Physiological and Phylogenetic Analysis of Burkholderia sp. HY1 Capable of Aniline Degradation

  • A new aniline-utilizing microorganism, strain HY1 obtained from an orchard soil, was characterized by using the BIOLOG system, an analysis of the total cellular fatty acids, and a 16S rDNA sequence. Strain HY1 was identified as a Burkholderia species, and was designated Burkholderia sp. HY1. GC and HPLC analyses revealed that Burkholderia sp. HY1 was able to degrade aniline to produce catechol, which was subsequently converted to cis,cis-muconic acid through an ortho-ring fission pathway under aerobic conditions. Strain HY1 exhibited a drastic reduction in the rate of aniline degradation when glucose was added to the aniline media. However, the addition of peptone or nitrate to the aniline media dramatically accelerated the rate of aniline degradation. A fatty acid analysis showed that strain HY1 was able to produce lipids 16:0 2OH, and 11 methyl 18:1 ${\omega}7c$ approximately 3.7-, 2.2-, and 6-fold more, respectively, when grown on aniline media than when grown on TSA. An analysison the alignment of a 1,435 bp fragment. A phylogenetic analysis of the 16S rDNA sequence based on a 1,420 bp multi-alignment sowed of the 16s rDNA sequence revealed that strain HY1 was very closely related to Burkholderia graminis with 95% similarity based that strain HY1 was placed among three major clonal types of $\beta$-Proteobacteria, including Burkholderia graminis, Burkholderia phenazinium, and Burkholderia glathei. The sequence GAT(C or G)${\b{G}}$, which is highly conserved in several locations in the 16S rDNA gene among the major clonal type strains of $\beta$-Proteobacteria, was frequently replaced with GAT(C or G)${\b{A}}$ in the 16S rDNA sequence from strain HY1.

A Study on Development of Structural Health Monitoring System for Steel Beams Using Strain Gauges (변형률계를 이용한 강재보의 건전도 평가 시스템 개발에 관한 연구)

  • This study aimed to develop a Structural Health Monitoring System for steel beams in the manner of suggesting and verifying a theoretical formula for displacement estimation using strain gauges, and estimating the loading points and magnitude. According to the results of this study, it was found that when a load of 160kN (56% of the yield load) was applied, the error rate of the deflection obtained with a strain gauge at the point of maximum deflection compared to the deflection measured with a displacement meter was within 2%, and that the estimates of the magnitude and points of load application also showed the error rate of not more than 1%. This suggests that the displacement and load of steel beams can be measured with strain gauges and further, it will enable more cost-effective sensor designing without displacement meter or load cell. The Structural Health Monitoring System program implemented in Lab VIEW gave graded warnings whenever the measured data exceeds the specified range (strength limit state, serviceability limit state, yield strain), and both the serviceability limit state and strength limit state could be simultaneously monitored with strain gauge alone.

Studies on the Development of Stevia (Stevia rebaudiana Bertoni M.) as a New Sweetness (신감미(新甘味) 자원식물(資源植物) Stevia의 개발(開發)에 관(關)한 연구(硏究))

  • The consumption of sugar in our diet increased so great that much U. S. dollars are spended to import sugar source from foreign contries. To replace a part of it we adopted mother plant and seeds of the well-known Stevia (Stevia rebaudiana Bertoni M.) originated from south America, from the Agricultural Experimental Station of Hokkaido, Japan, in 1974. In order to determine the growth condition of Stevia in our climate and soil condition, the results of survey and determination were as follows: 1. Germination rate of Stevia was averaged 33.6% at 35 th day after seedling and the maximum germination period was between 5 and 20 days after seedling. 2. The seedling rate of Stevia was about 8%. 3. Establishment of roots of Stevia propagated by cutting method was above 90% but only S-1 strain showed lower rate., 75%. 4. S-2 strain showed the best growth rate that was about 160cm by plant height and it was decreased by the following order : S-4, S-3, S-1, S-6 and S-5 strains. 5. S-4 strain resulted in 45 branches that was the highest number among strains tested and S-2 strain was the next. 6. The content of sweetening compound of Stevia, stevioside, was 5.3% in S-1, S-2, S-5 and S-4 strains but S-3 and $S^*$-6 strains were relatively low content, 4.9%. From the above results we obtained Stevia could be propagated by cutting method and the growth conditions including plant height, number of branches and content of stevioside were relatively better in S-2 and S-4 strains that were differentiated by the shape of leaf.

  • A shape optimization of flow guide is accomplished to minimize the wear rate of die in three-dimensional flat-die extrusion processes. In order to achieve the balanced flow and the uniformed distribution of the effective strain during the extrusion, a multi-objective optimization is implemented. During the process of optimization formulation, the flow balance and the deviation of strain is considered as constrained conditions. The proposed approach is applied to an extrusion of H section. Through the optimization, it has been confirmed that the wear rate of die can be minimized satisfying the constraint.

  • In this work, the effect of chemical treatments on surface properties of SiC was investigated in crack resistance properties of SiC/epoxy composites. The surface properties of SiC were determined by acid/base values and FT-IR measurements. Also the crack resistance properties of the composites were studied in critical strain energy release rate mode II ($G_{IIC}$) measurements. As a result, the acidically treated SiC had higher acid value than that of untreated SiC or basically treated SiC. The crack resistance properties of the composites had been improved in the specimens treated by acidic solution. These results were could be attributed to the acide-base intermolecular interaction between SiC and epoxy resin, resulting in increase of the degree of adhesion at interfaces.

  • Solid polymers exhibit rate-dependent deformation behavior such as nonlinear strain rate sensitivity and stress relaxation like metallic materials. Despite the different microstructures of polymeric and metallic materials, they have common properties with respect to inelastic deformation. Unlike most metallic materials, solid polymers and shape memory alloys (SMAs) exhibit highly nonlinear stress-strain behavior upon unloading. The present work employs the viscoplasticity theory [K. Ho, 2011, Trans. Mater. Process. 20, 350-356] developed for the pseudoelastic behavior of SMAs, which is based on unified state variable theory for the rate-dependent inelastic deformation behavior of typical metallic materials, to depict the curved unloading behavior of polyphenylene oxide (PPO). The constitutive equations are characterized by the evolution laws of two state variables that are related to the elastic modulus and the back stress. The simulation results are compared with the experimental data obtained by Krempl and Khan [2003, Int. J. Plasticity 19, 1069-1095].

Influence of strain rate on the acoustic emission signal characteristics in corrosive environment (부식환경하에서 음향방출신호 특성에 미치는 변형률속도의 영향)

  • The study was performed to study the effects of strain rate on acoustics emission( AE) during bulging test in corrosive environmentsynthetic sea water. The strain rates used were in the range $4 \times 10^{-6}S^{-1}$ to $1 \times 10^{-4} \times S^{-1}$ and the parameters used to evaluate AE signal characteristics were AE hit and amplitude. It can be observed that the cumulative AE hit and average amplitude during fracture process increase highly at decreasing strain rates while the equivalent fracture strain and the crack length of circumferencial direction become decrease. The peak point of AE signal characteristic parameters approach to the first half of test. When the average amplitude per unit equivalent fracture strain was above 20dB, it was definitly observed stress corrosion cracking phenomena. Additional, we knew that the AE test had the possibility to evaluate SCC susceptibility with various strain rates.

Mechanical Behavior of Shape Memory Fibers Spun from Nanoclay-Tethered Polyurethanes

  • This study examined the effect of nanoclays on the shape memory behavior of polyurethane (PU) in fibrous form. A cation was introduced into the PU molecules to disperse the organo-nanoclay (MMT) into poly($\varepsilon$-caprolactone) (PCL)-based PU (PCL-PU). The MMT/PCL-PU nanocomposites were then spun into fibers through melt-processing. The shape memory performance of the spun fibers was examined using a variety of thermo-mechanical tests including a new method to determine the transition temperature of shape memory polymers. The MMTs showed an improved the fixity strain rate of the MMT /PCL- PU fibers but a slight decrease in their recovery strain rate. This was explained by the limited movement of PU molecules due to the presence of nanoclays. The shape memory performance of the MMT/PCL-PU fibers was not enhanced significantly by the nanoclays. However, their recovery power was improved significantly up to a strain of approximately 50%.

Analytical Study on Interface Debonding of Reinforced Concrete Beams Strengthened with Carbon Fiber Sheet(CFS) (탄소섬유쉬트로 보강된 철근콘크리트보의 계면박리에 대한 해석적 연구)

  • The purpose of this study is to analyze the interface debonding of RC beams strengthened by carbon fiber sheet(CFS). The behavior of damaged RC beams strengthened with CFS is analytically investigated next using linear elastic fracture mechanics(LEFM) approach and the finite element method. The study includes an investigation of the separation mode by interface fracture of the strengthening materials due to the interfacial shear and normal stresses. The numerical method is presented to obtain the value of interfacial fracture parameter such as the strain energy release rate. Based on the results of this study, it is found that the critical case occurs when the interfacial cracks occur within a short region of the flexural crack. The CFS strengthening has not an adequate factor of safety against interfacial debonding of CFS. Furthermore, for the thicknesses of the adhesive studied[1mm~3mm], it is no noticeable effect on the strain energy release rate.

The Blanching Effects on the Drying Rates and the Color of Hot Red Pepper (고추의 건조율 및 색도에 미치는 Blanching 효과)

  • To examine the effects of blanching treatments on the drying of hot red pepper, the pilot scale hot-air dryer equipped with the weight sensor using strain gauge type load cell and strain amplifier was designed and manufactured. The drying characteristic curves of cut hot red pepper showed a settling down period followed by a constant rate period and falling rate period, but blanched hot red pepper showed only falling rate period. According as the blanching time and temperature rises, the drying rates and the capsanthin contents of hot red pepper fairly increased. Considering the drying rates and the color values of dried hot red pepper, we suggest the desirable blanching condition of hot red pepper should be water blanching at $80^{\circ}C$ for 3 mins.
