Extraction and effect of whitening agents from chinese plants (중국산 천연물에서 미백성분의 추출 및 효과)

  • In this work, extraction and purification of the possible whitening agents from the Chinese plants; Chrysanthemum morifolium Ramat (xizang cai ju hua), Rhodiola sachalinensis, and Terminalia chebula Retzius have been described. The chopped leaves of Chrysanthemum morifolium Ramat and Terminalia chebula Retzius were added to water and ethyl ether, respectively. Components were separated on a GS310 column ($21.5{\times}500mm$ i.d., $10-15{\mu}m$) and concentrated into four or three portions. The chopped leaves of Rhodiola salientness were added to methanol and separated and concentrated on a column ($C_{18}$ column, $3.9^{\circ}$�F8;300 mm i.d., $15{\mu}m$) into two parts. The whitening effects of extracts were examined by in-vitro melanin production assay, in melana and B16 cells at a concentration of $10{\mu}g/ml$. The ethyl acetate layer of Chrysanthemum morifolium Ramat showed 92% melanin inhibitory at $200{\mu}g/ml$, the extract of Rhodiola sachalinensis showed a whitening effect of about 60% melanin inhibitory, which was more efficient than the whitening effect of arbutin (45.6%). The methanol extract of Terminalia chebula Retzius inhibited melanin expression by 90% at $100{\mu}g/ml$; however, it was toxic to B16 melanoma cells.

  • This study was performed to compare the effects of immuno-modulating activities of Rhodiola sachalinensis A. Bor. fractionized by consecutive solvent separation. The Cytotoxicity of all fractionized extracts on human kidney cell (HEK293) was lower than crude extracts. Generally, the butanol and chloroform extracts showed less cytotoxicity on about 10.07% and 9.67% than the crude extracts. For human immune B and T cell growth, chloroform fraction showed the highest cell growth compared to the control. The secretion of cytokines (IL-6, $TNF-\alpha$) on human B and T cells were increased by adding chloroform extracts. Also, NK cell growth was significantly improved up to nearly 30% by adding the supernatant of B cell medium grown with the chloroform fraction. It was also found that chloroform fraction could yield higher nitric oxide production from macrophage than untreated control cells. Differentiation of HL-60 cells was increased up to 131.9% after treatment with chloroform fraction extracts, compared to the control. These results indicate that the chloroform fraction of R. sachalinensis have high immune activation activity than others fractions and the crude extracts, implying that this chloroform fractions could be used a new functional material.

Anticarcinogenic and Antioxidant Effects of Rhodiola sachalinensis (홍경천의 항산화 · 항발암 효과 연구)

  • In this study, we investigated the anticarcinogenic and antioxidative activities of Rhodiola sachalinensis (RS). Hexane (RSMH), ethylether (RSMEE), ethylacetate (RSMEA), butanol (RSMB), aqueous (RSMA) fractions and methanol extract (RSM) were screened for their growth inhibition effects using 3- (4,5-dimethyl-thiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay on HepG2, HeLa, MCF-7 and HT-29 cells. The anticarcinogenic effects of RSMEE was most significant when tested on MCF-7 and HepG2 cell lines at the concentration of $500{\mu}g/mL$ which resulted about $84\%\;and\;90\%$ on MCF-7 and HepG2 cells, respectively. The quinone reductase (QR)-inducing activity of RSMH on HepG2 cells was 3.5 times higher compared with the control at the concentration of $200{\mu}g/mL$. Antioxidative activities of RSM, RSMEE, RSMEA and RSMB showed about $80\%$ of electron donating activity (EDA) which were very similar to that of vitamin C as a control. We observed morphological changes of shrinking and the blebbing of HepG2 cancer cell membranes depending on the concentration of RSMEE.

Isolation and Identification of Antimicrobial Active Substances from Rhodiola sachlinensis (홍경천(Rhodiola sachlinensis)에서 항균성 물질의 분리 및 동정)

  • The antimicrobial substances from Rhodiola sachalinensis were extncted, isolated and identified. The highest level of antimicrobial activity and its yield were obtained in methanol extract. The minimum inhibition concentrations of the methanol extract were 500 $\mu\textrm{g}$mL on agar plate and 100 $\mu\textrm{g}$mL in broth media for four gram positive and four gram negative microbials. The methanol extract was fractionated by n-hexane, chloroform, ethyl ether, ethyl acetate, and butanol, orderly. The separate was developed on the TLC plate with different solvent system ratio of chloroform and methanol. Nine substances were isolated from chloroform and methanol mixture(9:1, v/v). Among them, three isolates showed antimicrobial activity. Three substances separated by HPLC were identified by GC/MS(EI) spectrum and $^1$H, /sup13/C-NMR spectrum. They were gallic acid, (-)-epicatechin and kaempferol. The antimicrobial activities of each substances were shown gallic acid, (-)-epicatechin, kaempferol orderly.

  • 홍경천의 간보호 효과를 알아보고자 사염화탄소를 투여하여 간손상을 유발시킨 흰쥐에 홍경천 물추출물을 투여한 후 간독성 보호효과를 알아본 결과, 혈청중 ALT, AST, LDH, ALP 활성도는 사염화탄소 투여에 의해 증가하였으나, 사염화탄소 투여 후 홍경천 물추출물의 투여로 유의적인 감소를 나타내었다. Total cholesterol, total lipid, triglyceride는 사염화탄소 투여에 의해 증가하였으나, 사염화탄소 투여 후 홍경천 물추출물의 투여로 유의적인 감소를 나타내었다. 한편 phospholipid는 사염화탄소만 투여한 CCL군과 사염화탄소 투여 후 홍경천 물추출물을 투여 한 군 모두 유의성 차이는 없었으나 홍경천 물추출물을 투여함으로 증가함을 알 수 있었다. HDL-cholesterol의 함량은 사염화탄소만 투여한 CCL군에 비해 사염화탄소 투여 후 홍경천 물추출물을 투여 한 RSLIII군에서 유의적으로 높은 수치를 보였다. LDL-cholesterol은 사염화탄소만 투여 한 군과 사염화탄소를 투여후 홍경천 물추출물을 투여한 군간 유의적 차이를 확인할 수 없었다 이상의 결과에서 사염화탄소 투여로 각종 효소 활성도 및 지질의 함량이 증가되었는데 이는 사염화탄소 투여로 간세포에 손상이 유발되었음을 알 수 있었고, 사염화탄소 투여 후 홍경천 물추출물을 투여한 군에서 사염화탄소 투여로 증가된 각종 효소 활성도 및 각종 지질의 함량을 저하시키므로서 홍경천 물추출물이 손상된 간기능을 회복시킬 수 있을 것으로 사료된다.

Growth and Utility of Rhodiola sachalinensis in Baekdu Mountain II. Activities of Superoxide Dismutase in Portions of the Seedlings (백두산 자생참돌꽃의 생육과 이용 II. 유식물의 부위별 superoxide dismutase 활성)

  • Superoxide dismutase (SOD) activities were investigated from the portions of shoots and roots in accordance with developmental stage and in response to environmental stress and antioxidants using Rhodiola-seedlings. The rates of SOD activities were revealed highly at the portion of roots and its tip of seedlings in the latter stages rather than the initial stages. SOD activities of seedlings in the initial stages treated with sodium chloride and cadmium as environmental stressors showed the decrease by 15 and 30% with respect to the control, respectively. However, in spite of stressor-treatments, the activities in the roots were increased according to the growth period showing a maximum rate of up to 45%. Also, SOD activities of the seedling treated with ascorbic acid as a antioxidant were increased by 46% of control value, but this was similar to the rate revealed in the presence of stressors. These results suggest that SOD activities in Rhodiola-seedlings may be related with the important defence-system against injurious environments.

  • Rhodiola sachalinensis has been used as a traditional medicine in Asia. We were germination in vitro seedling of grow naturally in Chang bai Moutain. And callus induction from leaf segments, treatmented plant regeneration in plant growth regulators (Auxins and cytokinins). We investigated optimal conditions for efficient plant regeneration through callus induction and shoots formation on medium with various kinds of growth regulators. Callus induction and adventitious shoots formation was achieved when cytokinin and auxin combinated to this experiment. Especially, there was the highest callus induction rates when we were used to 1 mg/L kinetin and 2 mg/L NAA $(98\%)$, Adventitious shoots formation wear obtained difference rate when cytokinin alone 1 mg/L BA $(96.6\%)$. And regenerated plantlet was acclimatized and transplanted to the soil, showed $100\%$ survival.
