  • Brine shrimp bioassay has been frequently utilized as a basic screening method for detecting a broad range of bioactive compounds from natural products. Each methanol, chloroform, and water extract of thirty-eight crude drugs were screened in the brine shrimp bioassay, and a number of crude drugs exhibited toxicity against brine shrimp nauplii.

  • We in anticancer drug development from natural resources have conceived and used a wide variety of experimental screening systems to support our efforts during the past 20 tears. Screens have been devided to address targets at the molecular, biochemical and cellular levels, both in vivo and in vitro. Screens have been essential for the experimental evaluation of the products from natural sources. In this congress, antitumor screening methods for deveol[ment of new drugs from natural sources and evaluation of their crude drugs are discussed.

  • The aqueous and methanolic extracts of 110 crude drugs were screened for hyaluronidase inhibitory activity using a microplate assay. Among them, MeOH extract of 15 crude drugs inhibited more than 80% of hyauluronidase activity at the concentration of 5mg/ml. The active principles of Anemarrhenae Rhizoma, Rhei Rhizoma, Ephedrae Herba, Pteropi Faeces and Ginseng Radix alba were transferred into organic solvents.

  • One hundred and seventeen crude drugs were screened for DNA-binding activity in vitro. The DNA-methyl green assay is a useful biochemical screen for the detection or development of biologically active compounds which bind to DNA. This assay is based upon the fact that free methyl green undergoes rapid spontaneous molecular rearrangement to its colorless carbinol base so that the liberation of the dye from DNA by displacement can be followed spectrophotometrically as a decrease in absorbance at 65Onm. Seven methanolic extracts of crude drugs including Cinnamomi Cortex spissus, Coicis Semen, Coptidis Rhizoma, Perillae Semen, Plantaginis Semen, Polygalae Radix and Zanthoxyli Fructus showed less than 2,000${\mu}$g/ml as their $IC_{50}$ values. The active principles of Plantaginis Semen and Zanthoxyli Fructus were transferred into organic solvents, which showed the $IC_{50}$ values with 588 and 574${\mu}$g/ml, respectively. These fractions have been selected for isolation of biologically active constituents.

  • Brine shrimp toxicity of water extracts of twenty three crude drugs was tested by the brine shrimp bioassay. Eight crude drugs were shown to have 100% mortality in $1000{\;}{\mu}g/ml$. They were fractionated by Amberlite XAD-2 column chromatography and were tested by the brine shrimp bioassay. Methanol fraction of Rhei Rhizoma $(LC_{50}=49.60{\;}{\mu}g/ml)$ and Cantharis $(LC_{50}=54.08{\;}{\mu}g/ml)$ were shown to have potent brine shrimp toxicity in this test.

  • Melanin biosynthesis inhibitors are useful not only for the materials used in cosmetics as skin-whitening agents but also for the remedy of hyperpigmentation. In order to find the new skin-whitening compounds from the natural products. screening of tyrosinase inhibitory activity in vitro has been carried out.   • In vivo antioxidant activities were screened over 30 kinds of crude drugs which are most frequently prescribed in oriental medicine. Of these, only Ginseng Radix, Cimicifugae Rhizoma, Zingiberis Rhizoma(steam dried), Alismatis Rhizoma, and Liriopes Tuber were shown to be positive in the activity.

  • Anti-inflammatory activity of crude drugs was evaluated by the albumin stabilizing activity test, according to the screening method of Mizushima et al., upon the randomly selected samples of 63 genus, 106 families, 123 species of plant. Almost every plant belonging to the families Araliaceae, Umbelliferae and Liliaceae showed strong stabilizing activity on the heat denaturation of bovine serum albumin, suggesting the presence of anti-inflammatory components in the plants.

Antifungal Actions of Crude Drug Water Extracts on Candida albicans(I) (Candida albicans에 대한 생약의 항진균성에 관한 연구(I))

  • Some crude drugs in ancient literatures have been used as traditional therapeutic agent of leucorrhea mainly caused by Trichomonas vaginalis and Candida albicans. Sixty six kinds of crude drugs in ancient literatures and ten constituents were selected as sample drugs. Trichomycin standard was tested to compare with the above drugs. To determine the anti-fungal effect of these drugs on Candida albicans Yu 1200, a test organism, screening test was conducted. Antifungal activities of crude drug water extracts were observed by means of two test methods : firstly through the agar slant method and secondly the counting chamber method which was used for acknowledged drug agents upon the result of the agar slant method. And in order to improve the fungicidal effect, the organisms were stained with 0.02% methylene blue solution. The results of the above test indicated that Fritillariae Rhizoma has antifungal action in the concentration of 310mcg/ml, Coptidis Rhizoma in 620mcg/ml, Meliae Cortex, Scutellariae Radix both in 5,000mcg/ml. Baicalin, catechol among the pure isolated constituents inhibited in the range of 50mcg/ml. This score was based on 50% inhibition in comparison with amounts of control organisms. Rhei Rhizoma, Mori Radicis Cortex, Linderae Radix, and Amomi globosi Fructus showed the antifungal effect moderately in 5,000mcg/ml, and baicalein and pectolinarin in 50mcg/ml in the limit of between 35% and 50% antifungal activity. Staining with 0.02% methylene blue showed that any of the crude drug extracts was unable to stain the cells, but trichomycin in 0.86unit/ml able to stain 12% of the cells. This result means that crude drugs probably do not have fungicidal but fungistatic action.
