The Optimum Installation Angle of Reticulated Root Piles under Lateral Loads (횡방.향하중을 받는 그물식 뿌리말뚝의 최적 타설경사각)

  • In order to investigate the influence of installation angle of reticulated root piles(RRP) on their lateral load capacities, model tests of lateral loads on RRP with various installation angles $0^{\circ}\;, 5^{\circ}\;, 10^{\circ}\;, 15^{\circ}\;, 20^{\circ}\;,and 25^{\circ}$ are carried out. One set of RRP consists of 12 piles which are installed in circular patterns forming two concentric circles, each of which has 6 piles. Each pile made of a steel bar of 5mm in diameter and 350mm in length, is coated with sand until the bar has the diameter of 6.5mm. According to the test results, RRP's response is travily influenced by the displacement level. At low displacement level(1m), lateral load capacity increases as the installation angle is increased. However, the value of the optimum installation angle decreases as the displacement level is increased. In fact, it is found to be $17.5^{\circ}$ at 6mm lateral displacement. The ratios of the lateral resistances for the optimum installation angles to those for the vertical RRP decrease as the lateral displacements are increased. Thus the effect of slant ins angle of RRP is expected to be reduced at higher level of lateral displacement.

  • This paper deals with six degree-of-freedom HILS(hardware-in-the-loop-simulation) of KSR-III rocket using a TAFMS(three axis flight motion simulator). This TAFMS HILS test is accomplished before main HILS tests in order to verify the control stability in the presence of TAFMS dynamic effects. The TAFMS HILS test includes initial attitude holding tests for INS initial alignment procedures, timer synchronization tests with an auxiliary lift-off signal, real-time calibration tests using an external thermal recorder, open-loop TAFMS operating tests, and final closed-loop TAFMS HILS tests using the TAFMS attitude measurements as inputs to the closed control loop. The HILS tests are accomplished for several flight conditions composed with nominal flight condition, TWD effect added condition, slosh modes and/or bending modes existing condition, and windy condition, etc.

  • The complexity of the bacterial recombination system is a barrier for the construction of bacterial mutants for the further functional investigation of specific genes. Several protocols have been developed to inactivate genes from the genus Pseudomonas. Those protocols are complicated and time-consuming and mostly do not enable easy construction of multiple knock-ins/outs. The current study describes a single and double crossover-recombination system using an optimized vector-free allele-exchange protocol for gene disruption and gene replacement in a single species of the family Pseudomonadaceae. The protocol is based on self-ligation (circularization) for the DNA cassette which has been obtained by overlapping polymerase chain reaction (Fusion-PCR), and carries an antibiotic resistance cassette flanked by homologous internal regions of the target locus. To establish the reproducibility of the approach, three different chromosomal genes (ncRNA31, rpoN, rpoS) were knocked-out from the root-associative bacterium Pseudomonas stutzeri A1501. The results showed that the P. stutzeri A1501 mutants, which are free of any plasmid backbone, could be obtained via a single or double crossover recombination. In order to optimize this protocol, three key factors that were found to have great effect on the efficiency of the homologous recombination were further investigated. Moreover, the modified protocol does not require further cloning steps, and it enables the construction of multiple gene knock-in/out mutants sequentially. This work provides a simple and rapid mutagenesis strategy for genome editing in P. stutzeri, which may also be applicable for other gram-negative bacteria.

Dynamic Modeling of Autonomous Underwater Vehicle for Underwater Surveillance and Parameter Tuning with Experiments (수중정찰용 자율무인잠수정의 운동 모델링 및 시험을 통한 계수 조정)

  • This paper presents the dynamic model of an AUV called HW200 for underwater surveillance. The mathematical model of HW200 is briefly introduced, considering its shape. The maneuvering coefficients were initially estimated using empirical formulas and a database of vehicles with similar shapes. A motion simulator, based on Simulink of Mathworks, was developed to evaluate the mathematical model of the vehicle and to tune the maneuvering coefficients. The parameters were finely tuned by comparing the experimental results and simulated responses generated with the simulator by applying the same control inputs as the experiment. The velocity of HW200 in the tuning process was fixed at a constant forward speed of 1.83 m/s. Simulations with variable speed commands were conducted, and the results showed good consistency in the motion response, attitude, and velocity of the vehicle, which were similar to those of the experiment even under the speed variation. This paper also discusses the feasibility of its application to a model-based integrated navigation system (INS) using the auxiliary information on the velocities generated by the model.

Fumonisin $B_1$-induced Alteration of Sphingolipid Metabolism in $LLC-PK_1$ Cells ($LLC-PK_1$ 세포에서의 퓨모너신 $B_1$에 의해 유도된 스핑고리피드 대사)

  • The purpose of this study was to determine the effect of sulfinpyrazone on fumonisin $B_1$-induced elevation of free sphingoid bases in LLC-$PK_1$ cells. Fumonis ins are a family of mycotoxins produced by the fungi Fusarium moniliforme which is common contaminant in corn. Fumonisins are also potent inhibiors of sphingosine and sphinganine N-acyltransferases (ceramide synthases), key enzymes in sphingolipid metabolism resulting in the elevation of free sphinganine. The cytosolic concentration of fumonisin B1 was known to be closely proportional to the elevation of free sphinganine in LLC-PK1 cells [Yoo, H.-S., Norred, W.P., Wang, E., Merrill, A.H., Jr., and Riley, R.T. (1992) Toxicol. Appl.Pharmacol. 114. 9-15]. Sulfinpyrazone, an anion transport inhibitor, reduced the elevated level of free sphinganine resulting from fumonisin B1 inhibition of de novo sphingolipid biosynthesis. Fumonisin B1 at a concentration of 20${\mu}$M showed approximately 120pmol/$10^6$ cells relative to 3-10pmol/$10^6$ cells in control cultures, and sulfinpyrazone at a concentration of 200${\mu}$M partially reversed the increased level of free sphinganine induced by fumonisin $B_1$ down to normal level after exposure to fumonisin $B_1$ for 8 to 24hr. However, the reduced effect of sulfinpyrazone on the fumonisin $B_1$-induced elevation of intracellular sphinganine was not shown after 24hr. Fumonisin $B_1$ exposure to LLC-PK1 cells for 36 and 48hr showed approximately 74 and 80pmol per $10^6$ cells relative to 82 and 76pmol,respectively, in fumonisin $B_1$ plus sulfinpyrazone-treated cultures. Sulfinpyrazone-induced less elevation of free sphinganine in confluent cells after exposure to fumonisin $B_1$ suggested that either sulfinpyrazone may block the availability of fumonisin $B_1$ to cells or act on the fumonisin $B_1$ interaction with ceramide synthase.

  • Nowadays, as going to information society based knowledge, the informations are acquired, processed, serviced based digital environment. In surveying field, the trend have been changed from the analog foundation to the digital foundation. Also, aerial photogrammetry is being changed from analog aerial photogrammetry to digital aerial photogrammetry. In this paper, the analysis of accuracy is performed for the comparison of traditional aerial photogrammetry with digital aerial photogrammetry usign UltracamX in AT and Block Adjustment. As the results, Bundle adjustment in digital aerial photogrammetry with GPS/INS have more advantages than traditional independent adjustment in analog aerial photogrammetry. Digital aerial photogrammetry contributes the higher accuracy in AT and block adjustment more than analog aerial photogrammetry.

  • Recently, DC-DC converters significantly increase the total losses as rising switching frequency. Trnditional soft switching technique for reducing switching losses even increase voltage/Clment stress of switch In this paper, Resonant circuit for soft switching is connected in parallel with power stage and only operates just before tum-on of the main sWItch. Therefore, ills doesn't affect the total circuit QI'||'&'||'pound;ration. The object of tIns paper is to make the linearized equivalent loss mxleIs. and to analyze the total losses by experiment. ZVT-PWlvI converter designed with 170-260[V] input, 400[V] 5[A] output, and 100[kHz] switching frequency is tested respectively with 500[W], 1[kW], 1.5[kW], and 2[kW] loads. The total losses in input 220[V], 2[kW] load are analyzed by usirm the linearized equivalent loss models.

Inhibitory Effect of Caffeine on Carbachol-Induced Nonselective Cationic Current in Guinea-Pig Gastric Myocytes

  • In gastrointestinal smooth muscle, muscarinic stimulation by carbachol (CCh) activates nonselective cation channel current ($I_{CCh}$) which is facilitated by intracellular [$Ca^{2+}$] increase. Caffeine is widely used in experiments to mobilize $Ca^{2+}$ from intracellular stores. This study shows a strong inhibitory effect of caffeine on $I_{CCh}$ in guinea-pig gastric myocyte. In this study, the underlying mechanism of the inhibitory effect of caffeine was investigated. $I_{CCh}$ was completely suppressed by the addition of caffeine (10 mM) to the superfusing solution. Inhibition of $I_{CCh}$ by caffeine was not related to the intracellular cAMP accumulation which was expected from the phosphodiesterase-inhibiting effect of caffeine. The blockade of $InsP_3-induced$ $Ca^{2+}$ release by heparin had no significant effects on the activation of $I_{CCh}$. When the same cationic current had been induced by intracellular dialysis of $GTP[{\gamma}S]$ in order to bypass the muscarinic receptor, the inhibitory effect of caffeine was significantly attenuated. The results of this study indicate that both intracellular signalling pathways for $I_{CCh}$, proximal and distal to G-protein activation, are suppressed by caffeine. A major inhibition was observed at the proximal level.

  • The road safety depend on many road factors like vertical alignment, horizontal alignment and road cross section angle. These data are hardly to get with drawings, and the real data are differ from drawings because of road pavement overlay, etc. To get these data, so many time and cost are needed, moreover it is dangerous work in heavy traffic road. In this study we obtained the road safety data with RoSSAV(Road Safety Survey & Analysis Vehicle) of Korea Institute of Construction Technology in accordance with traffic flow, and make analysis of road safety with the vertical alignment, horizontal alignment and road cross section angle data. We derived the safety improvement method in Young-dong accident prone spot and described detail method in this paper.

Influence of Vehicle Vibration on Track Geometry Measurement (차량 진동이 궤도 선형 측정에 미치는 영향)

  • Track maintenance works based on track geometry recordings are essential to enhance the safety and comfort of railway transportation. Usually, the track irregularity has been measured by a special inspection trains which all were imported from abroad. Because the inspection train speed is limited under 160 km/h, it takes a long time to inspect railways and there is difficulty in daytime operation. To solve this problem, we started to develop a track geometry measuring system (TGMS) with measurement speed up to 300 km/h which can be installed in commercial vehicles such as HEMU-430X. In this paper, we introduce a newly developed inertial TGMS and propose two inertial navigation system (INS) algorithms (method A, B) for measuring track geometry. In order to investigate advantage and disadvantage of each algorithm, we performed vibration test of the TGMS, which was simulated by 6-axis shaking table. Through the vibration test, we analyzed the influence of vehicle vibration on the TGMS which will be installed on bogie frame. To the vibration test, two methods satisfied the required accuracy of track geometry measurement under the level of the actual vehicle vibration of HEMU-430X train. Theoretically, method A is sensitive to vehicle vibration than method B. However, HEMU-430X's bogie vibration frequency range is out of interest range of measurement system. Therefore, method A can also apply the HEMU-430X train.
