  • 59 strains of Salmonella enterica subspecies enterica serovar Typhimurium (S. Typhimurium) were isolated from pigs in Gyeongbuk province, collected from 2011 to 2016. The isolates were investigated for the presence of antimicrobial resistance and multi drug resistance patterns. All 59 S. Typhimurium isolates were resistant to at least one of 10 antibiotics used in this study, 100% of S. Typhimurium isolates from pigs were resistant to two or more antimicrobials. As many as 5 isolates of isolates from pigs were resistant to 8 of 10 antimicrobials tested in this study. The ACSTNaGmKNaCf, ACSTGmAuKT/S, ACSTGmKCfT/S resistance phenotype was observed in 3.4%, 3.4%, 1.7% of the 59 isolates, respectively.

In Vitro Effects of Essential Oils from Ostericum koreanum against Antibiotic-Resistant Salmonella spp

  • The essential oil fraction of Ostericum koreanum was analyzed by GC-MS. Inhibiting activities of this oil and its main components were tested by the broth dilution assay and disk diffusion test against one antibiotic-susceptible and two resistant strains of Salmonella enteritidis and S. typhimurium, respectively. The GC-MS analysis revealed thirty-four compounds; the main components were $\alpha$-pinene (41.12%), $\rho$-cresol (17.99%) and 4-methylacetophenone (7.90%). The essential oil of O. koreanum and its main components were significantly effective against the tested antibiotic-susceptible strains as well as against the resistant strains of the two Salmonella species, with MICs (minimum inhibitory concentrations) ranging from 2 mg/mL to 16 mg/mL. The anti-Salmonella effects of the oils were dose-dependent on $M\"{u}ller-Hinton$ agar plates in this experiment. Additionally, checkerboard titer test results demonstrated significant combined effects of streptomycin and O. koreanum oil or cresol, one of the main components of this oil, against the two streptomycin resistant strains of S. typhimurium, with FICIs ranging from 0.12 to 0.37.

Characterization of Drug-Resistant Salmonella enterica Serotype Typhimurium by Antibiograms, Plasmids, Integrons, Resistance Genes, and PFGE

  • Forty-seven Salmonella Typhimurium (33 zoonotic, 14 clinical) strains were tested for antimicrobial resistance using the standard disk diffusion method. The presence of relevant resistance genes and class 1 integrons were investigated by using PCR. Pulsed-field gel electrophoresis (PFGE) and plasmid profiling were carried out to determine the genomic diversity of Salmonella Typhimurium. Approximately 57.4% of the S. Typhimurium strains were multidrug resistant (MDR) and showed high resistance rates to tetracycline (70.2%), sulfonamides (57.4%), streptomycin (53.1%), ampicillin (29.7%), nalidixic acid (27.6%), kanamycin (23.4%), chloramphenicol (21.2%), and trimethoprim (19.1%). Resistance towards cephalosporins was noted for cephalothin (27.6%), cephradine (21.2%), amoxicillin clavulanic acid (17.0%), and cephalexin (17.0%). Resistance genes, $bla_{TEM}$, strA, aadA, sul1, sul2, tetA, tetB, and tetC, were detected among the drug-resistant strains. Thirtythree strains (70.2%) carried class 1 integrons, which were grouped in 9 different profiles. DNA sequencing identified sat, aadA, pse-1, and dfrA genes in variable regions on class 1 integrons. Thirty-five strains (74.4%) were subtyped to 22 different plasmid profiles, each with 1-6 plasmids (2.0 to 95 kb). PFGE subtyped the 47 strains into 39 profiles. In conclusion, high rates of multidrug resistance were found among the Malaysian Salmonella Typhimurium strains. The emergence of multidrug-resistant Salmonella Typhimurium to cephalosporin antibiotics was also observed. The strains were very diverse and no persistent clone was observed. The emergence of MDR Salmonella Typhimurium is a worldwide problem, and this report provides information for the better understanding of the prevalence and epidemiology of MDR S. Typhimurium in Malaysia.

Anti-Salmonella Activity of Lemongrass Oil Alone and in Combination with Antibiotics

  • The effects of Cymbopogon citratus essential oil (Lemongrass oil) and its main component, citral (84.30%), on antibiotic-susceptible and -resistant strains of Salmonella enteritidis and S. typhimurium were assessed. C. citratus oil and citral significantly inhibited all strains of the two Salmonella species examined, with minimum inhibiting concentrations (MICs) ranging from 0.5 mg/ml to 8.0 mg/ml. The combined effects of C. citratus oil and citral (84.30%) were evaluated using a checkerboard microtiter assay. Essential oil fractions of C. citratus and citral exhibited strong synergistic or additive effects with streptomycin or kanamycin against S. typhimurium strains with fractional inhibitory concentration (FIC) indices in the range of 0.28 to 1.00. In conclusion, a combination of streptomycin and lemongrass oil or its main component, citral, may be useful for reducing the minimum effective dose of antibiotic required for the treatment of resistant S. typhimurium infections.

초생추 유래 Salmonella속균의 생물학적 특성 (Studies on Salmonella isolated from chicks)

  • This paper deals with the distribution, reservoir and mode of spread of infection on 6 hatcheries in Taegu, Kyungpook and 5 broiler farms in Kyungpook during the period from June 1991 to June 1992. Isolated Salmonella were examined for serotypes, biotyping of Salmonella(S) typhimurtum, antibiotic susceptibility and some biochemical characteristics. Forty two Salmonella strains were isolated from 42(2.7% of 1,577 caecal samples of chicks, and their serotypes were S typhimurium 10, S typhimurium var Copenhagen 5, S infantis 4, S thompson 3, and untypable 20. The isolation rate of Salmonella varied from 0 to 5.1% in 6 hatcheries and that of Salmonella from 5 broiler farms was 10. 5%. Biotypes of 10 S typhimurium and 5 S typhimurium var Copenhagen strains isolated from chicks of hatcheries and broiler farms were biotype 2(86.6%), 8(6.7%), and 10a(6.7%), and 26i(6.7%) according to Duguid's scheme. Antibiotic susceptibility test of Salmonella isolated were performed by agar dilution method, using 9 antibiotics as follows: ampicillin(Am), chloramphenicol(Cm), gentamicin(Gm), kanamycin(Km), nalidixic acid(Na), rifampicin(Rf), streptomycin(Sm), sulfadimethoxine(Su), and tetracycline(Tc), All the strains were sensitive to RF. But 8 strans(23.8%) were resistant to one or more drugs and the most common resistance patterns of transferred R plasmids were SmSuGm and SmSu. Among 42 isolates, one had transferable citrate utilizing plasmid. S typhimurium and S typhimurium var copenhagen strains were resistant to killing by 90% normal guinea pig serum.

Ampicillin과 Chloramphenicol 내성 Salmonella typhimurium 분리의 증가 (Increased Isolation of Ampicillin and Chloramphenicol Resistant Salmonella typhimurium)

  • Salmonella typhi infection, which was the most frequent enteric infection in Korea, has been decreasing, while the infection of other serogroups of Salmonella has been increasing since the later part of 1970s. In 1986, the number of serogroup B isolated by us increased to 46, which corresponds 21.1% of all enteropathogenic bacteria isolated from stool specimens. Salmonella isolates resistant to antimicrobial agents were extremly rare in Korea, in the 1970s. However, 7 of 13 serogroup B isolates showed resistance to ampicillin or to chloramphenicol in 1984. Among the serogroup B isolates in 1986, 71.7% and 69.6% were resistant to ampicillin and to chloramphenicol respectively. The minimum inhibitory concentrations of ampicillin and chloramphenicol against these isolates were >$128\;{\mu}g/ml$ and $128\;{\mu}g/ml$ respectively.

  • Salmonella Typhimurium is a virulent pathogen for human and animal. We studied serotypes, biochemical characteristics, and antimicrobial resistance of S. Typhimurium isolated from diseased pigs in Gyeongbuk province over 1998 to 2008. One hundred sixteen isolates were identified as S. Typhimurium by biochemical characteristics and serotypes from 90 farms. The biochemical characteristics of S. Typhimurium isolates was production of $H_2S$, indole-negative, fermentation of mannitol, dulcitol, sorbitol, inositol, rhamnose, and maltose, and ornithine decarboxylase. At antimicrobial susceptibility test, the majority of isolates were highly susceptible to amoxicillin/clavulanic acid, cefepime, ciprofloxacin, while were highly resistant streptomycin, cephalothin, enrofloxacin, nalidixic acid, apramycin, chloramphenicol, tetracycline. The isolates were divided into 65 resistant patterns and 47 of the isolates were shown as a DT104 ASSSuT resistant phenotype.

In Vitro Selection of RNA Aptamer Specific to Salmonella Typhimurium

  • Salmonella is a major foodborne pathogen that causes a variety of human diseases. Development of ligands directly and specifically binding to the Salmonella will be crucial for the rapid detection of, and thus for efficient protection from, the virulent bacteria. In this study, we identified a RNA aptamer-based ligand that can specifically recognize Salmonella Typhimurium through SELEX technology. To this end, we isolated and characterized an RNase-resistant RNA aptamer that bound to the OmpC protein of Salmonella Typhimurium with high specificity and affinity ($K_d$ ~ 20 nM). Of note, the selected aptamer was found to specifically bind to Salmonella Typhimurium, but neither to Gram-positive bacteria (Staphylococcus aureus) nor to other Gram-negative bacteria (Escherichia coli O157:H7). This was evinced by aptamer-immobilized ELISA and aptamer-linked precipitation experiments. This Salmonella species-specific aptamer could be useful as a diagnostic ligand against pathogen-caused foodborne sickness.

항생제 내성 및 감수성 Salmonella typhimurium 균주에 대한 개똥쑥 지상부 정유와 Kanamycin의 병용효과 (In vitro Effects of Essential Oils from the Aerial Parts of Artemisia annua L. Against Antibiotic-Susceptible and -Resistant Strains of Salmenella typhimurium)

  • The essential oil fraction from the aerial parts of A. annua was analyzed by GC-MS. As the results, caryophyllene oxide (11.7%), caryophyllene (7.54%), camphor (7.32%), 1,8-cineol (4.98%), and borneol (3.99%) were confirmed as the main components of the oil fraction. The effects of this oil and its main components on antibiotic-susceptible and -resistant strains of Salmonella enteritidis and S. typhimurium were assessed. A. annua oil fraction significantly inhibited all strains of the two Salmonella species examined, with minimum inhibiting concentrations (MICs) ranging from 2.0 mg/ml to 8.0 mg/ml. Among the main components of the oil, borneol and camphor showed relatively strong inhibiting activity with MICs between 1.0 mg/ml and 4.0 mg/ml. The MICs of caryophyllene and caryophyllene oxide were higher than 16 mg/ml. The combination effects of the oils with kanamycin were evaluated using a checkerboard microtiter assay. Against S. typhimurium KCCM11862 and CCARM8009 strains, the oil fraction of A. annua, camphor, and 1,8-cineol exhibited significant synergistic with kanamycin with fractional inhibitory concentration (FIC) indices in the range of 0.085 to 0.375. In conclusion, a combination of kanamycin and A. annua oil or its main component, camphor, and cineol, may be useful for reducing the minimum effective dose of antibiotic required for the treatment of resistant S. typhimurium infections.

닭과 돼지에서 분리한 Salmonella Typhimurium의 항생제 내성 특성 분석 (Characteristics of antimicrobial resistant Salmonella Typhimurium isolates from chicken and pigs)

  • Fifty and forty two Salmonella enterica subspecies enterica serovar 52 Typhimurium (ST) strains were isolated from chicken and pigs, respectively, collected from markets throughout Korea from 2008 to 2011. The isolates were investigated for the presence of antimicrobial resistance and multi-drug patterns. All 50 ST isolates from chicken and 42 ST isolates from pigs were resistant to at least one of 13 antibiotics used in this study, 92.0% of ST isolates from chicken and 88.1% of ST isolates from pigs were resistant to three or more antimicrobials. As many as 3 isolates of ST isolates from chicken were resistant to 11 of 13 antimicrobials tested in this study. Only one isolate of ST isolates from pigs was resistant to 10 of 13 antimicrobials. The ACSSuT resistance phenotype was observed in 34% of the 50 isolates and 23.8% of the 42 isolates. Especially, 1 isolate from pigs had the ACSSuTAu. The high rate of antimicrobial-resist and multi-drug resistant (MDR) ST isolation may give rise to crucial public health problems. Therefore, control of antimicrobial use, and continuous monitoring of antimicrobial resistance and MDR patterns among Salmonella isolates in chicken and pig farms is necessary to ensure public health.
