각종 전처리 및 건조 방법이 건조 채소류의 품질에 미치는 영향 (Effect of Various Pretreatments and Drying Methods on the Quality of Dried Vegetables)

  • 호박 고지, 고구마 줄기, 토란대, 및 도라지를 citric acid 용액 침지, 아황산염 용액 침지, 또는 황 훈중 등의전처리를 하여 천일 건조하거나 서로 다른 온도(50, 70, 90, $150^{\circ}C$)에서 열풍 건조한 후에, 실온 저장 중에 곰팡이 발생, 건조 상태 및 수화 복원 상태에서의 관능적 품질을 조사하였다. 호박 고지는 수분 함량이 15% 이하, 고구마 줄기는 20% 이하, 토란대는 25% 이하, 도라지는 15% 이하로 건조하여 PE film에 포장하여 실온에 저장했을때 3 개월여 동안 곰팡이가 발생하지 않았다. 전처리. 방법으로는 채소를 건조 전에 아황산염에 침지하거나 황훈중 처리했을 때 곰팡이 발생이 저지되었다. 엽록소를 함유한 채소는 전반적으로 열풍 건조한 경우가 천일건조한 경우에 비하여 현저히 관능적 품질이 우수했으며, 아황산염 용액에 침지하여 oven에 건조할 경우에 다른 처리 제품보다 관능적 품질이 대체적으로 좋았다. 도라지의 경우는 황 훈증이나 아황산 용액 침지가 건조 후의 제품의 품질에 다소 좋은 영향을 보였으나 건조방법은 관능적 품질에 큰 영향을 미치지 않았다. 건조온도는 도라지를 제외하고는 대체척으로 건조 실험에 사용한 온도 중에서 가장 낮은 $50^{\circ}C$에서 가장 좋은 품질의 건조 채소 제품을 얻을 수 있었으며, 모든 품목에 대하여 $105^{\circ}C$에서 건한 제품은 관능적 품질이 매우 떨어졌다. 채소틀 낮은 온도에서 건조할 때 채소의 복원율이 높았으며 복원 후외 관능적 품질도 채소의 건조 온도가 낮을수륵 좋았다.

생쥐배아의 동결보존에 관한 실험적 연구 (The Experimental Study on Cryopreservation of Mouse Embryo)

  • Objectives: This study was carried out to evaluate the effects of embryonic stage, cryoprotectant, and freezing-thawing method on the rates of survival and development of the cryopreserved mouse early embryo and finally to establish the cryopreservation method of surplus embryos obtained during assisted reproductive technology (ART). Materials and Methods: Two to eight cell embryos were obtained from oviducts of mated $F_1$ hybrid female mice superovulated by pregnant mare's serum gonadotropin (PMSG) and human chorionic gonadotropin (hCG). Two-step 1,2-propanediol (PROH), dimethylsulfoxide (DMSO) and 4-step PROH DMSO were used as cryoprotectant and dehydration and rehydration method of embryos, and slow-cooling or rapid-cooling method was used as frozen program. The survival rates of embryos were measured after thawing and rehydration, and the developmental rates of embryos were compared and observed during culturing embryos for 24, 48, 72, 96 hrs. Results: As for the survival and development rates of embryos according to embryonic stage, the survival rate of 2 cell stage in PROH and DMSO was significantly higher than 4-8 cell (64.5% versus 62.1 %,79.7% versus 73.2%) (p<0.01, p<0.01), but the development rates of 4-8 cell embryos in PROH and DMSO were significantly higher than 2 cell embryos for whole culture period (p<0.01) and the development rates of 4-8 cell embryos in PROH were significantly higher than 2 cell embryos in DMSO (p<0.01). As for the survival and development rates of embryos according to cryoprotectant, the survival rate of 2 cell embryo in DMSO was significantly higher than that in PROH (74.4% versus 64.5%) (p<0.01), whereas the development rate of embryos was not differ till 24 hrs. The developmen1 rate from morular to hatching blastocyst, however, was significantly higher in PROH than in DMSO during 48 hr (p<0.01). The survival rate of 4-8 cell embryo was 62.1% in PROH and 73.2% in DMSO. The development rates of embryo in PROH were significantly higher for whole culture periods (p<0.01, 0.05). In respect to the effect of freezing and thawing program on the survival and development rates of embryos, method of slow cooling and rapid thawing was more effective than that of rapid cooling and rapid thawing. Conclusions: The survival rate of embryo in 2 cell stage was higher than in 4-8 cell stage, and PROH appears more effective cryoprotectant than DMSO because PROH showed better development rates of embryos in 2 and 4-8 cell stage. Moreover, slow cooling and rapid thawing method was considered as the best cryopreservation program.

Effects of Alcohol Intake on Body Fluid Balance and Fat Mobilization After Exercise Induced Dehydration

  • To examine the effects of alcohol consumption on body fluid restoration and fat mobilization following exercise induced dehydration, nine healthy collegiate men ($24{\pm}2yrs,\;177{\pm}5cm,\;72{\pm}8kg,\;10.5{\pm}2.3%$ body fat) underwent three experiments. In each experiment, subjects ran on a treadmill to reduce individual body mass to $2.2{\pm}0.1%$ and consumed one of three beverages containing 0, 4, or 8% alcohol over 60 min followed by 4 hr of resting recovery. They consumed approximately 150% of weight loss $(2053{\pm}204,\;2091{\pm}149,\;and\;1943{\pm}295mL)$ and content of alcohol was $9.9{\pm}1.0(0%),\;71.9{\pm}5.1(4%)$, and $132.2{\pm}20.1g$ (8% trial). Body weight, urine volume and samples, blood samples, and thirst sensation were measured five times; at baseline, immediately after exercise, and 0, 1st, and 4th hr of recovery. Blood alcohol concentration after ingestion was $0.0{\pm}0.0(0%),\;0.1{\pm}0.02(4%)$, and $0.2{\pm}0.03%$ (8% trial). No differences in blood sodium and potassium concentrations, and urine specific gravity were noticed over time periods and trials. Thirst sensation tended to be elevated in all trials immediately after exercises and urine output was elevated during the recovery. The magnitude of changes in these variables was proportional to the alcohol concentrations, but not statistically significant. While serum osmolality was not different among trials and time periods in 0 and 4% trials, it was higher during recovery than the baseline in the 8% trial (P<0.01). Triglycerides did not change throughout the time period and among trials. Free fatty acids were elevated after exercise in all trials and 4th hr of recovery in 0% (P<0.05). Subjects' net body fluid balance at 4th hr of recovery was negatively maintained and proportional to alcohol concentrations. Only 8% trials showed a significant reduction at 1st and 4th hr of recovery compared to 0 hr. The results suggested that diuretic effect of alcohol after moderate level of dehydration appeared dose dependent, but beverage containing alcohol up to 4% did not induce impaired rehydration than alcohol free drinks. Alcohol effects on fat mobilization during recovery appeared to be minimal and the mechanism is unclear.

건조 방법에 따른 예비열처리 당근의 저장 중 품질 특성 (The Quality Properties of Dried Carrots as Affected by Blanching and Drying Methods during Storage)

  • 대표적 근채류인 당근을 이용하여 데치기 및 열풍, 감압 및 동결 건조방법에 따른 저장 중 품질변화를 관찰하였다. 열풍 및 감압에서의 건조율 비교 시 데치기한 당근(BC)는 데치기를 하지 않은 당근(RC)보다 빠르게 건조되었으며. 재수화에 의한 복원률은 모든 건조방법에서 RC가 BC보다 높았다. Carotene은 열풍과 감압건조 후 상온 저장 시 BC가 RC보다 잘 유지되었다. 갈변은 모든 건조방법에서 BC가 RC보다 낮게 나타나 데치기에 의해 억제됨을 알 수 있었으며, 동결건조 당근이 가장 덜 갈변되었다. 당근의 경도는 건조 후에 크게 감소하였으나, 열풍건조와 감압건조 당근은 유사한 값을 보였으며 동결건조 당근에 비해 높은 값을 나타내었다. BC와 RC의 경도 비교의 경우에는 모든 건조방법에서 BC가 RC보다 현저히 높게 나타났으며 저장 중 거의 일정하게 유지되었다. 또한 모든 건조당근에서 BC가 세포구조 및 미세구조가 더욱 안정하며 일정하여 데치기의 효과를 확인할 수 있었다.

Lysozyme의 Liposome 미세캡슬화와 유출 촉진 (Liposome-Microencapsulation of Lysozyme and Its Stimulated Release)

  • Liposome내에 효소를 포집시켜서 식품가공공정에 응용하기 위하여 모델 효소로서 Iysozyme을 선택하여 dehydration-rehydration법으로 미세캡술화시킬 때 조제 조건이 포집효율에 미치는 영향을 검토하고 외부 환경 인자의 변화 및 유출 촉진제의 첨가가 liposome 내의 효소의 유출에 미치는 영향을 연구하였다. MLV 현탁액을 bath sonicator에서 100 kHz로 2시간 초음파 처리했을 때 1ozyme의 포집효율이 90.9%로 가장 높았다. 초기 효소 첨가량을 증가시킴으로써 단위 지방질당 포집 효소량은 거의 비례적으로 증가하였으나 포집효율은 감소하여 1osozyme 50 mg/100 mg soy lecithin일 때 68% 수준까지 급격히 감소하였으나, 그 이상의 실험농도 범위에서는 매우 완만하였다. 효소를 포집한 liposome은 $37^{\circ}C$, pH 5.9에 저장하였을때에 가장 안정하였고 산성일수록 유출속도는 현저히 증가하였으며 일정한 속도로 지속적으로 유출되었다. Tween 80 1.0%와 흔합했을 때 초기 24시간 동안에는 급격히 유출되었으나 시간이 경과할수록 완만히 유출되어 72시간 경과 후 85%까지 유출되었다. 또한 $Ca^{2+}$ ion을 25-5OmM첨가했을 때 초기 1시간동안 급격히 유출되었으나 그 이후에는 완만히 유출되었다.

실리콘 하이드로겔 렌즈의 반복적 건조로 인한 렌즈형태 및 자·타각적 증상의 변화 (The Change in the Parameters of Silicone Hydrogel Lens and Objective/Subjective Symptoms induced by Repetitive Dryness of Lens)

  • 목적: 본 연구에서는 반복적인 건조에 의해 유발되는 실리콘 하이드로겔 렌즈의 형태 변화 및 착용시의 자 타각적 증상의 변화에 대해 알아보고자 하였다. 방법: 재질 혹은 두께가 다른 실리콘 하이드로겔 렌즈를 4회씩 건조하고 수화시킨 후 전체 직경과 베이스커브의 변화를 비교하였으며, 건조되었던 실리콘 하이드로겔 렌즈를 착용한 후 착용자의 자각적 증상 및 비침입성 눈물막 파괴시간과 순목횟수를 측정하였다. 결과: 반복되는 건조와 수화로 인하여 모든 실리콘 하이드로겔 렌즈의 전체 직경과 베이스커브가 증가하였다. 전체 직경과 베이스커브가 변하는 정도는 재질에 따라 각기 상이하였으며 두꺼운 렌즈에서는 건조수화 횟수에 따라 불규칙적인 변화가 나타났다. 실리콘 하이드로겔 렌즈의 재질이나 두께에 관계없이 건조수화 과정을 거친 렌즈를 착용하였을 때에는 착용자의 비침입성 눈물막 파괴시간은 감소하였으며, 순목횟수는 증가하였다. 또한, 반복적인 건조수화는 렌즈의 착용감에도 영향을 미쳐 가려움, 통증, 자극감, 이물감, 건조감이 증가하는 경향이 있었다. 결론: 본 연구 결과를 통하여 반복적인 건조수화로 실리콘 하이드로겔 렌즈의 형태 변화 및 이에 따른 착용감 저하 유발을 확인하였으므로 건조한 환경에 대하여 강한 내구성을 가지는 실리콘 하이드로겔 콘택트렌즈의 개발 필요성을 제안할 수 있다.

Effect of Dehydration and Rehydration of the pH-Sensitive Liposomes Containing Chimeric gag-V3 Virus Like Particle on Their Long-term Stability

  • One of the practical limitations with the use of liposomes for delivery of the pharmaceutical substances such antigens is that liposomes are relatively unstable in storage. In order to extend the stability of liposome in storage without affecting their functional activity, solution-type liposomes were dehydrated to form a structurally intact dry liposomes. Comparative immunological evaluation was carried out for both dry and solution-type liposomes containing gag-V3 chimera, consequently it was found that dry liposomes elicited both humoral and cellular response as efficiently as solution-type liposemes did against the same gag-V3 antigen. Especially, long-term stability of the liposomes was remarkably enhanced by the dehydration made to loposomes without a significant change in its ability to elicit immune response in vivo. These results indicate that dry pH-sensitive liposome may become an effective delivery and adjuvant system for general vaccine development.

제올라이트광물을 이용한 폐수중의 중금속제거 (Removal of Heavy Metal Ions in Wastewater Using Zeolite Minerals)

  • All the applications of natural zeolites make use of one or more of their physical and chemical properties: adsorption, ion-exchange and related molecular sieve properties, dehydration and rehydration, and siliceous composition. Accordingly, the applications of zeolite have been carried out in the various aspects because of its large cation exchange capacity and adsorption properties. In this paper, the adsorption effect of heavy metal ions in wastewater on zeolite mineral by batch adsorption process is studied. The amounts of adsorbed ions were variable by original pH and ionic concentration, especially original pH of solution had an important effect on the adsorption. In case of low pH solution, e.g. below 3.0, clinoptilolite adsorbed $Pb^{2+}$ ,$ Cd ^{2+ }$ , $Cu^{2+}$ and $Zn^{ 2+}$ , but mordenite almost did not adsorb except $Pb^{2+}$ . Under the same conditions, these ions were more adsorbed on clinoptilolite than on mordenite mineral. The velocity of adsorption was relatively fast and it was confirmed by shaking test that the equilibrium of adsorption could be attained in about one hour. The species of exchangeable cation of zeolite had an effect on its removing ability and zeolite of the sodium-exchanged type was the best.

Vital Wheat Gluten 의 제조 (Vital Wheat Gluten by Hot Air Drying)

  • Wet gluten 에 salt 및 소량의 산(酸)을 가하여 상압하(常壓下)에서 $6^{\circ}C$로 열풍건조하여 vital gluten 을 제조하였다. 이때 salt 및 산(酸)은 gluten의 점착성을 저하시켜 표면적을 크게하고 열내성(熱耐性)이 생기게 하여 건조를 쉽게 하였다. 1) Salt 농도는 높을수록 glulen의 건조가 용이하나, quality 는 저하되었다 salt 는 $5{\sim}10%$가 좋을 것같다. 2) Salt 외에 HCI을 0.12%첨가한 결과 건조 시간은 거의 반으로 단축되었고, glulen의 질(質)은 저하되지 않았다. 3) 산(酸)은 hydrochloric, acetic, phosphoric, lactic acid 중(中)에서 hydrochloric acid 건조 시간을 가장 적게 하였으며 질(質)은 별 차이 없으므로 hydrochloric acid 첨가가 가장 좋았다.

The Effects of a Fluid Intake Intervention for Elders in Long-term Care Hospitals

  • Purpose: This study was done to evaluate the effects of a fluid intake intervention on increasing fluid intake and ameliorating dehydration status in elders admitted to long-term care hospitals. Methods: A nonequivalent control group, pretest and posttest design was used. The experimental group of 39 participants received the 4-week intervention while the control group of 38 participants received routine care. Outcome variables were daily fluid intake and physiological indexes such as blood urea nitrogen and creatinine ratio (BUN/Cr), urine specific gravity (USG), and urine color. Results: After the intervention to increase fluid intake, there were statistically significant increases in daily fluid intake, normal BUN/Cr, and USG in the experimental group. However, a statistically significant improvement in normal urine color was not found for either group. Conclusion: The findings of this study demonstrated that the fluid intake intervention improved hydration status of the experimental group participants. Consequently, it was confirmed that the intervention is considered to be effective in preventing dehydration which occurs frequently in older adults in long-term care facilities and, thus this intervention may contribute to preventing various health issues resulting from dehydration.
