 대하여 항균력을 가지며 이의 MIC는 $300\;{\mu}g/ml$이었다.

Diversity of Endophytic Fungi from Different Verticillium-Wilt-Resistant Gossypium hirsutum and Evaluation of Antifungal Activity Against Verticillium dahliae In Vitro

  • Cotton plants were sampled and ranked according to their resistance to Verticillium wilt. In total, 642 endophytic fungi isolates representing 27 genera were recovered from Gossypium hirsutum root, stem, and leaf tissues, but were not uniformly distributed. More endophytic fungi appeared in the leaf (391) compared with the root (140) and stem (111) sections. However, no significant difference in the abundance of isolated endophytes was found among resistant cotton varieties. Alternaria exhibited the highest colonization frequency (7.9%), followed by Acremonium (6.6%) and Penicillium (4.8%). Unlike tolerant varieties, resistant and susceptible ones had similar endophytic fungal population compositions. In three Verticillium-wilt-resistant cotton varieties, fungal endophytes from the genus Alternaria were most frequently isolated, followed by Gibberella and Penicillium. The maximum concentration of dominant endophytic fungi was observed in leaf tissues (0.1797). The evenness of stem tissue endophytic communities (0.702) was comparatively more uniform than the other two tissues. Eighty endophytic fungi selected from 27 genera were evaluated for their inhibition activity against highly virulent Verticillium dahliae isolate Vd080 in vitro. Thirty-nine isolates exhibited fungistasis against the pathogen at varying degrees. Seven species, having high growth inhibition rates (${\geq}75%$), exhibited strong antifungal activity against V. dahliae. The antifungal activity of both volatile and nonvolatile metabolites was also investigated. The nonvolatile substances produced by CEF-818 (Penicillium simplicissimum), CEF-325 (Fusarium solani), CEF-714 (Leptosphaeria sp.), and CEF-642 (Talaromyces flavus) completely inhibited V. dahliae growth. These findings deepen our understanding of cotton-endophyte interactions and provide a platform for screening G. hirsutum endophytes with biocontrol potential.

Effective Microwell Plate-Based Screening Method for Microbes Producing Cellulase and Xylanase and Its Application

  • Cellulase and xylanase are main hydrolysis enzymes for the degradation of cellulosic and hemicellulosic biomass, respectively. In this study, our aim was to develop and test the efficacy of a rapid, high-throughput method to screen hydrolytic-enzyme-producing microbes. To accomplish this, we modified the 3,5-dinitrosalicylic acid (DNS) method for microwell plate-based screening. Targeted microbial samples were initially cultured on agar plates with both cellulose and xylan as substrates. Then, isolated colonies were subcultured in broth media containing yeast extract and either cellulose or xylan. The supernatants of the culture broth were tested with our modified DNS screening method in a 96-microwell plate, with a $200{\mu}l$ total reaction volume. In addition, the stability and reliability of glucose and xylose standards, which were used to determine the enzymatic activity, were studied at $100^{\circ}C$ for different time intervals in a dry oven. It was concluded that the minimum incubation time required for stable color development of the standard solution is 20 min. With this technique, we successfully screened 21 and 31 cellulase- and xylanase-producing strains, respectively, in a single experimental trial. Among the identified strains, 19 showed both cellulose and xylan hydrolyzing activities. These microbes can be applied to bioethanol production from cellulosic and hemicellulosic biomass.

In-vitro Antimicrobial Activity Phytochemical and Cytotoxicity of Methanolic Fruits Extract of Capsicum frutescent

  • Capsicum frutescen is known in Sudan, is one of the most commonly used pepper species in cooking and in Sudanese folk medicine. The present study was conducted to investigate antimicrobial (bacteria and fungi) and cytotoxicity (Brine Shrimp Lethality Test) of methanolic extract of Capsicum frutescen (fruits). The extract have been tested in the present study to investigate the in vitro potential effects against Gram positive, Gram negative bacteria and fungi. The selected organisms were Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumonia and Candida albicans using the cup plate agar diffusion method. The methanol extract of Capsicum frutescen (fruits) exhibited inhibitory effects against Escherichia coli with zone of inhibition (23 mm) and Klebsiella pneumonia with zone of inhibition (17 mm). The phytochemical screening revealed the presence of Tannins, Saponin, Alkaloids, Anthroquinoles and Terpenoids. The Cytotoxicity of methanolic extract of Capsicum frutescens was $LD_{50}$ $64.68{\mu}g/ml$. The activity and presence of compounds known to be biologically active are a validation for the use of Capsicum as a food ingredient and as a therapeutic element of traditional medicine.
