옥수수 (Zea mays) 뿌리의 알데히드 산화효소와 생장에 미치는 텅스텐산 나트륨의 영향 (The Effect of Sodium Tungstate on the Aldehyde Oxidase and the Growth in the Primary Root of Maize (Zea mays))

  • 몰리브덴 보조인자 형성을 방해하는 텅스텐산 나트륨이 옥수수 뿌리에서 알데히드 산화효소의 활성과 생장에 미치는 영향을 조사하였다. 다른 식물에서 보고된 바와 같이 옥수수 뿌리에서도 텅스텐산 나트륨은 그 농도가 증가됨에 따라 알데히드 산화효소의 활성을 억제하였는데, 억제 활성은 식물체에 직접 처리한 경우에만 나타나고 추출된 효소에 처리하였을 때에는 효과가 없었다. 텅스텐산은 알데히드 산화효소의 활성화를 억제하는 물질임에도 불구하고, Western분석에 의하면 알데히드 산화효소 단백질의 함량을 감소시키는 것으로 나타나 반응산물이 효소함량을 증가시키는 양성 되먹임 조절관계를 나타내었다. 텅스텐산 나트륨은 효소활성을 억제하는 농도에서 옥수수 원뿌리의 생장과 곁뿌리발생을 억제하였지만 굴중성 반응에는 영향이 없었다. 전자의 두 반응은 옥신 절대함량에 의존하고 후자는 상대량에 의존하므로 텅스텐산 나트륨에 의한 옥신 함량 변화로 관찰된 결과들의 설명이 가능할 것으로 사료되었다. 그러나 뿌리의free IAA의 함량 변화는 검출되지 않았다. 옥신 함량 조절에는 강력한 항상성 기작이 관여하므로 IAA conjugate분해와 nitrilase에 의한 생합성 증가 등 결과 설명에 적용 가능한 내용들을 논의하였다.

  • Effects of illumination with xenon or sulfur lamp on growth of tomato seedlings were studied. Results showed no obvious differences in net photosynthetic rate and apparent quantum efficiency. In comparison with xenon lamp, illumination with sulfur lamp slightly reduced ChI a and ChI b content in leaves, relatively shortened epicotyl and hypocotyl length, but significantly increased anthocyanin content of leaves and promoted root growth of tomato seedling plants.

Isolation and Identification of Phosphate Solubilizing Bacteria from Chinese Cabbage and Their Effect on Growth and Phosphorus Utilization of Plants

  • Phosphate solubilizing bacteria (PSB) were isolated from the rhizosphere of Chinese cabbage and screened on the basis of their solubilization of inorganic tricalcium phosphate in liquid cultures. Ten strains that had higher solubilization potential were selected, and they also produced indole-3-acetic acid, 1-aminocyclopropane-1-carboxylate (ACC) deaminase, and siderophores. The strains were identified to be members of Pseudomonas, by 16S rDNA sequence analysis. Seed bacterization with PSB strains increased the root elongation and biomass of Chinese cabbage in seedling culture, although they had no effect on phosphorus uptake of plants. The plant growth promotion by PSB in this study could be due to the production of phytohormones or mechanisms other than phosphate solubilization, since they had no effect on P nutrition.

Molecular and Morphological Characterization of Endophytic Heterobasidion araucariae from Roots of Capsicum annuum L. in Korea

  • A species of Heterobasidion was encountered during a diversity study of endophytic fungi from healthy root tissues of chili pepper (Capsicum annuum L.) in Korea. The fungal species (CNU081069) was identified as Heterobasidion araucariae based on phylogenetic analyses of the internal transcribed spacer and translation elongation factor gene sequences. Morphological descriptions of the endophytic isolate matched well with the previous references and supported the molecular identification. The fungus Heterobasidion araucariae CNU081069 is new to Korea.

  • Pb, Zn tolerance of phaseolus multiflorus was investigated, based on the elongation of root and stem, pollen Germination and progeny quality in various pb, zn concentrations. The result obtained by water culture showed that the growth of roots and steams of phaseolus multiflorus from pb-zn mine site is less inhivited than that of the control site. The flower of phaseolus multiflorus from which pollen was taken were grown without added pb, zn and percent germination of pollen observed in a range of pb, zn concentrations. The percent germination of pollen from pb-zn mine site was higher than the control site. phaseolus multiflorus collected at a pb-zn mine site and the control site was site were grown at different pb, zn concentrations, its progeny was retreated with same concentrations of pb-zn mine site was more vigorous than the control site. thus, pb-zn tolerance was able to expressed in both pollen and sporophytes.

Rapid Propagation through Tissue Culture of Cudrania tricuspidata, Medicinal Plant

  • An effective rapid propagation method was established through in vitro cultures of the medicinal plant, Cudrania tricuspidata. In vitro plantlets were obtained from in vitro germinated seeds. The various levels of cytokinins (BAP, Kinetin and TDZ) were tested on multiple shoot formation from plantlets. BAP (1.0 mg/l) treatment induced highest number of multiple shoots. Single shoot cultures gave higher initial shoot numbers than 5 shoots per culture. Among the various culture media, the shoot elongation was optimal on 2 MS basal medium without growth regulators. The IAA (2.0 mg/l) treatment induced highest number of roots. IBA (2.0 mg/l) treatment more promoted in vitro root growth than other concentrations. Rooted shoots were transferred directly to small pots with an artificial soil and successfully acclimatized.

Zinc Chloride Toxicity on Free Proline and Organic Acids in Germinating Rice Seed

  • The study was conducted to find the critical concentrations of zinc toxicity and to determine the changes of the contents of free proline and organic acids with treatment of different zinc chloride concentrations during rice germination and seedlings grown for seven days. The concentration of zinc chloride, 140 ppm, inhibited root elongation as much as 46 times compared with the control, and the germination rate was also decreased in all treatments of zinc chloride, showing that the germination rate decreased more with increasing concentrations of zinc chloride. Its rate was only 13% with treatment of 140 ppm zinc chloride. The content of free proline with treatment of zinc chloride, 140 ppm, was highest about 4,873 $\mu$M at 3 days compared with the control. Malic acid concentration with treatment of zinc chloride, 140 ppm, increased to approximately 4 times compared to the control. Citric and succinic acid content were also slightly increased in all treatments of zinc chloride.

국내콩 형질전환 기술개발 (Development of genetic transformation method of Korean soybean)

  • Current status of soybean transformation method in Korera was reviewed with recent publications. Most frequently used method for genetic transformation was Agrobacterium-mediated transformation on cotyledonary node which is most popular method used in foreign country. In addition to this, various methods such as sonicationmediated transformation, in planta transformation, and transformation on meristem tissue of germinating seed, have been tried in Korea, even though their efficiencies on repeatability and stability were relatively low. Based on the promising results developed recently by reviewer, several important considerations for successful soybean transformations were suggested. They are 1) proper genotype screening, 2) targeting transformation on exact point, 3) multiple shoot formation, 4) efficient selection pressure, 5) successful shoot elongation, 6) efficient root formation. These are the basic requirements for stable and highly efficient soybean transformation of Korean soybean.

Comments on a Case Study on Engineering Failure Analysis of Link Chain

  • The article by Tae-Gu Kim et al. conducted elastic FE modeling, which was inappropriate for fracture of elastic-plastic chain material (11.3% of elongation). FE analysis results and the findings in the fracto-graphic analysis did not tally but contradicted each other. The article identified "incorrect installation"/bending forces as the root cause while FE results of the chain under bending forces showed very low stresses at fracture locations but the highest stress in the middle of shank of the chain. The article's "step-like topographies indicating the fracture due to bending moment rather than uniaxial tension" lacked scientific support. The load value carried by each chain section under bending/incorrect installation was only half of that under tension, thus the article using same load value in FE simulation comparison for bending and tension was incorrect. The real cause of the chain fracture was likely improper checking the lifted load or/and using the wrong chain with much lower safety working load.

토양의 답압에 대한 맥류의 생육반응 (Reaction of Growth of Barley to Soil Stamping)

  • 토양의 답압이 맥류의 생육에 미치는 영향을 조사하여 다음과 같은 결과를 얻었다. 1. 토양의 답압에 의하여 초형은 포포형으로 되고 초장의 신장은 억제되며 분벽수가 증가하고 뿌리는 근수, 근경이 증대하며 심근성으로 되고 근중이 증가하였다. 2. 토양의 답압에 의하여 엽연소 함량이 높아지고 절단엽편의 수분감손량도 증가하였다. 3. 토양의 답압에 의하여 엽에서 생성하는 Ethylene 량이 증대하였다. 4. 토양의 답압에 의하여 하위절간의 생장이 억제되어 도복되기 어려운 생육형으로 되고 수량이 증대하였다. 이상의 결과로부터 맥류 답압의 효과는 맥류 그 자체에 대한 직접적인 효과와 토양을 답압하는 것에 의한 효과가 있음을 알 수 있으며, 또한 토양 답압의 효과는 엽에서 생성하는 Ethylene 량의 증대로 보아 토양 답압이 맥류 뿌리에 대하여 물리적 자극을 주고 있다는 가능성이 시사되었다.
