Complete Chloroplast Genome Sequence of Korean Endermic Species, Pseudostellaria longipedicellata

  • Pseudostellaria Pax (Caryophyllaceae) is a small genus distributed in temperate region. It consists of 25 species presenting high diversity in Asia. Pseudostellaria longipedicellata S. Lee, K. Heo & S. C. Kim was first announced as new species in 2012. Morphological characters of P. longipedicellata are closely related to those of Psedusotellaria palibiniana and Psedusotellaria okmotoi. These are distinguished from P. longipedicellata by shorter pedicel and puberulent pedicels, respectively and by being distributed allopatically between P. longipedicellata and rest of species. The complete chloroplast genome of P. longipedicellata was successfully rescued from raw reads generated by HiSeq2000. Its total length is 149,626 bp consisting of four regions: large single copy (LSC) region (81,292 bp), small single copy (SSC) region (16,984bp), and inverted repeats (IRs; 25,765 bp per each). It contained 126 genes (81 coding DNA sequence (CDS), eight rRNAs, and 37 tRNAs); 18 genes (seven CDS, four rRNAs, and seven tRNAs) are duplicated in inverted repeat regions. The overall GC content of P. longipedicellata is 36.5% and in the LSC, SSC, and IR regions were 34.3%, 29.3%, and 42.4%, respectively. Based on phylogenetic analysis of chloroplast genomes of P. longipedicellata and relatives species presents clear phylogenetic positions of Pseudostellaria genus. This chloroplast genome will be an important sequence resources for further researches of Pseudostellaria genus.

The Role of IMO for the Eradication of IUU Fishing (국제해사기구의 IUU 어업 퇴치 기여 및 전망)

  • The illegal, unreported and unregulated fishing can be properly regulated only if fishing activities, transhipment and landing of illegally caught fish, and processing of fish at sea involved in it are integrally managed. To do this, flag States, port States, coastal States, market States and relevant regional fisheries management organizations shall closely cooperate. In addition, fishing boats involved in IUU fishing often ignore requirements of safety of navigation so that they could be exposed to more causes of incidents and could have less chances to be rescued. Then, it seems to be necessary for the regulation on IUU fishing to include a certain consideration of strengthening safety of those ships. International Maritime Organization(IMO) has developed and implemented international rules for the safety of shipping. IMO has cooperated with Food and Agricultural Organization on IUU fishing since 2000 and the third joint meeting is scheduled in November 2015. This paper reviews the recommendations adopted in the previous meetings, and the measures actually taken as a result of the cooperation and, in particular, focus on the additional roles of IMO for the eradication of the IUU fishing.

Image Matching Algorithm for Thermal Panorama Image Construction Adaptable for Fire Disasters (화재상황에서 적용가능한 열화상 카메라의 파노라마 촬영을 위한 동일점 추출 알고리즘)

  • In a fire disaster in a tunnel, people should be rescued immediately using the information obtained from cameras or sensors. However, in heavy smoke from a fire, people cannot be clearly identified by a mounted CCTV, which is only effective in a clear environment. A thermal camera can be an alternative to this in smoky situations and is capable of detecting people from their emitted thermal energy. On the other hand, the thermal image camera has a smaller field of view than an ordinary camera due to its lens characteristics and temperature error, etc. In order to cover a relatively wide area, panoramic image construction needs to be implemented. In this work, a template-based similarity matching algorithm for constructing the panorama image is proposed and its performance is verified through experiments. This scheme provides guidelines for coping with difficulty in image construction, which requires an exact correspondence search for two images in cases of heavy smoke.

A77 1726 Inhibit NO-induced Apoptosis via PI-3K/AKT Signaling Pathway in Rabbit Articular Chondrocyte

  • Leflunomide is an immunomodulatory agent used for the treatment of rheumatoid arthritis (RA). Leflunomide known as a regulator of iNOS synthesis which largely decreases NO production in diverse cell type. However, the effect of leflunomide on chondrocyte is still poorly understood. In our previous studies, we have shown that direct production of Nitric oxide (NO) by treating chondrocytes with NO donor, sodium nitroprusside (SNP), causes apoptosis via p38 mitogen-activated protein kinase in association with elevation of p53 protein level, caspase-3 activation. In this study, we characterized the molecular mechanism by which A77 1726 inhibit apoptosis. We found that A77 1726 inhibit NO-induced apoptosis as determined by MTT (Thiazolyl Blue Tetrazolium Bromide) assay and DNA fragmentation. The inhibition of apoptosis by A77 1726 was accompanied by increased PI-3 kinase and AKT activities. So, inhibition of phosphatidylinositol (PI)-3kinase with LY294002 rescued apoptosis. Triciribine, the specific inhibitor of AKT, also abolished anti-apoptotic effect. Our results indicate that A77 1726, the active metabolite of leflunomide, mediates NO-induced apoptosis in chondrocytes by modulating up-regulation of PI-3 kinase and AKT.

Management of an Open Comminuted Fracture of the Metacarpus using Circular External Skeletal Fixation in a Korean Water Deer (Hydropotes inermis argyopus) (고라니에서 발생한 손허리뼈 개방 복합골절의 원형 외부골격고정법 적용)

  • A Korean water deer was rescued after being hit by a motor vehicle. Clinical and radiographic examination revealed a grade III open fracture of the right metacarpus with severe soft tissue injury. The water deer underwent closed reduction of the fracture, which was stabilized by the application of the Ilizarov method of 4-ring circular external skeletal fixation (CESF) as a minimally invasive technique. The water deer was successfully released into the wild. CESF was advantageous for the stabilization of open comminuted fracture and the management of a severely contaminated wound in a Korean water deer. CESF can be used successfully in wild deer with good tolerance and minimal complications.

  • A Eurasian jay (Garrulus glandarius) with lethargy, dyspnea, and poor physical condition was rescued and referred to the Animal Health Center, Seoul Zoo, Korea. The jay's eyelids were sealed with dark-colored hard scabs and it died the day after arrival. At necropsy, diphtheritic membranes on the tongue and oral mucosa were apparent, suggesting avian poxvirus infection based on the appearance of the oral diphtheritic lesions. PCR was conducted using a tissue sample to confirm the causative agent. The jay was diagnosed with an avian pox viral infection. This report describes the first case of a natural avian pox infection in a Eurasian jay in Korea.

Auranofin Suppresses Plasminogen Activator Inhibitor-2 Expression through Annexin A5 Induction in Human Prostate Cancer Cells

  • Auranofin has been developed as antirheumatic drugs, which is currently under clinical development for the treatment of chronic lymphocytic leukemia. Previous report showed that auranofin induced apoptosis by enhancement of annexin A5 expression in PC-3 cells. To understand the role of annexin A5 in auranofin-mediated apoptosis, we performed microarray data analysis to study annexin A5-controlled gene expression in annexin A5 knockdown PC-3 cells. Of differentially expressed genes, plasminogen activator inhibitor (PAI)-2 was increased by annexin A5 siRNA confirmed by qRT-PCR and western blot. Treatment with auranofin decreased PAI-2 and increased annexin A5 expression as well as promoting apoptosis. Furthermore, auranofin-induced apoptosis was recovered by annexin A5 siRNA but it was promoted by PAI-2 siRNA. Interestingly, knockdown of annexin A5 rescued PAI-2 expression suppressed by auranofin. Taken together, our study suggests that induction of annexin A5 by auranofin may enhance apoptosis through suppression of PAI-2 expression in PC-3 cells.

Management of a Severe Cervical Degloving Injury in a Bean Goose (Anser fabalis serrirostris)

  • A wild juvenile bean goose (Anser fabalis serrirostris) was rescued after sustaining an extensive degloving injury around the neck and chin region. Except for the degloving injury, physical and radiographic examination detected no other trauma-induced abnormality. On the day of presentation to the hospital, the patient underwent extensive debridement and suturing. A twice-daily force feeding with diluted parrot weaning food was required during the hospitalization due to severe anorexia. On day 18, the bean goose was moved to the aviary to undergo rehabilitation and feeding by itself. Management of the severe degloving injury of the cervical region with medical intervention and nutritional assistance was successful in this case, and the results of this case shows the proper management can be implemented to produce satisfactory outcomes.

MK-801-induced learning impairments reversed by physostigmine and nicotine in zebrafish

  • Previous studies have demonstrated that N-methyl-D-aspartate (NMDA) receptors and acetylcholine receptors are related to learning and memory in rat and mice. In this study, we examined the effects of MK-801, a non-competitive NMDA receptor antagonist, on learning and memory in zebrafish using a passive avoidance test. We further tested whether or not nicotine, a nicotinic acetylcholine receptor agonist, and physostigmine, an acetylcholinesterase inhibitor, reverse the effects of MK-801. Crossing time was increased significantly in the training and test sessions for the controls. When 20 ${\mu}M$ MK-801 was administered prior to the training session, the crossing time did not increase in either session. The MK-801-induced learning deficit was rescued by pretreatment with 20 ${\mu}M$ physostigmine, and crossing time was increased in the training and test sessions compared to the MK-801-treated zebrafish. Further, the MK-801-induced learning deficit was prevented by pretreatment with 20 ${\mu}M$ nicotine, and crossing time was increased in the training session but not in the test session. These results show that MK-801 induced a learning deficit in zebrafish that was prevented by pretreatment with nicotine and physostigmine.

Use of G gene-deleted single-cycle viral hemorrhagic septicemia virus (VHSV) for delivery of nervous necrosis virus (NNV)-like particles

  • Vaccines based on single-cycle viruses that are replication-incompetent due to knockout of replication-related structural gene(s) are more immunogenic than inactivated or subunit vaccines and can be used as delivery vehicles for foreign antigens without concerns on the reverting to virulent forms. The aim of this study was to develop a delivery vehicle for nervous necrosis virus (NNV)-like particles (VLPs) using G gene deleted single-cycle VHSV (rVHSV-𝚫G). Recombinant single-cycle VHSVs carrying NNV capsid protein gene between N and P gene of rVHSV-𝚫G genome (rVHSV-𝚫G-NNVCap) were rescued by reverse genetic technology. The successful expression of NNV capsid protein in cells infected with rVHSV-𝚫G-NNVCap was demonstrated by Western blot analysis, and the production of NNV VLPs in infected cells was confirmed using an electron microscopy. The results suggest that single-cycle VHSVs can be used as a safe delivery vehicle for NNV VLPs, and can be extended to other pathogens for the development of prophylactic vaccines.
