Characterization of starch and gum arabic-maltodextrin microparticles encapsulating acacia tannin extract and evaluation of their potential use in ruminant nutrition

  • Objective: The use of tannin extract and other phytochemicals as dietary additives in ruminants is becoming more popular due to their wide biological actions such as in methane mitigation, bypass of dietary protein, intestinal nematode control, among other uses. Unfortunately, some have strong astringency, low stability and bioavailability, and negatively affecting dry matter intake and digestibility. To circumvent these drawbacks, an effective delivery system may offer a promising approach to administer these extracts to the site where they are required. The objectives of this study were to encapsulate acacia tannin extract (ATE) with native starch and maltodextrin-gum arabic and to test the effect of encapsulation parameters on encapsulation efficiency, yield and morphology of the microparticles obtained as well as the effect on rumen in vitro gas production. Methods: The ATE was encapsulated with the wall materials, and the morphological features of freeze-dried microparticles were evaluated by scanning electron microscopy. The in vitro release pattern of microparticles in acetate buffer, simulating the rumen, and its effect on in vitro gas production was evaluated. Results: The morphological features revealed that maltodextrin/gum-arabic microparticles were irregular shaped, glossy and smaller, compared with those encapsulated with native starch, which were bigger, and more homogenous. Maltodextrin-gum arabic could be used up to 30% loading concentration compared with starch, which could not hold the core material beyond 15% loading capacity. Encapsulation efficiency ranged from $27.7%{\pm}6.4%$ to $48.8%{\pm}5.5%$ in starch and $56.1%{\pm}4.9%$ to $64.8%{\pm}2.8%$ in maltodextrin-gum arabic microparticles. Only a slight reduction in methane emission was recorded in encapsulated microparticles when compared with the samples containing only wall materials. Conclusion: Both encapsulated products exhibited the burst release pattern under the pH conditions and methane reduction associated with tannin was marginal. This is attributable to small loading percentages and therefore, other wall materials or encapsulation methods should be investigated.

항균활성 천연물질을 이용한 반추위 메탄저감용 친환경 첨가제 개발 (Development of an Environmental Friend Additive Using Antibacterial Natural Product for Reducing Enteric Rumen Methane Emission)

  • 본 연구는 천연물질에서 유래한 반추위 메탄저감용 친환경 첨가제 개발을 위해 각기 다른 종균을 이용하여 발효한 세신 추출물의 항균활성, 항산화활성 및 in vitro 반추위 발효시험을 체계적으로 실시하였다. 접종된 균주들의 성장효율을 알아보기 위해 실시한 생균수측정 결과 L. curvatus NJ40 균주에서 유의적으로 높은 균주성장을 나타냈다(p<0.05). 항균활성측정 결과는 대조구 대비 L. curvatus NJ40 및 L. plantarum NJ45 균주와 발효된 세신 추출물이 병원균에 대한 항균효과를 나타내는 것으로 조사되었다. In vitro 반추위 발효실험에 세신 및 발효 세신 추출물을 적용한 결과, 휘발성 지방산 생성량 대비 반추위 메탄 저감효과가 나타났다. 특히 반추위 미생물 활력 및 사료이용 효율을 대표할 수 있는 휘발성 지방산 생성효율에 대한 부정적 효과 없이, 오히려 휘발성지방산 생성효율을 향상시키면서 반추위 메탄 저감효과를 가져올 수 있는 것으로 나타났다.

Effects of Combination of Nitrate with ${\beta}$1-4 Galacto-oligosaccharides and Yeast (Candida kefyr) on Methane Emission from Sheep

  • The objective of the present study was to determine whether ${\beta}$1-4 galacto-oligosaccharides (GOS) and Candida kefyr combined with nitrate as manipulators could suppress rumen methanogenesis without nitrate poisoning in sheep. Four rumen fistulated wethers were allocated to a $4{\times}4$ Latin square design. Nitrate (1.3 g $NaNO_3$ $Kg^{-0.75}$body weight) with and without GOS and Candida kefyr were administered into the rumen through fistula as a single dose 30 min after the morning meal. GOS and Candida kefyr were supplemented by sprinkling onto the feed and through rumen fistula, respectively. The four treatments consisted of saline, nitrate, nitrate plus GOS and nitrate plus GOS plus Candida kefyr. Physiological saline was used as the control treatment. Compared to saline treatment, the administration of nitrate alone resulted in a very marked decrease in rumen methanogenesis and an increase in rumen and plasma nitrite production and blood methaemoglobin formation consequently causing a decline in oxygen consumption, carbon dioxide production and metabolic rate. When compared to nitrate alone, the simultaneous administration of nitrate with GOS decreased nitrite accumulation in rumen and plasma and nitrate-induced methaemoglobin, while retaining low methane production. However, GOS could not fully restore metabolic parameters reduced by nitrate. When compared to the simultaneous administration of nitrate with GOS, the simultaneous administration of nitrate with GOS plus Candida kefyr lowered rumen methanogenesis to a negligible level, but did not decrease rumen and plasma nitrite accumulation as well as blood methaemoglobin formation. Thus, these results suggest that combination of nitrate with GOS may be a potent manipulator to suppress rumen methanogenesis with abating the hazards of nitratenitrite toxicity in ruminants.

Conjugated Linoleic Acid in Rumen Fluid and Milk Fat, and Methane Emission of Lactating Goats Fed a Soybean Oil-based Diet Supplemented with Sodium Bicarbonate and Monensin

  • A metabolic study was conducted with four ruminally-cannulated lactating goats (Saanen, 29 weeks lactation, 65${\pm}$5 kg) in a 4${\times}$4 Latin square design with 4 dietary treatments. The goats were fed a basal mixed diet consisting of 80% concentrate and 20% chopped rye grass hay (DM basis, CON). The goats were also fed the CON diet supplemented with soybean oil at a 5% level of the concentrate (SO), the SO diet supplemented with 0.5% of sodium bicarbonate (SO-B) or the SO-B diet supplemented with 30 ppm monensin (SO-BM). The goats were housed in individual pen and the study was conducted for 8 weeks. An increased molar proportion of propionate (C3) was observed at 1 h (p<0.003) and 6 h (p<0.029) post-feeding from all the supplemented diets. Calculated methane emission was markedly decreased prior to morning feeding (p<0.01), and at 1 h (p<0.05) and 6 h post-feeding (p<0.05) in goats fed the supplemented diets. All the supplements increased (p<0.0001) cis9, trans11-CLA content in rumen fluid. Concentrations of both cis9, trans11-CLA (p<0.0001) and trans10, cis12-CLA (p<0.026) were also increased in the milk fat of lactating goats fed the supplemented diets. The SO-B and SO-BM diets further increased CLA content in goat milk compared to the SO diet. All supplements increased unsaturated (UFA, p<0.002), monounsaturated (MUFA, p<0.002) and polyunsaturated fatty acids (p<0.014) and reduced SFA to UFA ratio (p<0.023). The concentration of MUFA was even greater (p<0.002) for SO-BM than for the SO-B diet. In conclusion, feeding soybean oil (5% of concentrate) to lactating goats was a useful way to improve milk fat and to improve fatty acid profile in the milk by increasing potentially healthy fatty acids such as CLA. Supplementation of sodium bicarbonate or sodium bicarbonate with monensin to the soybean oil-based diet increased CLA content further in goat milk. Supplementation of soybean oil may be an effective method to reduce methane emission in lactating goats.

Effects of Levels of Feed Intake and Inclusion of Corn on Rumen Environment, Nutrient Digestibility, Methane Emission and Energy and Protein Utilization by Goats Fed Alfalfa Pellets

  • The effect of high and low level of feed intakes on nutrient digestibility, nutrient losses through methane, energy and protein utilization by goats fed on alfalfa (Medicago sativa L.) pellets based diets was investigated in this study. Twelve castrated Japanese goats were employed in two subsequent digestion and metabolism trials. The goats were divided into three groups, offered three diets. Diet 1 consisted of 100% alfalfa pellet, Diet 2 was 70% alfalfa pellet and 30% corn, and Diet 3 was 40% alfalfa pellet and 60% corn. The two intake levels were high (1.6 times) and low (0.9 times) the maintenance requirement of total digestible nutrients (TON). Rumen ammonia nitrogen ($NH_3$-N) level of Diet 1 was lower (p<0.001) compared to Diets 2 and 3, but the values were always above the critical level (I50 mg/liter), The pH values of rumen liquor ranged from 6.02 to 7.30. Apparent digestibility of nutrient components did not show differences (p>0.05) between the two intake levels but inclusion of corn significantly altered the nutrient digestibility. Diet 3 had highest (p<0.001) dry matter (DM), organic matter (OM), ether extract (EE) and nitrogen fee extract (NFE) digestibility followed by the Diet 2 and Diet 1. The crude protein (CP) digestibility values among the three diets were in a narrow range (70.1 to 70.8%). Crude fiber (CF) digestibility for Diet 3 was slight higher (p>0.05) than that for other two diets. When alfalfa was replaced by corn, there were highly significant (p<0.001) increases in DM, OM, EE and NFE apparent digestibility and a slight increase in the CF digestibility (p>0.05). There were no differences (p>0.05) in energy losses as methane ($CH_4$) and heat production among the diets but energy loss through urine was higher for the Diet 1. The total energy loss as $CH_4$ and heat production were higher for the high intake level but the energy loss as $CH_4$ per gram DM intake were same (0.305 kcal/g) between the high and low intake level. Retained energy (RE) was higher for Diet 3 and Diet 2. Nitrogen (N) losses through feces and urine were higher (p<0.001) for Diet 1. Consequently, N retention was lower (p>0.05) for Diet 1 and higher in Diets 3 and 2. It is concluded that inclusion of corn with alfalfa increased the metabolizable energy (ME) and RE, and retained N through reducing the energy and N losses. The high level of intake reduced the rate of nutrient losses through feces and urine.

The effect of feeding high fat diet to beef cattle on manure composition and gaseous emission from a feedlot pen surface

  • Background: Dietary manipulation is a common practice to mitigate gaseous emission from livestock production facilities, and the variation of fat level in the diet has shown great influence on ruminal volatile fatty acids (VFA) and enteric methane generation. The changes in dietary fat levels influence rumen chemistry that could modify manure nutrient composition along with odor and gaseous emissions from manure management facilities. Methods: A field experiment was carried out on beef cattle feedlots to investigate the effect of four levels of dietary fat concentrations (3 to 5.5 %) on the manure composition and gaseous emissions (methane-$CH_4$, nitrous oxide-$N_2O$, carbon dioxide-$CO_2$ and hydrogen sulfide-$H_2S$) from the feedlot pen surface. The experiment was carried out over a 5-month period from June to October during North Dakota's summer-fall climatic condition. Air and manure sampling was conducted five times at a 20-30 day intervals. Results: Overall, this research indicated that fat levels in diet have no or little effect on the nutrient composition of manure and gaseous emission from the pens with cattle fed with different diet. Though significant variation of gaseous emission and manure composition were observed between different sampling periods, no effect of high fat diet was observed on manure composition and gaseous emission. Conclusions: It can be concluded that addition of fat to animal diet may not have any impact on gaseous emission and manure compositions.

Evaluation of Megasphaera elsdenii supplementation on rumen fermentation, production performance, carcass traits and health of ruminants: a meta-analysis

  • Objective: This study was conducted to evaluate the use of Megasphaera elsdenii (M. elsdenii) as a probiotic on rumen fermentation, production performance, carcass traits and health of ruminants by integrating data from various related studies using meta-analysis. Methods: A total of 32 studies (consisted of 136 data points) were obtained and integrated into a database. The parameters integrated were fermentation products, rumen microbes, production performance, carcass quality, animal health, blood and urine metabolites. Statistical analysis of the compiled database used a mixed model methodology. Different studies were considered random effects, while M. elsdenii supplementation doses were considered fixed effects. p-values and the Akaike information criterion were employed as model statistics. The model was deemed significant at p<0.05 or had a tendency to be significant when p-value between 0.05<p<0.10. Results: Supplementation with M. elsdenii increased (p<0.05) some proportion of fermented rumen products such as propionate, butyrate, isobutyrate, and valerate, and significantly reduced (p<0.05) lactic acid concentration, acetate proportion, total bacterial population and methane emission. Furthermore, the probiotic supplementation enhanced (p<0.05) livestock production performance, especially in the average daily gain and body condition score. Regarding the carcass quality, hot carcass weight and carcass gain were elevated (p< 0.05) due to the M. elsdenii supplementation. Animal health also showed improvement as indicated by the lower (p<0.05) diarrhoea and bloat incidences as well as the liver abscess. However, M. elsdenii supplementation had negligible effects on blood and urine metabolites of ruminants. Conclusion: Supplementation of M. elsdenii is capable of decreasing ruminal lactic acid concentration, enhancing rumen health, elevating some favourable rumen fermentation products, and in turn, increasing production performance of ruminants.

Changes in Microbial Diversity, Methanogenesis and Fermentation Characteristics in the Rumen in Response to Medicinal Plant Extracts

  • This study evaluated the in vitro effect of medicinal plant extracts on ruminal methanogenesis, four different groups of methanogens and ruminal fermentation characteristics. A fistulated Holstein cow was used as a donor of rumen fluid. Licorice and mugwort extracts (Glycyrrhiza uralensis and Artemisia capillaris, 0.5% and 1% of total substrate DM, respectively), previously used as folk remedies, were added to an in vitro fermentation incubated with buffered-rumen fluid. Total gas production in Glycyrrhiza uralensis extract treatment was not significantly different between treatments (p<0.05) while total gas production in the Artemisia capillaris extract treatment was lower than that of the control. Artemisia capillaris extract and Glycyrrhiza uralensis extract reduced $CH_4$ emission by 14% (p<0.05) and 8% (p<0.05), respectively. Ciliate-associated methanogens population decreased by 18% in the medicinal plant extracts treatments. Medicinal plant extracts also affected the order Methanobacteriales community. Methanobacteriales diversity decreased by 35% in the Glycyrrhiza uralensis extract treatment and 30% in the Artemisia capillaris extract treatment. The order Methanomicrobiales population decreased by 50% in the 0.5% of Glycyrrhiza uralensis extract treatment. These findings demonstrate that medicinal plant extracts have the potential to inhibit in vitro ruminal methanogenesis.

할로겐 화합물의 첨가가 반추위 발효성상과 메탄생성에 미치는 영향 (Effects of Halogenated Compounds on in vitro Fermentation Characteristics in the Rumen and Methane Emissions)

  • 본 연구는 할로겐 화합물의 첨가가 in vitro 상의 반추위 발효성상과 메탄생성에 미치는 영향에 대한 효과를 규명하고자 실시하였다. Italian rye grass 및 배합사료를 6:4의 비율로 급여한 반추위 cannula가 시술된 홀스타인에서 반추위액을 채취하여 사용하였고, 채취된 반추위액은 분쇄된 timothy (대조구; C)에 bromochloromethane (BCM구), 2-Bromoethanesulfonic acid (BES구), 3-Bromopropanesulfonic acid (BPS구), chloroform (CLF구) 및 Pyromellitic diimide (PMDI구)의 5가지 할로겐 화합물을 각각 1 ppm씩 첨가하여 in vitro 배양하였다. pH는 6.72에서 6.25 정도로 배양시간이 경과함에 따라 낮아지는 경향을 나타내었고, 배양 48시간에는 처리구간 차이가 없었다. 배양 48시간 후의 총 가스 발생량은 처리구간 유의적인(p<0.05) 차이는 없었고, BPS구를 제외한 모든 처리구에서의 메탄 발생량은 대조구에 비해 유의적으로(p<0.05) 감소하였다. 배양 12시간에서의 총 휘발성 지방산 및 propionic acid의 발생량은 처리구가 대조구에 비해 유의적으로(p<0.05) 높았다. 본 실험의 결과, 할로겐 화합물의 첨가는 반추위 내의 pH, 건물 소화율, 미생물 수 및 총 가스 발생량의 발효 성상에는 영향을 주지 않으면서 메탄의 발생량이 감소 되었다. 실험 결과를 종합해 보면, 할로겐 화합물 첨가는 반추위 내 pH, 가스 발생량, 반추위 미생물 성장량 및 propionic acid 모두 증가하였으며, 반추위내 메탄생성을 억제하였다. 앞으로 할로겐화합물과 다른 메탄억제 물질과 혼합하여 반추위 내 메탄생성 억제에 관한 구체적인 연구가 필요한 것으로 생각된다.

질산염 화합물 함유 식물 추출물이 in vitro 반추위 발효성상과 메탄 발생에 미치는 영향 (Effects of Nitrate-rich Plant Extracts on the in vitro Ruminal Fermentation and Methane Production)

  • 본 시험은 질산염함유 식물 추출물을 이용하여 in vitro 반추위 발효성상 및 반추위 메탄 발생에 미치는 영향을 알아보기 위해 수행되었다. 공시재료는 감자, 당근, 배추, 상추 및 시금치 추출물을 사용하였으며, 발효 기질은 2mm로 분쇄된 timothy 0.3g을 사용하였다. In vitro 실험은 조사료(timothy) 및 배합사료를 6:4의 비율로 급여한 반추위 cannnula가 시술된 한우 암소에서 채취하였다. 반추위액과 McDougall's buffer를 1:2의 비율로 혼합한 발효액을 0.3g timothy와 식물 추출물이 담긴 50mL serum bottle에 혐기상태로 20mL 분주한 뒤, 39℃에서 9, 12, 24, 48시간 동안 발효하였다. pH는 발효 시간이 증가함에 따라 점점 감소하며, 6.31~6.96으로 정상범위였다. 건물 소화율은 9시간 배추 추출물구에서 유의적으로 낮았고, 암모니아 농도는 감자, 배추, 상추 추출물구에서 대조구에 비해 첨가구에서 유의적으로(p<0.05) 낮았으며, 반추위 미생물 성장량은 발효 24시간대의 당근 추출물구에서 높았다(p<0.05). Acetate와 propionate 농도는 대조구에 비해 첨가구에서 유의적으로 낮았으며(p<0.05) butyrate 농도는 첨가구간에 발효시간별로 각각 다른 양상을 보였다. 총 가스 발생량은 발효 12시간 및 24시간대에서 배추, 상추, 시금치 추출물구에서 유의적으로(p<0.05) 낮았으며, 메탄 발생량은 감자, 배추, 시금치 추출물구가 대조구에 비해 유의적으로(p<0.05) 낮았고, 이산화탄소 발생량 또한 첨가구에 비해 대조구에서 유의적으로(p<0.05) 낮았다. 결과적으로 본 시험에 사용된 5종의 질산염 함유 식물 추출물은 반추위 발효에 악영향을 미치지 않으면서 메탄저감 효과를 나타내었다. 특히 배추 추출물은 반추위내 메탄발생을 저감시킬 수 있는 식물추출물로서 가능성이 높은 것으로 사료된다.
