Isolation of Deletion Mutants by Reverse Genetics in Caenorhabditis elegans

  • Obtaining mutant animals is important for studying the function of a particular gene. A chemical mutagenesis was first carried out to generate mutations in C. elegans. In this study, we used ultraviolet-activated 4,5',8-trimethylpsoralen to induce small deletion mutations. A library of mutagenized worms was prepared for recovery of candidate animals and stored at $15^{\circ}C$ during screening instead of being made into a frozen stock library. In order to isolate deletion mutations in target genes, a polymerase chain reaction (PCR)-based screening method was used. As a result, two independent mutants with deletions of approximately 1.0 kb and 1.3 kb were isolated. This modified and improved reverse genetic approach was proven to be effective and practical for isolating mutant animals to study gene function at the organismal level.

  • In July, 2000, A 71-year-old man complained of these symptoms: frequent defecation, urgent need to defecate, fecal leakage, anal pain, flatus, and diarrhea after surgery for colorectal cancer. The symptoms are caused specifically by injury to the bowel and sphincter function. The patient suffered physically and psychologically; but, there was no special treatment in western medicine. Therefore, we treated him with electrical acupuncture for the management of sphincter tone and the alleviation of anal pain. We also treated him with herbal medicine for the recovery of strength and gastro-colic function. As a consequence of these treatments, the bowel disorders improved immediately and successfully, even though there were some limitations. So, We report this case with a brief review of related literatures.

Verapamil [anticalcium agent]의 심근 보호작용Langendorff씨 장치하의 심근 보호 작용 (Effects of Myocardial Protection of Verapamil in Serum: Under Langendorff Apparatus in Ischemic Arrest Heart)

  • This study was evaluated the metabolic, physiologic and histologic effects of myocardial protection of verapamil[isoveratril]on isolated rat hearts to 90 minutes of ischemic arrest. Heart was perfused with a modified Kreb’s Henseleit bicarbonate buffer with glucose and arrested with retrograde coronary perfusion by glucose insulin[GI], potassium and verapamil. Mean aortic systolic pressure, heart rate, coronary flows were measured and morphologic changes were examined during working heart perfusion. Perfusion and arrest were controlled four groups subjected 60 isolated rat hearts. Four groups hearts reperfused during 40 minutes after 90 minutes global ischemia for physiologic recovery. 15 hearts of four groups were assayed to histological morphologic changes. GI treated hearts recovered less than 28% of function and changed more than 80% of mitochondria of control group. Verapamil hearts[0.2, 0.1 gm/kg] recovered more than 88% of function and permitted the maintenance of continuous cellular level of Serum Glutamic Oxalaxetate Transaminase[SGOT], but declined 28% of Phosphate Kinase[CP], GI treated heart showed widespread evidence of extensive damage of mitochondria. The damage was that interstitial huge edema are present and there was contraction band formation within the swollen cells. The verapamil and potassium group were not found morphologic change compared with control group. Their functions were shown that metabolic and physiologic action of verapamil-group lasted 20 minutes longer than potassium group.

The Influence of Hardwood Interspecific Competition on Stand Structure and Dynamics for Loblolly Pine Plantations

  • The purpose of this study is to investigate the effects of hardwood competitions in stand structure and dynamics by applying prediction models for unthinned loblolly pine (Pinus taeda L.) plantations. A parameter recovery procedure for the Weibull distribution function based on four percentile equations was applied to develop diameter distribution prediction models. Four percentiles of the cumulative diameter distribution prediction equations were predicted as a function of quadratic mean diameter plus competin hardwood trees perhectare varibales. According to the results of this study. it was found that as the amount of competing hardwood trees increased, diameter distributions in terms of stand structure dynamics tended to be more skewed to the right. Therefore, the influence of non-planted hardwood trees interspecific competitoin on planted loblolly pines showed negative effects on the stand structure and dynamics.

  • The knee joint is composed of 3 skeletons that is the femoral bone, the tibial bone, and the patella bone. The tibiofemoral pint and patellofemoral pint act with the meniscus, so these function that is maintain the stabilities by the surrounding soft tissue is complex. The protection mechanism(muscle tension) of the surrounding muscles for the joint disease(Arthritis) limits consistently the motion of the pint to decrease the internal pressure of the joint, and these muscle tension acts with abnormal function for the surrounding tissue and the joint, sometimes the contracture is developed, if the joint with disease is not recovery or treated within early time. So we worked out efficient orthopedic local taping for the patient who is complained of the knee pint pain using the literature investigation about the anatomical structure and the biomechanics of the knee pint for the muscle and the pint problem esp, the rotation of the tibia, the dislocation of the patella, and the motion of the meniscus that is developed due to tension of surrounding muscles of the knee pint. And application of the pint mobilization, the stretching, and the muscle strengthening exercise for the pint will become successful treatment for the joint disease.

슬관절 수술 후 재활에 대한 Continuous Passive Motion 치료와 한의 병행치료의 체계적 문헌 고찰 (A Systemic Review of Korean Medical Treatment Combined with Continuous Passive Motion Therapy on the Knee Joint Postoperative Rehabilitation)

  • Objectives The objective of this study is to report the evidence that continuous passive motion (CPM) therapy combined with Korean Medicine is effective for knee injuries. Methods We searched 2 Korean medical electronic databases, and 2 Chinese electronic databases to find randomized controlled clinical trials (RCTs) that treat knee injuries with Korean Medicine combined with CPM therapy. We selected studies by screening. We extracted data from selected RCTs, and analyzed RCTs by using Cochrane's Risk of bias criteria. Results 386 studies are searched, and 7 RCTs are selected. According to the study, several kinds of Korean Medicine such as hydrotherapy, decoction, acupuncture, moxibustion, and manual therapy are used with CPM therapy for the knee joint postoperative rehabilitation. We found the encouraging evidence that Korean Medicine and CPM combined therapy is more effective than single CPM therapy by alleviating pain, increasing range of motion, and improving knee function, etc. However, the risk of bias in most RCTs was judged to be uncertain. Conclusions Combination of Korean Medicine and CPM therapy can compensate the defect of single CPM therapy, and promotes the recovery of knee joint function.

바이오매스 순환단지조성을 위한 거버넌스 구축방안 연구 (A Study of the Efficient Planning of Governance for Building Biomass Circulation Estate)

  • This research estimates the necessity of a better governance plan on the purpose of fulfillment energy recovery by building resource recycling system for biomass resources and waste resources that derive from agricultural and mountain village areas. The utilization of new renewable energy technology which uses waste and biomass sources diverse as variety of resources, collecting method, operator etc. and is structurally complicated the formation of policy is also very difficult. There is failure because of the problems which occurs from the policy led by government. Biomass Town Development Project should be made through the central government and the local government integrated support system and should be formed a consultative group in order to process the project mutually with these two department including the experts from the related areas. This consultative group, while government organizations carry out the hub function of strategic knowledge management, should carry out the control tower function to be able to be net working transfer the information with the cooperation of private and government so vitalize the communication area among the related actors. And to be able to increase the participation rate of the local people the consistent and various educations should be given so a smooth business promotion progress will be desired through the change of perception and coactive participation of people.

The Effects of Group Exercise, Manual Therapy and Home Exercise on Pain, Range of Motion and Function in Patient with Adhesive Capsulitis

  • Purpose: The purpose of this study was to determine the effects of group exercise, manual therapy, and a home exercise program on VAS (visual analogue scale), ROM (range of motion) of shoulder, and functional outcome in adhesive capsulitis. Methods: The study was conducted with 45 patients. All subjects were randomly assigned to 3 groups: group exercise (n=15), manual therapy (n=15), home exercise (n=15). The subjects performed an intervention program 3 times a week for 4 weeks a total of 12 times. Outcome measurements were VAS, ROM, and Constant-Murley score outcome measures. Measurements were taken four times, at baseline, 4 weeks later, 8 weeks later, and 12 weeks later. Results: VAS, ROM of the shoulder, and functional outcomes showed improvement in all groups at the final follow-up (p<0.05). VAS and ROM showed greater improvement with group exercise and manual therapy than home exercise, however, there were no significant differences between the two groups (p>0.05). Functional outcomes showed the greatest improvement in group exercise at the final follow-up. This improvement was greater than with manual therapy or home exercise (p<0.05). Conclusion: Group exercise and manual therapy were more effective for improving pain, ROM, and function than home exercise in adhesive capsulitis. In particular, group exercise is more effective in functional recovery than other physiotherapy interventions.

Modification of Retinal Function by Hypothermia and Hyperthermia

  • Temperature-dependent electroretinogram responses were investigated in the dark adapted bullfrog eyes within the physiological temperature range 0-40$\^{C}$. In hypothermic process(25→0→25$\^{C}$), the amplitude of b-and c-wave decreased with lowering the temperature again. Both b-wave amplitude and threshold responses were maximal around 15$\^{C}$ during the temperature increment. Upon warming to room temperature again (25$\^{C}$), the b-wave amplitude was approximately doubled as compared to that of control without temperature changes. During the hyperthermic process (25→40→25$\^{C}$), however, the responses decreased with warming, and the wave amplitude failed to recover by cooling to 25$\^{C}$ again. As describe above, the recoveries of ERG in both processes show the striking difference. The hypothermia induces the amplification of the b-wave, that is, enhances the retinal function with the temperature recovery toward room temperature. While the hypertherima produces the decrease of the b-wave even though recovered to room temperature, which indicates an irreversible retina. The morphological alteration is shown both hypothermic and hyperthermic process, such as an appearance of large vacuoles and degenerating outer segments, more intense in hyperthermia, similar to light induced damage.

Preservation of Facial Nerve Function Repaired by Using Fibrin Glue-Coated Collagen Fleece for a Totally Transected Facial Nerve during Vestibular Schwannoma Surgery

  • Recently, the increasing rates of facial nerve preservation after vestibular schwannoma (VS) surgery have been achieved. However, the management of a partially or completely damaged facial nerve remains an important issue. The authors report a patient who was had a good recovery after a facial nerve reconstruction using fibrin glue-coated collagen fleece for a totally transected facial nerve during VS surgery. And, we verifed the anatomical preservation and functional outcome of the facial nerve with postoperative diffusion tensor (DT) imaging facial nerve tractography, electroneurography (ENoG) and House-Brackmann (HB) grade. DT imaging tractography at the 3rd postoperative day revealed preservation of facial nerve. And facial nerve degeneration ratio was 94.1% at 7th postoperative day ENoG. At postoperative 3 months and 1 year follow-up examination with DT imaging facial nerve tractography and ENoG, good results for facial nerve function were observed.
