To identify the effects embryo developmental stage and gelrite concentration on plant regeneration in seed culture of rice, mature and immature seeds of rice were cultured on the $N_{6}$ medium supplemented with2 mg/$\ell$ 2.4-D and different levels of gelrite(0.2~1.0%). The calli formed immature embryos were produced more plants than those from mature embryos. The maximum frequency of plant regeneration was achieved in the culture of the calli of immature embryos which was harvested at the 21$^{th}$ day after pollination. The plant regeneration on the medium with gelrite was more accelerate than that on the medium with agar. The highest frequency(55%) of plant regeneration was obtained from the calli transferred to the medium with 6g/$\ell$ gelrite.
Rice embryo is more abundant than endosperms in nutrients such as proteins, lipids, and vitamin B1. In this study, we constructed p53 plasmid that could be expressed in a plant system, and investigated optimal germination conditions in a variety of media. For construction of p53 plasmid, we performed p53 amplification from pCDNA-p53, subcloned to TA cloning vector, and then reconstructed into pGEM-CaMV plant expression vector. On the other hand, we prepared a variety of imbibition buffers and complete media for efficient germination of the rice embryo. Imbibition buffers prepared with different concentrations of salt or detergent showed no significant effect on germination efficiency. We prepared further culture media, such as solid agar, liquid media, and paper towel to establish the optimal conditions. Rice embryo showed germination rates of more than 70% in the solid medium, more than 60% in the paper towel medium, but less than 25% in liquid media, although germination rate did not differ with varying concentrations of salt and sucrose in culture media. Under the optimal germination conditions, we introduced the p53 plasmid using imbibition method, and finally detected human p53 gene expression in the germinated rice embryo. This method might present a novel, practical approach for evaluating efficient gene expression utilizing imbibition method in rice embryo.
An attempt was made to link rice embryo proteins to DNA sequences and to understand their functions. One hundred of the 700 spots detected on the embryo 2-DE gels were microsequenced. Of these, 28% of the embryo proteins were matched to DNA sequences with known functions, but 72% of the proteins were unknown in functions as previously reported (Woo et al. 2002). In addition, twenty-four protein spots with 100% of homology and nine with over 80% were matched to ESTs (expressed sequence tags) after expanding the amino acid sequences of the protein spots by Database searches using the available rice EST databases at the NCBI (http://www/ncbi.nlm.nih.gov/) and DDBJ (http://www.ddbj.nig.ac.jp/). The chromosomal location of some proteins were also obtained from the rice genetic map provided by Japanese Rice Genome Research Program (http://rgp.dna.affrc.go.jp). The DNA sequence databases including EST have been reported for rice (Oryza sativa L.) now provides whole or partial gene sequence, and recent advances in protein characterization allow the linking proteins to DNA sequences in the functional analysis. This work shows that proteome analysis could be a useful tool strategy to link sequence information and to functional genomics.
The amino acid pattern of embryo and endosperm of brown rice different in specific gravity was investigated using Jinheung (local leading temperate variety) and IR667-Suweon 213 (high-yielding newly bred tropical variety). 1. Embryo of IR667 (higher protein rice) showed lower protein, and lower lysine or essential amino acid per protein than that of Jinheung (lower protein rice). 2. In both embryo and endosperm nitrogen recovery as amino acids was highest in middle class of specific gravity and lowest in low class indicating that abundancy of non-protein nitrogen in low class and decomposition of amino acids by starch in high specific gravity class. 3. In both embryo and endosperm IR667 showed abundancy in order of glutamic acid, aspartic acid while Jinheung showed glutamic acid, arginine, suggesting varietal difference in nitrogen metabolism. 4. In both IR667 and Jinheung least amino acid was histidine and next leucine in embryo but histidine and next threonine in endosperm, suggesting organ difference in nitrogen metabolism.
This experiment was designed to get some information on the parts in which necrosis occurs first in the embryo of rice, and on the phenomenon of necrosis that develops into other parts. Norin No.6 harvested in 1967 was used for this experiment and necrosis appearance was investigated by the T.T.C. method. The results obtained are summarized as follows; 1) Necrosis of the embryo in rice occurred in the plumule initially. 2) It was observed that the necrosis developed from the plumule to the radicle. 3) Finally, it was shown that necrosis developed from the surface tissue of the embryo to the inside tissue.
The effect of germination on hydration and germination properties, and on the changes of fatty acids and amino acids profiles of a brown rice 'Keunnun' (KN) with a large embryo was compared to 'Ilpumbyeo' (IP) with a normal embryo. A rapid germination up to 24 hr was observed in both brown rice cultivars, afterward decreased with germination time. At 60 hr, the KN ($86.0{\pm}4.24%$) showed slightly lower germination capability than the IP ($97.0{\pm}1.41%$). Lower water uptake during germination was also found in the KN ($1.22{\pm}0.02\;g$) compared to the IP ($1.59{\pm}0.05\;g$). Major fatty acids were palmitic acid, oleic acid, and linoleic acid accounting for more than 95% of total fatty acids. The most abundant amino acid in both types was oleic acid, which was decreased during germination, whereas palmitic and linoleic acids were increased. Eight amino acids were detected, and a remarkable increase in ${\gamma}-amino$ butyric acid (GABA) during germination was observed. The KN was characterized with higher tasty amino acids of aspartic acid, glutamic acid, glycine, and alanine.
This study was performed to investigate the influence of giant embryo feeding on hypoglycemia in streptozotocin-induced diabetic rats. Streptozotocin -induced diabetic rats were fed four kinds of experimental diets such as corn starch diet as a control, polished rice diet, brown rice diet and giant embeyonic rice diet for 6 weeks. The body weight gain diabetic of control was signaficantly suppressed compared to that of the normal. The supplementation of giant embryo increased the surivival rate. Feeding of giant embryo appeared to have ameliorating effects on diabetic symptoms including features such as polyphasia, polyuria. Giant embryo shortened gastrointestinal transit time and increased total fecal weight, total fecal dry weight and fecal water content compared with diabetic control. Giant embryo showed fasting blood glucose lowering effect compared with diabetic control. The disaccharidase activities in proximal part of intestine such as maltase sucrase and lactase in giant embryo feeding groups were lower than diabetic control. The results of this study show that giant embryo supplementation may have a beneficial Veffect on the hypoglycemia may be useful in the diet therapy for diabetic.

BACKGROUND/OBJECTIVES Oxidative stress is a major cause of cancer. This study investigated the effects of the ethanol extracts from germinated and non-germinated Keunnunjami rice, a blackish-purple pigmented cultivar with giant embryo, on selected human cancer cell lines and on the antioxidant defense system of mice fed with a high-fat diet. MATERIALS/METHODS: High fat-fed mice were orally administered with either distilled water (HF) or extracts (0.25%, w/w) from brown (B), germinated brown (GB), Keunnunjami (K), and germinated Keunnunjami (GK) rice. RESULTS: In comparison with the brown rice extract, Keunnunjami extract showed higher anticancer effect against cervical and gastric cell lines but lower anticancer activity on liver and colon cancer cells. Mice from the HF group showed significantly higher lipid peroxidation and lower antioxidant enzyme activities than the control group. However, the oxidative stress induced by high-fat diet markedly decreased in B, GB, K, and GK groups as compared with the HF group. CONCLUSIONS: Germination may be an effective method for improving the anticancer and antioxidative properties of Keunnunjami rice and extracts from germinated Keunnunjami rice may serve as a therapeutic agent against cervical and gastric cancers and oxidative damage.
This study was carried out to find out the properties of rice grain with embryo part. Obong-byeo. The embryo part on the rice remained by 96.7% in Obongbyeo after complete removal of the brans by common milling method. In nutritional composition. the content of lipid. vitamin B$_1$ and niacin were greater in Obongbyeo than Nagdongbyeo as a check variety. In particular. the contents of vitamin B$_1$ and niacin were about 1,5 times higher. Increased weight and feed effect of rat raising with milled rice were 107 and 172% in Obongbyeo and 101 and 125% in Nagdongbyeo. respectively. Eating quality of Obongbyeo was better than Nagdongbyeo.
The purpose of this study was to determine the effect of different feeding ratios of whole crop barley silage on the embryo production in Hanwoo donors. All donors were basically fed 2.5 kg concentrate daily. Donors were divided into three groups according to the different feeding of forage; hay 70% and rice straw 30% (control, n = 21), whole crop barley silage 80% and rice straw 20% (T1, n = 25), and whole crop barley silage 60% and rice straw 40% (T2, n = 23) fed based on TDN 6.70/ BW 500 kg. All Hanwoo donors received a CIDR together with injections of 1 mg estradiol benzoate and 50 mg progesterone ($P_4$, Day 0). Four days later, they were superovulated with 28 mg FSH twice daily IM in decreasing doses over 4 days. Then donors received 2 doses of $PGF_2{\alpha}$ (25 and 15 mg) with the 5th and 6th injections of FSH on Day 6. CIDR were withdrawn at the 6th FSH injection and the donors received $100\;{\mu}g$ GnRH 36 h after the second $PGF_2{\alpha}$ injection. The donors were artificially inseminated twice, at 8 and 24 h after GnRH, and embryos were recovered 7 or 8 days after the 1st insemination. The flush rate of the donors following positive superovulation responses did not differ among groups (76.2~96.0%, p>0.05). The number of corpus luteum (CL) at embryo recovery also did not differ among groups (10.6~14.0, p>0.05). Furthermore, the mean numbers of total ova (9.4, 10.5 and 12.0) and transferable embryos (5.3, 12.0 and 6.5) did not significantly differ among the control, T1 and T2 groups, respectively (p>0.05). However, mean concentrations of serum $P_4$ of the T1 (64.2 ng/ml) and T2 groups (55.7 ng/ml) were higher than that of control group (43.3 ng/ml, p<0.01), while serum cholesterol concentrations in the control (105.8 mg/dl) and T2 groups ($96.9\;{\pm}\;mg/dl$) were significantly lower than in the T1 group (121.1 mg/dl, p<0.05). Conclusively, whole crop barley silage can be fed a good substitute for hay forage for Hanwoo donors. Furthermore the ratios of whole crop barley silage 60% and rice straw 40% might be more worthful for embryo production.
