Changes of Gill Structure and Identification of Genes by Muddy Water Exposure in Cyprinus carpio (잉어(Cyprinus carpio)에서 탁수 노출에 의한 아가미 미세구조 변화와 유전자 확인)

  • Present study aimed to investigate morphological change and gene expression in the gill of Cyprinus carpio after exposure against muddy water caused by riverbed disturbance with various rearing condition. The gill of C. carpio showed abnormal shapes in its secondary lamellae and a rough surface with impure debris after exposed to muddy water for 80 days. In addition, the gills showed the edema, the exfoliation of epithelial cell, and the fusion of the secondary lamellae. Using 20 ACPs, 24 differentially expressed genes (DEGs) exposed to muddy water for 80 days were identified. 17 genes among them were up-regulated, while 7 genes were down-regulated in preterm deliveries. A BLAST searches revealed that 3 genes were revealed known genes as calcium transporter 1 (TRPV6) mRNA, macha mRNA for putative puroindoline b protein, and Efnb3 protein-like. Therefore, it is considered that Efnb3 gene from gill would be a useful indicator for neurobehavioral changes in fish influenced by muddy water.

The Effects of Temperature, Starvation, and Kind of Foods on Growth, RNA/DNA Ratios and Protein Contents during the Early Life Stages of Olive Flounder (Paralichthys olivaceus)

  • The effects of temperatures, starvation, and kind of foods on growth, RNA/DNA ratio and protein contents during metamorphosis and early juvenile stage of olive flounder (Paralichthys olivaceus) were examined. During metamorphosis, warm-acclimated fish showed higher RNA and DNA content than those of the cold-acclimated fish, excepting H stage (28 DAH) at which the ratio was higher at cold temperature. RNA/DNA ratio during metamorphosis showed similar values at two temperatures tested. However, after 42 DAH warm-acclimated juveniles had higher DNA content compared with cold-acclimated fish, resulted in marked decreases in RNA/DNA ratios. Higher RNA content at H stage of cold-acclimated fish was consistent with an increase in protein content. Growth of fish rearing at warm temperature was higher than those of fish at cold temperature during all experiments. In starvation experiment, contents of DNA, RNA and protein significantly decreased. Even though there were no significant differences in total length (TL) and body weight between the live mysid-fed and artificial pellet-fed fish at 35 mm TL, both RNA/DNA and protein/DNA ratios of the former group was significantly higher than those of the latter due primarily to lower DNA content of the live mysid-fed group. The results from this study suggest that temperature, starvation and kind of foods should be considered when RNA/DNA ratio applied to assessing the cultured larval and juvenile fish condition.

  • It is said that the focus of managing venture business is currently moving from technology competition to management competition. By the way, the software venture business(SVB) has some weak points in its structural composition and itematization and no professional personnel in other several sections except technology development section. In addition, such basic functions as technology and R & D, finance and accounting, marketing required to the management of business are concentrated on only one man, its representative director. Therefore, this study aims to provide the basic data useful to the establishment of governmental policy in information and communication, to the rearing of the SVB by a local government related to the software, and to the administration of SVB by investigating the actual conditions. This study attempts to examine the literature on venture business and software industry, and its management with a questionnaire about the actual conditions of managing the SVB. The questionnaire is given to 527 local enterprises belonging to the Software Industry Association and to 171 enterprises in the Software Center. This study compromises the characteristics of the SVB, the actual conditions of its technology and R & D, finance and accounting, and marketing. The characteristics of the SVB are classified into categories such as the stage of its growth(the stage of its seed and start-up, the stage of tis development and growth, the stage of its stability and maturity) and the main business(the system integration, the software development for contract, the package software development service, the software-related service). Additionally, the study attempts to analyze positively the actual condition of its management after classified by the areas of business profile, its general management, its technology development, its finance and accounting, and its marketing The result of this study is found that the SVB has a lot of troubles in part of marketing and finance & accounting activity as well as general management. The SVB realizes the importance of the technology development rather than that of management activity including marketing activity. So we expect this study can assist the SVB to establish the business guidelines for own management plans.

Effect of Feeding Frequency on Growth and Body Composition of Juvenile River Puffer, Takifugu obscurus in Winter season (동절기 배합사료 공급 횟수가 황복(Takifugu obscurus) 치어의 성장과 체조성에 미치는 영향)

  • To survey the most feeding frequence of formulated diet as food for cultured puffer in winter season, we performed a rearing test using juveniles of river puffer, Takifugu obscurus (body weight 15.0 g) for 120 days. The feeding frequencies were set up as 2times/1day, 2times/2days, 2times/3days and 2times/4days. We tested triplicately the experiment and investigated survival rate, daily food intake (DFI), feed efficiency (FE), condition factor (CF), daily growth rate (DGR). Consequently, growth was increased following to an increasing of feeding frequency, was the fast in 2times/1day of feeding frequency, and was the slowest in 2times/4days (p<0.05). DFI and CF were increased following to an decreasing of feeding frequency and was the highest in 2times/4days of feeding frequency. FE was decreased following to an decreasing of feeding frequency, and was the highest in 2times/1day of feeding frequency. In a proximate carcass composition at the final day, moisture and crude lipid contents were the lowest in 2times/4days, and in survival, there was not any significant difference among experimental groups. Therefore, we concluded that the 2times/2days are the best of feeding frequency for economical benefit of river puffer culture in winter season.

Nutritional Value of Mealworm, Tenebrio molitor as Food Source

  • Nutrition value of mealworm, Tenebrio molitor was analyzed due to increasing demand of usage as a protein source for domestic animals and even further for human consumption. The purpose of the present work was to determine the chemical compostion of the Tenebrio molitor larvae, adult that were maintained under standard condition for further usage of mass-rearing system and its exuvium, and excreta. Tenebrio molitor, larvae, adult, exuvium and excreta contained 46.44, 63.34, 32.87, and 18.51% protein respectively, suggested that even excreta could be used as an additional supplement in food recycling process. This protein was also rich in amino acids such as Isoleucine, leucine and Lysine which all met the nutritional value recommended by the Food and Agriculture Organization. Fatty acid composition was detected with high component of oleic acid (C18:1), along with linoleic acid (C18:2) and palmitic acid (C16) in all adult, larvae, exuvium and excreta. These oleic acid (C18:1), linoleic acid (C18:2) and palmitic acid (C16) components were the same or even highly contained in excreta of mealworm 22.29, 47.19 and 19.17% respectively. Longer chains of unsaturated fatty acids consisted of two to three double bonds are known as healthy product was recognized in large amount. These results show new ways to consume mealworms and its waste for animal and human consumption.

  • Objectives: This study was performed to analyse the effects of Sweet Bee Venom(Sweet BV-pure melittin, the major component of honey bee venom) on the central nervous system in rats. Methods: All experiments were conducted at Biotoxtech Company, a non-clinical studies authorized institution, under the regulations of Good Laboratory Practice (GLP). Male rats of 5 weeks old were chosen for this study and after confirming condition of rats was stable, Sweet BV was administered in thigh muscle of rats. And checked the effects of Sweet BV on the central nervous system using the functional observational battery (FOB), which is a neuro-toxicity screening assay composed of 30 descriptive, scalar, binary, and continuous endpoints. And home cage observations, home cage removal and handling, open field activity, sensorimotor reflex test/physiological measurements were conducted. Results: 1. In the home cage observation, there was not observed any abnormal signs in rats. 2. In the observation of open field activity, the reduction of number of unit areas crossed and rearing count was observed caused by Sweet BV treatment. 3. In the observation of handling reactivity, there was not observed any abnormal signs in rats. 4. In the observation of sensorimotor reflex tests/physiological measurements, there was not observed any neurotoxic signs in rats. 5. In the measurement of rectal temperature, treatment of Sweet BV did not showed great influences in the body temperature of rats. Conclusions: Above findings suggest that Sweet BV is relatively safe treatment in the central nervous system. But in the using of over dose, Sweet BV may the cause of local pain and disturbance of movement. Further studies on the subject should be conducted to yield more concrete evidences.

The Molecular Biomarker Genes Expressions of Rearing Species Chironomus riparious and Field Species Chironomus plumosus Exposure to Heavy Metals (실내종 Chironomus riparious와 야외종 Chironomus plumosus의 중금속 노출에 따른 분자지표 유전자 발현)

  • Chironomous is aquatic insect belonging to order Diptera, family Chironomidae. Their larval stage can be found mainly in aquatic benthic environment, hence good model organism to study environmental toxicology assessments and consider as useful bio indicators of contamination of the aquatic environment. In this study, Chironomus Heat Shock Proteins, Cytochrome 450, Glutathione S-transferase, Serine-type endopeptidase gene expressions were compared between polluted field areas (Chironomus plumosus) and under laboratory conditions (Chironomus riparious) to investigate molecular indicators for environmental contaminant stress assessment. Heavy metal (Al, Fe, Mn, Cu, Cr, Zn, Se, Pb, As, Cd) concentrations in sediments collected from three study areas exceeded the reference values. Moreover, HSPs, CYP450 and GST gene expression except SP for C. plumosus showed higher expression than C. riparious gene expression. Similar gene expression pattern was observed in C. riparious that exposed environment waters up to 96 h when compared to C. plumosus exposed to waters that grown in lab conditions. In summary, this comparative gene expression analysis in Chironomous between field and laboratory condition gave useful information to select candidate molecular indicators in heavy metal contaminations in the environment.

Effect of Three Microalgal Species on Gonadal Development and Sex Maturation of Ark Shell, Scapharca broughtonii (미세조류 3종에 따른 피조개 Scapharca broughtonii의 생식소 발달과 성숙)

  • Even though ark shell, Scapharca broughtonii is commercially important species, its artificial seed production is not yet fully developed. This study was carried out to understand the effect of three microalgal species on gonadal development and sex maturation of S. broughtonii in terms of broodstock management and food organism. Isochrysis galbana, Phaeodactylum tricornutum and Tetraselmis tetrathele were supplied to S. broughtonii broodstock in single or mixed. And condition index, gonadal development, sexual maturation and survival of the broodstock were analysed. After 45 rearing days, frequency of ripe stage of gonadal phases, rate of induced sexual maturation and survival of S. broughtonii broodstock fed mixed diet with 3 microalgal species or single diet of T. tetrathele were the highest.

Development of ICT based Automated Detection & Propagation System for Accidents in Agricultural Machinery (농기계 안전사고 시 자동상황전파를 위한 ICT기반 시스템 개발)

  • Recently, the aging of agricultural society has caused a rapid increase in safety accidents in -agricultural machinery. Sometimes, the wounded may be left unattended resulting serious situation. In order to solve these problems, ICT technology is used to detect and inform the accident quickly when a safety accident such as overturning, collision or other accidents occurs during the operation or moving the agricultural machinery, such as cultivator, rearing machine, and tractor. A system capable of minimizing the amount of data is required. In this paper, an ICT - based automatic accident detection & propagation system is proposed for the agricultural machinery accident such as colliding crashes or overturning of agricultural machinery. The proposed system enables quick rescue by sending a text message automatically to family, acquaintances, hospitals and 119 in the event of an agricultural accident.

Comparison of the Bacterial and Fungal Colonies from Rana dybowskii which Collected from Inside and Outside Frog Farms and Identification of the Bacteria from the Tadpoles (개구리 증양식장 내·외부에서 채집된 북방산개구리(Rana dybowskii)로부터 검출된 세균과 곰팡이 콜로니 수의 비교 및 유생으로부터 확인된 세균 규명)

  • There are many ongoing studies of infectious diseases as the major factor responsible for global declining of the amphibian population. Although some point out the amphibian rearing facilities like frog farms as one of the important sources of harboring and spreading amphibian infectious pathogens in the wild, there have been few related studies in South Korea. In this study, we investigated the bacterial and fungal colonies on the skin and in the internal organs of frogs and tadpoles collected inside and outside of Dybowski's brown frog farms in Inje, Goesan, and Gongju to compare the difference according to the region and between inside and outside the farm. We also intended to classify the bacteria collected from the tadpoles into species by analyzing 16s rDNA gene sequences. The result showed that the number of bacterial colonies found in the skin and gut of frogs and the number of fungal colonies found in the skin and liver of frogs collected in Goesan was significantly greater than those in the frogs in Inje. However, there was no difference between the frogs collected inside and outside of farms in both regions. In the case of tadpoles, the number of fungal colonies in the tadpoles collected from Gongju was greater than that in the tadpoles collected from Inje. The comparison of inside and outside frog farms showed that there were more bacterial colonies on the skin of the tadpoles collected from inside than outside the frog farm in Inje and more bacterial colonies in the organs of the tadpoles collected from outside than inside the farm in Gongju. The frogs with higher condition factor (body weight/snout-vent length*100) showed fewer bacterial colonies on the skin and fewer fungal colonies in the heart, but there were no significant relationships in tadpoles. We identified the total of 15 genera and four phyla of bacteria, but the difference according to regions and between inside and outside farm was not evident. The result of this study indicates that the different conditions according to the locality of farm and between inside and outside farm cause the difference in the population sizes of bacterial and fungal colonies and that it can affect the overall health condition of Dybowski's brown frogs in the farm. Moreover, the result suggests that effective disease control in the facility is greatly necessary to ensure successful operation of amphibian rearing facility and to prevent the possible spread of diseases from the facility to the wild.
