The objectives of this study are understanding on the type and the characteristic of the architecture in the saltern. At result, there are warehouse of the salt, residence of salt man and office building. These architectures are clearly different from Korean traditional architecture and constructed in special area (a saltern), on special purpose. These are not excellent architectural form, but materials and forms of these architectures are reasonably suitable for the regional characteristic and the function. Today, the architecture in the saltern, are gradually disappear, therefore the researches and the memories are meaningful in this study.
We studied the diversity of the halophilic archaea and bacteria in crystallizer ponds of a Korean solar saltern by analyzing 16S rRNA gene libraries. Although diverse halophilic archaeal lineages were detected, the majority (56%) were affiliated with the uncultured and cultured Halorubrum group. Halophilic archaea that have been frequently observed in solar saltern environments previously, such as Halogeometricum, Halococcus, Haloarcula, and Haloferax, were not detected in our samples. The majority of clones (53%) belonged to the Cytophaga-Flavobacterium-Bacteroides and ${\alpha}-,\;{\gamma}-,\;and\;{\delta}-Proteobacteria$ groups, with 47% of the clones being affiliated with ${\gamma}-Proteobacteria$. We also identified new ${\delta}-Proteobacteria$-related bacteria that have not been observed in hypersaline environments previously. Our data show that the diversity of the halophilic archaea and bacteria in our Korean saltern differs from that of solar salterns found in other geographic locations. We also showed by quantitative real-time PCR analysis that bacteria can form a significant component of the microbial community in solar salterns.

The community dynamics of benthic diatoms in the hypersaline environment are investigated to advance our understanding how salinity impacts marine life. Diatoms were sampled in the two salterns encompassing salt Ponds, ditches, and seawater reservoirs (n = 11), along the salinity gradient (max = 324 psu), and nearby tidal flats (n = 2). The floral assemblages and distributions across sites and stations showed great variations, with a total of 169 identified taxa. First, not surprisingly, higher diversity of benthic diatoms was found at natural tidal flats than salterns. The saltern diatoms generally showed salinity dependent distributions with distinct spatial changes in species composition and dominant taxa. Biota-environment and principal component analysis confirmed that salinity, mud content, and total nitrogen were key factors influencing the overall benthic community structure. Some dominant species, e.g., Nitzschia scalpelliformis and Achnanthes sp. 1, showed salinity tolerance / preference. The number of diatom species at salinity of >100 psu reduced over half and no diatoms were found at maximum salinity of 324 psu. The highest salinity for the observed live diatoms was 205 psu, however, a simple regression indicated a theoretical salinity threshold of ~300 psu on the survival. Finally, the indicator species were identified along the salinity gradient in salterns as well as natural tidal flats. Overall, high species numbers, varying taxa, and euryhaline distributions of saltern diatoms collectively reflected a dynamic saltern ecosystem. The present study would provide backgrounds for biodiversity monitoring of ecologically important microalgal producers in some unique hypersaline environment, and elsewhere.

In July 2018, solar saltern samples were collected from Siheung, Gyeonggi-do Province to obtain unrecorded haloarchaea in Korea. The samples were suspended in a 20% NaCl (w/v) solution, and serial dilution was performed in fresh DB Characterization media No. 2. The strains isolated in this study showed at least 98.7% sequence similarity or more compared to the previously reported. Finally, 16 haloarchaeal strains, which were not reported in Korea but validly published under the International Code of Nomenclature of Prokaryotes (ICNP), were obtained from a solar saltern in Siheung. These 16 isolates were allocated to the orders Halobacteriales and Haloferacales. The 10 Halobacteriales strains were classified into the family Halobacteriaceae and Haloarculaceae. Each family belonged to three genera, respectively. The other six Haloferacales belonged to the families Haloferacaceae and Halorubraceae. Each family belonged to genus genus, respectively. Collectively, the unrecorded haloarchaeal strains belonged to two orders, four families, and eight genera. During the research, the possibility of discovering previously unknown species in domestic solar saltern was established. Gram-staining, cell morphology, physiological and basic biochemical parameters, and phylogenetic analysis were all performed in this study and are described in detail for each strain.

Comprehensive chemical analysis of extracts and fractions of marine actinomycete strains led to the discovery of a new minor secondary metabolite, salternamide E (1), from a saltern-derived halophilic Streptomyces strain. The planar structure of salternamide E (1) was elucidated by a combinational analysis of spectroscopic data including NMR, MS, UV, and IR. The absolute configuration of salternamide E (1) was determined by circular dichroism spectroscopic analysis. Salternamide E displayed weak cytotoxicity against various human carcinoma cell lines.

Halophilic bacteria are promising reservoirs for halotolerant enzymes that have gained much attention in biotechnological applications due to their remarkable activity and stability. In this study, 62 halophilic bacterial strains isolated from a solar saltern were screened for the production of various extracellular enzymes. The results revealed that 31 strains (50%) were positive for amylase production while 26 strains (41.9%) were positive for protease. Further, 22 strains (35.48%) exhibited β-glucosidase activity and only 17 (27.41%) demonstrated lipase activity. Of the investigated halophiles, ten strains growing in the presence of ≥15% NaCl (w/v) were selected and identified based on their 16S rRNA gene sequences as Halomonas meridiana, Salinivibrio costicola, Virgibacillus oceani, Virgibacillus marismortui, Marinobacter lipolyticus, Halobacillus karajensis, Salicola salis, Pseudoalteromonas shioyasakiensis, Salinicoccus amylolyticus, and Paracoccus salipaludis. Therefore, the present study highlights the diversity of the culturable halophilic bacteria in a Mediterranean solar saltern, harboring various valuable halotolerant enzymes.
한국 근해와 염전으로부터 40 균주의 색소 생성 호염세균을 분리하여, 16S rDNA PCR-RELP와 16S rDNA 염기서열 분석을 통하여 다양성을 파악하였다. 분리된 호염성 색소 생성 세균들은 Pseudoalteromonas, Photobacterium, Vibrio, Halobavillus, Bacillus, Paracoccus, Salinicoccus, Tenacibaculum, Flavobacterium의 다양한 속의 세균 종이었다. 해양에서 분리한 색소생성 호염 세균의 $80\%$ 이상이 그람음성인 Pseudoalteromonas 속이었으며, 염전에서 분리한 세균의 대부분은 그람양성의 Halobavillus속 세균이었다. 또한 Salinicoccus 속에 속하는 신종 가능성이 있는 균주 KK7을 분리하였다.
대부분의 호염성 고세균은 고염환경으로 알려진 천일염전(solar saltern), 염호수(salt lake)를 비롯한 다양한 환경에서 서식하고 있다고 알려져 있다. 본 연구에서는, 한국의 대표적인 고염환경으로부터 분리배양을 통하여 호염성 고세균의 특성 분석을 실시하였다. 호염성 미생물들을 분리하기 위하여, 고염배지를 제작하고, 총 7개의 순수 배양체를 확보하였다. 분리된 호염성 미생물들의 16S rRNA 유전자 서열에 대한 계통학 및 상동성 분석을 실시하여 Halorubrum 속의 미생물 4종, Halogeometricum 속의 미생물 1종, Halobacterium 속의 미생물 1종, Haloarcula 속의 미생물 1종을 각각 확보 할 수 있었다. 이들 호염성 고세균들은 모두 그람 음성균이며, nitrate를 전자수용체로 사용한 혐기적 조건에서 성장이 관찰되지 않았다. 또한, 분리된 모든 미생물들은 12-30% (w/v, NaCl) 염분을 필요로 하였다. 본 연구는 국내의 호염환경으로 부터 호염성미생물의 분리기술 및 관련 연구에 대한 기초적 정보를 제공하며, 국내의 다양한 극한환경에서 서식하는 미생물 배양체 확보를 통하여 국내 생물자원 확보에 기여할 것으로 기대한다.
본 연구에서는 토판염전 결정지에 서식하는 세균군집의 계통학적 다양성을 분석하고 culturomics법에 기반하여 고도 호염균의 다양성을 확보하고자 하였다. 토판염전 내 세균밀도를 조사한 결과, 직접검경법에 의한 생균수는 평판배양법에 비해 $10^3{\sim}10^4$ 배 이상 높은 계수치를 나타내어 배양이 곤란한 세균(viable but non-culturable bacteria, VBNC)이 다수 존재해 있음으로 판단되었다. 토판염전 결정지 내 세균군집 다양성 해석을 위해 배양비의존적 방법인 pyrosequencing 분자기법을 이용하였다. 세균군집의 경우 1,778 OTUs, 다양성 지수 6.16로 나타났으며, 18문46강85목140과 243속으로 확인되었다. Archaea군집은 643 OTUs, 다양성 지수 4.95로 3문6강7목7과 38속이 분포해 있음이 확인되었다. 고도 호염균 생육에 적합한 배양배지 및 배양조건을 고려한 총 59가지의 다양한 배양 방법을 이용하여 137균주를 순수 분리하였다. 분리된 고도호염균의 16S rRNA 유전자 분석결과, 총4문11속의 다양한 계통군으로 확인되었으며 호염성 archaea 계통군 Haloterrigena 속과 haloferax 속이 culturomics법을 통해 성공적으로 분리되었다. 고도 호염균 다양성 확보를 위해 culturomics법이 매우 효과적임을 밝혔다.
C-1화합물은 고염분성 환경의 혐기적인 퇴적층에서 관찰되며, 이 퇴적층의 표면과 수면에는 호기성 메틸영양미생물의 잠재적인 서식지가 된다. 염전과 갯벌에서 채취한 토양 시료를 접종원으로 하여 메탄올을 탄소원과 에너지원으로 공급하고 염분농도에 따라 계대배양한 후 메탄올 산화 세균 성장 조건을 살펴 본 결과, 메탄올 산화 세균이 성장 할 수 있는 염분의 최대 농도는 20% 조건이었다. 변성 구배 젤 전기영동 (Denaturing gel gradient electrophoresis)을 이용하여 농화배양액 내 미생물 군집구조를 분석한 결과, 메탄올 산화 미생물인 Methylophaga 관련 세균이 우점하는 것으로 나타났다. 정량 PCR결과 고세균이 세균의 1-10%로 존재하는 것으로 나타났다. 세균용 항생제 실험결과, 메탄올 산화가 억제되어 고세균이 메탄올 산화에 관여하지 않는다는 것을 추정할 수 있었다. 이번 연구를 통해, 메틸영양세균이 고염분환경(염분 농도 20%까지)에서도 C-1 화합물을 산화할 수 있음을 확인 할 수 있었다.
