Folk Plants in the Inland of Northern Area in Gangwon-do (강원도 내륙 북부지역의 민속식물)

  • This study was carried out to classify the folk plants in inland of northern region of Gangwon-do. According to the survey results from 35 places of 9 counties and cities from April to October, 2010, the folk plants in inland of northern region of Gangwon-do consisted of a total 261 taxa; 73 families, 185 genera, 225 species, 2 subspecies, 30 varieties, and 4 form. Among the investigated 261 taxa, 3 Korean endemic, 11 rare plants and 9 naturalized plants were also included. The use by usage of 261 taxa was: 378 times; Edible, 53 times; Medicinal, 8 times; Dye, 7 times; Ornamental, 6 times; Fuel, 5 times; Spice, 3 times; Aroma, 2 times; Nectar, 41 times; Others, respectively, so the edible use is the highest. The most useful part was the leaf, followed by whole plant and root. The consistency comparison between the scientific name and the local name were the highest in the 20's and the lowest in 80's.

  • This study was carried out to classify the folk plants in southern region of Gangwon-do. According to the survey results from 1,103 survey sheets of 83 residents at 32 places of 9 counties and cities from March to October, 2010, the folk plants in the southern region of Gangwon-do consisted of a total 311 taxa; 92 families, 222 genera, 274 species, 2 subspecies, 29 varieties, and 6 form. The use by its usage was: 227 taxa; Edible, 174 taxa; Medicinal, 14 taxa; Ornamental, 5 taxa; Oil, 5 taxa; Spice, 4 taxa; Dye, 1 taxa; Fiber, 1 taxa; Aroma, 1 taxa; Fuel, 32 taxa; Others, respectively, so the edible use is the highest. The most useful part was the leaf, followed by root and fruit. The consistency comparison between the scientific name and the local name were the highest in the 50's and the lowest in 80's.

Influence of Lime and Phosphate Application on Amide and Ureide Nitrogen of Soybean Plants and Soil Microorganisms (석회(石灰)와 인산시용(燐酸施用)이 대두식물체중(大豆植物體中) Amide태(態) 및 Ureide태(態) 질소(窒素)와 토양미생물상(土壤微生物相)의 변화(變化)에 미치는 영향(影響))

  • A pot experiment was conducted to find out the effects of lime and phosphate application on the changes of number of soil microorganisms, indigenous Rhizobium japonicum, nodule formation, and ureide-and amide-N in leaf and stem exudate of soybean plant under uncultivated hillside red earth in very low pH value, organic matter, available phosphate, and cation exchange capacity. The results obtained were summarized as follows : 1. The plant height, stem length, root dry weight and nodule weight were significantly increased with the application of lime and phosphate application than that of control plot. 2. The concentration of amide-N in soybean plant at the 45 days after sowing was obtained as high in order of control>lime> lime+phosphate while the concentration was obtained in order of Iime+phosphate>lime> control at flowering stage 3. However, concentration of ureide-N in the soybean leaf at the 45 days after sowing was obtained as high in order of control>lime>lime+hosphate while reversed concentration was obtained in stem. 4. The number of soil microorgan isms were increased with increase of pH value, available phosphate and soil exchangeable cation. 5. Significantly negative high correlation were obtained with the concentration of Al, Fe in soil and the concentration of amide-and ureide-N in soybean plant at flowering stage while positive correlation was obtained with plant growth and the concentration of ureide-N.

Development of Analysis Method of Caffeine and Content Survey in Commercial Foods by HPLC (HPLC를 이용한 카페인의 분석법 개발 및 시판 식품중 함유량 조사)

  • A simple and practical method for determination of caffeine in foods was developed. The analysis of caffeine was performed by reverse phase high performance liquid chromatography using a ${\mu}-Bondapak\;C_{18}$ column at isocratic condition with methanol-acetic acid-water(20 : 1 : 79) on UV detector at 280 nm. The clean-up and extraction of caffeine in samples were based on a simple pretreatment using a Sep-Pak $C_{18}$ cartridge. Recovery rates obtained with this method for cider, candy, cookie, milk, ice cream and persimmon leaf tea were 99.23%, 99.50%, 99.17%, 99.37%, 98.93% and 99.10% respectively. And the detection limit of caffeine was $0.1\;{\mu}g/mL$. With this method, the range of caffeine contents extracted from coffee, green tea, black tea, Oolong tea(tea bag), soft drinks, ice cream, milk and commercial confectionery were $3.38{\sim}37.50\;mg/g,\;16.30{\sim}26.10\;mg/g,\;10.80{\sim}16.65\;mg/g,\;11.25\;mg/g,\;0.06{\sim}0.11\;mg/g,\;0.04{\sim}0.44\;mg/g,\;0.04{\sim}0.39\;mg/g\;and\;0.10{\sim}1.80\;mg/g$, respectively. But caffeine was not detected in the other tea such as Acanthopanax sessiliflorum tea, Angelica gigas tea, Angelica tea, Arrow root tea, Duchu'ng tea, Dunggulle tea, Ganoerma lucidum tea, Ginger tea powder, Persimmon leaf tea, Ssanghwa tea and Cocoa mix powder.

Changes in Tomato Growth and Productivity under Different Night Air Temperatures (야간온도에 따른 토마토 생육 및 생산성 변화)

  • This study aimed to investigate the effect of night temperature on the productivity, growth, and fruit characteristics of tomatoes and set the optimal night air temperature. Tomatoes (Solanum lycopersicum Mill. cv. Superdoterang and cv. Rapito) were grown at 5, 10, 15, and $20^{\circ}C$ during night time and ventilation temperature point during day time set $27^{\circ}C$. Depending on night air temperature, plant height, leaf area, fresh and dry weight of each organ, leaf, stem, and root were changed. Condition of high night temperature resulted in promotion of tomatoes growth with high dry weight and fresh weight. However, the dry weight under night temperature $20^{\circ}C$ decreased, especially, in 'Superdoterang'. On changes of yield, 'Rapito' showed higher productivity than 'Superdoterang' and the highest productivity point for nigh temperature is $13.32^{\circ}C$ for 'Superdoterang' and $14.25^{\circ}C$ for 'Rapito'. Although the total yield decreased from the point, the daily productivity increased with increasing night temperature, the highest point for nigh temperature is $14.45^{\circ}C$ for 'Superdoterang' and $16.46^{\circ}C$ for 'Rapito'. High temperature deceased the fruit weight and increased total soluble solid content in fruits. It is concluded that the night air temperature changes productivity of tomato and the temperatures between productivity and growth velocity have differences depending on different cultivars. scores than the other beef jerky samples (p<0.05).

Effect of Polymer, Calcium, Perlite and Chitosan in Soil Organic Amendment on Growth in Perennial Ryegrass (유기질 토양개량재에서 고분자 중합체, 칼슘, 펄라이트 및 키토산이 퍼레니얼 라이그래스의 생장에 미치는 효과)

  • The study was carried out to investigate the effects of polymer, calcium, perlite and chitosan on the growth of perennial ryegrass (Lolium perenne L., PR) and to provide a basic information needed for their practical application when establishing garden, parks, athletic field and golf courses with these materials. A total of 24 treatment combinations were applied in the study. Treatments were made of water-swelling polymer (WSP), calcium, perlite and chitosan mixed in soil organic amendment (SOA). Germination rate, turfgrass coverage, turfgrass density and top growth were evaluated in PR under greenhouse conditions. Significant differences were observed for these growth characteristics among the treatments. Turfgrass density and plant height, evaluated on a weekly basis, varied with time after seeding. A proper mixing rate of WSP was considered to be lower 3% for the growth of PR with an exception of being below 6% for turfgrass density. Germination rate and early survival capacity were greatly influenced by calcium and chitosan among the elements of calcium, perlite, and chitosan. But there was little effect by perlite. Calcium and chitosan were most effective one for turfgrass density and coverage, respectively. Top leaf-growth was influenced by all three elements, but the greatest effect was highly linked with calcium. Chitosan was very effective in early germination and vertical leaf growth, as compared with the others. Future studies are required for measuring the effect of WSP, calcium, perlite and chitosan on the turf growth characteristics in root zone mixtures of sand+SOA before a practical field use.

Antioxidant Activity of Different Parts of Lespedeza bicolor and Isolation of Antioxidant Compound (싸리나무(Lespedeza bicolor) 부위별 추출물의 항산화 활성 및 항산화물질 분리)

  • In this study, total antioxidant properties of extracts from different parts of Lespedeza bicolor were determined using techniques of measuring 1,1-diphenyl-2-picryl hydrazyl/2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)-radical scavenging activity and total phenolic contents. The total antioxidant activities of leaf, stem and root extracts from various solvents (water, 50, 70, 100% ethanol, and hot-water) indicated that 50 and 70% ethanol extracts have high radical scavenging activities and phenolic contents. A systematic approach was used to determine the total antioxidant activity of different solvent fractions of the Lespedeza bicolor extracts, partitioning with chloroform, ethyl acetate, n-butanol, and water, and the ethyl acetate fraction was found to have the strongest antioxidant activity. Antioxidant assay-guided isolation was carried out to isolate potential antioxidant compounds. The ethyl acetate fraction of the leaf extract was subjected to silica gel, LH-20 and RP-18 column chromatography successively, and afforded compound 1, which was identified as eriodictyol by NMR and MS analysis, after which its antioxidant activity was determined.

Photomixotrophic Growth of Solanum tuberosum L. in vitro with Addition and Omission of Organic Materials at Thee Initial Sucrose Levels in the Medium (세 수준의 자당이 첨가된 배지에서 유기물의 첨가 유무에 따른 Solanum tuberosum L.의 기내 광혼합영양생장)

  • The most commonly used inorganic nutrient compositions such as Murashige & Skoog medium have been optimized for heterotrophic growth. Therefore, they may not be optimal for photomixotrophic and photoautotrophic growth of plantlets. In photomixotrophic micropropagation, emdium sugar level is often lowered, while light and $CO_2$ levels in vessel are raised, and chlorophyllous explants are used to facilitate photosynthetic carbon acquisition. In a factorial experiment effect of addition (+) and omission(_) of organic materials (OM, 0.5 g ${\cdot}$ $m^{-3}$ each of thiamine, nicotinic acid and pyridoxine and 100 ${\cdot}$ $m^{-3}$ myo-inositiol) combined with three sucrose levels (0, 15, and 30 kg ${\cdot}$ $m^{-3}$) was tested on the growth of potato plantlets. Each of nodal cuttings with a leaf was cultured on 0.1${\times}$$10^{-4}m^{-3}$) MS agar ( 8 kg ${\cdot}$ $m^{-3}$) medium (pH 5.80 before autoclave) in glass test tubes (100 mm${\times}$25mm) capped with a sheet of transparent film with a 6 mm diameter gas permeable filter (5.1 air exchanges ${\cdot}$$h^{-1}$). Cultures were maintained in a room for 27 days at $23^{\circ}C$, 50% RH, 350-450${\mu}mol\;{\codt}\;mol^{-1}CO_2$, 16 h ${\cdot}$ $d^{-1}$ photoperiod at 13${\mu}mol\;{\codt}\;m\;{\codt}\;s^{-1}$ PPFD provided by white cool fluorescent lamps. Growth of potato plantlet in the +OM and -OM treatments were similar, while medium pH was 0.2 scale lower in the latter. Dry weight, % dry matter, and stem diameter enhanced, while shoot to root dry weight ratio, leaf area, chlorophyll concentration per gram dry weight, and medium pH decreased with increasing initial sucrose level. Interaction between OM and sucrose levels was observed in shoot length and medium pH. Results indicate that OM can be omitted from the medium without detrimental effect while addition of sucrose was beneficial for the photomixotrophic growth of potato plantlets under raised light and $CO_2$.

Methods of Application and Beneficial Effects of Silicate-Coating Rice Seeds (볍씨의 규산코팅방법에 따른 이용특성과 육묘효과)

  • A new silicate coating technology was developed which reduces the impact of dust and loosening during seeding compared to existing silicate-coatings (Seed/Si/Zeolite), and therefore can lower the production costs of rice cultivation. In this method, 100 g of rice seed is coated with 18 mL of liquid silicic acid and then dressed with a mixture containing 80 g of dolomite and 5 g of iron. To determine the most effective method of application and ensure that seedlings developed healthily, a series of experiments were carried out. Infected seeds scattered in seedling boxes and pots (soil and hydroponic) were coated dry, without disinfection. In comparison to the seed which were not treated with the silicate-coating, the new seed (A) were 1.84 times heavier in weight, and were also improved in terms of coating strength and coating color. Compared to the seedlings grown from the non-coated seed, those grown from the new silicate-coated seed were of significantly higher quality (weight/length) and had erect, dark greenish leaves, which are ideal plant characteristics. This was most likely due to increased silicate uptake. The symptoms of bakanae disease in the non-coated seed peaked after 38 days to 54.2%, whereas the control value was 68.8% in the new silicate-coated seed (A). In the infected seedlings grown from the new silicate-coated rice seed, subnormal macro-conidia, namely, a sickle shape spore without a septum; a straight oblong shape spore without a septum and with a thick cell wall; and inter-septal necrosis of a normal spore were detected. It is believed that the strong alkalinity of silicic acid have acted as unfavorable conditions for pathogenicity. In seedlings grown from the new silicate coated rice seed under hydroponic conditions without nutrients, normal root activity and growth was maintained without leaf senescence. Therefore, it was possible to reduce the rate of fertilization. In the future, a new silicate-coated rice seed was required for the study of minimal nutrition for anti-aging of seedlings.

  • In an effort of determine the metabolism and bio-synthesis of nitrogen, was studied at variance of souble nitrogens, free amino acids and total alcohol soluble amino acids during the growth of younger soybean plants, and saybean divides into cotyledon and embryonic organ (shoot and root system) in this experiment. 1. In the soluble nitrogen of soybean, ratio of increase and decrease in the amino acids of them was displayed the near phenomena both cotyledon and embryonic organ of soybean. But, in the 17th days after seed germination, that is the developmeatal stage of adult leaf of soybeans, was appear the maximum value. It has been suggested that the stage of first half period of growth as boundary the stage of adult leaf development which indicated clear morphologically, at the younger soybean plants, is the step that nitrogen assimilation. 2. It was investigated the amino acids of seventeen kinds in the seed state, but at the third days after seed germination, was investigated the amino acids of nineteen kinds. Ultimately, it appears the translocation from cotyledon to embryonic organ in the distribution of amino acid, the nineth day which differentiation begining day of embryonic organ, then after, it happen the variation of number of inspected amino acid in the cotyledon and embryonic organ but only the variance changes in the distribution and quantitative aspects. Especially, the most conspicuous fact was indicated the accumulation of Asparagine, that is the phenomena of Asparagine-accumulation was constitute, not with standing no fertilization from outside. It may be concluded from the results of this investigation that the difference of special phenomena of soybean from the embryo of other plans. 3. In the initial stage of differentiation at embryonic organ number of inspected amino acid was very few, and then, it was slightly appeared the increase-phenomena in the number of them. It was that the amino acid inspecting the initial stage was translocated from the cotyledon. It is suggested that the intermediate-metabolism of amino acid was constituted on the basis of above the result. 4. The phenomena of increase and decrease of total alcohol soluble amino acid were essentially identical to the water soluble amino acid of soybean, but it was appeared the severe difference of amounts in both of them.
