Electrophysiological and Morphological Classification of Inhibitory Interneurons in Layer II/III of the Rat Visual Cortex

  • Interneuron diversity is one of the key factors to hinder understanding the mechanism of cortical neural network functions even with their important roles. We characterized inhibitory interneurons in layer II/III of the rat primary visual cortex, using patch-clamp recording and confocal reconstruction, and classified inhibitory interneurons into fast spiking (FS), late spiking (LS), burst spiking (BS), and regular spiking non-pyramidal (RSNP) neurons according to their electrophysiological characteristics. Global parameters to identify inhibitory interneurons were resting membrane potential (>-70 mV) and action potential (AP) width (<0.9 msec at half amplitude). FS could be differentiated from LS, based on smaller amplitude of the AP (<∼50 mV) and shorter peak-to-trough time (P-T time) of the afterhyperpolarization (<4 msec). In addition to the shorter AP width, RSNP had the higher input resistance (>200 $M{Omega}$) and the shorter P-T time (<20 msec) than those of regular spiking pyramidal neurons. Confocal reconstruction of recorded cells revealed characteristic morphology of each subtype of inhibitory interneurons. Thus, our results provide at least four subtypes of inhibitory interneurons in layer II/III of the rat primary visual cortex and a classification scheme of inhibitory interneurons.

Cloning and Expression of pcbCD Genes in Escherichia coli from Pseudomonas sp. DJ-12 (Pseudomonas sp. DJ-12의 pcbCD 유전자의 클로닝과 Escherichia coli에서의 발현)

  • The pcb genes of Pseudomonas sp. DJ-12 coded for the catabolism of polychlorinated biphenyl (PCBs) and biphenyl. The products of the pcbCD genes were 2,3-dihydroxy-4'-chlorobiphenyl dioxygenase and meta-cleavage product (MCP) hydrolase, which acted on degradation of 2,3-dihydroxy-4'-chlorobiphenyl to 4-chlorobenzoate. The pcbCD genes were cloned in E. coli XLl-Blue, and then the pcbD gene was further subcloned. As a metabolite transformed from 2,3-dihydroxybiphenyl by the cloned cell of E coli CU103, benzoate was detected by the resting cell assay. The enzyme activities of 2,3-dihydroxybiphenyl dioxygease and MCP hydrolase produced in the cloned cells E. coli CU103 and CU105 were about 17 and 3 times higher than those of Pseudomonas sp. DJ-12, respectively.

Effects of Cryopreservation on Ca2+ Signals Induced by Membrane Depolarization, Caffeine, Thapsigargin and Progesterone in Boar Spermatozoa

  • Although the fertilizing ability of spermatozoa is greatly reduced after freezing, complete understanding of alterations induced by cryopreservation has not been elucidated. The present study evaluates the effects of cryopreservation on the $Ca^{2+}$ handling of boar spermatozoa using several sperm activators. Intracellular $Ca^{2+}$ signals from single spermatozoa were measured using confocal $Ca^{2+}$ imaging of unfrozen samples and of other spermatozoa after having been frozen. Elevation of the external $K^{2+}$ concentration elicited a three times larger $Ca^{2+}$ increase in fresh spermatozoa than in cryopreserved spermatozoa. Caffeine elicited $Ca^{2+}$ transients with some oscillations in the fresh spermatozoa, but not in the thawed spermatozoa. Depletion of the $Ca^{2+}$ store with thapsigargin induced a rapid rise in $Ca^{2+}$ in the control but generated a smaller increase of $Ca^{2+}$ after thawing. Exposure to progesterone induced a biphasic rise of the $Ca^{2+}$ level in the fresh spermatozoa only. Sperm viability was reduced by cryopreservation. Resting $Ca^{2+}$ levels in fresh and cryopreserved spermatozoa were similar. Longer incubation (2.5 h) of thawed spermatozoa partly recovered the $Ca^{2+}$ response to the interventions. These results suggest that cryopreservation reduces the responsiveness of spermatozoa to depolarization, modulators of the internal $Ca^{2+}$ store and progesterone in terms of the $Ca^{2+}$ signal, thus providing a possible mechanism for reduced fertility observed in cryopreserved boar spermatozoa.

  • Purpose: Alterations in the structure and function of vascular smooth muscle cells (VSMCs) are important in cardiovascular disease and maintaining chronic hypertension. Chronic hypertension is associated with changes in vascular smooth muscle tone. The spontaneous or myogenic tone of a blood vessel reflects the ability to adapt smooth muscle tone to changes in transmural pressure. However, the intracellular signaling mechanisms involved in myogenic tone are not fully understood. Methods: Here, we investigated the relationship between mitogen-activated protein kinases (MAPKs) and phosphatidylinositol-3 kinase (PI3K) in isometric contraction and enzymatic activity using muscle strips from rats made hypertensive with aldosterone-analogue deoxycorticosterone acetate (DOCA) salts. Results: Changes in myogenic tone and intracellular $Ca^{2+}$ concentration ($[Ca^{2+}]_i$) were different after physiological salt solution (PSS) in normotensive and hypertensive rats. The myogenic tone and quiescent phosphorylation induced by the PSS treatment were inhibited by 10 ${\mu}$M PD098059, an extracellular-regulated protein kinase 1/2 (ERK1/2) inhibitor, and 10 ${\mu}$M wortmannin, an inhibitor of PI3K, in hypertensive rats. Conclusion: The development of DOCA-induced hypertension is associated with altered isometric contractions and $[Ca^{2+}]_i$ via changes in activation of ERK1/2 and PI3K after DOCA-salt treatment. Therefore, ERK1/2 and PI3K activity affect hypertension and may be suitable targets for physical therapy in cardiovascular disease.

  • The effects of Korean Red Ginseng (KRG) in rats submitted to exhaustion exercise have been studied, by measuring different enzymatic and hematological parameters in plasma and muscle. KRG powder was daily administered to 15 male Wistar rats for a period of two weeks. Another group of 15 rats with the same characteristics were administered physiological saline. Both groups were divided as follows: 5 control. 5 exercised till exhaustion and 5 recovered for 48 h after exhaustion. The following results were obtained for the groups treated with KRG in rapport to those treated with saline: - Higher endurance to running, - Increase of the osmotic resistence of red blood cells and higher presence of reticulocytes. - Lower triglyceride levels in plasma. - Increase non statistically significant of urea levels in plasma, - Lower non statistically significant hypoglycemia after exhaustion exercise. - Decrease of liver glycogen after exercise and faster recovery of the resting - level. - Protective effect on tissular damage produced by exhaustion exercise - Lower LDH activity in all studied muscles. only statistically significant in the WG.

  • Reproductive biology of the naked-headed goby, Faronigobius gymnauchen was investigated by means of histological methods. The ovary was consisted of several ovarian lamellae and the oogonia originated from the inner surface of the ovarian lamella. The testis was seminiferous tubule One in internal structure. Seminiferous tubule was consisted of many testicular cysts which contained numerous germ cells in a same developmental stage. The size of group maturity was 4.5 cm intotal length. Gonadosomatic index(GSI) of the female and male was the highest in June and July, respectively. Reproductive cycle could be classified into the growing ($January{\~}March$), maturation ($April{\~}May$), ripe and spent (June{\~}July$), and recovery and resting ($August{\~}December$). Oocyte development was group-synchronous, and yolk nucleus was observed in the early growing oocyte.

  • Rhodopseudomonas sphaeroides K-7 evolves large quantities of molecular hydrogen under anaerobic and light illuminated conditions in the presence of utilizable organic compounds as electron donors. Photoevolution of molecular hydrogen was strictly dependent on light as the activity of nitrogenase in this organism. Both of these were inhibited to the nearly same extent at varying concentrations of ammonium ion which also depressed nitrogenase synthesis. In the reaction mixtures devoid of molybdenum ion which is known as the component of nitrogenase, hydrogen evolution also decreased similarly like nitrogenase activity. Photoevolution of molecular hydrogen appeared to have no relationship with hydrogenase activity and bacteriocholophyll content and it was markedly inhibited under the atmosphere of $C_2$H$_2$, $N_2$ or $O_2$. The results strongly indicate that hydrogen evolution by R. sphaeroides K-7 might be catalyzed by nitrogenase. Both hydrogen evolution and nitrogenase activity were largely influenced by the nutritional history of the resting cells. From which we propose that glutamate might play an important role in the regulation of nigrogenase activity in vivo.

  • Whole cells of Methylosinus trichosporium OB3b, the obligate methylotroph, were used to produce propylene oxide from propane. This strain has methane monooxygenase, which catalyzes the conversion methane to methanol and can catalyze also the conversion propane to propylene oxide. Optimal condition for the production of propylene oxide was investigated in resting-whole cell system. The optimal pH and temperature was 7.0 and $35^{\circ}C$ respectively. The end product, propylene oxide, didn't inhibit the production of propylene oxide and was not further metabolized in reaction mixture. The addition of methane metabolites (methanol, formaldehde and formic acid) to the reaction mixture stimulated formation of propylene oxide by $3{\sim}4$ times, and methanol was the most effective especially. Under the optimal conditions, the 14.2 mM of propylene oxide was produced after incubation of 60 min. and the conversion ratio of propane to propylene oxide was approximately 8%.

  • For the purpose of investigating seasonal changes of germinal centers of splenic lymph nodules and of blood lymphocytes of ducks, the spleens of 8 ducks from a flock were observed histologically every season; 2 cases in spring (March and April), 2 in summer (July and August), 2 in autumn (October and November) and 2 in winter (December and January). Blood cells of 8 ducks from the same flock also were counted during the summer (from July to August) and autumn (from October to November) The results obtained were summarized as follows: 1. There were seasonal changes in the germinal centers of lymph nodules, that is, the germinal centers were formed in spring and disappeared in autumn. In summer these were at the stage of transition from formation in spring to disappearance in autumn. In winter, on tile other hand. the process was reversed from the stage of disappearance in autumn to the stage of formation in spring. 2. The germinal center of splenic lymph nodule was encapsulated with a fibrous capsule which disappeared concommitantly with its germinal center. 3. The percentage and absolute value of lymphocytes in autumn were higher than those in summer, the fact that seemed to be not in agreement with Flemming's view that the lymphocytic clear germinal center may be functionally at the stage of lymphocyte formation, but in agreement with Maximow's view that the large lymphocytic clear germinal center is functionally at the stage of resting and medium-sized lymphocytic germinal center may be functionally at the stage of lympocyte formatiom.

Enhanced 2,5-Furandicarboxylic Acid (FDCA) Production in Raoultella ornithinolytica BF60 by Manipulation of the Key Genes in FDCA Biosynthesis Pathway

  • The compound 2,5-furandicarboxylic acid (FDCA), an important bio-based monomer for the production of various polymers, can be obtained from 5-hydroxymethylfurfural (HMF). However, efficient production of FDCA from HMF via biocatalysis has not been well studied. In this study, we report the identification of key genes that are involved in FDCA synthesis and then the engineering of Raoultella ornithinolytica BF60 for biocatalytic oxidation of HMF to FDCA using its resting cells. Specifically, previously unknown candidate genes, adhP3 and alkR, which were responsible for the reduction of HMF to the undesired product 2,5-bis(hydroxymethyl)furan (HMF alcohol), were identified by transcriptomic analysis. Combinatorial deletion of these two genes resulted in 85.7% reduction in HMF alcohol formation and 23.7% improvement in FDCA production (242.0 mM). Subsequently, an aldehyde dehydrogenase, AldH, which was responsible for the oxidation of the intermediate 5-formyl-2-furoic acid (FFA) to FDCA, was identified and characterized. Finally, FDCA production was further improved by overexpressing AldH, resulting in a 96.2% yield of 264.7 mM FDCA. Importantly, the identification of these key genes not only contributes to our understanding of the FDCA synthesis pathway in R. ornithinolytica BF60 but also allows for improved FDCA production efficiency. Moreover, this work is likely to provide a valuable reference for producing other furanic chemicals.
