This study examined the effect of pine needle distillate (Pinus densiflora Sieb. et Zucc) on the immune system and hematological parameters. C57BL/6 male mice weighing 20 ~21 g were divided into 3 groups and intraperitonially injected with either 200 $\mu$L of saline (control), 50% diluted (P50) or 100% pine needle distillate (P100) once a day for 24 days. At the end of the experiment, the mice were anesthetized by ether and peripheral blood was collected from the femoral artery and the spleen was excised. Spleen weight decreased significantly (p<0.001) in the pine needle groups compared to the control group. The blood was used for a complete blood count and flow cytometrical analysis after immunofluorescence staining. The pine needle distillate dose-dependently decreased the CD4$^{+}$/CD8 sup +/ ratio (p <0.05), and showed a tendency to increase the mean FSC (forward scatter) values of the CD8$^{+}$T cells, while decrease the values of the CD4$^{+}$T cells. There were no significant differences in WBC, RBC and platelet counts among the three groups, but hemoglobin and hemoglobin-related parameters and platelet volume increased and red blood cell volumes decreased with the administration of the pine needle distillate. These results suggest that the pine needle distillate may have immunosuppressive effects.
The aim of this study was to evaluate gamma-aminobutyric acid (GABA)-induced erythropoietin (EPO) and EPO-receptor expression in human Hep3B cells and Sprague Dawley (SD) rats during hypoxia. Expression levels of EPO, EPO-R mRNA, Janus kinase-2 (JAK-2), vascular endothelial growth factor (VEGF), hypoxia inducible factor-1 (HIF-1), and HIF-2 in response to GABA treatment were evaluated in cell lines. SD rats were randomly divided into 5 groups of 8 rats each, and GABA was orally administered; the groups were the normal control (NC), hypoxia-exposed (G0), as well as the GABA 1 mg/100 g body weight (BW) GABA treated group (G1), 5 mg/100 g BW GABA treated group (G5), and 10 mg/100 g BW GABA treated group (G10) with hypoxia. We analyzed EPO levels and red blood cell counts in rat blood and EPO gene expression in kidney tissue. EPO and VEGF mRNA levels in Hep3B cells exposed to hypoxia were significantly increased and further increased after GABA treatment. However, the expression of EPO-R and JAK-2 mRNAs were not affected by GABA, but hypoxia-induced HIF-1 and HIF-2 mRNA expression was inhibited by GABA. In the kidney tissue of rats exposed to hypoxia, the expression level of EPO mRNA was greatly increased, but levels in the GABA treatment groups significantly decreased. EPO levels in the serum showed the same significant trend, but the red blood cell counts were not significantly different. These findings demonstrate that HIF-1 and HIF-2 activation increase EPO expression in Hep3B cells exposed to hypoxia. However HIF decreased by GABA addition and VEGF increased significantly.
A 60-day experiment was conducted to determine the effect of feeding and starvation on the survival, growth, and blood parameters of red sea bream Pagrus major. The starved group was not fed during the first 32 days of the experiment and was then fed for 28 days. The growth rate of the starved fish group was significantly lower than that of the fed fish group. Starvation resulted in growth retardation and reductions in final body weight, growth rate, specific growth rate, and condition factor, whereas the fed fish group grew well and maintained a good condition. The growth rate of the starved group that was refed was higher than that of the fed group. Red blood cells, hematocrit, and hemoglobin showed no significant differences between the fed and starved groups. The cortisol and glucose levels of the fed group of juveniles were higher than those of the starved group. The cortisol levels of the starved group of adults were higher than those of the fed group, whereas the glucose levels of the starved group were lower than those of the fed group. These results suggest that the survival, growth, and hematological parameters used to reflect starvation and feeding in this study provide a useful index of physiological response and survival rate in red sea bream.
Wattle length, width, and area were measured to classify bilateral asymmetries in four lines of chickens. The lines were the S26 generation of White Leghorns selected for high (HAS) or low (LAS) response to sheep red blood cells and sublines in which selection had been relaxed for three generations (high antibody relaxed [HAR] and low antibody relaxed [LAR]). Antibody titers (AB) were greater for HAS than for HAR with both greater than for LAS and LAR which while different for males did not differ for females. The low antibody lines were heavier and reached sexual maturity at younger age than the high antibody lines. In general, wattle length, width, and area were greater in the low than high antibody lines. In 24 comparisons for bilaterality 18 exhibited fluctuating asymmetry and 6 exhibited directional asymmetry with 5 of the 6 being for wattle length. There was not a clear pattern for changes in degree of asymmetry when selection was relaxed for 3 generations. For females, the relative asymmetry (RA) of wattle area was larger ($p{\leq}0.05$) for HAR than for LAR and not different from the selected lines and relaxed lines. There were no differences among lines for RA of wattle length and width of females and wattle length, width, and area of males.

The effect of a solute concentration difference on the osmotic transport of water through the semi-permeable membrane of a simple cell model is investigated. So far, most studies on osmotic phenomena are described by simple diffusion-type equations ignoring all fluid motion or described by Stokes flow. In our work, as the governing equations, we consider the coupled full Navier-Stokes equations which describe the fluid motion and the full transport equation that takes into account of convection and diffusion effects. A two dimensional finite difference model has been developed to simulate the velocity field, concentration field, and semi-permeable membrane movement. It is shown that the cell swells to regions of lower solute concentration due to the uneven water flux through the semi-permeable membrane. The simulation is applied on a red blood cell geometry and the relevant results are presented.
The immunomodulating activity of Kp, an antitumor protein-polysacchanide preparation from the shake-cultured mycelia of Phellinus linteus, was investigated in ICR mice subcutaneously implanted wit $1\times10^6$ cells of sarcoma 180. The mice were intraperitoneally administered with Kp at a does of 100 mg/kg once daily for five consecutive days starting from 24 hrs after the tumor implantation. Ten days after the last injection, the mice were immunized with $1\times10^7$ or $4\times10^8$ sheep red blood cells (SRBC) and five days later, the antibody-forming immune response were assessed by direct hemolytic plaque assay. To an immunization does of $1\times10^7$ SRBC, the Kp-treated mice elicied a successful humoral immune response despite the turmor-burden and produced $259\times10^3$ plaque-forming cells (PFC)/spleen, while the corresponding tumor-bearing control mice showed virtually no reponse $(2.0\times10^3$ PFC/spleen) (the stimulation index=129.5). However, to an immunization dose of $4\times10^8$ SRBC, both of the control mice and Kp-treated mice showed almost the same level of strong humoral immune response. From these data it is clear that Kp effectively restores the humoral immune response of the turmor-bearing ICR mice.
To investigate the nature of typhoid cells, three cases of clinically, serologically and histopathologically proven typhoid lesions of the small intestine and regional lymph nodes were studied light and electron microscopically, Light microscopically, typhoid cells were swollen mononuclear cells characterized by abundant amount of eosinophilic cytoplasm and frequent phagocytoses of red blood cells, bacterial clumps and other tissue debris. These cells were pyronin negative, Electron microscopically, these cells showed marked and diffuse dilatation of RER cisternae and disappearance of ordinary cytoplasmic organelles, but frequent phagocytosed materials, The meaning and reason of RER cisternal dilatation and reduction of cytoplasmic organelles were discussed, and are regarded as degenerative process due to bacterial endotoxin. Although there was hot enough cytoplasmic organelles to pinpoint the origin of typhoid cells, active phagocytosis and evidences against being either plasmacytic or lymphocytic nature favored retuculoendothelial nature of the typhoid cells.
In this study, we investigated the biological damage and stress responses induced by ion beam (proton beam) irradiation as a basis for the development of protective measures against space radiation. We examined the biological effects of proton beam produced by MC-50 cyclotron at KIRAMS on the cultured cells and mice. The proton beam energy used in this study was 34.9 MeV and the absorption dose rate for cells and mice were $0.509Gy\;sec^{-1}$ and $0.65Gy\;sec^{-1}$, respectively. The cell survival rates measured by plating efficiency showed the different sensitivity and dose-relationship between CHO cells and Balb/3T3 cells. HGPRT gene mutation frequency in Balb/3T3 was $15{\times}10^{-6}Gy^{-1}$, which was similar to the reported value of X-ray. When stress signaling proteins were examined in Balb/3T3 cells, $I{\kappa}B-{\alpha}$ decreased markedly whereas p53, phospho-p53, and Rb increased after proton beam irradiation, which implied that the stress signaling pathways were activated by proton beam irradiation. In addition, cellular senescence was induced in IMR-90 cells. In the experiments with C57BL/6 mouse, the immune cells (white blood cells, lymphocytes) in the peripheral blood were greatly reduced following proton beam irradiation whereas red blood cells and platelets showed relatively little change. These results can be utilized as basic data for studying the biological effects of proton beam using MC-50 cyclotron with respect to proton therapy research as well as space radiation research.
The aim of our study is to determine maximum surgical blood order schedule (MSBOS) in each surgical operation through analyzing usage of blood products at Bundang Jesaeng General Hospital. We investigated the amount of transfused red cells for each operation and calculated crossmatching-to-transfusion ratio (C/T ratio) and MSBOS. This was accomplished by referring to the Laboratory Information System program during 1 year from January through December 2003. Coronary artery bypass surgery and decompressive craniectomy showed the highest MSBOS in our hospital. The average C/T ratio was 3.2 and excessive reservations for blood products have been made for many operations. From this study, guidelines for the optimal blood ordering for each surgery were suggested.

Background: The accumulation of unpaired α-globin chains in patients with β-thalassemia major may clinically create ineffective erythropoiesis, hemolysis, and chronic anemia. Multiple blood transfusions and iron overload cause cellular oxidative damage. However, α-tocopherol, an antioxidant, is a potent scavenger of lipid radicals in the membranes of red blood cells (RBCs) of patients with β-thalassemia major. Purpose: To evaluate the effects of α-tocopherol on hemolysis and oxidative stress markers on the RBC membranes of patients with β-thalassemia major. Methods: Forty subjects included in this randomized controlled trial were allocated to the placebo and α-tocopherol groups. Doses of α-tocopherol were based on Institute of Medicine recommendations: 4-8 years old, 200 mg/day; 9-13 years old, 400 mg/day; 14-18 years old, 600 mg/day. Hemolysis, oxidative stress, and antioxidant variables were evaluated before and after 4-week α-tocopherol or placebo treatment, performed before blood transfusions. Results: Significant enhancements in plasma haptoglobin were noted in the α-tocopherol group (3.01 mg/dL; range, 0.60-42.42 mg/dL; P=0.021). However, there was no significant intergroup difference in osmotic fragility test results; hemopexin, malondialdehyde, reduced glutathione (GSH), or oxidized glutathione (GSSG) levels; or GSH/GSSG ratio. Conclusion: Use of α-tocopherol could indirectly improve hemolysis and haptoglobin levels. However, it played no significant role in oxidative stress or as an endogen antioxidant marker in β-thalassemia major.
