  • An efficient regeneration protocol for stem disc culture of Populus euramericana, which is important species for bioenergy resource in agroforestry, was established. The number of explants that were obtained and the number of explants that regenerated varied with the genotypes. However, in all the genotypes, stem disc culture produced more regenerated shoots than did in axillary bud culture. A comparison of the effects of cytokinin type and concentration on shoot regeneration in different explants (i.e., petiole, leaf, and root segments of P. euramericana) revealed that a concentration of $0.002mg\;l^{-1}$ thidiazuron (TDZ) used on petiole segments resulted in the greatest shoot regeneration (95.83%). The hormonal requirements for the greatest shoot regeneration in the three explant types varied. Different concentrations of $AgNO_3$ and $CoCl_2$ were added separately to the medium to stop the yellowing and subsequent necrosis of the regenerated shoots. Lower concentrations (3 and $5mg\;l^{-1}$) of these compounds improved shoot regeneration and elongation, compared with the control. The in vitro-regenerated shoots were transferred to rooting medium and subsequently acclimatized. The highly efficient regeneration system of P. euramericana reported here can be used for mass propagation of this recalcitrant for regeneration, economically important tree species.

  • To investigate the effects of light intensity on the growth, and the critical minimum light intensity for growing of Cornus controversa seedlings at the stage of cotyledon, hypocotyl elongation, cotyledon expansion, the times of leaves appearance, dry weights of each organ, and specific leaf area(SLA) were measured on a growth chamber with several light intensity gradients(385, 32, 17, 8, and $5{\mu}mol\;m^{-2}s^{-1}$). There was a positive correlationship between the size of cotyledon and the biomass of cotyledon and total seedling. Hypocotyl was more elongated under relatively low light intensities, such as 32, 17, 8, and $5{\mu}mol\;m^{-2}s^{-1}$ than under $385{\mu}mol\;m^{-2}s^{-1}$ light intensity, however, dry weight of the hypocotyl was adverse. As the light intensities decreased, the leaf appearance was delayed and the number of leaves decresed. In addition, leaves did not appear under $8{\mu}mol\;m^{-2}s^{-1}$ and $5{\mu}mol\;m^{-2}s^{-1}$ light intensity. Although cotyledons were more fully expanded under 32 and $17{\mu}mol\;m^{-2}s^{-1}$ light intensities than $385{\mu}mol\;m^{-2}s^{-1}$ light intensity, the dry weights of cotyledons were greater under the high light intensity. The dry weight of cotyledon, hypocotyl, root and leaves showed a decreased pattern with decreasing light intensities, but root to shoot(hypocotyl+leaves) ratio rapidly increased. Roots did not develop below $8{\mu}mol\;m^{-2}s^{-1}$ light intensity. In conclusion, the results showed that the critical minimum light intensity for growing of Cornus controversa seedlings was above $17{\mu}mol\;m^{-2}s^{-1}$ light intensity.

  • This study was conducted to find suitable methods for screening organic resources useful for compost. Twenty-seven industrial and domestic sludges were collected from various cities and industrial areas. Contents of organic matters in the sludges were in the range of 79.3-98.0%, and the contents were much higher than the regulation level (60%) for raw materials of compost. Contents of total nitrogen were in the range of 0.8-2.6%. Contents of Fe and Al were very high. Content of HEM was highest in textile sludge ($257mg\;kg^{-1}$) and the contents in the others were in the range of $12.6-90.3mg\;kg^{-1}$. Content of PAHs was lowest in food sludge ($739.1{\mu}g\;kg^{-1}$ and pulp-mill sludge had the highest PAHs content ($3461.8{\mu}g\;kg^{-1}$). $Microtox^{(R)}$ $EC_{50}$ values were higher in the sludges which were classified as a possible material in composting after analysis and investigation. Lettuce root elongation and $EC_{50}$ values were relatively lower in pulp-mill sludge, sewage sludge 3 (Large city), food sludge and leather sludge. Therefore, mineral nutrients, heavy metals, organic compounds (HEM, PAHs, PCBs), and bioassay ($Microtox^{(R)}$ $EC_{50}$, Relative root elongation test) are recommended to be included in the screening system of raw material of compost in addition to the current regulation with organic matter and 8 heavy metals.

  • The results of ground vegetation experiment conducted at completely denuded forestland in the mudstone region are summerized as follows: On the reaults of soiling quantity the effect of soiling was observed where depth of soiling over 10 cm was practiced, and a plot where treated with 15cm soiling and without fertilizer showed poor growth and it was even worser than the plot where soiling was practiced only 1 cm in thikness but applied adequate amount of fertilizers. The depth of slits between 30cm and 40cm showed no significant difference in the effect. A plot where covered with vegetation bag showed somewhat better results in seed loss and early growth but no differences observed in the fall result over the none covered plot. And then, it is recommendable to have soiling over 10cm in thikness with slit of 30cm and 30cm in depth and to apply 30 gram of fertilizer (22;22:11, 50 gram) per slit. On various soiling materials trial there were no striking differences in the effect of soiling between weathered granite soil, wheathered tuffs soil and weathered mudstone soil. In the treatment with various green materials, a plot treated with straw mat showed a significant difference at 1 percent. The results show that weathered mudstone soil is effective to use as soiling materials and straw mat treatment was better. On forest fertilization trial, in the mudstone region where red and black pine trees already existing at a rate of 2,000-3,000 trees per hectare had applied 110kg of compound fertilizers (9:12:3 and 22:22:11) per hectare basis in terms of plant nutrient. As a result, the difference in effect between the compound fertilizers was not found however the leaf color and leaf length of the fertilizer added plot showed darker and longer at 30 percent over the no fertilizer received plot. Compound fertilizers, 14:37:12 and 9:12:3 were applied to alder trees at a rate of 20 gram and 40 gram per tree in terms of plant nutrient and a remarkable growth accelerantion was observed where 40 grams of plant nutrient applied. The effect difference between the compound fertilizers was not found. On investigation of tree root elongation, forty years old red pine trees showed only 15cm tap root elongation through mudstone while black pine had 23 cm tap root elongation. The total length of supporting root elongtion of red and black pines showed 20 and 13 meters, respectively. The tap roots of Black locusts were not able to elongate through mudstone, however, the supporting roots tended to develop to the underneath of pine tree where some moisture content is available. Black locusts And grown on the residual soil of mudstone normally die between 8 to 10 years. The red pine trees show flat in tree shape while black pine had triangle in the shape. With the results it can be said that in an artificial reforestation in denuded forest land of the mudstone region the adequate slit and enough amount of fertiliizer application must be provided for the succesful performance of the program. On integrated experimental results of 1972. for the establishment of ground vegetation on the completely denuded forest land in mudstone region, soiling could be effectively practiced with weathered mudstone soil and it would not specially necessiate to have either weathered granite or tuffssoil for the soiling. And the soiling depth should be more than 10 cm in thickness. Among green materials used the straw mat proved to be the most effective reatment. Three major factors which enable to establish ground vegetation by the shortest period of time: A. Physical improvement of soil is necessary to breakdown of the horizontal cracks sushas Slit, contour line plot, seeding hole and etc., and soiling with weathered mudstone soil. B. Chemical improvement of soil: is needed sufficient amount of fertilizer application 300~400kg ha, $N+P_2O_5+K_2O$), and increased production of ground covering and expedite resolution of the vegetation (ground vegetation, fallen leaves and twigs). C. Complete establishment of the basic structure for the erosion control (Prevention of surface soil erosion)

  • In this study, we compared the levels of methylotrophic bacterial community diversity in the leaf samples of 19 rice cultivars collected from three regions of Korea. Nineteen pink pigmented isolates showing characteristic growth on methanol were obtained. Physiological and biochemical characters of each isolate were examined according to methods described in Bergey's Manual of Systematic Bacteriology. When phylotypes were defined by performing numerical analysis of 37 characteristics, four distinct clusters were formed. The two reference strains, Methylobacterium extorquens AM1 and Methylobacterium fujisawaense KACC10744 were found to group under cluster IV and cluster III respectively. Cluster I diverged on the basis of nitrate reduction and four isolates showed tolerance upto 0.5 M NaCl concentrations. Two strains in cluster I and III were found to possess methane utilizing properties. Most of the isolates in all the four clusters utilized monosaccharides, disaccharide and polyols as carbon source. When the isolates were subjected for indole-3-acetic acid (IAA) analysis in the presence of L-tryptophan, only 8 isolates exhibited IAA production. In addition, the nitrogen source in the medium was found to influence the IAA production. Addition of $(NH_4)_2SO_4$ in the medium led to a 2 to 30 fold increase in the indole synthesis. However, $KNO_3$, $NH_4NO_3$ and $NH_4Cl$ substitution did not significantly stimulate the synthesis of IAA in the growth medium. Result of gnotobiotic root elongation assay significantly increased roots and shoots lengths, and number of lateral roots, which is mediated by IAA production in the culture medium. The rice seedlings primary roots from seeds treated with methylotrophic isolates were on average 27 to 56% longer than the roots from seeds treated with the uninoculated seeds. In addition, application of different high concentrations of authentic IAA ($400g\;mL^{-1}$) to roots of rice seedlings inhibited root growth. However, the IAA concentration from 10 to $200g\;mL^{-1}$, IAA promoted root growth of rice seedlings. These results suggest that bacterial IAA plays a major role in the development of the host plant root system.

  • The studies were conducted to obtain the basic information of the effects of pollutants on plant species and to select the plant species showing specific responses to the pollutants. For these purposes, paraquat, ammonium, and cadmium as a source of oxidative stress, nitrogen toxicity, and heavy metal toxicity respectively were treated to the plant species. Among the tested plants, Lamiaceae, Brassicaceae, and Caryophyllaceae were tolerant to paraquat, whereas Poaceae and Asteraceae were sensitive. Especially Mosla dianthera of Lamiaceae, Hemistepta lyrata and Aster pilosus of Asteraceae, and Paspalum thunbergii of Poaceae showed higher tolerance than others. Paraquat resistance was related with life style, overwintering capacity, so perennial and biennial species showed higher tolerance than annual species. In response to ammonium, Poaceae showed higher resistance while Fabaceae and Caryophyllaceae showed sensitiveness. Weed species having tolerance to ammonium were Echinochloa crus-galli var. praticola, Panicum dichotomiflorum, Setaria glauca, Chenopodium album, and Solanum nigrum, while Mosla dianthera, Arenaria serpyllifolia and Perilla frutescens var. japonica showed sensitiveness. In the response of plant species to cadmium, Digitaria sanguinalis, Amaranthus lividus showed higher resistance, whereas Galinsoga parviflora, Plantago asiatica, Ambrosia trifida, and Paspalum thunbergii showed sensitiveness. The injured degree on germination stage by pollutants did not related with injured degree on matured stage. During germination, the root elongation was more sensitive than shoot elongation by pollutants, paraquat, ammonium, and cadmium.

  • To investigate effects of inorganic $(GeO_2)$ and organic (Ge-132) germanium (Ge) on seed germination and on early stage growth of plane and the uptake characteristics, various concentrations (0, 10, 25, 50, 100 mg $L^{-1}$) of Ge to popular vegetables such as leaf mustard, chinese cabbage and pak-choi, respectively, were treated. On seed germination, no significant effect was observed in both inorganic and organic Ge treatments except 100 mg $L^{-1}$ treatment of inorganic Ge. Exogenous inorganic Ge ($10{\sim}100$ mg $L^{-1}$ treatments significantly inhibited the early root elongation growth of all plants. However, slight enhancement of early shoot elongation was detected in low concentrations (10 and 25 mg $L^{-1}$) of Ge in the leaf mustard and chinese cabbage plants. Organic Ge treatments significantly stimulated the 개ot and shoot growth at the 10, 25 and 50 mg $L^{-1}$ treatments. Ge was accumulated linearly in the vegetables as both inorganic and organic Ge concentrations were increased. Interestingly, total contents of Ge in plants with Ge-132 treatments were $2\sim4.5$ times more than those with inorganic Ge treatments in all concentrations. At 25 mg $L^{-1}$ treatment of Ge, contents of Ge in vegetables are following: in leaf mustard, inorganic Ge: 0.37 mg $g^{-1}dw$ and organic Ge: 1.47 mg $g^{-1}dw;$ in the chinese cabbage, inorganic Ge: 0.4 mg $g^{-1}dw$ and organic Ge: 0.86 mg $g^{-1}dw;$ in the pak-choi, inorganic Ge: 0.33 mg $g^{-1}dw$ and organic Ge: 0.70 mg $g^{-1}dw$, respectively. These results showed organic Ge is much better on early stage seedling growth and on germanium accumulation of vegetables than inorganic Ge.

  • This study was carried out to increase the rate of plant regeneration from embryogenic callus of A. keiskei on MS medium supplemented with ethylene inhibitors. When leaflet, petiolule, and petiole of A. keiskei were cultured on MS medium supplemented with 2, 4-D, callus was well induced from leaflet segments at 2.0 ppm 2, 4-D. Shoot elongation of plantlets and shooting from embryogenic callus of A. keiskei were best on 2, 4-D-free medium supplemented with 2 ppm $AgNO_3$ or 10 ppm $CoCl_2{\cdot}6H_2O$, but it was suppressed on the medium containing 1 ppm 2, 4-D with $AgNO_3$ or $CoCl_2{\cdot}6H_2O$. Root elongation of plantlets from embryogenic callus was best on 2, 4-D-free medium supplemented with 1 ppm $AgNO_3$ or 5 ppm $CoCl_2{\cdot}6H_2O$, but rooting from embryogenic callus was none on the medium containing 1 ppm 2, 4-D with $AgNO_3$ or $CoCl_2{\cdot}6H_2O$. Fresh weight of plantlets from embryogenic callus was heaviest on 2, 4-D-free medium supplemented with 2 ppm of $AgNO_3$ or $CoCl_2{\cdot}6H_2O$, while it was heaviest on the medium containing 1 ppm 2, 4-D with 1 ppm $AgNO_3$ or 2 ppm $CoCl_2{\cdot}6H_2O$.

  • This study was conducted to elucidate the effect of light sources and explant types on in vitro shoot multiplication and rooting of a rare and endangered plant Abeliophyllum distichum. Both apical buds and axillary buds were used as explants under 4 different light sources, cool white florescent light (F), 100% blue light-emitting diode (LED) (B), 50% blue and 50% red LED mixture (BR), and 100% red LED (R). Clear difference was observed in terms of shoot proliferation by light sources types but not by position-dependent explant types. Multiple shoot induction rates were enhanced under both B and BR light sources. Spontaneous rooting was induced in shoot induction medium under B light source. Both the rates of rooting and numbers of roots per explant were higher in apical bud explants compared to axillary bud explants. Interestingly R light source stimulated shoot elongation but inhibited root development. Therefore, our results suggest that the use of apical bud explants under B or BR light sources is suitable for in vitro micropropagation of a rare and endangered plant species, Abeliophyllum distichum.

  • An efficient micropropagation was established by using axillary bud explants from two-year-old tree(Echinosphorea koreensis Nakai), which has been known as a rare and endangered species. Among various basal media tested, DKW medium was shown to be the best for axillary shoot elongation. The addition of both BA and TDZ to the medium induced 6 to 10 shoots per explant during eight weeks of culture, without showing any abnormal morphology at the shoot proliferation stage. However, high concentration of TDZ(>0.05 mg/L) appeared to cause hyperhydration on either leaf or shoot at the later developmental stage. Approximately 20% of shoots produced roots by the addition of 1.0 mg/L NAA but not by IBA($0.2{\sim}1.0$ mg/L). Ex vitro micro-cuttings were better source for root induction; up to 58.6% of the micro-cuttings rooted when 100 mg/L IBA was applied to the soil(vermiculite). More than 90% of plantlets with roots were successfully acclimatized and grew normally in the field. Therefore, we suggest that this endangered tree species can be effectively micropropagated by axillary bud culture system developed in this study.
