Cytokine and antibody responses of reactivated murine toxoplasmosis upon administration of dexamethasone

  • Toxoplasma gondii has been shown to result in life-threatening encephalitis in immunocompromised patients after reactivation of dormant parasites. In order to obtain information on immune responses related to this phenomenon, BALB/c mice were infected with 25 cysts of the 76K strain of T. gondii, then, treated orally with dexamethasone (Toxo/Dexa-treated group) in order to reactivate the chronic toxoplasmosis. None of the T. gondii-infected mice died during the experimental periods, whereas the Toxo/Dexa-treated mice evidenced a significant attenuation of survival periods. Toxoplasma-specific IgG2a, IgA and IgM titers in sera were significantly depressed in the Toxo/Dexa-treated mice; however, the IgG1 sera titers were similar to those seen in the Toxoplasma-infected mice. The percentages of CD4+ and $CD8\alpha+$ T cells in the Toxo/Dexa-treated mice were significantly reduced 2 weeks after dexamethasone treatment. $IFN-\gamma$ and IL-10 production levels in the Toxo/Dexa-treated mice were depressed significantly, whereas IL-4 production was increased temporarily. The expression levels of the Toxoplasma-specific P30 and B1 genes were found to have been increased in the Toxo/Dexa-treated mice in comparison with the Toxoplasma-infected mice. Collectively, the findings of this study demonstrate that reactivation of murine toxoplasmosis as the result of dexamethasone treatment induced a depression in Th1 immune responses, whereas Th2 immune responses were not significantly influenced.

Montelukast Reduces Serum Levels of Eosinophil-Derived Neurotoxin in Preschool Asthma

  • Purpose: Several markers for eosinophilic inflammation have been proposed to predict response to asthma treatment. However, definitive criteria for treatment decisions have not yet been established. We investigate a potentially useful relatively non-invasive biomarker, eosinophil-derived neurotoxin (EDN), to predict favorable responses to budesonide or montelukast, common treatment for children with asthma. Methods: Young children (1 to 6 years old) were enrolled in this randomized, parallel, 2-group, open-label trial. Criteria for eligibility included: 1) being symptomatic during the run-in period; and 2) having a serum EDN (sEDN) level ${\geq}53ng/mL$, with positive specific immunoglobulin E to house dust mite. Eligible patients were randomly placed into 2 groups: the BIS group received budesonide inhalation suspension (BIS) 0.5 mg once daily; the MONT group received montelukast 4 mg once daily. Ineligible patients were invited to receive montelukast 4 mg once daily (OBS group). Treatment period was 12 weeks. Results: Asthma control days increased significantly in the BIS and MONT groups (P < 0.000) over the 12-week study period. There was no significant change in sEDN in the BIS group but there was a significant decrease in the MONT group (P < 0.000). Patients in the OBS group with high EDN levels (> 53 ng/mL) showed a significant decrease due to MONT treatment (P = 0.023). Rescue medication usage significantly decreased in the BIS and MONT groups (P < 0.000). Conclusions: EDN is a useful relatively non-invasive biomarker for predicting responses to montelukast and budesonide treatment of preschool children with beta2-agonist responsive recurrent wheeze and multiple-trigger wheeze (Trial registry at UMIN Clinical Trials Registry, UMIN000008335).

  • Three horticultural herbaceous plants and a natural herbaceous plant were tested to determine the growth responses, biomass and uptake of cadmium(cd), lead(pb) by application of cd and pb soil treatment in pot culture. The ecological effects on the growth of the plants were investigated to determine the tolerance for the heavy metal pollutants cd and pb. the marginal concentrain of cd treatment on the growth of the each plant was below the 1, 000 ppm treatment of cd. The marginal concentration of pb treatment was below the 1, 000ppm treatment of pb in cultivation of salvia splendens ker., celosia cristata l. and below the 3, 000ppm treatment of pb in cultivation of portulaca grandiflora hook., sedum saramentosum bunge. the resistance for cd of sedum saramentosum bunge, celosia criastata l., portulaca grandiflora hook. and salvia splendens ker. was in the listed order. The resistence for pb was in order of sedum saramentosum bunge, portulaca grandiflora hook. Salvia splendens ker.and celosia criastata l.stems. The flowering of portulaca grandiflora hook. was sustained in the pb 1, 000ppm treated group only. The higher the concentration of pb in the soil cultivated the plants was, the less the content of leaf chlorophy11 in each plant was. The number of stomata per unit leaf area was the highest in salvia splendens ker. and in order of celosia l., sedum saramentosum bunge., portulaca grandiflora hook., the higher the cd and pb concentration of cd and pb treatment was, the more the concent of cd and pb in the part of each plant increased. the content of cd and pb in the stems of salvia spiendens ker. was the highest in the 1, 000ppm-treated ground and in order of the roots, the leaves and the flowers.

Acclimation responses of Tamarix chinensis seedlings related to cold stress

  • The purpose of this study was to investigate the acclimation responses of Tamarix chinensis to cold stress. We evaluated the acclimation responses by measuring biomass, daily elongation rate, chlorophyll content, and total soluble carbohydrate content. The plant samples comprised leaves from seedlings of 2 different ages (8 and 12 weeks); the leaves were collected 0, 2, and 4 weeks after cold treatment. We found that the cold-treated samples showed reduced daily elongation rates and chlorophyll content. Further, these samples showed more than 8-fold increase in the total soluble carbohydrate content. However, the seedling ages did not have a significant influence on the growth of cold-treated seedlings. On the basis of these findings, we can conclude that T. chinensis seedlings aged less than 1 year old show acclimation to cold stress by accumulating soluble carbohydrates. This study may help us understand how T. chinensis seedlings acclimatize to their first cold season.

MtMKK5 inhibits nitrogen-fixing nodule development by enhancing defense signaling

  • The mitogen-activated protein kinase (MAPK) signaling cascade is essential for a wide range of cellular responses in plants, including defense responses, responses to abiotic stress, hormone signaling, and developmental processes. Recent investigations have shown that the stress, ethylene, and MAPK signaling pathways negatively affect the formation of nitrogen-fixing nodules by directly modulating the symbiotic signaling components. However, the molecular mechanisms underlying the defense responses mediated by MAPK signaling in the organogenesis of nitrogen-fixing nodules remain unclear. In the present study, I demonstrate that the Medicago truncatula mitogen-activated protein kinase kinase 5 (MtMKK5)-Medicago truncatula mitogen-activated protein kinase 3/6 (MtMPK3/6) signaling module, expressed specifically in the symbiotic nodules, promotes defense signaling, but not ethylene signaling pathways, thereby inhibiting nodule development in M. truncatula. U0126 treatment resulted in increased cell division in the nodule meristem zone due to the inhibition of MAPK signaling. The phosphorylated TEY motif in the activation domain of MtMPK3/6 was the target domain associated with specific interactions with MtMKK5. I have confirmed the physical interactions between M. truncatula nodule inception (MtNIN) and MtMPK3/6. In the presence of high expression levels of the defense-related genes FRK1 and WRKY29, MtMKK5a overexpression significantly enhanced the defense responses of Arabidopsis against Pseudomonas syringae pv. tomato DC3000 (Pst DC3000). Overall, my data show that the negative regulation of symbiotic nitrogen-fixing nodule organogenesis by defense signaling pathways is mediated by the MtMKK5-MtMPK3/6 module.

Inhibitory Effects of Polyphenol-Rich Fraction Extracted from Rubus coreanum M on Thoracic Aortic Contractility of Spontaneously Hypertensive Rats

  • The purpose of the present study was to investigate whether polyphenol-rich fraction extracted from fruit wine of Rubus coreanum M (PCRC) can affect the contractility of the thoacic aortic strips isolated from spontaneously hypertensive rats (SHRs), and to clarify its mechanism of action. PCRC (200-800 ${\mu}g/mL$) concentration-depenedently blocked phenylephrine (10 ${\mu}M$)-induced contractile responses of the isolated aortic strips of SHRs. PCRC (400 ${\mu}g/mL$), added in to bath medium, also depressed the contractile active tension evoked by both phenylephrine (3 and 10 ${\mu}M$) and high potassium (25 and 56 mM). In the simultaneous presence of PCRC (400 ${\mu}g/mL$) and L-NAME (a selective inhibitor of NO synthase, 300 ${\mu}M$), the contractile responses evoked by phenylephrine and high $K^+$ were recovered to considerable level of the corresponding control contractility compared with those effects of PCRC-treatment alone. However, in the simultaneous presence of indomethacin (10 ${\mu}M$, a selective cyclooxygenase inhibitor) and PCRC (400 ${\mu}g/mL$), they were not affected. In the endothelium-denuded aortic strips by CHAPS-treatment, PCRC did not affect the contractile responses induced by phenylephrine or high potassium. Interestingly, PCRC (1.0, 3.0 and 10.0 mg/kg/30 min, i.v., respectively) dose-dependently suppressed norepiphrine-induced vasopressor responses in anesthetized SHRs. Collectively, we concluded that PCRC causes vasorelaxation in the thoracic aortic strips with intact endothelium of SHRs at least partly by the increased NO production through the activation of NO synthase of vascular endothelium, but not through the activation of cyclooxygenase. These results suggest that PCRC might be helpful to prevent or alleviate cardiovascular diseases, including hypertension.

Floridoside suppresses pro-inflammatory responses by blocking MAPK signaling in activated microglia

  • Inflammatory conditions mediated by activated microglia lead to chronic neuro-degenerative diseases such as Alzheimer's, Parkinson's, and Huntington's diseases. This study was conducted to determine the effect of floridoside isolated from marine red algae Laurencia undulata on LPS (100 ng/ml) activated inflammatory responses in BV-2 microglia cells. The results show that floridoside has the ability to suppress pro-inflammatory responses in microglia by markedly inhibiting the production of nitric oxide (NO) and reactive oxygen species (ROS). Moreover, floridoside down-regulated the protein and gene expression levels of iNOS and COX-2 by significantly blocking the phosphorylation of p38 and ERK in BV-2 cells. Collectively, these results indicate that floridoside has the potential to be developed as an active agent for the treatment of neuro-inflammation.

잣버섯 균사체로부터 분리한 수용성 단백다당체 Lepidan의 면역 증가 작용 (Enhancement of Immune Responses by a Water Soluble Proteoglycan, Lepidan from Lentinus lepideus)

  • In this study, we investigated the immunopotent activities of lepidan, a water soluble proteoglycan from Lentinus lepideus. To examine whether lepidan may affect nonspecific immune responses, we determined the effect on the production of nitric oxide (NO). Lepidan effectively increased the NO production in IFN-${\gamma}$ and LPS triggered RAW 264.7 cells. To further demonstrate the evidence that lepidan activates various immune cells, we determined the alkaline phosphatase activity, plaque-forming cell number and secretion of interleukine-4 (IL-4) and granulocyte/macrophage-colony stimulating factor (GM-CSF) after lepidan treatment in murine splenocytes. The results showed that lepidan increased alkaline phosphatase activity and the number of plaque-forming cells, which indicates that lepidan can lead B lymphocytes to late stage of differentiation. Also, when the splenocytes were cultured with lepidan for 48 hr, the level of IL-4 and GM-CSF in the supernatant dramatically increased. Taken together, these data suggest that lepidan is a biological response modulator that is able to activate immune responses.

흰쥐의 척수손상 후 천궁처리에 의한 피질척수로 축삭재생 반응시 Glial cells의 역할 (The Role of Glial Cells in Regenerative Responses of the Injured Corticospinal Tract Axons in Rats Treated with Cindii Rhizoma)

  • 목 적 : 천궁은 중추신경계 신경조직의 손상을 감소시키는 데에 유효한 것으로 보고되었다. 따라서 손상 척수신경에서 천궁에 의한 각각의 신경조직의 반응성을 조사하기 위하여 흰쥐의 손상된 척수 신경에 천궁 추출물을 처리하여 관찰하였다. 방 법 : 흰쥐의 척수조직에 타박손상을 가한 후 천궁을 처리하고 손상부위의 GAP-43과 Cdc2 및 Erk1/2 단백질의 수준, 축삭 및 성상세포의 수, 아교상혼의 정도를 각각 확인하여 손상부위 및 피질척수로의 재성장 여부를 관찰하였다. 결 과 : 천궁처리에 의하여 손상부위에서는 GAP-43과 Cdc2 및 Erk1/2 단백질의 수준이 증가하였다. 손상척수 부위에서 성상세포의 수는 천궁처리에 의하여 비손상대조군에 비해 증가하였다. CSPG 단백질에 의하여 확인된 아교상흔의 정도는 천궁처리에 의하여 감소하였다. Dil에 의한 정방향 염색에 의하여 피질척수로를 확인하였으며, 측방축삭의 싹돌기는 천궁처리에 의하여 손상부위 상단부 및 하단부에 각각 증가하였다. 고분자미세관을 손상척수부위에 이식하여 미세관내부로 축삭 및 비신경세포의 이주양상을 분석한 결과 천궁처리에 의하여 축삭 및 성상교세포의 이주증가를 확인하였다. 결론 : 천궁이 손상척수신경부위에서 비신경세포 특히 성상세포의 활성화에 기여함을 알 수 있었다. 이러한 활성화는 손상 피질척수로 축삭의 재성장 반응을 증가시키는 효과가 있는 것으로 판단된다.

Hesperetin suppresses LPS/high glucose-induced inflammatory responses via TLR/MyD88/NF-κB signaling pathways in THP-1 cells

  • BACKGROUND/OBJECTIVES: Unregulated inflammatory responses caused by hyperglycemia may induce diabetes complications. Hesperetin, a bioflavonoid, is a glycoside in citrus fruits and is known to have antioxidant and anticarcinogenic properties. However, the effect of inflammation on the diabetic environment has not been reported to date. In this study, we investigated the effect of hesperetin on proinflammatory cytokine secretion and its underlying mechanistic regulation in THP-1 macrophages with co-treatment LPS and hyperglycemic conditions. MATERIALS/METHODS: THP-1 cells differentiated by PMA (1 µM) were cultured for 48 h in the presence or absence of hesperetin under normoglycemic (5.5 mM/L glucose) or hyperglycemic (25 mM/L glucose) conditions and then treated with LPS (100 ng/mL) for 6 h before harvesting. Inflammation-related proteins and mRNA levels were evaluated by enzyme-linked immunosorbent assay, western blot, and quantitative polymerase chain reaction analyses. RESULTS: Hesperetin (0-100 µM, 48 h) treatment did not affect cell viability. The tumor necrosis factor-α and interleukin-6 levels increased in cells co-treated with LPS under hyperglycemic conditions compared to normoglycemic conditions, and these increases were decreased by hesperetin treatment. The TLR2/4 and MyD88 activity levels increased in cells co-treated with LPS under hyperglycemic conditions compared to normoglycemic conditions; however, hesperetin treatment inhibited the TLR2/4 and MyD88 activity increases. In addition, nuclear factor-κB (NF-κB) and Acetyl-NF-κB levels increased in response to treatment with LPS under hyperglycemic conditions compared to normoglycemic conditions, but those levels were decreased when treated with hesperetin. SIRT3 and SIRT6 expressions were increased by hesperetin treatment. CONCLUSIONS: Our results suggest that hesperetin may be a potential agent for suppressing inflammation in diabetes.
