In Vitro Mass Propagation and Soil Adjastment of Zanthoxylum piperitum var. inerme Makino through Apical Meristem Culture (生長點 培養에 依한 민초피나무(Zanthoxylum piperitum var. inerme Makino)의 器內 大量 增殖 및 土壤 活着)

  • This study was conducted to investigate the effect of growth regulators and medium composition on the growth of each stage in apical meristem culture for mass propagation of Zanthoxylum piperitum var. inerme Makino. The source material, shoot tip segments were taken from three-years old graft trees. Apical meristems were cultured in vitro on basal MS, GD, WS, half strength MS(1/2MS) and half strength GD(1/2GD) media supplemented with various concentrations for growth regulators(BA, IBA) and inorganic nutrients. The results summarized are as follows: 1. In culture establishment stage, ratio of culture establishment was 96.7% and the best resuit was obtained using MS medium supplemented with 1.0mg/l BA and 0.2mg/l IBA. 2. In shoot multitication stage, both shoot multiplication and growth were achieved in average 5.6cm. These results were obtained on in MS medium supplemented with 1.0mg/l BA and 0.2mg/l IBA. 3. In roothing stage, phloroglucinol(PG) acted as IBA synergist in root initiation. The most faverable combinations for root development was half-strength MS medium supplemented with 162mg/l PG and 0.2mg/l IBA, and ratio of rooting was 58.0%. 4. In Vitro formed plantlets were transplanted to paper pots in greenhouse with 85% of relative humidity. 96% of survival rate was obtained from artificial soil mix having same volume of sand, vermiculite, peat, and soil.

  • The cautious blasting works had been used with emulsion explosion electric M/S delay caps. Drill depth was from 3m to 6m with Crawler Drill ø70mm on the calcalious sand stone (soft-moderate-semi hard Rock). The total numbers of fire blast were 88 round. Scale distance were induces 15.52-60.32. It was applied to propagation Law in blasting vibration as follows. It's divided into under loom and over 100m distance because the frequency is verified by the distance from blast site. Empirical equation of cautious blasting vibration is as follows. And Drilling pattern on the vibration levels, it is necessary to carry out more tests.

  • This study was undertaken to know the possibility of in vitro propagation of Stevia through axillary bud culture and the results indicated that: (1) Addition of NAA (0.01-0.05 mg/l) alone on Murashige-Skoog basal medium promoted shoot differentiation and growth rate. And also additional of kinetin of 0.5-1.0 mg/1 alone showed the same trend as that of NAA: (2) Addition of both NAA (0.01-0.05 mg/l) and kinetin (0.5-1.0mg/l) to MS medium promoted better shoot formation. (3) Shoot differentiation and growth were better on the full salt strength of MS medium (1X MS) than that of half strength ( $\frac{1}{2}$MS), while their effects were reversed for root differentiation

  • This experiment was carried out to obtain the basic information on cultivation of Gentiana scabra BUNGE. Optimum temperature for germination, planting time effect of chemicals on rooting for cutting propagation, effect of cutting on root yield, and content of gentiopicroside were investigated. The germination ratio was highest at $25^{\circ}C$ but lower at $10,\;15,\;20\;,30^{\circ}C.$ The optimum planting time was March 20 the cutting in May and NAA treatment were effective on rooting for cutting propagation. Inhibition of flowering by cutting was effective for increasing root yield but not effective for gentiopicroside content of root.

  • This research was carried out to establish the large quantity propagation system for Kalopanax pictus using root cutting. The results are as follows. As there was no significant difference in the promotion of adventitious shoot development from the root cuttings of Kalopanax pictus among kinds (IBA, NAA and Kinetin) and concentrations (0 to 2000 mg/L) of growth regulators, no application is recommended, which showed more than 90% of shoot development. Horizontal and vertical placement of rooting showed more than 93% of shoot occurrence, but the latter showed more desirable growth traits in the number of shoot development, number of leaf, petiole length, and leaf length and leaf width Polarity was observed and thus normal vertical placement of root cutting is preferred. However, root cuttings placed upside down showed the shoot development at the distal end, which indicates further sophisticated test is necessary to investigate this phenomenon.

  • To establish the culture practices for mass propagation of Epimedium koreanum, root propagules were preserved with different methods. Bud emergence rate and number of multiple buds were the highest in root segments obtained from roots with buds which were preserved in open-burial. Plant height, leaf area, and number of new buds were the highest in root segments obtained from roots preserved with stratification.

Growth of Salix gracilistyla Miq. Originated from Provenance in Gangwon-do, Korea for Bioenergy Resource

  • Salix gracilistyla has a characteristic of fast growth especially in the river side or infertile soil, showing a lot of lateral branches and a strong root system among the Salix. spp. On the basis of results observed, we provide several growth characteristics of S. gracilistyla among some selected provenances. In two-year results, root collar diameter ranged from 33.5 mm to 56.7 mm. Taeback showed the lowest value, and Jeongseon and Hoengseong were higher than the other areas in the traits above. Height growth was the lowest in Samcheok and the highest in Wonju. To examine growth and development of lateral branches is important because S. gracilistyla has a lot of lateral branches which account for high biomass production. Length of lateral branch showed high variation, in which the highest number was observed in Yeongwol and the lowest in Samcheok. Many lateral branches were shown in Wonju. Preliminary study on biomass resources evaluation, we analyzed correlation between volume and length, and volume and root diameter. High correlation was observed between root diameter and plant volume. And we compared the observed volume with the caculated volume by regression formula. To evaluate Salix species as biomass resources, it is needed to survey growth characteristics continuously and adopt selection of individuals or families of high biomass and caloric values by reliable data. In addition, Salix spp. can be also applicable to selection breeding regime because of feasible propagation and fast growth.

Comparison of Chemical Constituents of Upland Wasabia japonica Matsum Grown by Different Propagation Methods (번식방법(繁殖方法)에 따른 밭고추냉이의 성분비교(成分比較))

  • Fatty acid, amino acid and organic acid contents were analyzed by gas chromatography, amino acid analyzer and high pressure liquid chromatography, respectively, in order to compare the chemical constituents of upland wasabi plant propagated by seed and auxiliary bud. Total fatty acid content and fatty acid composition of upland wasabi were not affected by the propagation methods. Generally, fatty acid content of leaf was higher than that of other parts such as enlarged stem, petiole, peduncle and root. In fatty acid composition, leaf had highest content of linolenic acid, 60-63%, in plant propagated by both seed and auxiliary bud, followed by palmitic acid, oleic acid and linoleic acid in the order. Similarly, total amino acid content was not influenced by propagation methods but plant propagated by seed had higher amount of amino acid content in enlarged stem, petiole and root than that by auxiliary bud -propagated plant. A total of 17 amino acids including 7 essential amino acids were identified in both seed and auxiliary bud propagations. Like total fatty acid content and fatty acid composition, leaf contained high amount of amino acids, especially glutamic acid, asparatic acid and leucine. Organic acid contents were similar in both propagation methods. The major organic acid in upland wasabi was acetic acid (60.0-78.2%), followed by succinic acid (9.9-29.7%) and malic acid (2.9-7.9%). Maleic acid content was least (0.5-2.6%). The result indicates that content and composition of fatty acid, amino acid, and organic acid in upland wasabi were not influenced by propagation methods.

A Study of Pull-out strength increasement by root of grasses (식물뿌리에 의한 인발력 증가에 관한 연구)

  • In this study, a pull-out strength increasement of grasses was tested in field and the relationship between a weight of root and pull-out strength was established. The tested grasses were commmon reed, reed and sedge which were cultivated by mat-type like turf and used for revegetation of bank. The shear stress of soil at a section can be discribed as a function of root area and pull-out strength, therefore the result of this study will be used as a foundational data for reinforcing the shear stress of the revegetated bank. The heavier weight of root increased, the stronger shear stress was for all grasses. But the relationship between the weight and the shear stress were different by a kind of grass. The difference between common reed, sedge and reed is due to difference of growth and propagation. A reed propagates by subterranean stem and a root weight and pull-out strength are linearly increased by root and growth of subterranean stem.

Influence of Physico.Chemical Properties of Root Substrates on the Growth of Mother Plants and the Occurrence of Daughter Plants during the Propagation of 'Maehyang' Strawberry Using a Bag Culture System ('매향' 딸기 번식을 위한 플라스틱 백 재배시 상토 물리.화학성이 모주생육과 자묘 발생에 미치는 영향)

  • The objective of this research was to determine the influence of the physical and chemical properties of root substrates used during the production of 'Maehyang' strawberry propaguleson the growth of the mother plants and the rate of daughter plant formation. Plants were cultured in plastic bags containing six different formulations of root substrates composed of: a) 50% coir dust and 50% perlite (5:5 by volume, A), b) 60% coir dust and 40% perlite (6:4, B), c) 70% coir dust and 30% perlite (7:3, C), d) 70% coir dust and 30% coconut chip (7:3 D), e) 60% coir dust and 40% coconut chip (60:40, E), or f) 50% sphagnum peat and 50% vermiculite (50:50, F). All media formulations contained a moderate level of base fertilizers. Physical and chemical properties of each formulation were determined before plant establishment and after 120 days of stock plant culture and runner production. Total porosity (TP) and container capacity (CC) of all substrate formulations were higher than 85% and 55%, respectively, allowing a suitable range of air and water holding characteristics. Formulation F provided the highest TP and CC values among the all substrate modifications evaluated. Substrate formulations A, B, C and F had higher electrical conductivity (EC) and $NO_3{^-}$-N concentrations than formulations D and E, when determined before and after plant culture. Formulations A, B, C, and F, having higher EC readings, also performed better as root substrates thanthe formulations D and E in increasing fresh and dry weights of the runners as well as the production of daughter plants per plant. The 'Maehyang' strawberry plants grown in the formulation F had the highest tissue N content, followed by those grown in substrate B, A, C, or D for 120 days after transplanting. Formulation F also facilitated accumulation of higher tissue phosphorus (P) and copper (Cu) contents compared to other treatments. Results of this experiment suggest that the chemical properties, rather than physical properties, of root substrates had a major influence on the growth of mother plants and the occurrence of healthy daughter plants during the bag-culture phase of propagation.
