A study on the Effects of Rehmannia Radix and Eukmigihwangtang (EMGHT) on Antioxidation Activity in Aging Rats (숙지황(熟地黃)과 육미지황탕(六味地黃湯)이 노화과정(老化過程) 흰쥐에서의 항산화(抗酸化) 기전(機轉)에 미치는 영향(影響))

  • This experimental study was designed to verify the anti-aging efficacy of Eukmigihwangtang (EMGHT) and Rehmannia Radix, and determine the specific role and actions of Rehmannia Radix. Normal rat (2 months old), aging rat (8 months old), and pathologically induced rat (2 months old, injected 30mg/kg of streptozotocin) are observed to study the aging eliciting factors such as peroxide contents and enzyme activities. The following results were obtained in this study: 1. For the body weight changes, normal group given Rehmannia Radix showed decrease in the body weight compared to the control group, aging group given EMGHT and Rehmannia Radix showed significant decrease in the body weight, and STZ injected group showed suppression to the body weight loss when given EMGHT and Rehmannia Radix. 2. For the content changes in serum lipid peroxide, normal group showed increasing level as the rat gets older. Aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant decrease in the lipid peroxide level compared to the control group. Decrease was more prominant in the group given EMGHT. 3. For the changes in serum hydroxyl radical, normal group did not show significant changes, but aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant decrease in the hydroxyl radical level compared to the control group. Decrease was more prominant in the group given EMGHT. 4. For the changes in serum superoxide dismutase (SOD) activity, normal group did not show significant changes, but aging group given EMGHT and Rehmannia Radix showed significant increase in the SOD activity compared to the control group. STZ injected group given EMGHT and Rehmannia Radix showed significant decrease in the SOD activity compared to the control group. 5. For the content changes in hepatic lipid peroxide, aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant decrease in the lipid peroxide level compared to the control group. 6. For the changes in hepatic cytochrome P-450 activity, aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant decrease compared to the control group. Cytochrome b5 activity was significantly decreased only in the STZ injected group given EMGHT and Rehmannia Radix. 7. For the changes in hepatic aminopyrine demethylase and aniline hydroxylase activity, aging group given EMGHT and Rehmannia Radix showed significant decrease compared to the control group. STZ injected group given EMGHT and Rehmannia Radix showed significant increase in the aminopyrine demethylase activity, and showed significant decrease in the aniline hydroxylase activity compared to the control group. 8. For the content changes in hepatic protein bound-SH and nonprotein bound-SH, againg group and STZ injected group given EMGHT and Rehmannia Radix showed significant increase compared to the control group. 9. For the content changes in hepatic glutathione level, aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant increase compared to the control group. 10. For the changes in hepatic glutathione S-transferase activity, aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant increase and decrease, respectively, compared to the control group. 11. For the changes in hepatic glutathione reductase activity, aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant increase compared to the control group, while $\gamma$-Glutamylcystein synthetase activity did not show significant changes. 12. For the changes in hepatic superoxide dismutase activity, aging group and STZ injected group given EMGHT and Rehmannia Radix showed significant decrease compared to the control group. From the above results, the antioxidant effects of EMGHT and Rehmannia Radix were proved, as well as the role of Rehmannia Radix, a chief of EMGHT, was examined. In addition, since no change was reconized as the quantity of Rehmannia Radix and the order herbs increased, the reasonableness on EMGHT was proven with respect to its composition and quantity. Thus, the significance of EMGHT could be objectively exmined in terms of its composition and quantity. Considering animals used in the experiment, there were obvious changes in aging rats and pathologically induced rats than in normal rats. Consequently, it was noticeable that EMGHT and Rehmannia Radix were working selectively on the subjects.

Dehydration Kinetics of Rehmannia (Rehmannia glutinosa Liboschitz)

  • Sliced and whole root of rehmannia were dehydrated in a laboratory dryer at 40, 60, 80, and $100^{\circ}C$ to evaluate the kinetic parameters for dehydration of rehmannia. The drying curves of both samples were characterized by a falling-rate drying period only. Sliced rehmannia dried 1.1 to 3.1 times faster than whole root of rehmannia depending on drying temperature. Equilibrium moisture content (EMC) of rehmannia samples at the drying temperature tested were 0.069-0.078 g water/g dry solid, which was coincided with the monolayer moisture content (0.06 and 0.07 g water/g dry solid) evaluated from desorption isotherms using GAB (Guggenheim-Anderson-de Boer) model. A logarithmic model for thin layer drying was applied to evaluate the drying time to reach EMC ($t_{EMC}$) and drying constant (k). The effect of temperature on $1/t_{EMC}$ and k was described by the Arrhenius model with activation energy values of 32.56 and 47.14 kJ/mol determined using the former parameter, and 34.27 and 38.26 kJ/mol determined using the latter parameter for sliced and whole root of rehmannia, respectively.

Protective Effects of Rehmannia Glutinosa Extract and Rehmannia Glutinosa Vinegar against b-amyloid-induced Neuronal Cell Death (베타아밀로이드로 유도된 신경세포사멸에 대한 지황(地黃) 및 지황식초(地黃食醋)의 보호효과)

  • Alzheimer's disease, a representative neurodegenerative disorder, is characterized by the presence of senile plaques and neurofibrillary tangles accompanied by neuronal damages. b-Amyloid peptide is considered to be responsible for the formation of senile plagues that accumulate in the brains of patients with Alzheimer's disease. There has been compelling evidence supporting that b-amyloid-induced cytotoxicity is mediated through generation of reactive oxygen species. In this study, we have investigated the possible protective effect of Rehmannia glutihosaagainst b-amyloid-induced oxidative ceil death in cultured human neuroblastoma SH-SY5Y cells. SH-SY5Y cells treated with b-amyloid underwent apoptotic death as determined by morphological features and positive in situterminal end-labeling (TUNEL staining). Rehmannia glutinosawater extract, wine, and vinegar pretreatments attenuated b-amyloid-induced cytotoxicity and apoptosis. Rehmannia glutinosa vinegar exhibited maximum protective effect by increasing the expression of anti-apoptotic protein, Bcl-2. in addition to oxidative stress, b-amyloid-treatment caused nitrosative stress via marked increase in the levels of nitric oxide, which was effectively blocked by Rehmannia glutinosa. To further explore the possible molecular mechanisms underlying the protective effect of Rehmannia glutinosa, we assessed the mRNA expression of cellular antioxidant enzymes. Treatment of Rehmannia glutinosa vinegar led to up-regulation of heme oxygemase-1 and catalase. These results suggest that Rehmannia glutinosa could modulate oxidative neuronal cell death caused by b-amyloid and may have preventive or therapeutic potential in the management of Alzheimer's disease. Particularly, Rehmannia glutinosa vinegar can augment cellular antioxidant capacity, there by exhibiting higher neuroprotective potential.

Effect of Drying Method on Antioxidant Activity of Jiwhang (Rehmannia glutinosa)

  • Jiwhang (Rehmannia glutinosa), one of the most widely used medicinal herbs, was dried with various methods such as sun drying, hot air drying, vacuum drying, and freeze drying methods, and their effects on the antioxidant capacity in relation with the content of total phenolic compounds were studied with a steamed-and-dried rehmannia (sookjiwhang) for comparison. Generally, total phenolic contents decreased significantly by all of the drying treatments except the steamed-and-dried rehmannia, in which total phenolic contents increased 2.4 fold compared with fresh rehmannia. Content of verbascoside, a functional phenolic compound, was the highest in the freeze-dried rehmannia ($177.97{\pm}0.02\;{\mu}g/g$ d.m.) followed by vacuum-dried ($105.55{\pm}0.07\;{\mu}g/g$ d.m.), hot air-dried ($23.01{\pm}0.02\;{\mu}g/g$ d.m.), and sun-dried ($4.89{\pm}0.13\;{\mu}g/g$ d.m.) ones comparable to the fresh rehmannia ($80.15{\pm}1.26\;{\mu}g/g$ d.m.). Antioxidant capacity determined by both 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azinobis (3-ethyl-benzthiazoline-6-sulphonic acid) (ABTS) methods agreed with the result of total phenolic contents, that is, the antioxidant capacity was the highest in the steamed-and-dried rehmannia followed by fresh rehmannia, vacuum-dried, hot air-dried, sun-dried, and freeze-dried ones. Conclusively, the total phenolic contents and antioxidant capacity of rehmannia were greatly affected by the drying methods used.

Selection of disease resistant lines of Rehmannia glutinosa with mutant treatment

  • Rehmannia glutinosa is one of the most important medicinal crops in Korea. However, various plant pathogens including Fusarium sp. cause great damages and cause enormous economic losses. Therefore, this study was conducted to select Fusarium resistant plants by using mutagen-treated Rehmannia glutinosa. The plant material used was a native accession of Rehmannia glutinosa. As a result, among the Rehmannia glutinosa treated with various concentrations of EMS, R. glutinosa treated with 0. 03M EMS showed higher resistance against infection of F. oxysporum in pot tests, and Rehmannia glutinosa plants treated with 0.12M EMS showed higher resistance against infection of F. oxysporum in field tests.

The Incidence of Virus Diseases in Rehmannia glutinosa in Korea (국내 지황에 발생하는 바이러스병 발생 현황)

  • While rehmannia (Rehmannia glutinosa Libosch) was identified as a host of at least five viruses, including Rehmannia mosaic virus (ReMV), Youcai mosaic virus (YoMV), Broad bean wilt virus 2 (BBWV2), Plantago asiatica mosaic virus (PlAMV), and Rehmannia virus 1 (ReV1), viral incidence surveys have not been performed yet in rehmannia fields in Korea. In this study, we performed field surveys during 2017-2018 to investigate the incidence of 5 major viruses in rehmannia. A total of 145 symptomatic samples were collected from the rehmannia fields in major cultivation areas of Korea. Molecular diagnosis assays showed that all the collected leaf samples were infected with more than two viruses. Particularly, two species of Tobamovirus, ReMV and YoMV, were detected in all the samples. In addition, our analysis showed that the root stocks of 4 rehmannia cultivars were infected with at least two viruses. Since rehmannia is propagated by vegetative propagation, it is highly important to produce virus-free root stocks of rehmannia to control virus diseases in rehmannia.

Antioxidant Activities of Rehmannia glutinosa by Traditional Methods (생지황을 이용하여 전통방법으로 제조한 숙지황의 항산화 활성)

  • The objective of this study was to evaluate antioxidant activities of Rehmannia glutinosa (Raw Jihwang) by traditional method. The total phenol content of Rehmannia radix Preparata (the final cycle of Jihwang) was increased to 205%, compared with Rehmannia glutinosa. Antioxidant activities, determined by ferric-reducing antioxidant potential (FRAP), 2,2'-azinobis(3 ethybenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity, 1,1-diphenyl-2-picrydrazyl (DPPH) and hydroxyl radical scavenging activities, increased remarkably as the number of steaming-drying cycles increased. Especially, FRAP value increased 285%. Also, $IC_{50}$ values for DPPH and hydroxyl radical scavenging activities of the final 9th-cycling product, decreased 48.4% and 76%, respectively, compared with those of Rehmannia glutinosa. Our result was suggested that antioxidant activities of Rehmannia radix Preparata improve according to the increasing number of steaming-drying cycles.

The Effects of Rehmannia on Nephrectomy-induced Chronic Renal Failure Rats (생지황(生地黃)이 신절제술로 유도된 만성신부전 Rat에 미치는 영향(影響))

  • The aim of the study was to investigate recovery effects of Rehmannia, which has been used clinically for chronic renal failure therapy. Mice had 5/6 nephrectomy to induce chronic renal failure. The results were as follows: 1. The protein amount in urine per 24hrs of the Rehmannia-treated group was significantly reduced compared to the control. 2. The albumin amount in the blood of the Rehmannia-treated group significantly increased compared to the control. The creatinine. total-cholesterol, LDL-cholesterol and triglyceride levels in serum of the Rehmannia-treated group as compared to the control were significantly inhibited. 3. The structural change in kidney of the Rehrnannia-treated group was significantly inhibited compared to the control. 4. The factor (macrophage/monocyte antigen (ED-1), type IV collagen and angiotensin II type 1 ($AT_1$) receptor) of the Rehmanala-treated group was significantly inhibited compared to the control, which induced the structural change in kidneys. The above results suggest that Rehmannia partially improved kidney function.

The Effects of distilled Rehmannia glutinosa Herbal Acupuncture on the Heart Rate Variability(HRV) (생지황 약침이 정상인의 심박변이도(HRV)에 미치는 영향)

  • Objectives : We investigated the effects of distilled Rehmannia glutinosa Herbal Acupuncture on autonomic nervous system with the Heart Rate Variability(HRV) in adult man. as well as we tried to observe how distilled Rehmannia glutinosa Herbal Acupuncture on the balance of the autonomic nervous system. Methods : We investigated on 58 healthy volunteers consisted of 28 subjects in experiment(distilled Rehmannia glutinosa Herbal Acupuncture) group and 30 subjects in control(Normal Saline) group. Study form was a randomized, placebo-controlled, double-blind clinical trial. 28 subjects in experiment group were injected distilled Rehmannia glutinosa Herbal Acupuncture at $GB_{21}$(Kyonjong) and 30 subjects in control group were injected Normal Saline at $G_{21}$(Kyonjong). except of 2 subjects(in control group) who can't be measured and 6 subjects(2 in experiment group and 4 in control group) who move or make unforceable error during measuring. Finally 26 subject in experiment group and 24 subject in control group are studied. We measured HRV by PolyG-I on 7 times : before and after injection per 5 minutes during 30 minutes. The SPSS 10.0 for windows was used to analyze the data and the paired t-test(in group) and Student t-test(between two groups) were used to verify the result. Results : 1. After distilled Rehmannia glutinosa Herbal Acupuncture injection, Mean HRV is significantly low only for first 5 minute, SDNN is significantly high after 5 minute, Complexity is significantly low only from 20 minute to 25 minute. HRV index is significantly high for first 5minute and from 10 minute to after 10 minute. 2. HRV index of distilled Rehmannia glutinosa Herbal Acupuncture Group significantly increased from 25 minute to 30 minute, and pNN50 of Rehmannia glutinosa Herbal Acupuncture Group significantly decreased from 20 minute to 30 minute compared with those of Normal Saline group. 3. After distilled Rehmannia glutinosa Herbal Acupuncture injection, Ln(TP) and Ln(VLF) are significantly high after injection. Normalized LF is significantly high from 5 minute to 20 minute and from 25 minute and 30 minute. Normalized HF is significantly low from 5minute to 20 minute and from 25 minute and 30 minute. 4. Ln(TP) and Ln(VLF) of distilled Rehmannia glutinosa Herbal Acupuncture Group significantly increased from 20 minute to 25 minute compared with those of Normal Saline group. Conclusions : The results suggest that Rehmannia glutinosa Herbal Acupuncture in healthy adult man tend to activate the autonomic nervous system and parasympathetic nervous system compared to Normal Saline within normal range.

The Effects of Distilled Astragali Radix Herbal Acupuncture, Wild Ginseng Herbal Acupuncture and Rehmannia Glutinosa Herbal Acupuncture on Vital Sign;A Randomized, Placebo-controlled, Double-blind Clinical Trial (수종의 약침제제 견정혈 자입이 활력징후 변화에 미치는 영향;황기 약침, 산삼 약침, 생지황 약침을 중심으로)

  • Objectives: We investigated the safty of distilled Astragali Radix Herbal Acupuncture, Wild Ginseng Herbal Acupuncture and Rehmannia glutinosa Herbal Acupuncture on vital sign -BP, pulse, resperation-in adult man. Methods : We investigated on 106 healthy volunteers consisted of each 30 subjects in experiment (distilled Astragali Radix Herbal Acupuncture, Wild Ginseng Herbal Acupuncture and Rehmannia glutinosa Herbal Acupuncture) group and 30 subjects in control(Normal Saline) group. Study form was a randomized, placebo-controlled, double-blind clinical trial. 30 subjects in each experiment group were injected distilled Astragali Radix Herbal Acupuncture, Wild Ginseng Herbal Acupuncture, Rehmannia glutinosa Herbal Acupuncture at $GB_{21}$(Kyonjong) and 30 subjects in control group were injected Normal Saline at $GB_{21}$ (Kyonjong). except of 2 subjects(in control group) who can't be measured and 10 subjects(6 in experiment group and 4 in control group) who move or make unforceable error during measuring. Finally each 25. subject, subject, subject in experiment group and 24 subject in control group are studied. We measured resperation by PolyG-I and BP & pulse by electric manometer on 5 times : before injection per 5 minutes during and after injection per 10 minutes during 35 minutes. The SPSS 13.0 for windows was used to analyze the data and the Student t test(between two groups) were used to verify the result. Results : 1. After distilled Astragali Radix Herbal Acupuncture injection, Wild Ginseng Herbal Acupuncture and Rehmannia glutinosa Herbal Acupuncture , systolic BP is not significantly change in all experiment time. 2. After distilled Astragali Radix Herbal Acupuncture injection, Wild Ginseng Herbal Acupuncture and Rehmannia glutinosa Herbal Acupuncture , diastolic BP is not significantly change in all experiment time. 3. After distilled Astragali Radix Herbal Acupuncture injection, Wild Ginseng Herbal Acupuncture and Rehmannia glutinosa Herbal Acupuncture , pulse is not significantly change in all experiment time. 4. After distilled Astragali Radix Herbal Acupuncture injection, Wild Ginseng Herbal Acupuncture and Rehmannia glutinosa Herbal Acupuncture , resperation is not significantly change in all experiment time. Conclusions : The results suggest that distilled Astragali Radix Herbal Acupuncture, Wild Ginseng Herbal Acupuncture and Rehmannia glutinosa Herbal Acupuncture in healthy adult man don't influence vital sign within normal range. This result is derived from that distilled Astragali Radix Herbal Acupuncture, Wild Ginseng Herbal Acupuncture and Rehmannia glutinosa Herbal Acupuncture in healthy adult man are safety.
