A Study on the Solubilization and Physical Properties of Sanjoinine-A (산조인 알카로이드의 가용화 및 물성에 관한 연구)

  • The seeds of Zizyphus Jujuba have been used as an antianxiety agent for the treatment of insomnia from the earliest times. Sanjoinine-A, isolated from the seeds of Zizyphus Jujuba, have been found to have a minor tranquilizer activity. However this drug is poorly soluble in water. In order to increase the dissolution rate of sanjoinine-A, solid dispersions with PVP-MC and inclusion complex with ${\beta}-cyclodextrin$ were prepared and evaluated. All of these systems increased the dissolution rate of sanjoinine A comparing with sanjoinine-A free base. From pH-rate profile of sanjoinine-A at $60^{\circ}C$, it was found that sanjoinine A was relatively stable in acidic solution, but unstable in basic solution.

  • The effects of Sanjoinine-A, an alkaloid isolated from Zizyphus spinosus semens, on central nervous system and general pharmacology were studied. In summary, Sanjoinine-A depress the spontaneous locomotor activity without motor incoordination and it has slight analgesic effect. Those effects are qualitatively similar to that of diazepam but its potency is much lower than diazepam(20 times). Sanjoinine-A does not depress the electric or pentylenetetrazole induced convulsion. Those effects are dissimilar with that of diazepam. Sanjoinine-A slightly depress the spontaneous or acethylchollne induced motility of smooth muscles but degree of depressant effect was variable to tissues. Sanjoinine-A does not show any effects on digestive system, blood, kidney fuction and neural ganglion.

Sedative Activity of Aporphine and Cyclopeptide Alkaoids Isolated from the Seeds of Zizyphus Vulgaris var. Spinosus, and the Fruits and Stem Bark of Zizyphus Jujuba var. Inermis in mice (산조인 및 대추, 대추나무로부터 단리한 아포르핀과 환상 펩티드 알칼로이드의 생쥐에 대한 진정작용)

  • The objective of this study was to evaluate the sedative activity of four aporphine alkaloids (APA) and nine cyclopeptide alkaloids(CPA), which had been isolated from the seeds (sanjoin) of Zizyphus vulgaris var. spinosus, and the fruits and stem bark of Zizyphus jujuba var. inermis. The assessment of sedative activity was carried out, employing a hexobarbital-induced sleeping time method in mice. When the relative sedative potency of sanjoinine-A(CPA) was given as one unit, those of nuciferine (APA), lysicamine (APA), chlorpromazine (positive control), and sanjoinine -Ahl (an epimer of sanjoinine-A) were 13, 6.5, 5, and 3, respectively. The sedatvie activities of other CPAs were much lower than those of sanjoinine-A and -Ahl, and other APAs were not active. On heat treatment, nuciferine and lysicamine were degraded into some artifacts which exhibited no sedative activity, while sanjoinine-A was converted into sanjoinine-Ahl which showed more potent sedative activity. These results suggested that nuciferine and sanjoinine-A were major sedative components of native sanjoin, and that sanjoinine-A and its epimeric artifact, sanjoinineAhl were the active principles of roasted sanjoin. It provides a scientific basis for heat-processing (roasting) of this Oriental medicine.

Tranquilizer-like Effects of Sanjoinine A: Possible GABA/Benzodiazepine Receptors Complex Involvement

  • Zizyphi Spinosi Semen (ZSS) has been widely used for the treatment of anxiety and insomnia in Korea and China. This experiment was performed to know whether sanjoinine A, one of major alkaloid compounds of ZSS has anxiolytic and hypnotic effects through the GABAergic systems. Our results showed that administration of sanjoinine A increased open arm entries and spent time in open arm in the elevated plus-maze and increased head dips in hole board test. Different from traditional anxiolytic, diazepam, sanjoinine A itself did not decrease locomotor activity and strength level in mice. Furthermore, Sanjoinine A (0.5-2.0 mg/kg) prolonged sleeping time and reduced sleeping latency induced by pentobarbital in a dose-dependent manner similar to muscimol, a $GABA_A$ receptor agonist. Sanjoinine A (0.25-1.0 mg/kg) also increased sleeping rate and sleeping time in the combined administration at the sub-hypnotic dose of pentobarbital and showed synergic effects with muscimol in potentiating sleeping onset and enhancing sleeping time induced by pentobarbital. However, sanjoinine A itself did not induce sleeping at the higher dose. In addition, both of sanjoinine A and pentobarbital increased chloride influx in primary cultured cerebellar granule cells. Sanjoinine A decreased the $GABA_A$ receptor ${\alpha}$-subunit expression and increased ${\gamma}$-subunit expression, and had no effects on abundance of ${\beta}$-subunit in primary cultured cerebellar granule cells, showing different expression of subunits from pentobarbital. In conclusion, sanjoinine A shows anxiolytic-like effects and augments pentabarbital-induced sleeping behaviors through the modification of GABAergic systems.   • 진정성 cyclopeptide인 sanjoinine-A의 진정작용을 분자 수준에서 밝히고자 sanjoinine-A의 ionophore 활성과 칼슘 결합 단백질인 calmodulin과의 결합을 CD와 NMR을 이용하여 연구하였다 sanjoinine-A는 칼슘과 마그네슘에 대하여 ionophore 활성이 있었으며 calmodulin과 두 단계로 결합한다는 것을 알 수 있었다. sanjoinine-A는 칼슘의 유무에 관계없이 calmodulin과 결합하였으며 CD와 NMR 스펙트럼상에서 현저한 변화가 있었다. sanjoinine-A가 calmodulin과 결합시 생체 내에서 활성 상태인 칼슘결합 calmodulin에 비하여 a-helix 구조비율이 감소하였으며 칼슘결합 loop에서 수소결합을 하고있는 amide 수소들의 exchange rate가 현저히 빨라졌다. sanjoinine-A는 calmodulin과 결합하여 칼슘결합 loop 주위의 공간구조를 변형시켜 그 활성을 변화시키는 것으로 보인다.

  • Sedative principles of the seeds of Zizyphus vulgaris var. sphinosus have been characterized as sanjoinine-A (frangufoline), nuciferine and their congeners. Also, heat treatment of sanjoinine-A-produced a more active artifact, sanjoinine-Ahl, which provides a scientific basis for heat-processing (roasting) of this Oriental medicine.

  • A number of sedative alkaloids were isolated from Sanjoin(酸棗仁), the seeds of Zizyphus vulgaris Lamark var. spinosus Bunge (Rhamnaceae) which is an important Chinese medicinal material used to treat insomnia and sometimes to treat sleepiness. Those compounds were designated as Sanjoinine-A, B, C, D, etc. depending on the order of increasing polarity. Effects of the Sanjoin on the Rat Brain - Focused on Serotonin, Sleeping Time, Sleep EEG and Autonomic Activity - (산조인이 백서 뇌에 미치는 영향 - 세로토닌, 수면시간, 수면뇌파, 자동운동을 중심으로 -)

  • Objects : Sanjoin, the seeds of Zizyphus vulgaris var. spinosus has been used as the most important hypnotic agent in chinese medicine to treat insomnia. This research was performed in order to examine the effect of betulinic acid and sanjoinine-A which are components of Sanjoin. Method : Sleeping time, sleep recordings of EEG, EMG, serum serotonin level, and locomotor activity were measured in rats which received betulinic acid and sanjoinine-A as sleep induction material extracted from Sanjoin. Results : 1) Groups received betulinic acid, sanjoinine-A, and lorazepam showed increased sleep time than control group with saline. 2) Groups with betulinic acid, sanjoinine-A, lorazepam and saline recorded ${\beta}$-wave in sleep recordings of EEG. In EMG, there was no significant difference among all groups. 3) No significant difference in serum serotonin level among all groups was found. 4) In autonomic activity testing, groups of betulinic acid, sanjoinine-A, and lorazepam showed significantly more decreased in activity than saline group. In comparison of groups of betulinic acid and sanjoinine-A with a group of lorazepam, there was no significant difference. Conclusion : These results suggest that betulinic acid and sanjoinine-A have the sedative effect like lorazepam rather than sleep effect.

Binding of Sanjoinine-A (Frangufoline) to Calmodulin

  • A binding protein of radio-labeled sanjoinine-A (fangufoline) in rat brain cytoplasm was investigated, using an equilibrium dialysis technique. The labeled agent was bound to the cytosol fraction with two distinctly different types of sets in calclum ion-dependent manner. The bound protein was identified as calmodulin by dgel filtration of the sanjoinine-A bound cytosol fraction on a Sephadex G-75 column. Calmodulin was bound to sanjoinine-A bound at two sets of binding sites in the calculated as two at high affinity sites $(Kd=1.1\;mu{M)}$ and four at low affinity sites $(Kd=3.1\;\mu{M)}$.
