Studies were made to optimize the simultaneous saccharification and fermentation (SSF) of rice straw to produce butanol using Clostridium acetobutylicum KCTC 1037 and a cellulolytic enzyme preparation from Trichoderma viride. The fermentation was inhibited when the liquid enzyme preparation from Novo was used, whilst a successful fermentation was achieved in the SSF using the enzyme manufactured by Pacific Chemical Co. The minimum cellulase concentration for the successful fermentation of pure cellulose was found to be 4 IU/g of substrate used. Alkaline treatment was better method for the fermentation of rice straw by the system. SSF using 25% alkaline treated rice straw produced 150 mM butanol, 90 mM acetone. On the other hand, fermentation of ball milled rice straw was mainly acidogenic producing 98 mM acetate and 64 mM butyrate with less than 20 mM butanol. These results show that rice straw contains (a) specific inhibitor(s) for solventogenesis which is destroyed or soluble in alkali.
Three sheep fitted with cannulas in the rumen and the caecum were used in a $3{\times}3$ Latin square design to study the changes in ruminal and caecal microbial populations and their metabolite composition with ammoniated rice straw feeding. The 3 diets contained either 80% untreated rice straw (UTS) or ammoniated rice straw (ATS) and 20% formula feed. These were a control diet (C), a urea supplemented diet (U) containing urea at 1.1% and an ammoniated rice straw diet (AT). Data were analyzed by analysis of variance and means separated by the Student Neumann Kuel's multiple comparison. AT feeding increased ruminal bacterial counts, in particular cellulolytic bacterial counts (p < 0.05) which were 1.8, 2.4 and 7.0 (${\times}10^6/ml$ ruminal fluid) for C, U and AT, respectively. There was an increasing tendency (p < 0.10) in ruminal fungal population with U; values were 2.0, 5.2, 3.1 (${\times}10^3/ml$ ruminal fluid) for C, U and AT, respectively. Ruminal protozoa counts were not significantly (p > 0.05) altered with diets. Caecal total viable bacterial count with AT was about thrice the value with C. Total VFA concentration in the rumen was significantly increased (p < 0.025) (7.7 mmol/dl for C and 8.2 mmol/dl for AT) and correspondingly, pH lowered when AT was fed. Sheep on AT tended to produce less acetate and more butyrate in the rumen without significance (p > 0.05). Similar to the rumen, total VFA concentrations of 4.4, 3.8 and 5.2 mmol/dl were detected, respectively, for C, U and AT. Caecal ammonia-nitrogen concentrations were about six-fold of that in the rumen, though there were no differences (p > 0.05) among treatments.

The effect of soybean meal and barley supplementation of the utilization of rice straw was investigated. Balance trials were conducted with three Japanese Corriedale wethers fed rice straw supplemented with soybean meal and barley at three different levels of protein: low (40 g CP/d, LCP), medium (67 g/d, MCP) and high (94 g/d, HCP). In addition, all the supplements were formulated to contain the same amount of TDN (275 g/d). Voluntary intake of rice straw was not affected by any supplementation, while digestibility of organic matter in sheep given HCO diet was significantly higher (p<0.05) than those on LCP diet. Crude protein, neutral detergent fiber (NDF) and acid detergent fiber (ADF) digestibilities of MCP and HCP diets were significantly improved (p<0.05) over the LCP diet. Average daily gain of the animals under MCP and HCP diets were significantly higher (p<0.05) than those under LCP diet. Differences of rumen pH among the treatments were not significant, while concentration of rumen $NH_3-N$ was significantly higher (p<0.05) for HCP diet than for LCP and MCP diets. Total volatile fatty acids ($VFA_s$) and blood urea nitrogen (BUN) concentrations were significantly higher (p<0.05) in sheep fed MCP and HCP diets than those fed LCP diet, while plasma total protein concentration was not affected by any supplementation. Sheep fed MCP diet had a higher nitrogen retention than those fed LCP and HCP diets. It was concluded that rice straw was utilized better by sheep when SBM and barley were supplemented at the medium level of protein.

Objective: The goal of this study was to evaluate kenaf as a roughage source in vitro and its effects on meat quality of Hanwoo (Korean native) cattle. Methods: Three roughage materials, rice straw silage, ryegrass silage, and kenaf silage, were tested in a batch culture and feeding trial. Rumen fermentation parameters, including gas, pH, volatile fatty acid (VFA), and ammonia were analyzed. In the feeding trial, Hanwoo steers ($373.5{\pm}5.1kg$, n = 36, 11 month of age) were divided into three feeding groups (n = 12 each). Animals were fed with each silage and concentrate until the fattening stage. Results: Crude protein, ether extract, and non-structural carbohydrates were greater in kenaf silage. Total gas production was higher in ryegrass silage, followed by kenaf silage and rice straw silage (p<0.05). Total VFA and individual VFA (acetate, propionate, and n-butyrate) were greater in kenaf silage than rice straw silage (p<0.05). In vitro dry matter digestibility showed a similar trend to that of total gas and VFA production; it was higher in ryegrass silage and lower in rice straw (p<0.05). Throughout the feeding trial, the rice straw silage group showed significantly greater average daily gain than did the others (p<0.05). The feed conversion ratio in the group fed kenaf silage was significantly greater than that of others (p<0.05). No significant differences were observed in yield or quality traits, including carcass weight, ribeye area, backfat thickness, and scores for marbling, meat color, and fat color (p>0.05). Conclusion: The results indicated that no negative effects on growth performance and carcass characteristics occurred across treatments. Therefore, kenaf could be substituted for rice straw, which is most widely used as a roughage source in Korea.
This study aimed to assess the effects of neutral detergent fiber (NDF) from rice straw on blood metabolites and productivity of dairy cows in the tropics. Eight Holstein ${\times}$ indigenous (75-87.5% Holstein) were randomly allocated to dietary treatments according to a double $4{\times}4$ Latin square design. Contents of roughage NDF from rice straw were 12.8, 14.8, 16.8 and 18.8% dry matter (DM) and concentrate NDF content was 10.2% DM, so that levels of dietary NDF were 23, 25, 27 and 29% DM. Dietary treatments were isonitrogenous and isocaloric diets. The average maximum and minimum temperature humidity index during the experimental period were 85.6 and 76.9, respectively. Plasma glucose (p<0.05), dry matter intake and 4% fat corrected milk (p<0.01) decreased with increasing contents of roughage NDF from rice straw, while rectal temperature, serum urea, cortisol, low density lipoprotein-cholesterol (p<0.05), serum total protein and ${\beta}$-hydroxybutyrate (p<0.01) increased. Serum triglyceride, total cholesterol and high density lipoprotein-cholesterol were not affected (p>0.05). Decreased rectal temperature and concentrations of serum cortisol were associated with decreasing levels of ${\beta}$-hydroxybutyrate and serum urea, but with increased concentrations of plasma glucose, DMI and 4% fat corrected milk indicating that lactating cows fed lower fiber diets were in lower oxidative stress. Therefore higher intake and metabolic efficiency in dairy cows fed diets lower in roughage NDF from rice straw would be of an advantage in productive systems under tropical conditions.
A filamentous microorganism was isolated from completely rotten wood for the production of cellulolytic enzyme. The Trichoderma sp. FJ1 produced a large amount of cellulolytic enzymes, such as CMC, xylanase, ${\beta}-glucosidase$, and avicelase. For the production of the enzymes, when cellulolsic wastes were used as carbon sources of strain FJ1, rice straw showed higher enzyme activities than sawdust and pulp. The activities of CMC, xylanase, ${\beta}-glucosidase$, and avicelase were 2.95, 5.89, 0.45, and 0.12 U/ml in use of rice straw, respectively. To enhance production of the enzymes, the mixture substrate of rice straw and commercial cellulosic materials was investigated as carbon sources. The highest activities of CMCase, ${\beta}-glucosidase$, and avicelase were found in the mixture of rice straw and avicel, particularly rice straw:avicel (50:50), and the highest xylanase was obtained in the mixture ratio of 71:29. Bacto peptone addition of 0.1% showed enhanced production of the cellulolytic enzymes in which the activities of CMCase, xylanase ${\beta}-glucosidase$, and avicelase were 19.23, 27.18, 1.28, and 0.53 U/ml, respectively. The production of the enzymes using rice straw was efficiently induced in present of avicel and pulp containing high content of cellulose. Consequently, the filamentous microorganism, strain FJ1 utilized various cellulosic wastes as carbon sources and cellulases productivities were excellent compared to those of others strains reported previously, suggesting that the strain FJ1 will be expected as a favorable candidate for biological saccharification of cellulosic wastes in further.
Soil organic matter (SOM) plays an important role in the continuous production and environmental conservation in arable soils. In particular, the decomposition of organic matter in soil might promote soil organic matter and fertility due to the mineralization of N. In this study, to evaluate the effect of organic matter amendment on the C mineralization and N dynamic, $CO_2-C$ flux, extractable N and $N_2O$ emission were determined using closed chamber for 4 weeks at 10, 15, $20^{\circ}C$ of incubation temperature after the mixture of $2Mgha^{-1}$ rice straw compost and rye in sandy loam and clay loam. Regardless of soil texture, decomposition rates of rice straw compost and rye at $10{\sim}20^{\circ}C$ of incubation temperature ranged from 0.9 to 3.8% and 8.8 to 20.3%, respectively. Rye application in soil increased $NH_4-N$ and $NO_3-N$ content as well as the $N_2O$ emission compared to the rice straw compost. After incubation for 4 weeks, total C content in two soils was higher in rice straw compost than in rye application. In conclusion, application of rice straw compost and rye to soil was able to improve the soil organic matter and fertility. However, organic matter including the recalcitrant compounds like rice straw compost would be effective on the management of soil organic matter and the reduction of greenhouse gases in soil.

Objective: This study was to evaluate the fermentation dynamics, structural and nonstructural carbohydrate composition and in vitro gas production of rice straw ensiled with lactic acid bacteria and molasses. Methods: Fresh rice straw was ensiled in 1-L laboratory silos with no additive control (C), Lactobacillus plantarum (L), molasses (M) and molasses+Lactobacillus plantarum (ML) for 6, 15, 30, and 60 days. After storage, the silages were subjected to microbial and chemical analyses as well as the further in vitro fermentation trial. Results: All additives increased lactic acid concentration, and reduced pH, dry matter (DM) loss and structural carbohydrate content relative to the control (p<0.05). The highest organic acid and residual sugar contents and lignocellulose reduction were observed in ML silage. L silage had the highest V-score with 88.10 followed by ML silage. L and ML silage improved in vitro DM digestibility as compared with other treatments, while in vitro neutral detergent fibre degradability (IVNDFD) was increased in M and ML silage (p<0.05). M silage significantly (p<0.05) increased propionic acid (PA) content and decreased butyric acid content and acetic acid/PA as well as 72-h cumulative gas production. Conclusion: The application of ML was effective for improving both the fermentation quality and in vitro digestibility of rice straw silage. Inclusion with molasses to rice straw could reduce in vitro ruminal gas production.
Analytical methods were developed to determine cyclosulfamuron residues in soil, water, rice grain and straw using high-performance liquid chromatography (HPLC) with ultraviolet absorption detection. In these methods, cyclosulfamuron was extracted with aqueous $Na_2HPO_4$/acetone and acetone/methanol mixture from soil and rice samples respectively. Liquid-liquid partition coupled with ion-associated technique, Florisil column chromatography, and solid-phase extraction (SPE) were used to separate cyclosulfamuron from interfering co-extractives prior to HPLC analysis. For water sample, the residue was enriched in $C_{18}$-SPE cartridge, cleaned up in situ, and directly subjected to HPLC. Reverse-phase HPLC under ion-suppression was successfully applied to determine cyclo-sulfamuron in sample extracts with the detection at its ${\lambda}_{max}$ (254 nm). Recoveries from fortified samples averaged $87.8{\pm}7.1%$ (n=12), $97.3{\pm}7.2%$ (n=12), $90.8{\pm}6.6%$ (n=6), and $78.5{\pm}6.7%$ (n=6) for soil, water, rice grain and straw, respectively. Detection limits of the methods were 0.004 mg/kg, 0.001 mg/L, 0.01 mg/kg and 0.02 mg/kg for soil, water, rice grain and straw samples, respectively.
To verify the utilization of biomass as energy, the combustion characteristic has been studied by an experimental combustion furnace under an isothermal and non-isothermal combustion. The wood pellet, rice straw and rice husk were used as biomass samples in this work. The characteristics of emission gases, dusts and residues from biomass combustion have been analyzed and compared with those of reuse derived fuel (RDF). From isothermal combustion experiments, it was found that the incomplete combustion of rice straw was greater that that of rice husk, wood pellet and RDF. This is due to the fact that the combustion reaction rate of the rice straw was faster than that of other samples, and the oxygen concentration in rice straw combustion was rapidly decreasing. It was also found that $NO_{X}$ concentration of emission gas from wood pellet combustion was the lowest. From non-isothermal combustion experiments, it was found that all samples were burned before $900^{\circ}C$. Also, the temperature range of $NO_{X}$ emission was similar to that of CO emission, on the other hand, $SO_{2}$ was emitted at a higher temperature than that of CO emission.
