Comparison of Physicochemical Properties and Malonyl Ginsenoside Contents between White and Red Ginseng (백삼 및 홍삼의 이화학적 특성 및 말로닐 진세노사이드 함량 비교)

  • Korean ginseng (Panax ginseng C. A. Meyer) has been used as a traditional herbal medicine in East Asia and is very popular in the world, because of its health benefits. To comparison of pharmacological components and physiochemical properties between white and red ginseng from same body, we analyzed ginsenoside and malonyl ginsenoside, ash, crude lipid/protein, fatty acid, mineral contents, total/reducing sugar, and total phenolic and acidic polysaccharide contents. The general components did not show any significant difference between white and red ginseng. Whereas, the content of neutral ginsenoside $Rb_1$, $Rb_2$, Rc and Rd were higher in red ginseng than those of white ginseng. However, malonyl ginsenoside such as $m-Rb_1$, $m-Rb_2$, m-Rc and m-Rd in white ginseng were similar to neutral ginsenoside $Rb_1$, $Rb_2$, Rc and Rd in white ginseng and far higher than those of red ginseng. These results exhibit that malonyl ginsenosides were converted to neutral ginsenosides in steaming process for red ginseng. So, we suggest that malonyl ginsenoside are necessary to applies in ginsenoside analysis of Korean ginseng.

Quality Characteristics and Antioxidant Activity of Soy Sauce with Added Levels of Black Garlic Extract (흑마늘 추출액의 첨가 농도에 따른 간장의 품질특성 및 항산화 활성)

  • Purpose: In order to develop soy sauce and increase its functionality, by adding black garlic extract. Methods: We compared quality characteristics and antioxidant activity of 20 days aging black garlic soy sauce from mixed manufacturers with raw soy sauce (commercial fermented soy sauce) and different ratio of black garlic extract (65 Brix, 0.3-5.0%). Results: The salt content of the black garlic extract (0.3-5.0%) added soy sauce were 12.35-12.77%. The pH was lowered to 4.99, and acidity was increased to 2.12%, depending on the increase in black garlic extract added ratio. The crude protein and total nitrogen contents were 6.23-6.62% and 1.10-1.16%, respectively, and content of amino form nitrogen was 0.52-0.53%, without significant differences between experimental groups. Contents of reducing sugar and free sugars (fructose and glucose) tended to be higher with higher mixing ratios of black garlic extract. Total phenolic compounds and flavonoids contents were significantly increased in the 3% and 5% black garlic extract addition group. At concentrations of 62.5, 125, 250, 500, 1,000 and $2,000{\mu}g/mL$, the DPPH and ABTS radical scavenging activity of the polysaccharides isolated from control soy sauce and 5% black garlic extract added soy sauce showed higher correlation with their concentration. Conclusion: These results, confirmed that the soy sauce produced by adding more than 3% of black garlic extract has high antioxidant activity.

Correlations between Soil Environment and Bioactive Constituents of Namdo Garlic Harvested in the Non-volcanic Ash Soil Distributed Western Jeju (제주 서부지역 비화산회토양에서 수확한 남도마늘의 기능성 성분 함량 및 토양환경과의 상관관계 분석)

  • In this study, we determined the ingredient analysis of harvested garlic bulb and soil analysis of four garlic-cultivated regions in Jeju, being one of the major areas of Namdo garlic production. Soil pH and electric conductivity were 7.02 and 1.03 dS/m, respectively. Soil organic matter was 4.31%. The mineral elements (potassium, calcium, magnesium, sodium, iron, manganese, copper and zinc) of Namdo garlic cultivated soil were analyzed by ICP, and calcium was the most highly contained mineral with $14.67cmol_+/kg$ and in the decreasing order of magnesium ($2.25cmol_+/kg$), potassium ($1.51cmol_+/kg$). Soluble solid and total acidity were 7.60 oBrix and 0.49%, respectively. The mineral contents of garlic bulb were in order of potassium (12,728 ppm) > sulfur (7,778 ppm) > phosphorus (4,916 ppm) > magnesium (691 ppm) > calcium (359 ppm). The content of total phenolic, total flavonoid and reducing sugar were 71.14 mg GAE/100 g, 17.64 mg QE/100 g and 26.53 mg GE/g, respectively. Alliin and allicin were 8.78 mg/g and 2.10 mg/g, respectively. The Pearson's correlation coefficients between mineral contents of soil and garlic bulb are analyzed. Macronutrients of soil is correlated with macronutrients of garlic (positive) and micronutrients of garlic (negative) contents.

Comparison of Biochemical Composition and Antimicrobial Activity of Southern-Type Garlic Grown in the Eastern and Western Regions of Jeju (제주 동부와 서부지역 남도마늘의 생화학적 성분 및 항균활성 비교)

  • Components and physiological activity of crops are strongly affected by the natural environments of the growth regions. In this study, we carried out soil analysis and determined the allicin, alliin, total pyruvate, total phenol and flavonoid, and reducing sugar contents of garlic from garlic-cultivating regions in the eastern and western portions of Jeju Province. Significantly higher contents of macronutrients were found in the soil and garlic from the eastern region of Jeju compared to western region, but micronutrients were higher in the western region than eastern region. In addition, the alliin and total phenolic contents were higher in the western region than in the eastern region. Also, allicin, total pyruvate, total flavonoid content, and ${\alpha}$-glucosidase inhibitory activity of the eastern region samples were higher than those of the western region in Jeju. This study promotes our understanding of the different components of garlic according to the cultivation regions of Jeju.

Effect of Storage Conditions on the Chemical Characteristics of Traditional Kochujang (저장조건에 따른 전통고추장의 성분 특성 변화)

  • Effect of anti-microbial agents, such as alcohol, garlic, chitosan, K-sorbate, mustard, or pasteurization on the quality of traditional kochujang was investigated during storage at $30^{\circ}C$ for 24 weeks. The pH of kochujang decreased during storage and titratable acidity changed little in the group of mustard and garlic addition. Reducing sugar increased during storage, with higher values for K-sorbate of alcohol added groups than the other groups. Alcohol contents increased during storage with higher values in control or chitosan added group. Amino nitrogen contents of kochujang decreased gradually during storage and were lower in control or chitosan added group. Ammonia nitrogen contents were lower in alcohol, garlic, K-sorbate or mustard added kochujang. The content of total free amino acid increased slightly during storage and were higher in pasteurized, garlic or alcohol added kochujang. After 24 weeks of storage, the results of sensory evaluation showed K-sorbate, garlic or alcohol added kochujang were more acceptable than control in the sense of taste.

Quality of Soybean Paste (Doenjang) Prepared with Sweet Tangle, Sea Mustard and Anchovy Powder (다시마, 미역 및 멸치분말이 첨가된 된장의 품질 특성)

  • To improve the quality of traditional Doeniang, the sea tangle, sea mustard, or anchovy powder were added to soybean paste and matured for 90 days. The quality and sensory evaluation of the Doenjang were analysed. The addition of anchovy showed higher fat and protein contents in the Doenjang. In the mineral contents, the sweet tangle and anchovy addition significantly increased Mg and K contents. The reducing sugar content showed 9.4% in the Doenjang prepared with sweet tangle and anchovy powder than 6.0% of the control. However, the pH and acidity were not showed any significantly difference at the 5% level by Duncan's multiple range test. The amino nitrogen content was 618.7 mg% in the anchovy Doeniang that is higher than 531.1 mg% of the control. The addition of sweet tangle and anchovy to the Doenjang showed higher score in the sensory evaluation.

The effects of soil condition on the agronomic characters and aromatic substances in cultivated Codonopsis Lanceolata (토양조건이 더덕의 생육특성 및 향끽미물질 발현에 미치는 영향)

  • The experiment was carried out to determine the effects of soil condition on the agronomic characters and aromatic substances in cultivated Codonopsis Lanceolata. An increase of planting densities of Codonopsis Lanceolata. from 1 to 5 plant per pot resulted in the smaller root width, but root yield increased from 109.8g to 286.9g per 1/200pot. The width growth as soil water contents were higher in surplus soil water than that of cultivated deficit soil water. The contents of water, reducing sugar, crude protein and malic acid were higher in cultivated wild than in cultivated upland soil. The higher contents of volatile essential oils as 3-rnethylpentanoic acid and squalene was obtained at the normal soil moisture.

Quality and Antioxidant Characteristics according to Different Harvest Periods and Steaming Treatment of Apios (Apios americana Medikus) Cultivated in Korea (국내산 아피오스의 수확시기와 증자처리에 따른 품질 및 항산화 특성)

  • The quality and antioxidant characteristics of apios (Apios americana Medikus) according to different harvest periods and steaming treatment were investigated. The quality and antioxidant characteristics of apios were significantly different depending on harvest periods. Total starch contents was higher in 1st harvesting period as 62.32 g/100 g than other harvesting period. The water binding capacity and water solubility index was higher in 1st harvesting period as 228.65 and 11.29% than other harvesting period. The sucrose and total free sugar contents were 3.64~8.67 and 4.49~9.54 g/100 g, respectively. Total polyphenol and flavonoid contents of apios was the highest 2nd and 4th harvesting period at 4.21 mg GAE/g and 611.11 ㎍ CE/g, respectively. DPPH radical scavenging activity was higher in 1st harvesting period as 84.96 mg TE/100 g than other harvesting period, and decreased as the harvest periods were delayed. ABTS radical scavenging activity and ferric-reducing antioxidant power were 43.81~47.89 mg TE/g and 231.20~264.07 mM/100 g, and increased to 50.58~51.44 mg TE/g and 342.55~384.29 mM/100 g after heat treatment. As a result, it is thought that studies on change of quality and physicochemical characteristics according to cultivation characteristics should be preceded for cultivation stability of apios.

Microbial Diversity in the Enrichment Cultures from the Fermented Beverage of Plant Extract Using Ribosomal RNA Sequence Analysis (라이보좀 RNA 염기서열 분석을 이용한 집식배양된 식물추출물발효음료의 미생물 다양성)

  • A beverage was produced by the fermentation of mixed extracts from the various fruits, vegetables, algae, and medical herbs. The physicochemical properties of the fermented beverage of plant extracts (FBPE) and microbial diversity were analyzed in cultures enriched from FBPE using 16S and 26S rRNA gene sequence analyses. The pH, acidity, $^{\circ}brix$, reducing sugar, and alcohol contents of the FBPE were determined to be the 3.48, 1.68%, 70.0, 1,026 g/L, and 3.5%, respectively. The most abundant free sugar and organic acid in the FBPE were glucose (567.83 g/L) and tartaric acid (93.68 mg/L), respectively. Lactobacillus homohiochii was the predominant species in all enriched culture samples: 100% of the species in 0B (0% sugar) and 40B (40% sugar) libraries and 95.6% of 20B library (20% sugar). Lactobacillus fructivorans was detected in the 20B library. The predominant species in the samples of enrichment cultures collected from FBPE with three different sugar concentrations were: Candida zeylanoides (45.2%) in the 0Y library (0% sugar), Candida lactis-condensi (35.7%) and C. zeylanoides (35.7%) in the 20Y library (20% sugar), and C. lactis-condensi (38.1%) in the 40Y library (40% sugar). This result may provide a useful frame of reference for further analyses of microbial population dynamics in FBPE.

Physicochemical Properties and Antioxidant Activities of Steam-Dried Allium hookeri Root (증포 처리한 삼채 뿌리의 이화학적 특성 및 항산화 활성)

  • This study was carried out to investigate the effect of steam-drying method on physicochemical properties and antioxidant activities of Alliun hookeri root (AHR). Moisture, crude protein, crude fat, and crude ash contents of raw and steam-dried AHRs were 10.91~14.15%, 11.14~13.49%, 0.83~3.02%, and 7.55~8.98%, respectively. Sulfur contents of steam-dried AHRs were 2.0 and 2.2 times lower than that of raw AHR (0.51%), respectively. pH and total sugar contents of AHRs were reduced by steam-drying, whereas titrate acidity and browning intensity were increased. The L and b values of AHRs in Hunter's value were also reduced, but a value was increased by steam-drying. Among hot water extracts from raw and steam-dried AHRs, four times steam-drying showed the lowest $EC_{50}$ values (0.44, 9.01, and 0.48 mg/mL, respectively) in DPPH radical assay, ABTS radical assay, and reducing power, whereas four times steam-drying had the highest total phenolic content ($34.47{\mu}g/mg$) and browning intensity (2.05 and 0.20 at 280 and 420 nm, respectively). The antioxidant activities of hot water extracts from raw and steam-dried AHRs were closely correlated with their total phenolic contents and browning intensity, showing coefficient of determination ($R^2$) values higher than 0.87. From the results, we suggest that steam-drying method could be used as an effective process for increasing the antioxidant activity of AHR.
