Effect of Dietary Antimicrobials on Immune Status in Broiler Chickens

  • This study evaluated the effects of dietary anticoccidial drugs plus antibiotic growth promoters (AGPs) on parameters of immunity in commercial broiler chickens. Day-old chicks were raised on used litter from a farm with endemic gangrenous dermatitis to simulate natural pathogen exposure and provided with diets containing decoquinate (DECX) or monensin (COBN) as anticoccidials plus bacitracin methylene disalicylate and roxarsone as AGPs. As a negative control, the chickens were fed with a non-supplemented diet. Immune parameters examined were concanavalin A (ConA)-stimulated spleen cell proliferation, intestine intraepithelial lymphocyte (IEL) and spleen cell subpopulations, and cytokine/chemokine mRNA levels in IELs and spleen cells. ConA-induced proliferation was decreased at 14 d post-hatch in DECX-treated chickens, and increased at 25 and 43 d in COBN-treated animals, compared with untreated controls. In DECX-treated birds, increased percentages of $MHC2^+$ and $CD4^+$ IELS were detected at 14 d, but decreased percentages of these cells were seen at 43 d, compared with untreated controls, while increased $TCR2^+$ IELs were evident at the latter time. Dietary COBN was associated with decreased fractions of $MHC2^+$ and $CD4^+$ IELs and reduced percentages of $MHC2^+$, $BU1^+$, and $TCR1^+$ spleen cells compared with controls. The levels of transcripts for interleukin-4 (IL-4), IL-6, IL-17F, IL-13, CXCLi2, interferon-${\gamma}$ (IFN-${\gamma}$), and transforming growth factor${\beta}$4 were elevated in IELs, and those for IL-13, IL-17D, CXCLi2, and IFN-${\gamma}$ were increased in spleen cells, of DECX- and/or COBN-treated chickens compared with untreated controls. By contrast, IL-2 and IL-12 mRNAs in IELs, and IL-4, IL-12, and IL-17F transcripts in spleen cells, were decreased in DECX- and/or COBN-treated chickens compared with controls. These results suggest that DECX or COBN, in combination with bacitracin and roxarsone, modulate the development of the chicken post-hatch immune system.

  • 배 경: 액체환기가 손상된 폐에 긍정적인 영향을 주는 한 기전으로 perfluorocarbon(PFC)이 기도상피세포에서 chemokine발현을 억제할 수 있는 지를 관찰하기 위해 본 연구를 시행하였다. 방 법: 기도 상피세포로는 A549 세포주를, PFC로는 perfluorodecalin을, PFC의 노출은 Transwell$^{(R)}$배양접시의 lower chamber를 이용하여 시행하였다. PFC가 말초혈액 탄핵구층(peripheral blood mononuclear cell : PBMC)의 기능을 억제해서 A549세포의 chemokine 발현을 억제할 수 있는지를 관찰하기 위해서 PBMC를 분리하여 Transwell$^{(R)}$ 접시에서 배양하면서 lipopolysaccharide(LPS, 10 ${\mu}g/mL$)로 자극과 PFC의 노출에 따라 군을 나누었으며 24시간 후 그 배양 상층액을 포함한 conditioned media(CM)으로 24시간 동안 A549세포를 자극한 후 chemokine발현을 측정하였다. 또한 PFC가 직접 기도 상피세포의 기능을 억제할 수 있는 지를 관찰하기 위해서 A549 세포를 Transwell$^{(R)}$ 접시에서 배양하면서 interleukin-l$\beta$(IL-1$\beta$, 10 ng/ml), TNF-$\alpha$(10 ng/ml)로 각각 혹은 동시에 24시간동안 자극하면서 PFC노출여부에 따른 IL-8과 RANTES발현 정도를 비교하였다. Chemokine 발현은 IL-8과 RANTES의 단백에 대한 ELISA와 mRNA는 Northern analysis를 통하여 분석하였다. 결 과: 1. LPS로 자극한 PBMC의 배양상층액을 포함한 CM로 A549세포를 자극하였을 때 IL-8과 RNATES mRNA 발현과 면역반응성 단백 생성이 의미 있게 상승하였으나(p<0.05) PFC노출여부에 따른 유의한 차이는 관찰할 수 없었다. 2. TNF-$\alpha$와 IL-1$\beta$ 모두 A549세포에서 IL-8과 RANTES mRNA자발현과 면역반응성 단백 생성의 증가를 가져왔으나(p<0.05) PFC노출에 따른 유의한 차이는 관찰할 수 없었다. 결 론: 기도 상피세포의 chemokine발현 감소를 통한 항염증 작용은 액체환기시 보이는 염증반응 감소에 큰 기여를 하지 않을 것으로 생각되며 추후 액체환기시 관찰되는 염증반응의 감소의 기전에 대한 연구가 더 필요할 것으로 사료된다.

Evaporating Heat Transfer Characteristics of R-l34a in a Horizontal Smooth Channel

  • Convective boiling heat transfer coefficients were measured in a horizontal minichannel with R-l34a. The test section was made of stainless steel tube with an inner diameter of 3.0 mm and a length of 2m. It was uniformly heated by applying electric current directly to the tube. Local heat transfer coefficients were obtained for heat fluxes from 10 to $40kW/m^2$, mass fluxes from 200 to $600kgT/m^2s$, qualities up to 1.0, and the inlet saturation temperature of $10^{\circ}C$. The experimental results were mapped on Wojtan et $al.'s^(7)$ and Wang et $al.'s^(8)$ flow pattern maps. The nucleate boiling was predominant at low vapor quality whereas the convective boiling was predominant at high vapor quality. Laminar flow appeared in the flow with minichannel. The experimental results were compared with six existing two-phase heat transfer coefficient correlations. A new boiling heat transfer coefficient correlation based on the superposition model for refrigerants was developed with mean and average deviations of 10.39% and -3.66%, respectively.

g, r, i 필터에 대한 Yonsei-Yale Isochrones의 분석과 적용

  • $Y^2$(Yonsei-Yale) Isochrones은 Yale의 항성 진화 계산을 이용한 정밀한 등연령곡선으로 구상성단의 연구에 널리 사용되고 있다. 이번 연구에서는 $Y^2$-Isochrones를 Kurucz 모형을 이용한 색변환을 통해서 SDSS로 널리 알려진 ugriz 필터 체계에 대해 적용하고 실제 관측 결과와 비교함으로써 타당성을 검증하고자 한다. 우선 ugriz 필터의 등연령곡선을 제공하는 BaSTI와 DSEP, Padova 모델간의 비교를 통해 $Y^2$-Isochorones이 MSTO에서 g-r값이 다른 모델에 비해 0.05정도 큼을 확인하였다. 또한 CFHT 3.6m 망원경의 가시광 카메라 MegaCam으로부터 얻은 중원소 함량이 낮은 다섯 개의 구상성단 M15, M30, M53, NGC 5053, NGC 5466에 대한 g, r, i 필터의 색등급도에 각각의 등연령곡선을 적용하여 모델에 따른 구상성단의 특성을 분석하였다. $Y^2$-isochrones를 이용한 (g-r, r) CMD의 분석 결과, BaSTI와 DSEP 모델에 비해 0.1~0.3만큼 거리지수가 크고, 성단의 나이는 1~3Gyr 정도 어리게 측정됨을 확인할 수 있었다. 더불어 SDSS 관측으로부터 얻어진 구상성단에 대한 색등급도와 비교 분석과정을 수행하여, 다른 망원경에서의 사용가능 여부를 확인하였다. 이 연구를 통하여 각 모델에 따른 등연령곡선의 차이를 확인하고, 현재 여러 망원경에서 사용되고, 앞으로 여러 대형망원경에서 사용될 ugriz 필터 체계에 $Y^2$-Isochrones를 적용할 수 있는 타당성을 제시하고자 한다.

Panax ginseng-derived fraction BIOGF1K reduces atopic dermatitis responses via suppression of mitogen-activated protein kinase signaling pathway

  • Background: BIOGF1K, a fraction of Panax ginseng, has desirable antimelanogenic, anti-inflammatory, and antiphotoaging properties that could be useful for treating skin conditions. Because its potential positive effects on allergic reactions in skin have not yet been described in detail, this study's main objective was to determine its efficacy in the treatment of atopic dermatitis (AD). Methods: High-performance liquid chromatography was used to verify the compounds in BIOGF1K, and we used the (3-4-5-dimethylthiazol-2-yl)-2-5-diphenyltetrazolium bromide method to determine its cytotoxicity in RBL-2H3 and HMC-1 cell lines. RBL-2H3 cells were induced using both anti-DNP-IgE/DNP-BSA and calcium ionophore (A2187) treatments, whereas HMC-1 cells were induced using A2187 alone. To measure mast cell degranulation, we performed histamine (enzyme-linked immunosorbent assay) and β-hexosaminidase assays. To quantify interleukin (IL)-4, IL-5, and IL-13 levels in RBL-2H3 cells, we performed quantitative polymerase chain reaction (PCR); to quantify expression levels of IL-4 and IL-13 in HMC-1 cells, we used semiquantitative reverse transcription polymerase chain reaction (RT-PCR). Finally, we detected the total and phosphorylated forms of extracellular signal-regulated kinase, p-38, and c-Jun N-terminal kinase proteins by immunoblotting. Results: BIOGF1K decreased the AD response by reducing both histamine and β-hexosaminidase release as well as reducing the secretion levels of IL-4, IL-5, and IL-13 in RBL-2H3 cells and IL-4 and IL-13 in HMC-1 cells. In addition, BIOGF1K decreased MAPK pathway activation in RBL-2H3 and HMC-1 cells. Conclusions: BIOGF1K attenuated the AD response, hence supporting its use as a promising and natural approach for treating AD.

인간 간암세포주 HepG2에서 김 분획물의 항산화 활성을 통한 증식 억제 및 유전자 발현 양상 (Anti-proliferating Effects and Gene Expression Profiles through Antioxidant Activity of Porphyra yezoensis Fractions on Human HepG2 Cell Lines)

  • 김(Porphyra yezoensis, Laver)의 MeOH 추출에 의한 유기용매 별 분획물에서 폴리페놀 함량과 항산화 활성 및 간암세포주 HepG2의 세포증식 억제효과를 확인하였다. $CHCl_3$ 분획물의 폴리페놀 함량은 $10.34{\mu}g/mg$으로 물 분획물의 $13.08{\mu}g/mg$ 보다는 다소 적게 나타났으나, DPPH 자유라디칼 소거에 의한 전자공여능(EDA)에서 나타난 $ED_{50}$$16.96{\mu}g/ml$로 가장 높게 나타났다. $CHCl_3$과 EtOAc 분획물은 농도의존적으로 HepG2 세포의 증식을 억제하였으며, 특히 $900{\mu}g/ml$$CHCl_3$ 분획물을 24시간 동안 처리하여 90%의 세포증식이 억제되었다. 한편 $CHCl_3$ 분획물이 처리된 HepG2 세포의 유전자 발현 양상을 microarray로 확인하였다. P. yezoensis의 효능과 연관지은 gene ontology 분석으로 비타민 D 합성 과정, 항균작용에 대한 반응 및 영양물질에 대한 반응에 관련된 유의 유전자들을 탐색하였다. 유의 유전자로 IL6R와 CYP1A1를 선정하였고, 이들 유전자의 상위 조절자는 ARNT 유전자가 선정되었다. 또한 50 및 $100{\mu}g/ml$$CHCl_3$ 분획물이 처리된 HepG2 세포에서 IL6R와 CYP1A1 단백질의 발현과 상위 조절자인 ARNT의 활성을 Western blotting으로 확인하였다.

  • Two-phase flow boiling heat transfer of R-410A in horizontal small tubes was reported in the present experimental study. The local heat transfer coefficients were obtained over a heat flux range of 5 to 40 kW/$m^2$, a mass flux range of 170 to 600 kg/$m^2s$, a saturation temperature range of 3 to $10^{\circ}C$, and quality up to 1.0. The test section was made of stainless steel tubes with inner diameters of 0.5 and 3.0 mm, and lengths of 330 and 3000 mm, respectively. The section was heated uniformly by applying a direct electric current to the tubes. The effects on heat transfer of mass flux, heat flux, inner tube diameter, and saturation temperature were presented. The experimental heat transfer coefficient is compared with six existing heat transfer coefficient correlations. A new boiling heat transfer coefficient correlation based on the superposition model for R-410A in small tubes was developed with mean deviation of 10.13%.

  • Many countries concentrated on the space developments to enhance the national security and the people's quality of life. A space launch vehicle for accessing the space is a typical large complex system that is composed of the high-technology like high-performance, high-reliability, superhigh-pressure, etc. The project developing large complex system like space launcher is mostly conducted in the uncertain environment. To achieve a goal of the project, its success probability should be enhanced consistently by reducing its uncertainty during the life cycle: it's possible to reduce the project's uncertainty by performing the risk management (RM) that is a method for identifying and tracing potential risk factors in order to eliminate the risks of the project. In this paper, we introduce the risk management (RM) process applied for a Space Launch Vehicle R&D Project.

Increased Cytokine and Nitric Oxide Levels in Serum of Dogs Experimentally Infected with Rangelia vitalii

  • This study aimed to measure the levels of interferon-gamma (IFN-${\gamma}$), tumor necrosis factor-alpha (TNF-${\alpha}$), interleukin 1 (IL-1), interleukin 6 (IL-6), and nitrite/nitrate ($NO_x$) in serum of dogs experimentally infected with Rangelia vitalii. Twelve female mongrel dogs were divided into 2 groups; group A (uninfected controls) composed by healthy dogs (n=5) and group B consisting of dogs inoculated with R. vitalii (n=7). Animals were monitored by blood smear examinations, which showed intraerythrocytic forms of the parasite on day 5 post-infection (PI). Blood samples were collected through the jugular vein on days 0, 10, and 20 PI to determine the serum levels of IFN-${\gamma}$, TNF-${\alpha}$, IL-1, IL-6, and $NO_x$. Cytokines were assessed by ELISA quantitative sandwich technique, and $NO_x$ was measured by the modified Griess method. Cytokine levels (IFN-${\gamma}$, TNF-${\alpha}$, IL-1, and IL-6) were increased (P<0.01) in serum of infected animals. Serum levels of $NO_x$ were also increased on days 10 PI (P<0.01) and 20 PI (P<0.05) in infected animals. Therefore, the infection with R. vitalii causes an increase in proinflammatory cytokines and nitric oxide content. These alterations may be associated with host immune protection against the parasite.

Result As the study for standardization of EAV's measurement values according to constitution and sex and age, the questionnaire on QSCC and QSCC(I) and EAV were carried out to 603 general patients in east : west promotion health center affiliated to Kyung Hee Medical center. The following conclusions were reached as the result of statistical handling of the data of questionnaire and EAV. 1) Each standard scope of BAZ and CMP was suggested. 2) BAZ and CMP examined about constitution did not show any statistical difference. 3) The result of examination by sex on BAZ and CMP is such as followings. A meaningful difference was showed at the level of p<0.001 in HH, HEL, HFR, and FF in case of male on BAZ. meaningful difference were showed at the level of p<0.001 in R3, R4, R18, L3, L16 and L18 and at the level of P<0.01 In R5, R8, R12, L7, L9, L10, L11, and L13 and at the level of P<0.05 in R1, R2, R6, R11, R13, R14, R16, R17, R19, R20, L1, L2, L4, L5, L14, L17, and L19 in case of male on CMP. There was no statistical difference in R7, R9, R10, R15, L12, and L15. 4) In every measure point of both BAZ and CMP a meaningful difference was notified at the level of P<0.01 by age. 2. Suggestion 1)Environmental, examinee and examiner's factors which may cause error in EAV measurement should be excluded. 2)The factor of change by time and space should be clarified. 3)The correlation between disease and meridian should be learned. And scopes of BAZ and CMP according to disease should be studied.
