During the last three decades considerable advances has been made in goat embryo production and transfer technology. The Korean native black goat is the most useful domestic ruminant in this country for biological investigation and application because it has a lot of merits such as relatively short generation period (1 vs 2 year for a cow), easy of handling, well adaptation, high fertility, convenient and inexpensive. This article covers the methods of superovulation, estrus synchronization, embryo collection and transfer techniques, pregnancy diagnosis and subsequent pregnancy and kidding rates for the production of transgenic Korean native black goats. More than one hundred goat kids have been produced as a result of our transgenic goat project via microinjection of foreign gene into pronuclei, in vitro culture, transfer of various stages of fresh and frozen-thawed microinjected embryos into oviducts or uteri of recipient does. We have got two transgenic goats carrying a transgene targeting the expression of recombinant human granulocyte colony stimulating factor (hG-CSF) to the mammary gland so far. Since collection and transfer of embryos in this species is usually accomplished by laparotomy, exteriorization of the reproductive tract for surgical embryo collection leads to the formation of post-operative adhesions. Nonsurgical or laparoscopic technique to reduce adhesions from repeated surgeries has great advantages in improving embryo production and transfer especially from valuable donors. We will discuss this later.
본 연구에서는 hGM-CSF를 생산해 배지 내로 분바하도록 유전자 조작된 Nicotiana tabacum 세포를 수성이상계 내에서 배양하여 in situ recovery를 시도하였다. 특히 식물세포 자체의 생상에 큰 영향을 주지 않으면서도 일반 배지에서 회수한 단백질에 비해 많은 양을 생산할 수 있는 수성이상계 시스템을 선정하였다. 6% (w/w) PEG 20,000과 10% (w/w) dextran 2.000,000 을 이용하는 경우 최대 세포 농도가 18.6 g/L 로, 일반배지를 사용하는 경우 (15.7 g/L)에 비해 저해가 없음을 확인하였다. 또한 목적 단백질인 hGM-CSF의 생산에 있어서도 위의 시스템의 경우 dextran-rich phase인 아랫 상으로 분배됨으로 인해 회수가 쉬울 것으로 확인되었으며 그 생산량 또한 일반배지에서의 생산량 (1.5 ng/mL)에 비해 크게 다르지 않음을 확인할 수 있었다. 따라서 hGM-CSF의 회수를 위한 in situ recovery 시스템에 있어서 수성이상계의 이용 가능성을 확인하였다.

Mono PEGylated rhG-CSF (PEG-G-CSF) prepared by utilizing unique PEG was purified and characterized by cation-exchange chromatography. A unique, trimer-structured PEG was chosen for PEGylation of rhG-CSF among various PEG moieties. The in-vitro bioactivity, stability, and pharmacokinetics of mono-PEG-G-CSF were examined and compared to those of native rhG-CSF. Mono PEG-G-CSF exhibited reduced in-vitro bioactivity to native rhG-CSF but showed an excellent in-vivo bioactivity and stability. Furthermore, it showed markedly reduced clearance in rats, thereby increasing the biological half-life by about 4.5-fold compared to that of native rhG-CSF. The results suggest that this unique, trimer-structured 23 kDa PEG can provide advantages to improve the bioactivity of therapeutic proteins in clinical use.
This study was conducted to evaluate the repeated dose toxicity of DA-3030, a recombinant human granulocyte colony stimulating factor(rhG-CSF), in rats. DA-3030 was administered intravenously once a day for 4 weeks to 20 males and 20 females per group at doses of 0(control), 115 and 1150 $\mu\textrm{g}$/kg, and to 15 males and 15 females per group at doses of 1.15 and 11.5$\mu\textrm{g}$/kg. After the administration period, 5 males and 5 females per group in the 0,115 and 1150 $\mu\textrm{g}$/kg groups were placed on withdrawal for 2 weeks. Through-out the study, all the rats survived. The administration of DA-3030 induced, a marked increase in the number of peripheral neutrophils, elevation of serum alkaline phosphatase activity, and splenomegaly in the rats of both sexes receiving 115 or 1150 $\mu\textrm{g}$/kg. Histopathologic examination revealed extramedullary granulopoiesis in spleen and liver, and increase in the number of activated macrophages in spleen in rats of both sexes in 115 and 115 $\mu\textrm{g}$/kg groups, and increased M/E ratio in 11.5, 115 and 1150$\mu\textrm{g}$/kg groups. Most of the changes produced by DA-3030 were thought to be attributable to exaggerated pharmacological effect of the drug, and subsided or disappeared after the recovery period. Under the present condition, no effect dose of DA-3030 is estimated at 1.15 $\mu\textrm{g}$/kg/day.
hGM-CSF가 식물세포 현탁 배양을 통하여 생산이 가능한지를 조사하기 위하여 hGM-CSF를 포함하고 있는 A. tumerfaciens LBA4404를 가지고 상추에 형질전환시켰다. 형질전환된 상추로부터 캘러스를 유도하여 캘러스를 이용한 세포배양체계를 확립하였다. PCR과 Southern blot analysis 결과 상추에 hGM-CSF 유전자가 도입된 것을 확인하였으며, Northern blot analysis 결과 상추식물체에 hGM-CSF 유전자가 발현됨을 확인하였다. 현탁 배양 세포로부터 분비된 hGM-CSF를 ELISA를 이용하여 측정한 결과 149.0 $\mu\textrm{g}$/L가 생산됨 을 확인하였다 이러한 결과는 상추의 현탁 배양 세포가 hGM-CSF와 같은 치료용 단백질의 생산 숙주로 이용될 수 있음을 보여주었다.
CD11c와 CD80 및 CD86과 같은 보조 수용체는 주로 수지상 세포에서 발현되는 세포 표지 인자이다. 본 연구에서는 KG-1, HL-60, NB4 및 THP-1 세포와 같은 여러 종류의 백혈병 세포를 이용하여 이들 세포에 재조합 Flt-3 리간드를 처리하였을 때 수지상 세포의 표면 인자인 CD11c의 발현에 어떠한 변화가 있는가를 조사하였다. KG-1 세포뿐만 아니라 NB4세포와 HL-60 세포에서도 Flt-3 수용체가 발현됨을 확인하였으나 THP-1 세포에서는 이들 수용체가 발현되지 않았다. KG-1 세포를 Flt-3 리간드나 granulocyte macrophage-colony stimulating factor (GM-CSF)와 tumor necrosis factor (TNF)-$\alpha$를 섞은 배양액에서 배양하였을 때 세포 증식은 억제되었으며 CD11c 발현은 현저히 증가되었다. 그러나 Flt-3 리간드를 처리한 KG-1세포에서는 GM-CSF와 TNF-$\alpha$를 처리한 세포에서와는 다르게 major histocompatibility complex (MHC)-I 및 MHC-II의 발현은 증가되지 않았다. Flt-3 리간드는 HL-60 세포와 NB4 세포의 CD11c 발현도 증가시켰으나 THP-1 세포에서는 아무런 영향이 없었다. CD11c의 발현과 비교하여 CD11b의 발현은 Flt-3 리간드에 의하여 KG-1 세포에서는 약하게 증가하였으나 NB4 세포와 HL-60 세포에서는 증가되지 않았다. KG-1 세포를 Flt-3 리간드로 처리하였을 때 extracellular signal-regulated kinase-1/2 (ERK-1/2)와 p38-mitogen-activated protein kinase (p38-MAPK)의 단백질 인산화가 증가되었으며 Flt-3 리간드에 의한 CD11c 발현의 증가는 MEK의 억제제인PD98059에 의하여 사라짐을 확인하였다. 본 연구 결과는 Flt-3 수용체 리간드의 처리에 의하여 $CD34^+$ myelomonocyte분화 단계인 KG-1 세포와 promyelocyte 분화 단계의 백혈병 세포에서 수지상 세포와 유사한 세포 형으로 분화된다는 것을 보였고 Flt-3 수용체 리간드에 의한 이들 백혈병 세포의 수지상 세포유사 세포로의 분화는 ERK-1/2의 활성화에 의하여 일어날 수 있음을 보여 준다.
상처치유는 growth factor와 cytokine들의 복합적인 작용에 의해 일어나는 과정이다. GM-CSF는 호중구와 대식세포를 분화시키고 각질세포를 증식시키는 다면발현성 사이토카인으로 여러 세포에서 재조합되어 사용되고 있다. (주)핸슨바이오텍에서 개발한 rice cell 유래의 재조합 rrhGM-CSF는 다른 세포에서 유래된 GM-CSF들보다 탄수화물 측쇄가 많은 것이 특징이다. rrhGM-CSF의 창상치유효과를 알아보기 위하여 햄스터에 만성 당뇨를 유도하여 상처치유가 어려운 동물모델을 만든 후, 등쪽에 인위적으로 피부 전 층을 제거하여 창상을 유발하였다. 실험군에는 rrhGM-CSF가 포함된 연고를 적용하고, 대조군에는 base cream만을 적용하여 재상피화되는 과정과 피부조직의 구조 및 PCNA 반응을 통한 세포증식 정도를 비교하였다. 육안 및 조직학적 관찰 결과, rrhGM-CSF를 적용한 그룹의 재상피화 속도가 더 빨랐으며, PCNA 면역조직화학적 조사에서도 증식하는 각질세포의 수가 대조군보다 상대적으로 많은 것으로 나타남으로써, rrhGM-CSF에 의한 피부상처 치유 효과를 관찰할 수 있었다. 이러한 결과들로 미루어 보아 벼세포 유래의 rrhGM-CSF는 국소적인 피부 창상 치유에 효과적인 것으로 보이며, rrhGM-CSF의 풍부한 당화 사슬은 이 단백질이 상처부위에 안정된 상태로 기능을 나타냄으로써 효과를 오랫동안 지속시킬 수 있었던 것으로 사료된다.
목적 : 최근 항암 화학요법이나 두경부의 방사선치료 후 나타나는 백혈구감소증을 치료하기 위해 조혈세포 성장인자인 과립구 집락형성인자를 투여한 결과, 백혈군감소증과 더불어 구강 점막염의 완화가 몇몇 임상연구에서 보고되고 있다. 그러나 그 기전이나 적절한 투여 방법에 대해서는 뚜렷이 밝혀지지 않았으며, 다른 위장관 점막에 대한 보호효과에 대해서도 아직 알려진 바가 없다. 이에 저자는 생쥐 공장 점막에 대한 과립구 집락형성인자의 방사선 보호제로서의 가능성을 알아보고자 하였다. 대상 및 방법 : 실험 동물은 체중 20 g 내외의 생후 $4\~5$주된 BALB/c 생쥐 105 마리로 각각 정상 대조군, 과립구 집락형성인자 단독투여군(제 I 군 : $10\;{\mu}g/kg/$일, 제 II 군 $100\;{\mu}g/kg/$일), 방사선 단독조사군(7.5 Gy 또는 12 Gy 전신조사), 그리고 방사선조사와 과립구 집락형성인자 병행 처치군(G-CSF I 또는 II+7.5 Gy, G-CSF I 또는 II+12 Gy)의 아홉군으로 구분하였다. 과립구 집락형성인자는 과립구 집락형성인자 단독투여군 및 방사선조사와 과립구 집락 형성인자 병행처치군 모두에서 방사선조사 2일 전부터 24시간 간격으로 방사선조사일까지 3일 동안 매일 1회 피하 주사하였고, 방사선조사는 6 MV 선형 가속기를 이용하여 7.5 Gy와 12 Gy를 각 군별로 1회 전신조사하였다. 각 군별로 생쥐를 방사선조사 후 1, 3, 7일에 희생하여 공장을 적출하고 H&E 염색 및 PAS 염색을 시행한 후, 공장 조직 표본에서 소낭선의 수, 융모의 길이 및 조직학적 손상등급을 측정하고 조직학적 변화를 관찰하였다. 결과 : 방사선조사없이 과립구 집락형성인자 만을 투여한 군(제 I 군 : $10\;{\mu}g/kg/$ 및 제 II 군: $100\;{\mu}g/kg/$) 에서는 정상 대조군에 비해 생쥐 공장 점막 변화에 유의한 차이가 없었다(p>0.05). 7.5 Gy 및 12 Gy 방사선 단독조사군에서는 정상 대조군에 비해 공장 점막의 소낭선 수의 감소와 높은 조직학적 손상등급을 관찰하였다(p<0.05). 12 Gy 방사선조사와 과립구 집락형성인자 병행처치군에서는 공장 점막의 손상이 7.5 Gy 방사선 단독조사군에 비해 1일 군에서는 차이가 없었으나(p>0.05), 3일 군에서 유의하게 높은 소낭선 수 및 낮은 조직학적 손상등급을 관찰하였고, 7일 군에서 낮은 조직학적 손상등급을 관찰하였다(p<0.05). 12 Gy 방사선조사와 과립구 집락형성인자 병행처치군에서의 공장 점막 손상은 12 Gy 방사선 단독조사군에 비해 소낭선의 수와 융모의 길이, 조직학적 손상등급의 차이가 유의하지 않았으며(p>0.05), 두 마리를 제외한 대부분의 생쥐가 방사선조사 후 5일 이내에 모두 사망하였다. 결론 : 한계 선량 이내의 전신 방사선조사시 과립구 집락형성인자는 생쥐의 공장 점막에 대한 방사선보호작용이 있는 것으로 사료된다.
Recently three proteins, playing central roles in the bidirectional transport of urate in renal proximal tubules, were identified: two members of the organic anion transporter (OAT) family, OAT1 and OAT3, and a protein that designated renal urate-anion exchanger (URAT1). Antibodies against these transporters are very important for investigating their expressions and functions. With the cytokine gene as a molecular adjuvant, genetic immunization-based antibody production offers several advantages including high specificity and high recognition to the native protein compared with current methods. We fused high antigenicity fragments of the three transporters to the plasmids pBQAP-TT containing T-cell epitopes and flanking regions from tetanus toxin, respectively. Gene gun immunization with these recombinant plasmids and two other adjuvant plasmids, which express granulocyte/macrophage colony-stimulating factor and FMS-like tyrosine kinase 3 ligand, induced high level immunoglobulin G antibodies, respectively. The native corresponding proteins of URAT1, OAT1 and OAT3, in human kidney can be recognized by their specific antibodies, respectively, with Western blot analysis and immunohistochemistry. Besides, URAT1 expression in Xenopus oocytes can also be recognized by its corresponding antibody with immuno-fluorescence. The successful production of the antibodies has provided an important tool for the study of UA transporters.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
