Enhancement of Cell Migration by Corticotropin-Releasing Hormone (CRH) in Human Gastric Cancer Cell Line, MKN-28 (Corticotropin-Releasing Hormone (CRH)에 의한 인간 위암 세포(MKN-28)의 Migration 증가)

  • Background: Corticotropin-Releasing Hormone (CRH), an important regulator of stress response, has a potent immunoregulatory effect with the ability to promote the growth of various cancer through CRH receptor type 1 under stress. Although the metastasized cancers through cell migration are more aggressive than the primary cancers, little is known about the effect of CRH on cell migration. Gastric cancer is prone to metastasize to other tissues and it is reported that gastric cancer is response to various stresses such as oxidative stress. Herein, we studied the relationship between CRH and gastric cancer cell migration. Methods: We used gastric cancer cell line, MKN-28 and tested the CRH receptor type 1 expression on MKN-28 by RT-PCR. To examine the change in the ability of migration by CRH in MKN-28, cells were incubated with CRH and then migration ability was measured using a cell migration assay. Results: We confirmed that CRH receptor type 1 was expressed in MKN-28 and HaCaT cells. The migration ability of MKN-28 cells was increased by CRH in a time-, dose- dependent manner. Conclusion: These data suggest that CRH increases migration ability in gastric cancer cell line and that CRH may be a critical regulator in the metastasis of gastric cancer cell.

  • This research carried out to investigate the effects of application of hairy vetch instead of duck-releasing in a large scale of duck-releasing cultivation. The results are as followed; Cultivation of hairy vetch, a green manure crop, for the winter season seemed to be a possible natural fertilizer to supply crop mineral nutrients. Application of hairy vetch residues improved physical properties of soil and was possible to assure better productivity of agricultural products. Introduction of hairy vetch into a large scale of duck-releasing cultivation reduced the pollution of agricultural water.

Initial Adaptation of Released Black Sea Bream, Acanthopagrus schlegeli in Gamak Bay, Southern Coast in Korea (가막만 방류 감성돔 (Acanthopagrus schlegeli) 치어의 초기 적응)

  • The reared fries of black sea bream, Acanthopagrus schlegeli were released in Gamak Bay and initial adaptation to the environment was evaluated. Thirty thousand fries were marked with magnetic tag, released and recaptured and its stomach content were analysed; the neighboring zooplankton, beuthic algae and other young fishes were investigated. The fries were released on July 30, 2001 in summer season. Water temperature of surface and bottom were $24^{\circ}C\;and\;21^{\circ}C,$ respectively. $Chlorophyll\;\alpha$ content was 4.5 ${\mu}g/L$ with high primary production. Salinity was $32.0-32.5\%_{\circ}.$ Water depth of releasing site was about 30 m. The sea jungle was formed at the first point of releasing and it kept the fries for about 3 days in the bay with plenty of food organisms within 5 m depth. Stomach analysis of the fries revealed that real feeding started from the 15 days after releasing and full feeding took place after the tenth day.

Effects of Gardeniae Fructus on Corticotropin-Releasing Factor, c-fos and Tyrosine Hydroxylase in Forced Swimming Test (치자(梔子)가 강제수영부하시험에서 Corticotropin-Releasing Factor, c-fos와 Tyrosine Hydroxylase에 미치는 영향)

  • Objectives : The goal of this study was to investigative the effect of Gardeniae Fructus (GF)as antidepressant in the forced swimming test(FST) model rats. Methods : The expressions of corticotropin-releasing factor(CRF), tyrosine hydroxylase (TH) and c-fos were measured by immunohistochemical method at paraventricular nucleus(PVN), locus coeruleus (LC) and ventral tegmental area(VTA). Results : The duration of immobility in FST was significantly decreased in the GF 100mg/kg groups (p<0.05). CRF expression was significantly decreased at PVN in the GF 100 mg/kg and 400mg/kg treated group in comparison with the control group, respectively (p<0.01). c-fos expression was decreased at PVN in the GF 100 mg/kg treated group with no significance. TH expression was significantly decreased in the GF 100 mg/kg and 400mg/kg treated group in comparison with the control group, at LC and VTA respectively (p<0.001). Conclusion : According to the results, it can be considered that Gardeniae Fructus has antidepressant effect by showing the reduction of immobility in FST through the reduction of CRF, TH expression.

  • Objectives : The goal of this study was to investigate the effect of Moutan Cortex as antidepressant in forced swimming test(FST) model. Methods : The expressions of corticotropin-releasing Factor(CRF), c-Fos and tyrosine hydroxylase(TH) were measured with immunohistochemical method at paraventricular nucleus (PVN), locus coeruleus(LC) and ventral tegmental area(VTA). Results : The duration of immobility in the forced swimming test was significantly decreased in the Moutan Cortex 100 mg/kg treated group in comparison with the control group(p<0.01). CRF and c-Fos expressions at PVN were decreased in the Moutan Cortex 100mg/kg treated group in comparison with the control group. But only the expression of c-Fos was shown the significance(p<0.05). TH expressions at LC and VTA were significantly decreased in the Moutan Cortex 100mg/kg and 400mg/kg treated group in comparison with the control group(p<0.001). Conclusion : According to the results, Moutan Cortex has the antidepressant effect by showing the reduced immobility through the reduction of c-Fos expression at PVN and the reduction of TH expression at LC and VTA.

Preparation and Drug-releasing Properties of Chitosan-based Thermosensitive Composite Hydrogel

  • The novel chitosan-based thermosensitive hydrogels were prepared as control-releasing drug carriers. N-carboxyethyl chitosan (ACS) was synthesized by microwave heating for 1 h through Michael addition of CS to acrylic acid in a grafting yield of 52.97%, which was proved to be a faster and more efficient way than ordinary methods. 5-Fu was modified with formaldehyde to synthesize N,N'-Bis(hydroxymethyl)-5-fluorouracil (5-Fu-OH). Then an esterification was performed using ACS and 5-Fu-OH to give 5-Fu-ACS. The new thermosensitive hydrogels were prepared by adding sodium glycerophosphate to the solution of compounds under a certain constant temperature. Simultaneously, the hydrogels' swelling rate, in vitro drug release rate and thermosensitive were studied, and found that the 5-Fu-ACS composite hydrogel had more excellent releasing effect, higher drug loading and better thermosensitive.

Effects of teat dipping disinfectant containing biodegradable iodophor microspheres on preventing dry period intramammary infection in dairy cows (생분해성 iodophor microsphere 함유 유두침지 소독제의 건유기 젖소에서의 유방내 신규감염 예방 효과)

  • 1% iodophor loaded microspheres of PLGA (Poly[DL-Lactide-co-Glycolide]) were prepared by solvent evaporation method and were applied to the cows on dry period for evaluating it's preventive effects on intramammary infections. The morphology of the microspheres were evaluated using scanning electron microscopy and their releasing patterns were investigated. On investigating idophor releasing patterns of the microsphere, burst releasing pattern was detected until 2 days after in vitro incubation and sustained releasing was observed until 4 weeks. In field trial of teat dipping solution containing idophor loaded microspheres in dry cows showed significant preventive effects of intramammary infection caused by S. aureus, S. agalactiae, coagulase negative Staphylococci and coliform bacteria (p<0.05).

  • Growth hormone-releasing peptide-2 (GHRP-2, also named KP102) is a new hexapeptide of a series of synthetic growth hormone-releasing peptides (GHRPs) which stimulates the secretion of growth hormone (GH) in vitro and in vivo in several species including calf, sheep and pig. The GH-releasing activity of GHRP-2 is two to three times more effective than that of the original GHRP-6, and GHRP-1 in the rats and humans. To date, GHRP-2 seems to be the most potent member of the family of GHRPs. Since the GHRPs are short peptides (5-7 amino acid residues), they are synthesized easily and are not as readily degraded in plasma as GHreleasing hormone (GHRH). These features ameliorate their potential on domestic animals because of their chemical nature the GHRPs are efficacious when administered i.v. orally or orally. However, studies in cow, pig and sheep do not indicate such a close relationship between GHRH, somatostatin (SS) and GH, calling into question the general applicability of the human and rat models. Perhaps there is an important role for an endogenous GHRP in the regulation of GH secretion in domestic animals. This review provides an overview on the current knowledge of physiological role of GHRP-2 in domestic animals.

The Clinical Report on 3 Cases of Lateral Epicondylitis Treated with Musculotendinous Releasing Manual Therapy after Acupuncture Treatment (침치료 후 근건이완수기요법을 적용한 주관절 외상과염 환자 치험 3례)

  • We researched 3 patients diagnosed as Lateral epicondylitis (Tennis elbow). We considered it as tendinosis including irregular collagen fiber's pattern. So, acupuncture therapy and musculotendinous releasing manual therapy were used to treat the patients. We measured their pain and functional disorder by visual analogue scale (VAS), Patient-Rated Tennis Elbow Evaluation (PRTEE), and a dynamometer before and after treatment. As a result, VAS and PRTEE significantly reduced and grasping power was also improved. Therefore, we conclude that the acupuncture therapy with musculotendinous releasing manual therapy is an effective way to treat Lateral epicondylitis. But there are limits on this study due to insufficient number of cases, absence of control group and absence of follow-up after last treatment. Further studies will be needed.
