  • Caspases are a family of cysteine proteases that play an important role in the apoptotic pathway. Caspase-6 is an apoptosis effector that cleaves a variety of cellular substrates. The active form of the enzyme is required for use in research. However, it has been difficult to obtain sufficient quantities of active caspase-6 from Escherichia coli. In the present study, we constructed a caspase-6 with a 23-amino-acid deletion in the pro-domain. This engineered enzyme was expressed as a soluble protein in E. coli and was purified using affinity resin. In vitro enzyme assay and cleavage analysis revealed that the engineered active caspase-6 protein had characteristics similar to those of wild-type caspase-6. This novel method can be a valuable tool for obtaining active caspase-6 that can be used for screening caspase-6-specific substrates, which in turn can be used to elucidate the function of caspase-6 in apoptosis.

  • This study was done to examine adherence of oral bacteria to titanium dental implant and to know the effective prophylactic antibiotics using an in vivo model. Three samples each of the implant material were set in an acrylic resin flange and placed in the maxillary buccal sulcus of twenty volunteers. At 6- and 54-hour intervals, each sample was placed on blood agar plate (BAP) and chocolate agar, and then they were incubated and identified. Also antibiotic susceptibility test was performed. The results obtained mere as follows ; 1. The microorganisms were chain-like Gram positive cocci and staphyline Gram positive cocci, Gram positive bacilli in order of frequency were found at 6-hour and 54-hour samples by Gram staining. 2. Streptococci was found predominantly at both 6-hour and 54-hour samples, but number of streptococci was decreased as compared to 6-hour samples. 3. There was no difference in the bacterial species adherent to implant between 6-hour and 54-hour samples. 4. All the microbes were sensitive to AMC (amoxacillin clavulanic acid), chloramphenicol, quinolone and vancomycin in the antibiotic susceptibility test. Above results suggest that streptococcus are mainly adhered to titanium implant after implant was placed in the oral cavity and AMC is the most recommendable antibiotics to prevent the peri-implant inflammation.

  • In 10M chloride (4M HCl + 6M LiCl) solution, cobalt, but not nickel, formed complex anion (${CoCl_3}^-$), and this anion was extracted by a liquid anion exchanger with Amberlite LA-2. The ion exchange capacity was 2.175meq of cobalt complex per unit ml of Amberlite LA-2. Upon eluting the resin with 0.4M nitric acid, the cobalt complex was stripped and transfered into eluate quantitatively. By using this separation method in the chloride solution dissolved with 50mg of cobalt (II) and 500mg of nikel(II), recovery of cobalt were 99.6 percent.

  • Catalytic oligomerization of isobutene to produce triisobutenes has been performed over a cation-exchange resin (Amberlyst-35) by using commercial C4 feeds. The catalytic activity in the oligomerization was retained without deactivation up to 90 h of reaction in a simulated reaction feed without butadiene, but its activity was significantly affected by the presence of butadiene in commercial C4 feeds. The isobutene conversion with time-on-stream was significantly decreased in the presence of butadiene, indicating the catalyst deactivation by butadiene. However, the stable activity for trimerization was accomplished when the oligomerization was carried out after eliminating butadiene by hydrogenation of the feeds. This work demonstrates that butadiene plays a role as a catalyst poison on the solid acid catalyst, so that its removal in the reactant feed is essential for practical application of trimerization.

  • A new one-column chromatography process, analogous to a four-zone simulated moving bed (SMB), was presented. The basic principle of the process was identical to that of a four-zone SMB. The process consisted of one chromatographic column and four tanks, instead of the four columns in the four-zone SMB (1-1-1-1), and has been used for the separation of two amino acids, phenylalanine and tryptophan, using an ion exchange resin. The operating parameters for the one-column process and four-zone SMB were obtained from equilibrium theory. Computer simulations were used to compare the performances of the new one column process to that of the general four-zone SMB, using Aspen $Chromatography^{TM}$ v 11.1. The differences between the one-column and SMB processes in terms of the purities and yields of phenylalanine and tryptophan were less than 4 and about $6\%$, respectively. The lower purities of the one-column process were due to the loss of the developed concentration profiles in the column when the liquid was stored in tanks. The one-column process gave great flexibility, and would be useful for reconstructing an existing conventional chromatography process to one of a SMB.

  • The purpose of this study is to compare the biocompatibility of commercial purity Ti, Ti-6AI-4V and Ti-6AI-7Nb alloy specimens with and without surface treatment in mouse abdominal connective tissue in vivo. Each metal was implanted into specific abdominal subdermal tissue site of female mouse. After 4 weeks, the implants were removed and abdominal tissues were fixed, dehydrated and embedded in glycol methacrylate resin. And the tissues were histologically prepared for microscopical evaluation. It was characterized by the presence of connective tissue with fibrous capsule surrounding the implant. The fibrous tissue surrounding the implant was studied to determine the biocompatibility of implanted metals. The average thickness of the fibrous capsule formed around the implant was much thinner for the hydrogen peroxide added hydrochloric acid solution-treated specimen than for the others. The results of this evaluation indicate that modification of the surface properties of titanium and titanium alloy implants changes the biological properties in the abdominal connective tissue. In conclusion, these observations suggest that the proper surface treatment performed in the study is effective for the improvement of biocompatibility.

  • The curing of epoxy asphalt mixture depends on the chemical reaction of epoxy resin and the curing agent. The curing level of epoxy asphalt mixture needs to be predicted in order to decide traffic opening time and to establish further construction plans. In this study, chemical analysis of the prediction model was executed to expand the applicability of the previous prediction model. Consequently, the curing level prediction model of epoxy asphalt concrete mixture was proposed using the concentration ratio and the acid value ratio. According to the results of outdoor curing experiments, the final prediction model showed that the correlation coefficient is greater than 0.971. Precise prediction results of different composition epoxy asphalt were obtained by reflecting the chemical composition ratios in the curing level prediction model.

  • The antioxidative and protective activities of kefir, low-fat dry milk (NFDM) extract and fractions on SFME cells in serum-free medium were investigated. Kefir and low-fat kefir and NFDM extract were made by solubilizing the freeze dried powder forms in deionized water, filtering through glass prefilter, 12 ㎛ and 2 ㎛ membrane, and demineraling with chelating resin. Kefir, low-fat kefir and NFDM extract were fractioned into dialyzate and retentate by dialysis with membrane tube having the molecular cut-off of 3,500 Dalton. An antioxidative activity was analyzed by the in vitro model system using a linoleic acid. In the case of kefir an antioxidative activity was detected only in the retentate of kefir extract. On the other hand NFDM showed an antioxidative activity in extract, demineralized extract, dialyzate and retentate. The retentate of kefir extract had the higher antioxidative activity than that of NFDM extract. Kefir showed the protective effect of SFME cells in serum-free medium in extract, demineralized extract and retentate, but low-fat kefir didn't. NFDM had the similar protective effect on SFME cells as extract, demineralized extract and retentate of kefir.

  • For the selection of peptide specifically binding to $Pb^{2+}$, the biopanning with the commercially available Ph.D.-7 phage displayed heptapeptide library was carried out against $Pb^{2+}$ immobilized on a metal-chelating IDA (iminodiacetic acid) resin. After four rounds of screening against $Pb^{2+}$-IDA including negative selections against charged bead with metal ions other than $Pb^{2+}$ and uncharged bead, several candidate lead-binding phage peptides were initially determined based on the order of frequency from the screened phage clones. Of the selected phage peptide sequences, the peptide of the highest frequency, CysSerIleArgThrLeuHisGlnCys (CSIRTLHQC) also exhibited the strongest affinity for $Pb^{2+}$ in binding assays for individual phage clones. However, there was not a significant difference in $Pb^{2+}$ affinity between selected peptides when using synthetic heptapeptides corresponding to the displayed peptide sequences of phage clones.
