Analgesic Constituent from the Herba of Saururus chinensis ($L_{OUR.})B_{AILL.}$ (삼백초의 진통성분)

  • For the investigation of bioactive natural products with analgesic activity, we have evaluated various extracts of Saururi Herba (Saururaceae), which has been used in traditiona l medicine for edema, beriberi, jaundice, turbid urine, carbuncle and furuncle. The diethylether extract of this plant was found to show a significant analgesic effect on writhing syndrome in mice. Using bioactivity-guided separation of the diethylether extract, analgesic constituent, (8S, 8`R, 7R, 6`R)-2'-oxo-4,5: 4',5'-bis(methylenedioxy)-${\Delta}^{1,3,5,3'}$-8.8', 7.6', 2.O.5'-neolignan(sauchinone) was isolated and structurally identified by physico-chemical properties and spectroscopic evidences. This compound has good analgesic activity with lower toxicity, as compared to other antipyretic-analgesic drug(acetaminophen).

  • In our search for bioactive natural products with antitumor activity, we have valuated various extracts of Saururi Herba (Saururaceae), which has been used in traditional medici ne for edema, beriberi, jaundice, turbid urine, carbuncle and furuncle. The hexane extract of the aerial part of this plant was found to show a potent cytotoxicity against several kinds of cultured human solid tumor cell lines (AGS, A549, HCT15, SKOV3, HEP-3B) in vitro. Using cytotoxicity-guided chromatographic separation of the hexane extract, cytotoxic constituent: 10-aminomethyl-3-hydroxy-4-methoxyphenanthrene-carboxylic acid lactam, was isolated and structurally identified by physico-chemical properties and spectroscopic evidences.

Antioxidant and Anticancer Effects of Agrimony (Agrimonia pilosa L.) and Chinese Lizardtail (Saururus chinensis Baill) (짚신나물, 삼백초의 항산화와 항암활성 효과)

  • The antioxidant activities of Agrimony (Agrimonia pilosa L.) and Chinese lizardtail (Saururus chinensis Baill) according to extraction methods were measured. SOD-like activity showed greater antioxidant effects with ethanol solvent than those with water. Ethanol extracts of Agrimony leaves showed the highest SOD-like activity of 94.4%. SOD-like activity differed according to the extraction solvents. The contents of polyphenolic compounds were higher in water extracts than those in ethanol extracts. The contents were 161.4 mg for Agrimony roots, 100.2 mg for Agrimony leaves, and 79.1 mg for Agrimony stalks in order. EDA in Agrimony leaves that were highest among medicinal plants were 83.4% in the water extract and 81.7% in the ethanol extract. The anticancer effects of the extracts by water and ethanol from Agrimony and Chinese lizardtail were experimented. The growth of stomach cancer cells, SNU-719 was inhibited 94.5% by the hexane fractions of Agrimony and also the growth of liver cancer cells, Hep3B was inhibited 83.2% by the hexane fractions of Agrimony, while the growth of normal cell, DC2.4 was not affected.

Antimicrobial Activity of Coptis chinensis and Sophora flavescens against Streptococcus mutans ATCC 25175 (Streptococcus mutans ATCC 25175에 대한 황련과 고삼의 항균활성)

  • As part of our screening of anticariogenic agents from medicinal plants, the ethanol extracts of 25 herbs widely used in the fork medicine were tested for the antimicrobial activity against a cariogenic bacterium Streptococcus mutans ATCC 25175. The ethanol extracts of Coptis chinensis, Sophora flavescens, Glycyrrhiza uralensis, Mentha arvensis, and Saururus chinensis showed antimicrobial activities against S. mutans. The extracts of Coptis chinensis and Sophora flavescens among these medicinal plants showed significant antomicrobial activity against S. mutans. These results suggested that the extracts of Coptis chinensis and Sophora flavescens could be the potential source of antimicrobial agent against S. mutans ATCC 25175.

Antioxidative Activity of Medicinal Plants from Korean (한국산 한약재의 항산화 활성 효과)

  • This study was performed to determine antioxidative activity of solvent extracts from medicinal plant. As the result, methanol extract from Saurus chinensis Bail and Glycine soya exhibited superior free radical activity as well as reducing power. In particular, methanol extract from Saurus chinensis Bail showed high free radical activity with 130.08${\mu}g/mL$ as $IC_{50}$ value and strong reducing power at concentration of $400{\mu}g/mL$.

Saucerneol F, a New Lignan Isolated from Saururus chinensis, Attenuates Degranulation via Phospholipase Cγ1 Inhibition and Eicosanoid Generation by Suppressing MAP Kinases in Mast Cells

  • During our on-going studies to identify bioactive compounds in medicinal herbs, we found that saucerneol F (SF), a naturally occurring sesquilignan isolated from Saururus chinensis (S. chinensis), showed in vitro anti-inflammatory activity. In this study, we examined the effects of SF on the generation of 5-lipoxygenase (5-LO) dependent leukotriene $C_4$ ($LTC_4$), cyclooxygenase-2 (COX-2) dependent prostaglandin $D_2$ ($PGD_2$), and on phospholipase $C{\gamma}1$ ($PLC{\gamma}1$)-mediated degranulation in SCF-induced mouse bone marrow-derived mast cells (BMMCs). SF inhibited eicosanoid ($PGD_2$ and $LTC_4$) generation and degranulation dose-dependently. To identify the molecular mechanisms underlying the inhibition of eicosanoid generation and degranulation by SF, we examined the effects of SF on the phosphorylation of $PLC{\gamma}1$, intracellular $Ca^{2+}$ influx, the translocation of cytosolic phospholipase $A_2$ ($cPLA_2$) and 5-LO, and on the phosphorylation of MAP kinases (MAPKs). SF was found to reduce intracellular $Ca^{2+}$ influx by inhibiting $PLC{\gamma}1$ phosphorylation and suppressing the nuclear translocations of $cPLA_2$ and 5-LO via the phosphorylations of MAPKs, including extracellular signal-regulated protein kinase-1/2 (ERK1/2), c-Jun N-terminal kinase (JNK), and p38. Taken together, these results suggest that SF may be useful for regulating mast cell-mediated inflammatory responses by inhibiting degranulation and eicosanoid generation.

Hepatoprotective Constituents of Saururus chinensis Roots Against Tacrine-induced Cytotoxicity in Human Liver-derived Hep G2 Cells (삼백초근의 타크린으로 유발한 간 세포독성 보호 성분)

  • Five liglans, sauchinone (1), di-0-methyltetrahydrofuriguaiacin B (2), manassantin A (3), manassantin B (4) and saucerneol B (5), have been isolated from the MeOH extract of Saururus chinesis roots. The evaluation for protective effect of compounds 1-5 against tacrine-induced cytotoxicity in human liver-derived Hep G2 cells was conducted. Compounds 1,2, and 5 showed significant protective effects with the EC$_{50}$ values of74.2${\pm}$0.9, 111.3${\pm}$0.8,64.3${\pm}$0.8 ${\mu}$M, respectively. Silybin, one of the well-known hepatoprotective agents, used as a positive control, and also showed protective effect with an EC$_{50}$ value of 86.2${\pm}$0.5 ${\mu}$M.

The Efficacy of Saururus chinensis on Cervical Cancer Cells : The Inhibitory Effect on the Function of E6 and E7 Oncogenes of HPV Type 16 (삼백초 추출물의 자궁경부암세포 억제 효능)

  • Cervical cancer is one of the leading causes of female death from cancer worldwide with about 500,000 deaths per year. A strong association between certain human papilloma viruses (HPV type 16 and 18) and cervical cancer has been well known. An extract of Saururus chinensis, named as PE-46, has been used to investigate whether this agent has the ability of inhibiting the oncogenes E6 and E7 of HPV type 16. PE-46 inhibited the proliferation of human cervical cancer cell lines in a dose response manner. PE-46 also inhibited the in vitro binding of E6 and E6AP which are essential for the binding and degradation of the tumor suppressor p53. In addition, PE-46 inhibited the in vitro binding of E7 and Rb which is essential tumor suppressor for the control of cell cycle. The levels of mRNA for E6 and E7 were also decreased by PE-46. SiHa cells treated with PE-46 induced G0/G1 arrest, resulting in inhibition of growth. Our study showed that the PE-46 can inhibit the cervical carcinomas via both inhibition of bindings between oncogenes and tumor suppressors, and inhibition of G1longrightarrowS transition. PE-46 inhibited the oncogenecity of E6 and E7 of HPV 16 type, thus could be used as a putative modulating agent for the treatment of cervical carcinomas caused by HPV.
