Investigation of Filamentous Fungi Producing Safe, Functional Water-Soluble Pigments

  • The production of water-soluble pigments by fungal strains indigenous to South Korea was investigated to find those that are highly productive in submerged culture. Among 113 candidates, 34 strains that colored the inoculated potato dextrose agar medium were selected. They were cultured in potato dextrose broth and extracted with ethanol. The productivity, functionality (radical-scavenging activities), and color information (CIELAB values) of the pigment extracts were measured. Five species produced intense yellowish pigments, and two produced intense reddish pigments that ranked the highest in terms of absorbance units produced per day. The pigment extracts of Penicillium miczynskii, Sanghuangporus baumii, Trichoderma sp. 1, and Trichoderma afroharzianum exhibited high radical-scavenging activity. However, the S. baumii extract showed moderate toxicity in the acute toxicity test, which limits the industrial application of this pigment. In conclusion, P. miczynskii KUC1721, Trichoderma sp. 1 KUC1716, and T. afroharzianum KUC21213 were the best fungal candidates to be industrial producers of safe, functional water-soluble pigments.

  • In this study, to obtain isoindoline compounds with the high thermal resistance and reddish yellow color using an environmental friendly method without the use of any surfactants, isoindoline derivatives with different structures were added at various reaction temperatures for the synthesis and the product was then crystallized by controlling temperatures and times in autoclave. Chemical structures, particle sizes, color differences, and optical properties were evaluated by the means of FT-IR, FE-SEM, UV-Vis spectroscopy, PSA (particle size analyzer) and Zeta potential analyzer. The samples with an enhanced dispersibility, highly thermal resistance, uniform particle sizes were achieved possibly due to the addition of isoindoline derivatives into the crystallization processing mixtures. The color change trend was also observed depending upon synthesis conditions.

  • 3-Hydroxy-canthaxanthin, reddish purple needle shaped crystals, has been isolated from the abdominal skin of Bombina Orientalis. Its structure has been established by visible and IR spectral characteristics, chromatographic behaviors, and physical and chemical properties.

  • Scolelepis (Scolelepis) daphoinos is newly reported in Korean fauna. This species can be distinguished from its congeners by the following characteristics: the presence of reddish pigment patches on the posterior part of the prostomium, notopodial postchaetal lamellae that are partially fused to the branchiae, and the presence of only the bidentate hooded hooks. The morphological diagnosis and photographs of S. (S.) daphoinos are provided. The partial mitochondrial cytochrome c oxidase subunit I (COI), 16S ribosomal DNA(16S rDNA), and the nuclear 18S ribosomal DNA (18S rDNA) sequences from Korean specimens of S. (S.) daphoinos were determined. Species identification was supported by a comparison of DNA barcode sequences of COI and 16S rDNA with morphological examination from the specimens of type locality, China.

  • Two urostyloid ciliates, collected from brackish water in Korea, were identified as Diaxonella pseudorubra pseudorubra (Kaltenbach, 1960) Berger, 2006 and Pseudokeronopsis flava (Cohn, 1866) Wirnsberger, Larsen and Uhlig, 1987. The description was based on living, protargol impregnated specimens. These species are described as follows: Diaxonella pseudorubra pseudorubra: body size in vivo $145-230{\times}40-60\;{\mu}m$, elongated ellipsoidal in shape. Cytoplasm reddish and flexible. Adoral zone of membranelles occupied 30-40% of the body; composed of 33-44 membranelles; 1-3 frontoterminal cirri, 1-4 frontal row cirri, 4-6 buccal cirri, 6-10 transverse cirri. Midventral rows composed of 14-24 cirri, four left marginal rows, one right marginal row. Two kinds of cortical granules; the larger one is yellowish and the smaller one is reddish. Pseudokeronopsis flava: body size in vivo $150-210{\times}30-45\;{\mu}m$, elongated ellipsoidal shape. Cytoplasm yellowish and flexible. Adoral zone of membranelles occupied 25-30% of body; composed of 44-58 membranelles in number. Frontal cirri forming bicorona composed of 5-7 cirral pairs, 2-3 frontoterminal cirri, one buccal cirrus, and 2-3 transverse cirri. Midventral rows composed of 18-33 cirri, 34-53 left marginal cirri, and 40-58 right marginal cirri. Two kinds of cortical granules; the larger one is colorless and "blood-cell-shaped," and the smaller one is yellowish. Diaxonella pseudorubra pseudorubra is different from the most similar subspecies, D. pseudorubra pulchra, in cytoplasmic color and number of midventral cirri. Pseudokeronopsis flava is different from its most similar congeners in pigment granular color, number of bicorona, number of midventral cirri, and position of the contractile vacuole.

Prodigiosin Production From Serratia sp. PDGS120915 Isolated From Daeyeon Stream Water in Busan (하천에서 분리한 Serratia sp. PDGS120915의 프로디지오신 생산)

  • Prodigiosin is a red pigment characterized by a common pyrrolylpyrromethane skeleton. It is produced by Serratia marcescens, Vibrio psychroerythrus, Hahella chejuensis, etc. Prodigiosin has been reported to possess anticancer, immunosuppressant, antifungal antimalarial, and algicidal activities. However, despite prodigiosin's diverse range of activities, its production rate is significantly low and biosynthesis conditions are difficult. Consequently, the selling price is high, and its usability is limited. This study aimed to increase the efficiency of prodigiosin production according to the culture conditions of Serratia. In this study, a bacterial strain PDGS120915 producing prodigiosin was isolated from lightly contaminated stream water in Busan and identified as a strain of Serratia sp. based on 16S rDNA gene sequence analysis and physiological characteristics. The reddish pigment from PDGS120915 was directly extracted using acidified ethanol, and a characterization analysis confirmed that it was a prodigiosin compound. The optimal conditions for pigment production were 25℃, pH 7, and 0% NaCl concentration for a duration of 14 hr. Furthermore, by treating carbon and nitrogen sources, such as fructose and beef extract, respectively, prodigiosin production increased approximately six-fold and four-fold. Among the minerals tested, 0.1% KCl was found to be the most effective for prodigiosin production. Moreover, casein was identified as the most suitable source for prodigiosin production.

Redescription of Two Marine Ciliates (Ciliophora: Urostylida: Pseudokeronopsidae), Pseudokeronopsis carnea and Uroleptopsis citrina, from Korea

  • The morphology of the two marine urostyloid ciliates, Pseudokeronopsis carnea (Cohn, 1866) and Uroleptopsis citrina Kahl, 1932, in the family Pseudokeronopsidae, collected from the Yellow Sea, and the East Sea, Korea, respectively, were studied using live observation and protargol impregnation. Additionally, the small subunit ribosomal RNA (SSU rRNA) gene was sequenced. These two species are firstly recorded in Korea. The main diagnostic key is as follows. Pseudokeronopsis carnea: body outline elongate-elliptical, brown-reddish or orange-red in colour in vivo; bicorona of 16-24 frontal cirri; one buccal and two frontoterminal cirri; 7-10 transverse cirri; 5-7 dorsal kineties; two types of cortical granules (one orange-red pigment, mainly grouped around cirri and dorsal bristles, arranged in typical rubra-pattern; the other, colourless and blood-cell-shaped, and densely distributed); contractile vacuole in the posterior half of the cell on the left side, usually in posterior 1/3-2/5. Uroleptopsis citrina: body outline elongate-elliptical, lemon-yellow in colour in vivo; two types of cortical granules (one yellow pigment; the other, blood-cell-shaped, densely distributed); bicorona of 12-18 frontal cirri; 2-3 frontoterminal cirri; two midventral rows comprising 26-35 cirri (consisting of anterior paired cirri, non-paired single cirri, and posterior paired cirri); three dorsal kineties. In addition, the SSU rRNA sequences of the two species were compared with public database of these species and consequently, showed high similarity.

A Study on the Dyeing of Silk Fabrics Treated with Aplysia kurodai Extract (군소 추출색소를 이용한 견직물의 염색성에 관한 연구)

  • This study intends to identify applicability of natural dye extracted from Aplysia kurodai, aiming to standardization of Aplysia kurodai natural dyeing through scientific validation by analyzing characteristics of pigment elements to review dyeability, colorfastness and antibacterial activity. Such material as silk fabrics that is animal fiber were used for this purpose, and derived following summarized results. UV/VIS spectrum analysis on the pigment of Aplysia kurodai that was extracted from purple gland showed that maximum absorbtion was near 540 nm. The silk fabric optimal dyeing can be achieved at a temperature of $55^{\circ}C$ with a colorant concentration of 5%. Dyed at $55^{\circ}C$ with interval of 10~90 minutes for identifying dye uptake over time to observed slow increase of dye uptake over time, and equilibrium occurred at 50 minutes. For dye uptake according to pH, while dye uptake was superior in acidity, it decreased rapidly in a base after pH 7. For color changes according to pH variation, it was reddish purple in acidity and was purplish red in a base. For color changes according to mordanting method, more clear color change had been obtained when process with aluminium pre-mordanting than non-mordanting and post-mordanting. The colorfastness to light, perspiration and washing was 1, 4~5, and 3~4 ratings respectively. The silk fabric dyed with Aplysia kurodai demonstrated excellent antimicrobial activity to Staphylococcus aureus and Klebsiella pneumoniae. The Aplysia kurodai can be used as a new colorant for the natural dyeing of silk.

A Basic Research for Preservation of Works Exhibited in the Outdoor Sculpture Park - A Scientific Analysis of Painted Work 'Conversion' Exhibited in the Cheonmasan Sculpture Park -

  • Outdoor sculptures of modern art works are being damaged and deteriorated as they are exposed to the outdoor environment due to the nature of exhibition in the outdoor environment, but secure of basic data through the measures for conservation and advanced researches still remain in the early stage. The surface of "Conversion" which is exhibited in the Busan Cheonmasan Sculpture Park has been exfoliated and deteriorated due to outdoor exhibition for a long time, so systematic conservation and management of works are considered necessary. Prior to the conservation and management, this study conducted observation of cross section, analysis of inorganic components, FT-IR, Raman and Py-GC/Mass analysis to examine the nature and type of paints used for the work through a scientific analysis. As a result of analysis, paints used for the "Conversion" include paint mixed with silvery aluminium powder and white pigment, reddish paint mixed with toluidine red, bluish paint that mixed prussian blue and titanium white and mixture of phthalocyanine blue and titanium white. The result is expected to be used as basic data for selecting materials necessary for conservative treatment of and establishing a plan for conservative treatment of the "Conversion".

Natural Dyeing of Silk, Cotton and Rayon Fabrics using Tea Leaves -Focusing on Green Tea, Oolong Tea, Black Tea, Dark Tea- (차에 의한 천연염색 연구 -녹차, 우롱차, 홍차, 흑차를 중심으로-)

  • The objective of this study was to investigate dyeing characteristics of silk, cotton and rayon fabrics using dyeing solution extracted from different varieties of tea. Comparative results of CIE Lab values of 360 pieces of dyed fabrics were studied to quantify the effects of dye concentrations (100%, 150%, and 200%) and mordants (Fe, Cu, Sn, Al). In addition, the color difference values (${\Delta}E$) of the dyed and mordanted fabrics were evaluated. The results of the study were as follows: First, among all of the 360 pieces of silk, cotton and rayon fabrics, the best dyeing effects was observed on silk fabrics. Second, the dyeing effect of the eight different types of tealeaves belonging to green tea, oolong tea, black tea, and dark tea tended to enhance as the degree of fermentation increased. Third, when the fabrics were mordanted with Fe, Cu, Sn, Al mordants, various colors such as brown, brownish red, brownish yellow, gray, and reddish brown were obtained.
