Effects of Onion Juice on Toxicity of Lead in Rat (양파즙 투여가 Rat의 납 독성에 미치는 영향)

  • In this study the onion juice (2%) in diet fed rats simultaneously ingested lead acetate 100mg/ kg (OP group) showed more increased weight gain than single lead treated rats (P group). The OP group had also improved in the hemoglobin contents and biochemical analyzed values of blood including GPT, blood urea nitrogen and alkaline phosphatase, which were elevated in case of P group rats. The Pb content in the rats liver of OP group was lower than in the rats liver of P group. In the histopathological findings of liver cell OP group rats did not show any signs of liver damage as observed in P group rats that had degenerated hepatocytes, followed sinusoidal dilatation, perivascular hemorrhage and some necrosis of hepatic cells accompanied by increased Kuffer cell bearing dark brown pigment. In conclusion 2% onion juice diet in rat have somewhat antidotic effects on the lead intoxicated rats.

Effect of Ethanol Intake on Blood Component in Broiler Chicks (알콜섭취가 성장기 닭의 혈액성분에 미치는 영향)

  • This study was performed to investigate the influence of mixtures of 0(control), 1, 2 and 3% ethanol and water respectively on growth and various blood parameters of growing broiler chicks. At the end of the 7 weeks' experiment, body weight gain in 1% ethanol group and liver weight in 3% ethanol group were significantly higher than those of control group. It was found that the levels of red blood cell, hemoglobin, hematocrit, and serum protein were within normal ranges. Serum GOT and r-GTP activities were significantly increased in 2 and 3% ethanol groups compared with those in control group. But serum CPT activity was slightily decreased in all ethanol groups. Serum LDH activity was increased in all ethanol groups compared with that In control group. Serum alkaline phosphatase was not affected by the ethanol. Serum glucose concentration in 3% ethanol group was significantly lower than that in control group.

The Hepatotoxicity and Testicular Toxicity Induced by Arecoline in Mice and Protective Effects of Vitamins C and E

  • Arecoline is a major alkaloid of areca nuts which are widely chewed by southeast Asian and it manifests various toxic effects in different organs of human and animals. In this work, mature mice were treated by vitamins C plus E, arecoline, or both daily for four weeks. The results showed that arecoline significantly increased the levels of serum alkaline phosphatase (ALP), glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT) and significantly decreased the levels of reduced glutathione (GSH), glutathione-S-transferase (GST), superoxide dismutase (SOD) and catalase (CAT) in the liver tissues. Additionally, the body weight, testis weight, sperm counts, motility and normal sperms also were significantly decreased. The supplement of vitamins C and E can bring the activities of ALP and GPT to normal levels and partially restore the sperm counts compared to the arecoline-treated group but have no other positive effects. In conclusion, the vitamins C and E partially attenuated the arecoline-induced hepatotoxiciy but basically had on protective effects against the arecoline-induced testicular toxicity.

Effect of Prunus mume Extract on Gastric Secretion in Rats and Carbon Tetrachloride Induced Liver Damage of Rabbits (매실추출물이 흰쥐의 위액분비 및 사염화탄소로 유발시킨 가토의 간장장애에 미치는 영향)

  • The aim of this study is to investigate effects of Prunus mume extract on gastric secretion in rats and experimentally carbon tetrachloride-induced liver damage of rabbits. The groups of Prunus mume extract showed excellent increasing effect in total acidity and peptic activity and decreasing effect on gastric volume than the control group whereas clear differences were not found between sample adminstered group and control group in gastric pH. In liver function test Prunus mume extract exhibited rapid recuperation of liver function that is sGPT activity showed an apparant decreasing effect from the 6th day total cholesterol and alkaline phosphatase level from the 10th day respectively compared to the control group in carbon tetrachloride-intoxicated rabbits. It is, is suggested that Pruns mume extract can be used as the stomachics to promote the gastric digestion by increasing the peptic activity and health food to shorten the recovery time from liver diseases.

Chemopreventive Potential of Angelicae gigantis Radix Aqua-acupuncture Solution (당귀 약침액의 암예방 효과)

  • Angelicae gigantis Radix aqua-acupuncture solution (AGRAS) and Angelicae gigantis Radix water-extracted solution (AGRWS) were prepared and tested for their organ toxicities and chemopreventive potentials. The organ-toxicity of AGRAS to male ICR mice was studied by the measurements of glutamic oxaloacetic transaminase (GOT), glutamic pyruvate transaminase (GPT), lactate dehydrogenase (LDH) and alkaline phosphatase (ALP-s) activities after injection of AGRAS for 7 days. The activities of GOT GPT and LDH were decreased, but the activity of ALP-s was not changed with AGRAS. When AGRAS was administered once daily for 10 days before the tumor implantation, AGRAS exerted antitumor activity by inhibiting the growth of Ehrich ascites tumor cells (EATC) in viva. The inductions of quinone reductase (QR), glutathione (GSH) and glutathione S-transferase (GST) and inhibition of polyamine metabolism were tested for the chemopreventive potentials of AGRAS and AGRWS. AGRAS was potent inducer of QR activity in murine hepatoma Hepalclc7 cells. In cultured rat Ac2F cells, AGRAS was also significantly induced QR activity GSH levels were increased about 1.3 fold with AGRAS. In addition the activity of GST was increased about 2.5 fold with AGRAS at the concentration of $0.1{\;}{\times}{\;}$. The effects of AGRAS and AGRWS were tested on the growth of Acanthamoeba castellanii. Proliferation of Acanthamoeba castellanii in a broth medium was inhibited by AGRAS and AGRWS at the concentration of $1{\;}{\times}{\;}and{\;}5{\;}{\times}{\;}$, respectively: These results suggest that AGRAS has chemopreventive potential by inducing QR activity increasing GSH and GST levels and inhibition of polyamine metabolism.

Protective effects of Jucha-whan on liver injury of guinea pigs induced by $CCl_4$ and d-galactosamine (주차환(舟車丸)이 $CCl_4$ 및 d-galactosamine으로 유발된 흰쥐의 간손상(肝損傷)에 미치는 영향)

  • Objective : In order to investigate the curative effect of Jucha-whan on the protective liver of guinea pigs induced by $CCl_4$ and d-galactosamine, serum transaminase(GOT, GPT), lactic dehydrogenase(LDH), alkaline phosphatase(ALP), glutathione S-transferase(GST), superoxide dismutase(SOD) were used to measure enzyme activities and lipid peroxide level. Method : The subject animals were divided into 5 groups; a control group(untreated), a subject group(administered with 0.9% Saline solution), a sample I (500mg/kg administered), sample II group (1000mg/kg administered), positive control group(administered with 200mg/kg silymarine). Result : The inhibitory effects on the serum GOT, LDH, ALP, SOD and Lipid peroxide level activities in protective liver of mice induced by $CCl_4$ were noted both in the sample I group and sample II. The inhibitory effects on the serum GPT activities in protective liver of guinea pigs induced by $CCl_4$ were noted in sample II group, but it was not noted in the sample I. The inhibitory effects on the GST activities in protective liver of guinea pigs induced by $CCl_4$ were not noted in both sample I and sample group II. The inhibitory effects on the serum GOT, GPT activities in protective liver of guinea pigs induced by d-galactosamine were noted in both sample I and sample II, but it was not recognizable statistically. The inhibitory effects on the serum LDH activities in protective liver of guinea pigs induced by d-galactosamine were noted in sample II, but it was not noted in sample I group. Conclusion : According to the above results, it is considered that Jucha-whan has treatment effects on liver injury in guinea pigs induced by $CCl_4$ and d-galactosamine. So it is required to study about the actions of mutual relation of medicines and patho-mechanism through experiment.

Antitumor and Immunomodulatory Effects of Glycyrrhizae Radix Aqua-acupuncture Solution (감초 약침액의 항암 및 면역활성에 미치는 영향)

  • Glycyrrhizae Radix aqua-acupuncture solution (GRAS) and Glycyrrhizae Radix water-extracted solution (GRWS) were prepared and tested for organ toxicities, antitumor activities, and immunomodulatory effects. The organ-toxicity of GRAS to male ICR mice was studied by the measurements of glutamic oxaloacetic transaminase (GOT), glutamic pyruvate transaminase (GPT), lactate dehydrogenase (LDH), and alkaline phosphatase (ALP-s) activities after injection of GRAS for 7 days. The activities of GOT, GPT, LDH, ALP-s were decreased with GRAS. It was shown to possess considerable toxicity toward various tumor cell lines. Concentration of GRAS at 1.5g/ml and 3g/ml resulted in more than 80% inhibition of growth in Ehrlich ascites tumor cells (EATC), Hepa1c1c7, and HeLa cells. Toxicity of GRAS to A549 revealed that 68% inhibition of growth. GRWS at the concentration of 3g/ml showed more than 80% inhibition of growth with EATC, Hepalclc7, A549 and HeLa. In morphological study, the number of cells were decreased, and the shape of cells was round-form in EATC, Hepalclc7, A549 and HeLa cells with GRAS. Administration of GRAS inhibited the growth of EATC in vivo. Mice given EATC at 1.5g/ml or 0.3g/ml GRAS had 16.7% to 50% survival after 21 days. GRAS increased the proliferation of T and B cells and the cytolytic activity of purified T cell. The biosyntheses of nucleic acid and protein of EATC, Hepalclc7, A549 and HeLa cells were inhibited by GRAS.

Protective Effect of Pterocarpus santalinus on Galactosamine Induced Liver Damage

  • The present study was carried out to investigate the hepatoprotective effect of the extract of Pterocarpus santalinus Linn on acute hepatotoxicity induced in Wistar albino rats by a single dose of Galactosamine (400 mg/kg). Suspensions of methanolic extract of heartwood of P. santalinus (200 and 400 mg/kg) in 0.3% Carboxy Methyl Cellulose (CMC) were administered p.o. to experimental animals and hepatoprotective activity was monitored by estimating aspartate amino transferase (ASAT, GOT), alanine amino transferase (ALAT, GPT), alkaline phosphatase (ALP), total bilirubin (TB), lactate dehydrogenase (LDH), total cholesterol (TC), triglycerides (TGL), albumin, total protein (TP) levels. The methanolic extract significantly reduced the elevation of serum transaminases and alterations of biochemical parameters induced by hepatotoxin, and alleviated the degree of liver damage. The results were supported by histopathological studies of liver samples showing regeneration of hepatocytes in treated animals. Silymarin (25 mg/kg), a known hepatoprotective drug was used for comparison. Based on the results obtained, it can be concluded that P. santalinus exerts hepatoprotective activity and may serve as a useful adjuvant in several clinical conditions associated with liver damage.

Studies on the potential hazard of dichlorvos-resin vaporizing insecticide in human and domestic fowl (Dichlorvos 증기(蒸氣)의 지속적(持續的) 흡입(吸入)이 인체(人體) 및 가계(家鷄)에 미치는 영향)

  • Safety study of the continuous releasing dichlorvos-resin insecticide $(Mopari^{\circledR})$ was conducted in human volunteers and domestic fowls. For the purpose, the potential hazards in using the insecticide were observed in terms of the inhibition of plasma cholinesterase activity and the changes in the liver function (GOT, GPT, Alkaline phosphatase, Bilirubin, Thymol turbidity), the blood picture (RBC, WBC with differential count, Hemoglobin, Hematocrit and ESR) and the urine picture (sugar, albumin, pH and microscopic findings) in 40 healthy adult volunteers and 60 leghorn domestic fowls. In case of the human study the observation was continued for 2 months during the application of the insectiside ($1{\sim}3$ solid formulations/$30m^3$) in the living rooms of ordinary Korean dwelling houses or in the office. In the animal test, however, 1 to 5 solid formulations of the insecticide were applied in the fowl cage of $9.2m^3$ for 5 weeks. Any significant inhibition of the plasma cholinesterase activity was not observed in both the human volunteer and the fowl throughout the experimental period. And the liver function as well as the blood and urine pictures were also not changed after exposure to the vaporizing insecticide. It is considered from the result that the amount of dichlorvos released into the air by the continuous vaporizing dichlorvos-resin insecticide presents no significant hazardous effect on humans or animals in the present experimental condition.

Effect of Cadmium Dose Injection on Peroxidative Damage in Rat Liver (카드뮴 투여가 흰쥐 간조직의 과산화적 손상에 미치는 영향)

  • In order to investigate the liver damage and hepatic protective systems in cadmium(Dd) administered rats, five different levels of Cd were injected intraperitoneally to male rats of sprague-Dawley strains weighing $250{\pm}15g.$ Levels of daily Cd administration were 0(control), 0.625(A), 1.25(B), 2.5(C) and 5mg(D)/kg of body weight and single inhection per day was done for consecutive two days. With increasing Cd dosed, serum glutamic oxaloacetic transaminase, glutamic pyruvic transaminase and alkaline phosphatase activities were increased. And at the same time, hepatic reduced glutathione contents were decreased, whereas the levels of oxidized form were increased. Liver lipid peroxide levels of A, B, C and D groups were 1.1, 1.5, 1.8 activities and vitamin E contents were progressively reduced in accordance with the increase in Cd dose. However, liver superoxide dismutase activities were not different between control and A group although it was higher in B and lower in C and D groups compared with control.
